Influence of malting procedure on the isoflavonoid content of soybeans

Abstract:The goal of this study was to analyse, whether malting technique (consisting of seed hydration, germination and drying) can be used to modify concentration of various isoflavonoids in soybean seeds. Seeds of three soybean varieties were germinated by different lengths of time (from 24 to 120 h) and dried by two different methods, typically used to produce so-called 'light' and 'caramel' malts. It was determined, that malting decreases concentration of 7-O-β-D-glucosides such as daidzin, genisitin and glycitin, while at the same time increasing concentration of aglycones (daidzein, genistein and glycitein). Increasing time of the germination period increased concentration of aglycones. 'Caramel' type malts were characterised with higher concentration of most of the isoflavonoids (daidzin, daidzein, genistin, genistein and glycitein) than 'light' type malts. Results of this study suggest that soybean malts can be an interesting substrate in the production of various food products with increased aglycone content.

Soybean contains rich isoflavones, and these compounds have multiple health benefits, such as antioxidant, anti - inflammatory and anti - cancer effects. - **Types of isoflavones**: The main isoflavones include aglycones (such as genistein, daidzein and glycitein) and 7 - O - β - D - glucosides (such as genistin, daidzin and glycitin). - **Application of germination techniques**: Germination techniques are usually used for grains (such as barley), but in recent years they have also been applied to legume seeds to improve their nutritional and processing characteristics. ### Research methods: - **Materials**: Three different varieties of soybean seeds (Abaca, Abelina and Aurelina) were selected. - **Germination and drying**: 30 different malt samples were prepared through different germination times (24 hours, 48 hours, 72 hours, 96 hours and 120 hours) and two drying methods (continuous drying at 50 °C and programmed drying of caramel malt). - **Isoflavone extraction and analysis**: The content of isoflavones was analyzed by high - performance liquid chromatography (HPLC). ### Research results: - **Changes in aglycones and glucosides**: The germination technique significantly reduced the content of 7 - O - β - D - glucosides while increasing the content of aglycones. As the germination time extended, the content of aglycones further increased. - **Effect of different drying methods**: Compared with continuous drying at 50 °C, the drying method of caramel malt significantly increased the content of most isoflavones. - **Differences among different soybean varieties**: There are significant differences in the changes of isoflavone content during the germination process among different soybean varieties, which may be related to the activity and quantity of enzymes in different varieties. ### Conclusion: The results of this study indicate that the germination technique can be used as an effective method to increase the content of aglycones in soybean products, thereby producing foods rich in health - promoting components. In addition, the effects of different germination times and drying methods on the isoflavone content also provide important references, which are helpful for optimizing the processing technology of soybean products.
