Abstract:Three new esters of aspirin were synthesized and evaluated in vitro as potential prodrug forms of aspirin with the aim of depressing the gastrotoxicity of the drug by temporarily masking the carboxylic acid function. The esters, derived from N-(hydroxymethyl)acetamide, N-(hydroxymethyl)benzamide and ga-hydroxy-N-benzoylglycine benzyl ester, were all found to undergo a facile hydrolysis in aqueous solution of pH 0–8 with a quantitative regeneration of aspirin. The compounds were very easily cleaved at pH 7.4 but were more stable at lower pH values. Due to the great lability at pH 7.4 the compounds were also found to be cleaved quantitatively or predominantly to aspirin in the presence of human plasma rather than to the corresponding salicylate esters and hence salicylic acid.

Abstract:5-Bromo-2-hydroxy-3-(1-propionyl) benzoic acid methyl ester was synthesized by using salicylic acid as raw material.The mass of catalyst,molar ratio of methanol to salicylic acid in the esterification,the effects of the solvent,feeding mode and molar ratio of reactants in the C-acylation were respectively investigated. The product was characterized by IR and GC-MS.The result indicated that the optimum technical conditions were as follows:m(ion exchange resin):m(salicylic acid)=1.00%,n(methanol):n(salicylic acid) = 7:1,chloroform as solvent;ester was added into the chloroform solution of aluminum chloride;n(aluminum chloride):n(5- bromo-salicylic acid methyl ester):n(propionic anhydride) = 2:1:1.Under these conditions,the total yield of desired product was 74.52%.

Abstract:N-Methyl-4-aminobenzyl sulfonamide, a key intermediate for synthesis of the pharmaceutical substance Sumatriptan, was prepared throught four steps. 4-Nitrobenzyl chloride was first converted to 4-nitrobenzyl sulphonic acid by reacting with sodium hydrosulfite, then the sulphonic acid obtained was oxidized to 4-nitrobenzyl sulfonyl chloride by chlorine, and N-methyl-4-nitrobenzyl sulfonamide was produced by reacting with methylamine, in the end, the title product was obtained by reducing the nitro group to amino group with hydrazine hydrate in the presence of iron trichoride on active carbon as the catalyst. The yields of the four steps are 90%, 93%, 60% and 77%, respectively. The intermediates and product were characterized by GC-MS and H~1NMR.

Abstract:Acetylsalicylic acid commonly known as aspirin is the most widely used drug in the world today. Its analgesic, antipyretic, and anti-inflammatory properties make it a powerful and effective drug to relive symptoms of pain, fever, and inflammation. The purpose of this experiment was to synthesize aspirin by reacting salicylic acid and acetic anhydride in the presence of phosphoric acid to form acetylsalicylic acid. After synthesis, the sample of acetylsalicylic acid was purified by recrystallization and TLC analysis was utilized to check the purity of the sample. The actual yield of aspirin synthesized was 0.546 grams and the theoretical yield was 2.628 grams, thus the percent yield was 20.77% resulting in a percent error of 79.22%. The Rf values of crude product, salicylic acid, and recrystallized product were 0.590 and 0.897, 0.818, and 0.606 and 0.879 respectively. Due to these Rf values, it was brought to the conclusion that the crude and recrystallized aspirin were composed of both salicylic acid and acetylsalicylic acid.

Catalytic Synthesis of Aspirin with Sodium Acetate

Abstract:Catalytic synthesis of Aspirin over pretreated sodium acetate using salicylic acid and acetic anhydride as starting materials. The results showed when the amount of salicylic acid was 3.0 g, acetic anhydride 6 mL, sodium acetace3%(wt% of salicylic acid and acetic anhydride),55 ℃ reaction for 50 min,the yield of purified Aspirin was 81.9%.The product is characterized by IR and NMR spectrometry. The effects of dose catalyst-to-crude ratio, reaction temperature and reaction time on the yield of Aspirin are discussed.

