Abstract:Glycine betaine (GBT) is a nitrogenous osmolyte ubiquitous throughout the marine environment. Despite its widespread occurrence and significance in microbial cycling, knowledge of the seasonality of this compound is lacking. Here, we present a seasonal dataset of GBT concentrations in marine suspended particulate material. Analysing coastal waters in the Western English Channel, GBT peaked in summer and autumn but did not follow the observed maxima in total phytoplankton biomass or chlorophyll a . Instead, we found evidence that GBT concentrations were associated with specific phytoplankton groups or species, particularly in the summer when GBT correlated with dinoflagellate biomass. In contrast, autumn maxima corresponded with a period of rapidly changing salinity and nutrient availability, with potential contributions from some phytoplankton species and Harpacticoid copepods. This suggests distinct environmental drivers for different periods of the GBT seasonality. Building on evidence that GBT and dinoflagellate biomass peak in summer, concomitantly with low nutrients, we propose that GBT positively affects dinoflagellate fitness, allowing them to outcompete other plankton when inorganic nutrients are depleted. By using this assumption, we improved the performance of a marine ecosystem model to reproduce the observed increase in dinoflagellates biomass in the transition from spring to summer. This work sheds light on the interplay between phytoplankton succession, competitive advantage and changing environmental factors relevant to climate change. It paves the way for future multidisciplinary research aiming to understand the importance of dinoflagellates in key coastal ecosystems and their potential significance for methylamine production, compounds relevant for particle growth in atmospheric chemistry.

Isolation and quantification of alginate in choline chloride-based deep eutectic solvents

Abstract:Extraction of seaweed compounds using Deep Eutectic Solvents (DES) has shown high interest. Quantification, however, is challenging due to interactions with DES components. In this research work, three chemical separation techniques were investigated to isolate and quantify alginate from a set of choline chloride-based DES. While choline chloride served as the hydrogen bond acceptor (HBA); Urea, Ethylene Glycol, Propylene Glycol, Glycerol, Sorbitol, Xylitol and Glucose were used as hydrogen bond donors (HBD). DES containing sodium alginate were subjected to precipitation with sulfuric acid 0.2 M (pH 1.6), ethanol-water mixture (80 % v/v) and calcium chloride (1 % w/v CaCl2·2H2O). Alginate in precipitates was quantified and used to evaluate the performance of each separation technique. The highest recovery yields (51.2 ± 1.3 %) were obtained using the ethanol-water mixture followed by calcium chloride (45.7 ± 1.2 %), except for polyols (e.g. sorbitol). The lowest recovery yields were obtained with acid, with a particularly low recovery yield when urea was used as HBD (9.6 ± 1.3 %). Estimations of ManA/GulA ratios showed lower values for precipitates from DES compared to the ones obtained from water. This research shows ethanolic precipitation as a suitable method for alginate separation from the studied set of choline chloride-based DES.
Microbial dynamics and biogenic methane production responses to the addition of glycine betaine in shales

Abstract:Biogenic methane production depends on microbial community compositions in shale gas reservoirs, and glycine betaine plays an important role in methanogenic metabolic pathways. Previous studies have mainly focused on the microbial community dynamics in the water produced by shale hydraulic fracturing. Here, we used fresh shale as a sample and obtained the methane (CH4) and carbon dioxide (CO2) concentrations, microbial communities, and methanogenic functional gene numbers of solid and liquid groups in anaerobic bottles through gas chromatography, 16S rDNA sequencing (60 samples) and quantitative real-time PCR analysis in all culture stages. With glycine betaine addition, the total CH4 concentrations of the S1, S2 and Sw samples were 1.56, 1.05 and 4.48 times, while CO2 increased by 2.54-, 4.80- and 0.43-fold compared with samples without glycine betaine after 28 days of incubation, respectively. The alpha diversity was reduced when glycine betaine was added. The significant differences in bacterial community abundance at the genus level in samples with glycine betaine were Bacillus, Oceanobacillus, Acinetobacter, and Legionella. The bacterial and archaeal community changes implied that the addition of glycine betaine may promote CH4 production mainly by first forming CO2 and then generating CH4. The results of mrtA, mcrA, and pmoA gene numbers showed that the shale had great potential for producing methane. The addition of glycine betaine to shale changed the original microbial networks and increased the nodes and taxon connectedness of the Spearman association network. Our analyses indicate that the addition of glycine betaine enhances CH4 concentrations, causing the microbial network to be more complex and sustainable which supports the survival and adaptation of microbes in shale formations.

