Abstract:Heteroatom doping except oxygen is the main method to improve the performance of carbon quantum dots (CQDs), the application is also limited by unpredictable biological negative effects and environmental risks from heteroatoms (N/P/S, etc.). To maximize the role of oxygen while avoiding the negative effects of other heteroatoms, herein, we rationally designed and synthesized a new type of carbon-oxygen quantum dots (COQDs) with high fluorescent performance aim to realize pH sensing and reactive oxygen species (ROS) scavenging. The toxicity test results from four levels of bacterias, Hela cells, Daphnia Magna, and zebrafish indicate that the designed COQDs possess high biocompatibility and low environmental risk. Based on the opposing response of COQDs and Rhodamine 6G (Rh 6G) fluorescent signal to pH changes in the range of 6.0–9.0, a COQDs-Rh 6G complex was constructed by combining Rh 6G for ratiometric pH sensing, and a scanning colorimetric analysis model was further developed for the rapid pH judgment of commercially available waters. The proposed COQDs and pH sensing system were also successfully applied for bioimaging and intracellular pH analysis, respectively. More importantly, the designed COQDs also exhibit excellent ROS scavenging ability, which can be used as an effective antioxidant with good biocompatibility.

Abstract:Reactive oxygen and nitrogen species (ROS and RNS) produced by macrophages are essential for protecting a human body against bacteria and viruses. Micrometer-sized electrodes coated with Pt black have previously been used for selective and sensitive detection of ROS and RNS in biological systems. To determine ROS and RNS inside macrophages, one needs smaller (i.e., nanometer-sized) sensors. In this article, the methodologies have been extended to the fabrication and characterization of Pt/Pt black nanoelectrodes. Electrodes with the metal surface flush with glass insulator, most suitable for quantitative voltammetric experiments, were fabricated by electrodeposition of Pt black inside an etched nanocavity under the atomic force microscope control. Despite a nanometer-scale radius, the true surface area of Pt electrodes was sufficiently large to yield stable and reproducible responses to ROS and RNS in vitro. The prepared nanoprobes were used to penetrate cells and detect ROS and RNS inside macrophages. Weak and very short leaks of ROS/RNS from the vacuoles into the cytoplasm were detected, which a macrophage is equipped to clean within a couple of seconds, while higher intensity oxidative bursts due to the emptying of vacuoles outside persist on the time scale of tens of seconds.

Abstract:Five major reactive oxygen species (ROS) are generated in diseases including H2O2, (OH)-O-center dot, O-2(center dot-), ROO center dot, and O-1(2). Simultaneous detection of the five ROS with a single probe is crucial for a comprehensive understanding of the development and progression of many diseases, such as cancer and inflammatory diseases. However, currently reported detection systems are limited by targeting one ROS with one probe. This one-to-one detection mode may fail to sufficiently unveil the diseased state. In this study, we achieved simultaneous detection of all the five ROS with one probe (i.e., one-to-all detection), by designing a novel para-aminothiophenol (PATP) and hemin-decorated gold (Au/PATP/Hemin) nanoprobe. The design is principled by our discovery that PATP can react with (OH)-O-center dot, O-2(center dot-), ROO center dot, and O-1(2) by a radical oxidative coupling mechanism to form 4,4'-dimercaptoazobenzene (DMAB). The DMAB then elicited strong characteristic surface-enhanced Raman scattering (SERS) peaks at 1142, 1386, and 1432 cm(-1); which in turn enables direct detection of (OH)-O-center dot, O-2(center dot-), ROO center dot, and O-1(2) and indirect detection of H2O2 by hemin-catalyzed fenton reaction to convert H2O2 into (OH)-O-center dot. In two representative ROS-elevated mice models of tumors and allergic dermatitis, the Au/PATP/Hemin nanoprobe demonstrated its robust performance of monitoring tumor development and inflammation progression in a highly sensitive and quantitative manner.

Abstract:Reactive oxygen species (ROS) encompasses a collection of complicated chemical entities characterized by individually specific biological reactivities and physicochemical properties. ROS detection is attracting tremendous attention. The reaction-based nanomaterials for ROS “turn-on” sensing represent novel and efficient tools for ROS detection. These nanomaterials have the advantages of high sensitivity, real-time sensing ability, and almost infinite contrast against background. This review focuses on appraising nanotechnologies with the ROS “turn-on” detection mechanism coupled with the ability for broad biological applications. In this review, we highlighted the weaknesses and advantages in prior sensor studies and raised some guidelines for the development of future nanoprobes.