Abstract:Aspirin was polycondensed at various temperature-time combinations with or without the addition of a catalyst. Furthermore, several polycondensations of both the trimethylsilyl ester of aspirin, and o-trimethylsiloxybenzoyl chloride were conducted. Viscosity and GPC measurements indicated that from all three monomers only oligoesters having number average molecular weights (M̄n) ≤ 1 500 Da were obtained. FAB mass spectra of crude reaction products revealed that cyclic oligoesters (having DPs up to 7) were formed as the main reaction products under all circumstances. When ortho and meta trimethylsiloxybenzoyl chloride were copolycondensed with variation of the molar ratio, the molecular weights decreased with higher feed ratios of the salicylic acid derivative. Obviously, the salicylic monomer favored cyclization reactions at the expense of the chain growth. The predominant formation of cyclic oligoesters (with M̄ns up to 5 000 Da) was confirmed by MALDI-TOF mass spectra.

Abstract:Pharmaceutical synthesis commonly relies on a homogeneous acid catalyst, which is relatively simple in application yet problematic in product purification procedures. This study was an effort to investigate an alternative replacement for homogeneous acid catalysts with an acidified solid catalyst, developed from activated carbon. The activated carbon catalyst had been treated with sulfuric acid before it was applied to produce aspirin (acetylsalicylic acid) through catalytic acetylation route. Using atmospheric pressure reaction, conducted at 353 K, aspirin was successfully synthesized. When salicylic acid and acetic acid glacial were used as reactants, approximately 9-10 % reaction yield could be obtained. Replacement of acetic acid with acetic anhydride in the acetylation reaction produced an overall yield as high as 28 %.
Catalytic synthesis of aspirin by trifluoromethanesulfonic acid

Abstract:Trifluoromethanesulfonic acid was used as the catalyst for synthesis of aspirin from salicylic acid and acetic anhydride at 50 ℃.The effects of catalyst amounts,molar ratio of salicylic acid to acetic anhydride and reaction time on the product yield was investigated.The results showed that the optimum reaction conditions were found as follows:0.2% catalyst based on salicylic acid,molar ratio of salicylic acid to acetic anhydride 1∶ 2.0,and reaction time was 50 min.Under this condition,the yield of aspirin was 90.4%.

Abstract:Abstract The synthesis method of lysine aspirin was improved, and the new method was applied to the synthesis of similar new drugs. Aspirin is reacted with an aqueous solution of sodium hydrogen carbonate to prepare its sodium salt, and lysine was reacted with a sodium salt thereof in water to prepare lysine aspirin. The product was identified by melting point, infrared spectroscopy and high performance liquid chromatography; the cysteine salts of aspirin was prepared by this method, and the product was identified by micro melting point apparatus and infrared spectroscopy. Lysine aspirin was synthesized by the reaction of sodium acetylsalicylate with lysine. The author measured the melting point and infrared absorption spectrum of the cysteine salt sample of aspirin, which was initially determined to be possible. The new method offers the possibility to expand the production route of lysine aspirin, and also provides new ideas for the preparation of similar new drugs.

Abstract:OBJECTIVE To study and improve the synthesis of donepezil.METHODS 3, 4-dimethoxy-benzaldehyde was condensed with malomc acid in the presence of pyridine, then hydrogenated with Raney Ni to get3-(3,4-dimethoxy-phenyl)-propanic acid, which was cy-clized with poly-phosphoric acid (PPA) to yield 5,6-dimetrioxy-l-indenone (intermediate 1). N-benzyl piperidone was epoxidated with (CH3)2 SOCH2- to get epoxide, which was rearranged with magnesium bromide-etherate to get N-benzyl-4-formyl-piperidine (intermediate 2) . The two intermediates were condensed with NaOH, followed by selective hydrogenation with 5% Pd-C and hydrochlorination to yield donepezil. RESULTS The structures of intermediates and donepezil were identified by IR,1H-NMR and MS.CONCLUSION This method avoided using expensive reagents such as n-BuLi, IDA, and there was no need to use column chromatograph in all steps.
Condition investigation on the synthesis of Aspirin by vitamin C catalyst

Abstract:Objective:To synthesize Aspirin by using vitamin C as the catalyst,salicylic acid as the raw material and acetic anhydride as the acylating agent.Methods:The method of acetylation was used.Results:The synthesis conditions was determined,the temperature was 80℃,the reaction time was 40 min,the quantity of vitamin C was 0.2 g,the yield was 56.8%.Conclusion:The method is feasible,and the synthesis condition needs to be improved.