Abstract:AbstractDespite significant advances in chemical ecology, the biodistribution, temporal changes and ecological function of most marine secondary metabolites remain unknown. One such example is the association between choline esters and Tyrian purple precursors in muricid molluscs. Mass spectrometry imaging (MSI) on nano-structured surfaces has emerged as a sophisticated platform for spatial analysis of low molecular mass metabolites in heterogeneous tissues, ideal for low abundant secondary metabolites. Here we applied desorption-ionisation on porous silicon (DIOS) to examinein situchanges in biodistribution over the reproductive cycle. DIOS-MSI showed muscle-relaxing choline ester murexine to co-localise with tyrindoxyl sulfate in the biosynthetic hypobranchial glands. But during egg-laying, murexine was transferred to the capsule gland and then to the egg capsules, where chemical ripening resulted in Tyrian purple formation. Murexine was found to tranquilise the larvae and may relax the reproductive tract. This study shows that DIOS-MSI is a powerful tool that can provide new insights into marine chemo-ecology.

Abstract:Microalgae-based carbon capture and utilization (CCU) offers a promising negative emission technology that absorbs CO 2 and generates valuable biomass. However, heavy metal pollutants in industrial flue gases can compromise carbon sequestration efficiency and bioproduct quality. Spirulina was investigated as a model organism for CO 2 sequestration, using a modified Zarrouk's medium to integrate salt stress and an osmoprotectant to boost biomass yield under 15 % CO 2 . This approach enhances both biomass yield (380.83 mg L –1 d –1 )and resistance to lead toxicity. Analytical assessments revealed that glycine betaine (GB) supplementation drastically reduced lead accumulation, decreasing the extracellular and intracellular contents by 39.7 % and 60.7 %, respectively. A notable decrease in extracellular dissolved organic matter was also observed. Furthermore, transcriptomic analyses confirmed that GB treatment strengthened osmotic stress responses and suppressed metal ion transport. These findings enhanced the feasibility of microalgae-based CCU technologies, marking significant progress in sustainable algal biotechnology.

Abstract:The urgent need to remediate ocean acidification has brought attention to the ability of marine macrophytes (seagrasses and seaweeds) to take up carbon dioxide (CO2) and locally raise seawater pH via primary production. This physiological process may represent a powerful ocean acidification mitigation tool in coastal areas. However, highly variable nearshore environmental conditions pose uncertainty in the extent of the amelioration effect. We developed experiments in aquaria to address two interconnected goals. First, we explored the individual capacities of four species of marine macrophytes (Ulva lactuca, Zostera marina, Fucus vesiculosus and Saccharina latissima) to ameliorate seawater acidity in experimentally elevated pCO2. Second, we used the most responsive species (i.e., S. latissima) to assess the effects of high and low water residence time on the amelioration of seawater acidity in ambient and simulated future scenarios of climate change across a gradient of irradiance. We measured changes in dissolved oxygen, pH, and total alkalinity, and derived resultant changes to dissolved inorganic carbon (DIC) and calcium carbonate saturation state (Ω). While all species increased productivity under elevated CO2, S. latissima was able to remove DIC and alter pH and Ω more substantially as CO2 increased. Additionally, the amelioration of seawater acidity by S. latissima was optimized under high irradiance and high residence time. However, the influence of water residence time was insignificant under future scenarios. Finally, we applied predictive models as a function of macrophyte biomass, irradiance, and residence time conditions in ambient and future climatic scenarios to allow projections at the ecosystem level. This research contributes to understanding the biological and physical drivers of the coastal CO2 system.