Abstract:Living cells can rapidly adjust their metabolic activities in response to external stimuli, leading to fluctuations in intracellular temperature and reactive oxygen species (ROS) levels. Monitoring these parameters is essential for understanding cellular metabolism, particularly during dynamic biological processes. In this study, we present a bifunctional nanoprobe capable of simultaneous measurement of ROS levels and temperature within single cells. The nanoprobe features two individually addressable nanoelectrodes, with platinum (Pt) and nickel (Ni) coatings on both sides. At the tip, these two metal layers form a nano-thermocouple, enabling precise intracellular temperature measurements, while the Pt layer facilitates selective ROS detection. This dual functionality allows for real-time monitoring of cellular responses during synergistic chemo-photothermal therapy of cancer cells and zebrafish embryos subjected to mitochondrial toxic stress. Our results demonstrate that the nanoprobe effectively measures increases in temperature and ROS levels in HeLa cells undergoing chemo-photothermal therapy, as well as in chemically stimulated zebrafish embryos. By providing detailed analysis of submicrometer-scale temperature and ROS variations within living cells, this nanoprobe offers valuable insights into cellular processes and holds promise for early disease detection and drug development.
Dual Ligand–capped Gold Nanoclusters for the Smart Detection of Specific Reactive Oxygen Species

Abstract:Quantitative detection of different types of reactive oxygen species (ROS) is vital for understanding the crucial roles of them in biological processes. However, few researches achieved the detection of multiple types of ROS with one probe until now. Given this, we designed and prepared fluorescent gold nanoclusters capped by dual ligand bovine serum albumin and lysozyme (BSA-LYS-AuNCs), which could detect 3 specific types of ROS based on its different fluorescent responses to H2O2, •OH and ClO−, respectively. The limit of detection (LOD) of H2O2, •OH, and ClO− was as low as 0.82 μM, 0.45 μM, and 0.62 μM. Moreover, as an important ROS type, ClO− was detected with high sensitivity and low LOD by BSA-LYS-AuNCs. It was also proved that the crosslinking of protein mainly contributed to the unique fluorescent characteristics of the probe exposing to ClO−. Furthermore, the fluorescent probe achieved the smart detection of hROS (including •OH and ClO−) and wROS (the form of H2O2) in the real sample, which could also been applied specifically to the detection of antioxidants, e.g. ascorbic acid. The gold nanoclusters developed have high potential for the smart detection of multiple ROS in the body fluid of organisms.
A highly sensitive and selective artificial nanochannel for in situ detection of hydroxyl radicals in single living cell

Abstract:As the most aggressive reactive oxygen species (ROS), hydroxyl radical (•OH) can directly modulate the biological ion channel and interfere with the progression of diseases. Inspired by biological •OH-activated ion channel, we reported a novel •OH-regulated glass nanopore functionalized with protoporphyrin Ⅸ (PP) film. This system showed outstanding •OH selective response owing to the ultra-fast reaction between •OH and thiol derivatives. In this case, the PP film is responsible for the changing not only of wettability but also of the inner surface charge. The synergetic effect of the dual transitions can regulate the ion transportation within the nanochannels and enabled tremendous enhancement of responsive efficiency. The detection limit could be achieved down to 1.58 nM. Taking advantage of the excellent analytical performance and mechanical qualities of this glass nanopore, the changes of •OH in single living cells were in situ monitored. Together, this study is beneficial for exploring the role of •OH in pathological events and shows promising potential for biomedical research.

Abstract:Hydroxyl radicals (•OH) is a type of short-lived radicals which is the most aggressive reactive oxygen species due to its high reac-tivity to biomolecules. Dynamic measurement of •OH level in living cells is critical for understanding cell physiology and patholo-gy. In this manuscript, we prepare individual Ag-Au@PEG/RGD nanocages for in-situ determination of endogenous •OH at single-cell level, whose spectral shift rate correlate to the •OH concentration. The high-selective response to •OH relies on the specific oxidization of the conjugated PEG/RGD outside and the silver etching inside the nanocages that resulted in a significant LSPR signal and scattered color changes. The spectral red-shift rate of LSPR has a linear relationship with the logarithm of •OH concen-tration in range of 100 pM to 1 μM, suitable for the measurement of endogenous •OH. Thus, the individual nanocages were suc-cessfully used to monitor the dynamic intracellular •OH level of single tumor cells under oxidative stress. This strategy has great potential in promoting •OH mediated cell homeostasis and injury research.