Study on catalytic synthesis of aspirin with potassium dihydrogen phosphate

Abstract:Aspirin was synthesized from salicylic acid and acetic anhydride with potassium dihydrogen phosphate catalyst by ultrasonic oscillation.The optimum laboratory conditions obtained by orthogonal experiments were that 3.0 g salicylic acid,6.2 mL acetic anhydride(1 ∶3,mol/mol),0.5 g potassium dihydrogen phosphate,reacted 30～40 min at 75～80 ℃.The FTIR spectrum of the product was completely consistent with the standard spectrum.The potassium dihydrogen phosphate may be used repeatedly as catalyst with nearly 90% recovery.Comparing with the concentrated sulphuric acid catalysis,the potassium dihydrogen phosphate catalyst may be recycled,and has the advantages of security,environmental protection.This method is suitable for undergraduate teaching and industrial production.

Abstract:The aim of this work is to develop an environmentally friendly, safe, and simple route for realizing efficient preparation of aspirin. Here, inspired by enzyme synthesis in vivo, the aspirin synthesis has been realized by sub-nanoconfined esterification with directional flow and ≈100% conversion in an unprecedented reaction time of <6.36 s at 23 °C. Such flow esterification reaction is catalyzed by thermally transformed graphene oxide (GO) membranes with tailored physicochemical properties, which can be obtained simply through a mild annealing method. A possible mechanism is revealed by density functional theory calculation, indicating that the synergistic effect of spatial confinement and surface electronic structure can significantly improve the catalytic performance. By restricting reactants within 2D sub-nano space created by GO-based laminar flow-reactors, the present strategy provides a new route to achieve rapid flow synthesis of aspirin with nearly complete conversion.
Synthesis of Aspirin by Microwave Radiation

Abstract:Aspirin was synthesized through the method of microwave radiation with salicylic acid and acetic anhydride as raw materials.Catalytic effects of stronger sulfuric acid,aluminum chloride and anhydrous sodium carbonate were compared.The optimum synthetic conditions of aspirin were investigated.The results show that when aluminum chloride is used as catalyst,its amount is 3% of the salicylic acid mass,ratio of acetic anhydride and salicylic acid is 2.5∶1,microwave power is 200 W,radiation time is 60 s,yield of aspirin can reach 81.4%.

Abstract:Aspirin was used as the lead compound, and its structure was modified by galactosylation. Galactose-based aspirin prodrug was synthesized by using the α-d-glactopyranosyl bromide as glycosyl donor and aspirin as acceptor. The experimental method is simple and reproducible, has high yield and great practical value. The galactosylated aspirin prodrug was found to possess reduced cytotoxicity compared to aspirin, and selectively exhibited antiproliferative activity in the presence or in the absence of β-d-galactoside galactohydrolase with the approach of antibody-directed enzyme prodrug therapy (ADEPT) or the prodrug monotherapy (PMT).

The product is characterized by IR and UV spectrometry. This method had the application value.

Abstract:<p>Aspirin is a widely used medicine for a variety of indications. It is unique amongst non-steroidal anti-inflammatory drugs (NSAIDs) in that it causes irreversible acetylation of COX enzymes. Like all NSAIDs however, aspirin causes severe gastrointestinal side-effects, in particular with chronic administration. Prodrugs of aspirin have been proposed as a solution to these side-effects. However, identifying true prodrugs of aspirin, rather than salicylic acid, has proven challenging. This review details the challenges and highlights recent progress in the development of such prodrugs.</p>