Abstract:In Dunaliella tertiolecta, a microalga renowned for its extraordinary tolerance to high salinity levels up to 4.5 M NaCl, the mechanisms underlying its stress response have largely remained a mystery. In a groundbreaking discovery, this study identifies a choline dehydrogenase enzyme, termed DtCHDH, capable of converting choline to betaine aldehyde. Remarkably, this is the first identification of such an enzyme not just in D. tertiolecta but across the entire Chlorophyta. A 3D model of DtCHDH was constructed, and molecular docking with choline was performed, revealing a potential binding site for the substrate. The enzyme was heterologously expressed in E. coli Rosetta (DE3) and subsequently purified, achieving enzyme activity of 672.2 U/mg. To elucidate the role of DtCHDH in the salt tolerance of D. tertiolecta, RNAi was employed to knock down DtCHDH gene expression. The results indicated that the Ri‐12 strain exhibited compromised growth under both high and low salt conditions, along with consistent levels of DtCHDH gene expression and betaine content. Additionally, fatty acid analysis indicated that DtCHDH might also be a FAPs enzyme, catalyzing reactions with decarboxylase activity. This study not only illuminates the role of choline metabolism in D. tertiolecta's adaptation to high salinity but also identifies a novel target for enhancing the NaCl tolerance of microalgae in biotechnological applications.

Abstract:Abstract Submarine groundwater discharge (SGD) is a globally important process supplying nutrients and trace elements to the coastal environment, thus playing a pivotal role in sustaining marine primary productivity. Along with nutrients, groundwater also contains allochthonous microbes that are discharged from the terrestrial subsurface into the sea. Currently, little is known about the interactions between groundwater‐borne and coastal seawater microbial populations, and groundwater microbes' role upon introduction to coastal seawater populations. Here, we investigated seawater microbial abundance, activity and diversity in a site strongly influenced by SGD. In addition, through laboratory‐controlled bottle incubations, we mimicked different mixing scenarios between groundwater and seawater. Our results demonstrate that the addition of 0.1 μm filtered groundwater stimulated heterotrophic activity and increased microbial abundance compared to control coastal seawater, whereas 0.22 μm filtration treatments induced primary productivity and Synechococcus growth. 16S rRNA gene sequencing showed a strong shift from a SAR11‐rich community in the control samples to Rhodobacteraceae dominance in the <0.1 μm treatment, in agreement with Rhodobacteraceae enrichment in the SGD field site. These results suggest that microbes delivered by SGD may affect the abundance, activity and diversity of intrinsic microbes in coastal seawater, highlighting the cryptic interplay between groundwater and seawater microbes in coastal environments, which has important implications for carbon cycling.

Deciphering the Performance and Mechanisms of Glycolipids in Regulating Crop Growth in Coastal Saline-Alkali Soils: Perspectives on Soil Properties and Microbial Communities

Abstract:Coastal regions face significant challenges in crop production due to soil salinization. Rhamnolipids and sophorolipids are prototypical glycolipid biosurfactants that exhibit considerable promise in soil amelioration and crop growth regulation. Nevertheless, the efficacy and mechanisms of these glycolipids in coastal saline-alkali soils remain poorly understood. In this study, a pot experiment was carried out to examine the impact of rhamnolipids and sophorolipid supplementation at different doses (i.e., 50, 100, and 200 mg/kg) on oleic sunflower growth. Both biosurfactants at appropriate doses effectively reduced the soil alkalinity, elevated the levels and bioavailability of soil nutrients (i.e., available nitrogen, phosphorus, and organic matter), and improved enzyme activities (i.e., catalase, urease, beta-glucosidase, and alkaline phosphatase). Notably, the application of medium-dose rhamnolipids resulted in the most substantial enhancement of crop growth (up to 117 % increase in fresh weight). Besides, an appropriate dose of rhamnolipids enriched the beneficial microbiota (e.g., Glycomyces, Stachybotrys, and Mortierella) while inhibiting the growth of pathogens (e.g., Fusarium) in rhizosphere zones. Conversely, a high dose of sophorolipids accumulated plant pathogens in both the rhizosphere and bulk soils. Correlation-based network analysis and microbial function predictions revealed that the intervention of medium-dosed rhamnolipids enhanced the cooperation within bacterial and fungal communities and activated the community functions related to carbon and nitrogen cycling. Moreover, the increased soil enzyme activity was the main driver for improved crop growth in rhamnolipids-supplemented treatments. The findings provide new insights into the underlying mechanisms of glycolipids-assisted improvement of crop growth in coastal saline-alkali regions.
Highly Efficient Harvesting and Lipid Extraction of Limnetic Chlorella Sorokiniana SDEC-18 Grown in Seawater for Microalgal Biofuel Production

