Abstract:A broad range of N-sulfonyformamidines, widely used intermediates for drugs, were synthesized in moderate to excellent yields from 2-Pyridinemethanamine as N-source via Copper-catalyzed C-N cleavage. Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides, then reacted with N,N-dimethylformamide dimethyl acetal to obtain the corresponding N-Sulfonylformamidines analogs, during which pyridin-2-ylmethyl and sulfonyl groups were essential for the C-N bond cleavage. The current work presents a valuable complementarity to the synthesis of N-sulfonyformamidines as 2-pyridinemethanamine can provide the N source and sulfonyl chloride,s which could be original materials. Background: N-sulfonylamidines have gained considerable attention from schools and industries because of their unique bioactivity. Since Pinner's strategy, expanding the synthesis methods of N-sulfonylamidines has been the goal of many organic chemists over the past decades. Besides the crash reaction conditions and the participation of undesirable reagents, the production of N-sulfonylamidines commonly required unstable ammonia and azides as the source of nitrogen that hindered the further development and application of N-sulfonylamidine derivatives. Objective: The study aims to find a stable N source to replace NaN3 or NH3 to synthesize N-sulfonylamidines from sulfonyl chlorides. Methods: Firstly, N-(2-pyridinylmethyl)benzenesulfonamides were smoothly synthesized via 2-pyridinemethanamine and sulfonyl chlorides. Then the reaction conditions of N-(2-pyridinylmethyl) benzenesulfonamides and N,N-dimethylformamide dimethyl acetal (DMF-DMA) were screened and optimized. The reaction was processed in glycol at 80 degrees C for 8 hours with the addition of 5 mol% Cu(OAc)(2)center dot H2O as a catalyst. Results: Taking advantage of pyridin-2-ylmethyl, a scope of N-Sulfonylformamidines were synthesized from those N-(2-pyridinylmethyl)benzenesulfonamides under copper-catalyzed C-N bond cleavage. Conclusion: This ready synthetic method will be more of a promising inspiration for bioactive compound synthesis and drug development than for an innovative approach to synthesizing N-sulfonylformamidines.

Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. Abstract:A triflate salt-catalyzed nonhydrolytic method for the deacylation of N-acylsulfonamides and subsequent one-pot condensation of the newly formed sulfonamides with N,N-dimethylformamide dimethyl acetal to provide N-sulfonylamidines is presented. A range of aliphatic and aromatic N-acylsulfonamides bearing various N-acyl groups such as acetyl, propionyl, butyrl, isobutyryl, octanoyl, benzoyl, 2-phenylacetyl, and sterically hindered pivaloyl are readily transformed into the corresponding N-sulfonylamidines in good to excellent yields. A variety of functional groups including halogeno, keto, nitro, cyano, hydroxyl, ether, and carboxylic ester are tolerated intact.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:A ligand‐free Pd/Cu‐catalyzed aminosulfonylation of aryl iodides has been successfully developed. The protocol provides a straightforward approach to access a broad range of structurally intriguing sulfonamides in one pot, at low catalyst loading from the sulfur dioxide surrogate 1,4‐diazabicyclo[2.2.2]octane bis(sulfur oxide) (DABSO) and O‐benzoyl hydroxylamines under mild conditions. Even without additional ligands, the protocol reveals broad substrate scope for both partners and tolerates all kinds of functional groups. Furthermore, the proposed sulfinate intermediate could be critical for this ligand‐free catalytic transformation.

Abstract:Sulfonamides are prominently featured in organic synthesis and medicinal chemistry. Yet, a general synthetic platform for the modular construction of aryl sulfonamides remains elusive. Herein, a metal-free photoredox-catalyzed three-component construction of aryl sulfonamides via an aryl sulfonyl ammonium salt intermediate, which was generated in situ via a SET event, has been established. A variety of structurally diverse primary, secondary, and tertiary aryl sulfonamides were synthesized rapidly from abundant amines or sodium azide under mild conditions. Notably, the primary aliphatic amine, which remains challenging in the Cu-catalyzed protocols, worked well in this approach. Moreover, aryl sulfonyl fluoride can be accessed smoothly as well, using potassium hydrogen fluoride as the nucleophile. The potential utility of this conversion is demonstrated in the facile construction of three bioactive drug compounds. Preliminary mechanistic studies have revealed that the aryl sulfonyl radical and aryl sulfonyl ammonium salt are the key intermediates in this mechanistically innovative methodology.

Abstract:A versatile copper catalyzed one‐pot synthesis of diversely substituted pyrimidines directly from amides, N,N′‐dimethylformamide dimethylacetal (DMF−DMA) and enamines has been established. The reaction involved the two C−N bonds and one C−C bond formation by formal [2+1+3] annulation approach to pyrimidines. This protocol is based on the use of readily available primary amides, DMF−DMA and enamines to install di‐ and tri‐substituted pyrimidine structure with diverse functionality in one‐pot manner, which makes this strategy to be appealing for the medicinal chemistry.

Abstract:A palladium-catalyzed three-component reaction enabled the synthesis of N-formylanilines from aryl iodides, NaN(3), and oxalic acid. This one-pot process involves aminocarbonylation, the Curtius rearrangement, and reduction using oxalic acid as both a carbon monoxide and hydrogen donor. The method has a broad substrate scope, tolerates various functional groups, and delivers N-formylanilines in high yields, making it a green and useful synthetic approach.

Abstract:A new strategy for the sequential formation of aryl and amidyl C-N bonds is reported. Using trichloroethoxysulfonyl azide as a bifunctional nitrogen source, Ir-catalyzed aryl C-H sulfonamidation and subsequent desulfonative amide formation proceed effectively without any need of oxidants or coupling reagents. This protocol is suitable for readily available benzamides and stable carboxylates including primary, secondary, and tertiary alkyl, alkenyl, and phenyl carboxylates, thereby providing a direct and efficient method for the synthesis of biologically and chemically useful N-arylamides.