Abstract:Excessive or burst generation of reactive oxygen species (ROS) can induce oxidative stress, precipitating a range of critical illnesses, including cancers, Parkinson's disease and Ischemia-reperfusion injury. Conventional biological assays for ROS, involving discrete steps of capturing, labelling, and spectrometric detection, are complex and time-intensive. Moreover, their accuracy is substantially compromised by the short lifespan (microseconds to milliseconds) of ROS. Consequently, there is a pressing need for a rapid and efficient method that enables real-time detection. In this study, we have developed a printable, flexible ROS sensor based on a robust nanoenzyme composite by direct deposition of the paste onto a flexible polyethylene terephthalate (PET) substrate. This device demonstrated the fast and real-time responses to the hydrogen peroxide (mimetic agent) in the laboratory and to total ROS in sweat of an individual, exhibiting an outstanding current response to hydrogen peroxide across a broad concentration range of 0.01-10 mM, with a limit of detection (LOD) of 1.85 μM. The device's sensitivity to hydrogen peroxide (136.59 μA mM-1 cm-2), was found to be 1.5 to 10 times higher than that of sensors previously reported. Moreover, the IFRS device successfully identified instantaneous ROS levels in the sweat of adult males in vitro, with amperometric response increased 8 times after half an hour strenuous exercise, thereby exhibiting excellent selectivity, remarkable stability, and confirmed high biosafety. Overall, the IFRS provides a viable and practical solution for simple, expedited, and real-time ROS detection in the near future.

Abstract:Reactive oxygen and nitrogen species (ROS and RNS) are continuously produced in the cellular systems and are controlled by several antioxidant mechanisms. Here, we developed a straightforward, sensitive, and quantitative assay for the colorimetric and spectroscopic detection of various ROS and RNS such as H2O2, ·OH, (-)OCl, NO·, and O2(-) using glutathione-modified gold nanoparticles (GSH-AuNPs). A basic principle here is that the GSHs on the AuNP surface can be readily detached via the formation of glutathione disulfides upon the addition of ROS and RNS, and destabilized particles can aggregate to generate the plasmonic couplings between plasmonic AuNPs that trigger the red shift in UV-vis spectrum and solution color change. For nonradical species such as H2O2, this process can be more efficiently achieved by converting them into radical species via the Fenton reaction. Using this strategy, we were able to rapidly and quantitatively distinguish among cancerous and normal cells based on ROS and RNS production.

Abstract:Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are prominent metabolic products which show well-established significance. At relatively low concentrations, they play multifaceted roles in regulating a number of physiological processes. Overproduction of ROS/RNS contributes to the pathogenesis of a plethora of physiological disorders, including but not limited to cardiovascular diseases, neurodegenerative diseases, cancer. Electrochemistry have been extensively used for detecting and monitoring ROS/RNS, benefiting from their inherent advantages including fast response, low costs, real-time detection, high sensitivity and selectivity. This review focuses on three types of ROS/RNS (H2O2, O2-, NO) with emphasis on their electrochemical detection/monitoring respectively. We demonstrate the application of electrochemical strategies for ROS/RNS detection in body fluids, in vitro, and in vivo, outlining the hardware architecture and comparing analytical performance of these sensors. This review aims for a holistic view of limitations in existing ROS/RNS detection by comprehensively explaining the shortcomings of the current works in the hope of drawing attentions to the challenges of ROS/RNS electrochemical technologies. We pay particular attention to in vitro and in vivo sensors and extend our evaluation to suggest possible solutions. Specifically, this review focuses on the development of currently nanotechnologies, biomimetic engineering, 3D-culture methods and implanted sensors to provide a guideline for future works.