Abstract:The low efficiency and high costs involved in harvesting microalgae and extracting lipids have been major barriers toward large scale algae-based biofuel production. Here we demonstrate the feasibility of employing seawater as a substitute for freshwater in cultivating Chlorella sorokiniana SDEC-18. Strikingly, compared to freshwater, seawater was found to 1) enhance Chlorella autoflocculation, reaching 90 % settling efficiency within 2 h; 2) promote lipid accumulation, reaching about 60 % (0.19-0.28 g/L of lipid concentration) due to salinity stress; and 3) improve lipid-extraction efficiency, with full extraction achieved after only the second extraction step. The mechanisms for enhanced Chlorella harvest were revealed through the observation of extracellular polymeric substances using transmission electron microscopy after lanthanum fixation. Moreover, new insights into cell fragility under salinity stress have been obtained through our pioneering proposal of the involvement of Chlorella microtubules. The advances we have achieved in Chlorella harvesting and lipid extraction lay the foundation for a new generation of biofuel production.

Abstract:Fungal secondary metabolites constitute a rich source of yet undiscovered bioactive compounds. Their production is often silent under standard laboratory conditions, but the production of some compounds can be triggered simply by altering the cultivation conditions. The usage of an organic salt – ionic liquid – as growth medium supplement can greatly impact the biosynthesis of secondary metabolites, leading to higher diversity of compounds accumulating extracellularly. This study examines if such supplements, specifically cholinium-based ionic liquids, can support the discovery of bioactive secondary metabolites across three model species: Neurospora crassa, Aspergillus nidulans , and Aspergillus fumigatus . Enriched organic extracts obtained from medium supernatant revealed high diversity in metabolites. The supplementation led apparently to increased levels of either 1-aminocyclopropane-1-carboxylate or α-aminoisobutyric acid. The extracts where bioactive against two major foodborne bacterial strains: Staphylococcus aureus and Escherichia coli . In particular, those retrieved from N. crassa cultures showed greater bactericidal potential compared to control extracts derived from non-supplemented cultures. An untargeted mass spectrometry analysis using the Global Natural Product Social Molecular Networking tool enabled to capture the chemical diversity driven by the ionic liquid stimuli. Diverse macrolides, among other compounds, were putatively associated with A. fumigatus ; whereas an unexpected richness of cyclic (depsi)peptides with N. crassa . Further studies are required to understand if the identified peptides are the major players of the bioactivity of N. crassa extracts, and to decode their biosynthesis pathways as well.

Through analysis of their associated microbial communities, we found a variety of microbial metabolisms that may benefit the host, including nitrogen metabolisms, sulfur oxidation, and the production of B vitamins.

Abstract:Background: An economical strategy for producing microalgae as biofuel feedstock is driven by the freshwater and nutrients input. In this study, seawater was applied to limnetic algal cultivation and the behavior of algae in seawater media was observed including growth, lipid synthesis, and ultrastructure. To make seawater cater algae, a kind of wastewater, anaerobically digested effluent from kitchen waste (ADE-KW), was used as nutrient sources. Results: Pure seawater cannot support the growth demand of freshwater microalga, due to high salinity and lack of nutrients. However, it is the conditions triggered the algae to synthesize lipids of 60%, double of lipid content in standard medium BG11. Introducing 3 or 5% ADE-KW (volume percentage) into seawater made algal growth reach the level attained in BG11, while lipid content compared favourably with the level (60%) in pure seawater. This method achieved the goal of fast growth and lipid accumulation simultaneously with the highest lipid productivity (19 mg/L day) at the exponential stage, while BG11 obtained 10.55 mg/L day at the stationary stage as the highest lipid productivity, almost half of that in seawater media. Moreover, the condition for highest lipid productivity enlarged algal cells compared to BG11. Under the condition for highest lipid productivity, Chlorella sorokiniana SDEC-18 had enlarged cells and increased settling efficiency compared to BG11, which facilitated harvest in an energy saving way. Conclusions: The results suggested that combining seawater with ADE-KW to cultivate microalgae had a double function: nutrients and water for algal growth, and high salinity for stimulating lipid accumulation. If this technology was operated in practice, freshwater and non-waste nutrient consumption would be completely obviated.

Abstract:Dissolved organic carbon (DOC) release by seaweeds (marine macroalgae) is a critical component of the coastal oceans biogeochemical carbon cycle but is an aspect of seaweed carbon physiology that we know relatively little about. Seaweed-derived DOC is found throughout coastal ecosystems and supports multiple food web linkages. Here we discuss the mechanisms of DOC release by seaweeds and group them into passive (leakage, requires no energy) and active release (exudation, requires energy) with particular focus on the photosynthetic 'overflow' hypothesis. The release of DOC from seaweeds was first studied in the 1960's but subsequent studies use a range of units hindering evaluation: we convert published values to a common unit (μmol C · g DW-1 · h-1) allowing comparisons between seaweed phyla, functional groups, biogeographic region, and an assessment of the environmental regulation of DOC production. The range of DOC release rates by seaweeds from each phylum under ambient environmental conditions was: 0 – 266.44 μmol C · g DW-1 · h-1 (Chlorophyta), 0 – 89.92 μmol C · g DW-1 · h-1 (Ochrophyta) and 0 – 41.28 μmol C · g DW-1· h-1 (Rhodophyta). DOC release rates increased under environmental factors such as desiccation, high irradiance, non-optimal temperatures, altered salinity and elevated dissolved carbon dioxide (CO2) concentrations. Importantly DOC release was highest by seaweeds which were desiccated (<90 times greater DOC release compared to ambient). We discuss the impact of future ocean scenarios (ocean acidification, seawater warming, altered irradiance) on DOC release rates by seaweeds, the role of seaweed-derived DOC in carbon sequestration models, and how they inform future research directions.

Glycine betaine biosynthesis and catabolism in bacteroids of Rhizobium meliloti: effect of salt stress

Abstract:Summary: In medium of low or high osmolality, bacteroids isolated from Medicago sativa nodules induced by Rhizobium meliloti 102F34 rapidly catabolized [14C]choline. Trimethylamine was never detected and glycine betaine was the predominant product within the ethanol-soluble fraction of bacteroids subjected to salt stress (0·4 M-NaCl). Both choline oxidase and glycine betaine aldehyde dehydrogenase activities were characterized; the apparent K m values for choline and glycine betaine aldehyde were 2·0 and 0·4 mM, respectively. A 3 h incubation of the bacteroids in medium of high osmolarity, supplemented or not with choline, did not significantly modify the specific activity of the two enzymes. Similarly, salinization of the host plants for 2 weeks with 0·15 M-NaCl had only a slight effect on both enzymic activities. Thus, the choline—glycine betaine biosynthetic pathway was not modulated by the external osmotic pressure. Glycine betaine itself was actively degraded by bacteroids suspended in low-osmolarity medium, but the demethylation process producing sarcosine and glycine was extremely slow in bacteroids subjected to salt stress. Thus, high concentration of glycine betaine can be maintained in salt-stressed bacteroids.
Cytotoxicity and cell membrane interactions of choline-based ionic liquids: comparing amino acids, acetate, and geranate anions