Abstract:This Account describes a range of strategies for the development of fluorescent probes for detecting reactive oxygen species (ROS), reactive nitrogen species (RNS), and reactive (redox-active) sulfur species (RSS). Many ROS/RNS have been implicated in pathological processes such as Alzheimer's disease, cancer, diabetes mellitus, cardiovascular disease, and aging, while many RSS play important roles in maintaining redox homeostasis, serving as antioxidants and acting as free radical scavengers. Fluorescence-based systems have emerged as one of the best ways to monitor the concentrations and locations of these often very short lived species. Because of the high levels of sensitivity and in particular their ability to be used for temporal and spatial sampling for in vivo imaging applications. As a direct result, there has been a huge surge in the development of fluorescent probes for sensitive and selective detection of ROS, RNS, and RSS within cellular environments. However, cellular environments are extremely complex, often with more than one species involved in a given biochemical process. As a result, there has been a rise in the development of dual-responsive fluorescent probes (AND-logic probes) that can monitor the presence of more than one species in a biological environment. Our aim with this Account is to introduce the fluorescent probes that we have developed for in vitro and in vivo measurement of ROS, RNS, and RSS. Fluorescence-based sensing mechanisms used in the construction of the probes include photoinduced electron transfer, intramolecular charge transfer, excited-state intramolecular proton transfer (ESIPT), and fluorescence resonance energy transfer. In particular, probes for hydrogen peroxide, hypochlorous acid, superoxide, peroxynitrite, glutathione, cysteine, homocysteine, and hydrogen sulfide are discussed. In addition, we describe the development of AND-logic-based systems capable of detecting two species, such as peroxynitrite and glutathione. One of the most interesting advances contained in this Account is our extension of indicator displacement assays (IDAs) to reaction-based indicator displacement assays (RIAs). In an IDA system, an indicator is allowed to bind reversibly to a receptor. Then a competitive analyte is introduced into the system, resulting in displacement of the indicator from the host, which in turn modulates the optical signal. With an RIA-based system, the indicator is cleaved from a preformed receptor–indicator complex rather than being displaced by the analyte. Nevertheless, without a doubt the most significant result contained in this Account is the use of an ESIPT-based probe for the simultaneous sensing of fibrous proteins/peptides AND environmental ROS/RNS.

Abstract:Reactive oxygen species (ROS), produced during oxygen metabolism, participate in and regulate various life processes. It is of great significance to monitor ROS in biological organs to further study oxygen metabolism. Herein, an ultrasensitive sensing platform is developed with electrochemiluminescent (ECL) signalling by integrating bioactive magnetic beads (BMBs) on indium tin oxide (ITO) coated glass using a magnet. For the first time, AuNPs were successfully deposited on Fe3O4 NPs in situ by reduction of α-ketoglutaric acid (α-KG), therefore the electroactive protein, haemoglobin (Hb) or cytochrome C (Cyt C), was assembled on via covalent bonds. The protein can realize direct electron transfer (DET) and catalyse the redox of ROS, reaching a detection limit of 6.21 μM or 0.6 μM of H2O2. Also Au@Fe3O4 NPs efficiently enhanced the ECL of luminol, promoting the sensing ability for ROS. This simultaneous effect endows the platform with low LOD of ROS for 7.69 nM (Hb), or 1.97 nM (Cyt C). Finally, the feasibility and practicality of the sensing platform were verified by monitoring the ROS released from mouse myocardial tissue.

Abstract:Quantification of hydroxyl radical (•OH), one form of reactive oxygen species (ROS), plays critical roles in early diagnosis and treatment monitoring of various diseases. In this work, we report the development of a responsive nanoprobe for ratiometric fluorescence detection and imaging of •OH in macrophage polarization. The nanoprobe, BSA-CCA@LDH-SRB, was designed and prepared using coumarin 3-carboxylic acid (CCA) as the sensing unit for •OH, and sulforhodamine B (SRB) loaded on layered double hydroxide (LDH) served as the fluorescent reference component. The coupling of CCA to bovine serum albumin (BSA) and the loading of BSA-CCA on the surface of LDH enabled the nanoprobe for fluorescence detection of •OH with high sensitivity and minimal interference from other biomolecules, ions, and ROS. The emission of the prepared BSA-CCA@LDH-SRB at 444 nm emerged and the intensity was increased according to the concentration of •OH, while the emission at 580 nm was maintained, allowing the nanoprobe for ratiometric fluorescence (F444/580) detection of •OH. Loading of the BSA protein on LDH surface, the biocompatibility and colloidal stability of the LDH-based fluorescent nanoprobe were further improved, facilitating the detection of •OH generation in macrophage polarization stimulated by both biomolecules and physical ultrasound irradiation. This study thus offers a new nanoprobe as the tool for investigating •OH evolutions, advancing the biomedical investigations of macrophage polarization associated inflammation.