Abstract:Ionic liquids (ILs) have found diverse applications in research and industry. Biocompatible ILs, a subset considered less toxic than traditional ILs, have expanded their applications into biomedical fields. However, there is limited understanding of the toxicity profiles, safe concentrations, and underlying factors driving their toxicity. In this study, we investigated the cytotoxicity of 13 choline-based ILs using four different cell lines: Human dermal fibroblasts (HDF), epidermoid carcinoma cells (A431), cervical cancer cells (HeLa), and gastric cancer cells (AGS). Additionally, we explored the haemolytic activity of these ILs. Our findings showed that the cytotoxic and haemolytic activities of ILs can be attributed to the hydrophobicity of the anions and the pH of the IL solutions. Furthermore, utilising quartz crystal microbalance with dissipation (QCM-D) technique, we delved into the interaction of selected ILs, including choline acetate [Cho][Ac] and choline geranate [Cho][Ge], with model cell membranes composed of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC). The QCM-D data showed that ILs with higher toxicities exhibited more pronounced interactions with membranes. Increased variations in frequency and dissipation reflected substantial changes in membrane fluidity and mass following the addition of the more toxic ILs. Furthermore, total internal reflection fluorescence microscopy study revealed that [Cho][Ac] could cause lipid rearrangements and pore formation in the membrane, while [Cho][Ge] disrupted the bilayer packing. This study advances our understanding of the cellular toxicities associated with choline-based ILs and provides valuable insights into their mechanisms of action concerning IL-membrane interactions. These findings have significant implications for the safe and informed utilisation of biocompatible ILs in the realm of drug delivery and biotechnology.
Concurrent enhancement of biomass production and phycocyanin content in salt-stressed Arthrospira platensis: A glycine betaine- supplementation approach

Abstract:In industrial-scale cultivation of microalgae, salinity stress often stimulates high-value metabolites production but decreases biomass yield. In this research, we present an extraordinary response of Arthrospira platensis to salinity stress. Specifically, we observed a significant increase in both biomass production (2.58 g L-1) and phycocyanin (PC) content (22.31%), which were enhanced by 1.26-fold and 2.62-fold, respectively, compared to the control, upon exposure to exogenous glycine betaine (GB). The biochemical analysis reveals a significant enhancement in carbonic anhydrase activity and chlorophyll a level, concurrent with reductions in carbohydrate content and reactive oxygen species (ROS) levels. Further, transcriptomic profiling indicates a downregulation of genes associated with the tricarboxylic acid (TCA) cycle and an upregulation of genes linked to nitrogen assimilation, hinting at a rebalanced carbon/nitrogen metabolism favoring PC accumulation. This work thus presents a promising strategy for simultaneous enhancement of biomass production and PC content in A. platensis and expands our understanding of PC biosynthesis and salinity stress responses in A. platensis.

Abstract:As an essential nutrient, the organic water-soluble compound choline is important for human health. Choline is required for numerous biological processes, including the synthesis of neurotransmitters, and it is an important prerequisite for structural integrity and the functioning of cells. A choline-rich diet provides crucial choline sources, yet additional choline dietary supplements might be needed to fully meet the body's requirements. Dependent on the structure of choline in different sources, absorption and metabolism may differ and strongly impact the bioavailability of circulating choline. This study in healthy volunteers aimed to compare the pharmacokinetics of free choline and of selected choline metabolites between the single dose intake of phosphatidylcholine, present in SuperbaBoostTM krill oil, and choline bitartrate salt. Results demonstrate that albeit free choline levels in plasma were comparable between both choline sources, peak choline concentration was reached significantly later upon intake of SuperbaBoostTM. Moreover, the occurrence of choline metabolites differed between the study products. Levels of the biologically important metabolites betaine and dimethylglycine (DMG) were higher, while levels of trimethylamine N-oxide (TMAO) were substantially lower upon intake of SuperbaBoostTM compared to choline bitartrate.