Abstract:Reactive oxygen and nitrogen species (ROS and RNS) play important roles in various physiological processes (e.g. phagocytosis) and pathological conditions (e.g. cancer). The primary ROS/RNS, viz., hydrogen peroxide, peroxynitrite ion, nitric oxide, and nitrite ion, can be oxidized at different electrode potentials and therefore detected and quantified by electroanalytical techniques. Nanometer-sized electrochemical probes are especially suitable for measuring ROS/RNS in single cells and cellular organelles. In this article, we survey recent advances in the localized measurements of ROS/RNS inside single cells and discuss several methodological issues, including optimization of nanoelectrode geometry, precise positioning of an electrochemical probe inside a cell, and interpretation of electroanalytical data.

Selective and Sensitive Detection of Intracellular O2(•-) Using Au NPs/cytochrome C As SERS Nanosensors.

Abstract:A novel surface-enhanced Raman scattering (SERS) nanosensor was developed by modifying oxidized cytochrome c (Cyt c) on gold nanoparticles (Au NPs) for the sensitive and selective determination of intracellular superoxide anion radical (O2(•-)). On the basis of the differences in the SERS spectra between the oxidized and reduced form of Cyt c, this nanosensor could be employed to investigate O2(•-) concentration by measuring the SERS spectra of the reduced Cyt c. Using this SERS nanosensor, a detection limit of 1.0 × 10(-8) M for O2(•-) could be attained. Additionally, the selectivity of the SERS nanosensor for O2(•-) was examined, showing that other reactive oxygen species (ROS) and biologically relevant species did not influence the detection of O2(•-). More importantly, the nanosensor could be delivered to the living HeLa and normal human liver cells and permitted the concentration of O2(•-) to be monitored in real time and in a noninvasive manner, which indicates that this nanosensor will be suitable for the qualitative and quantitative analysis of O2(•-) in biosystems, thus leading to a greater understanding of oxidative-stress-related diseases at a cellular level.

Abstract:Reactive oxygen species (ROS), well-known as the highly reactive small molecules, play an important role in physiological and pathogenic events. Therefore, the detection of ROS receives a great deal of interest. With the rapid development of nanotechnology and nanomaterials, the nanomaterial-based chemiluminescence (CL) biosensors with good selectivity and high sensitivity have risen for tracing of the ROS. The nanomaterial-based CL biosensors not only provide the quantitative detection of ROS, but also the real-time generation dynamic of ROS. In this review, we summarized the applications of nanomaterial-based CL biosensors for tracing of ROS in recent years. Furthermore, we focused on the approaches for the design of nanomaterial-based CL biosensors for tracing of ROS, including the selection of nanomaterials and the basic mechanism for selective detection of ROS. Finally, we provided our thinking and predictions on exploring novel nanomaterial-based CL platforms. This review may open viable opportunities and inspirations for the researchers in analytical chemistry and material science to design new strategies for ROS tracing in a variety of biological environments.

Abstract:Ionic current rectification (ICR) based nanopipettes allow accurate monitoring of cellular behavior in single living cells. Herein, we proposed a 30 nm nanopipette functionalized with G-quadruplex DNAzyme as an efficient biomimetic recognizer for ROS generation at subcellular level via the changes of current-voltage relationship. Taking advantages of the ultra-small tip, the nanopipette could penetrate into a single living cell repeatedly or keep measuring for a long time without compromising the cellular functions. Coupled with precision nanopositioning system, generation of ROS in mitochondria in response to cell inflammation was determined with high spatial resolution. Meanwhile, the changes of aerobic metabolism in different cell lines under drug-induced oxidative stress were monitored continuously. We believe that the ICR-nanopipette could be developed as a powerful approach for the study of cellular activities via electrochemical imaging in living cells.
