Abstract:We present a high sensitivity single-monochromator Raman spectrometer which allows operation with a tunable laser source. The instrument is based on the modification of a commercial Raman spectrometer; such instruments operate with holographic Rayleigh filters which also act as a laser mirrors and are usually considered as inherently narrow-band. In our design, the two tasks are separated and the filter can be freely rotated without much effect on the light alignment. Since rotation shifts the filter passband, this modification allows tunable operation with efficient stray light filtering down to 150 1/cm. The design is optimized for single-wall carbon nanotubes, for which the performance is demonstrated using a tunable dye-laser source. The spectrometer thus combines the high sensitivity with the broadband characteristics of usual triple monochromator systems.

Abstract:We present an optical nanotrapping setup that exhibits enhanced efficiency, based on localized plasmonic fields around sharp metallic features. The substrates consist of laser-structured silicon wafers with quasi-ordered microspikes on the surface, coated with a thin silver layer. The resulting optical traps show orders of magnitude enhancement of the trapping force and the effective quality factor.

Tip-enhanced Near-Field Raman Spectroscopy with a Scanning Tunneling Microscope and Side-Illumination Optics

Abstract:Conventional Raman spectroscopy (RS) suffers from low spatial resolution and low detection sensitivity due to the optical diffraction limit and small interaction cross sections. It has been reported that a highly localized and significantly enhanced electromagnetic field could be generated in the proximity of a metallic tip illuminated by a laser beam. In this study, a tip-enhanced RS system was developed to both improve the resolution and enhance the detection sensitivity using the tip-enhanced near-field effects. This instrument, by combining RS with a scanning tunneling microscope and side-illumination optics, demonstrated significant enhancement on both optical sensitivity and spatial resolution using either silver (Ag)-coated tungsten (W) tips or gold (Au) tips. The sensitivity improvement was verified by observing the enhancement effects on silicon (Si) substrates. Lateral resolution was verified to be below 100 nm by mapping Ag nanostructures. By deploying the depolarization technique, an apparent enhancement of 175% on Si substrates was achieved. Furthermore, the developed instrument features fast and reliable optical alignment, versatile sample adaptability, and effective suppression of far-field signals.
Rapid theoretical method for inverse design on a tip-enhanced Raman spectroscopy (TERS) probe

Abstract:Tip-enhanced Raman spectroscopy (TERS) can provide correlated topographic and chemical information at the nanoscale, with great sensitivity and spatial resolution depending on the configuration of the TERS probe. The sensitivity of the TERS probe is largely determined by two effects: the lightning-rod effect and local surface plasmon resonance (LSPR). While 3D numerical simulations have traditionally been used to optimize the TERS probe structure by sweeping two or more parameters, this method is extremely resource-intensive, with computation times growing exponentially as the number of parameters increases. In this work, we propose an alternative rapid theoretical method that reduces computational loading while still achieving effective TERS probe optimization through the inverse design method. By applying this method to optimize a TERS probe with four free-structural parameters, we observed a nearly 1 order of magnitude improvement in enhancement factor (|E/E0|2), in contrast to a parameter sweeping 3D simulation that would take ∼7000 hours of computation. Our method, therefore, shows great promise as a useful tool for designing not only TERS probes but also other near-field optical probes and optical antennas.

Abstract:Optical tweezers have found widespread use in studies of biological macromolecules and in manipulation of microscopic objects, including biological cells and a variety of dielectric particles. But rapid progress over the last decade has demonstrated that optical tweezers also can be used as a powerful method for manipulation and control of plasmonic metal nanostructures. Here, we review our recent results in this area with a focus on the interaction between nanoparticles confined in an optical trap and applications in surface-enhanced Raman scattering spectroscopy.

Abstract:Tip-enhanced Raman spectroscopy (TERS) provides the sensitivity required to obtain the vibrational fingerprint of few molecules. While single molecule detection has been demonstrated in UHV experiments, the sensitivity of the technique in ambient, liquid and electrochemical conditions is still limited. In this work, we present a new strategy to increase the signal-to-noise in TERS by spatial light modulation. We iteratively optimize the phase of the excitation beam employing two different feedback mechanisms. In one optimization protocol, we monitor the spectral changes upon aberration correction and tight far-field focusing. In a second protocol, we use a phase-optimization strategy where TER spectra are directly used for feedback. Far-field tight focusing results in average signal enhancements of a factor of 3.5 in air and has no impact on TER signals obtained from solid/liquid interfaces. Using the TER spectrum as direct feedback, we obtain average signal enhancements between a factor of 2.6 in liquid and 4.3 in air. In individual cases, some bands increase by more than one order of magnitude in intensity upon spatial light modulation. Importantly, phase modulation in addition allowed the retrieval of bands that were initially not discernible from the noise. The proposed beam-modulation strategy can be easily implemented in existing TERS instruments and can help to push the detection limit of the technique in applications where the signal-to-noise level is low.

Abstract:Optical tweezers has emerged as a powerful tool in manipulating microscopic particles and in measuring weak forces of the order of a pico-Newton. As a result, it has found wide applications ranging from material science to biology. Dual-trap optical tweezers (DTOT) are of particular importance as they allow for two point correlation measurements as in molecular force spectroscopy, two-point active micro-rheology, etc. Here we report a novel design for a steerable DTOT setup which uses back-scattered light from the two traps for position detection. This is performed using a confocal scheme where the two detectors are placed at the conjugate points to the respective traps. This offers several significant advantages over current designs, such as, zero cross-talk between signals, single module assembly and robustness to thermal drift. Moreover, our design can be very easily integrated with standard microscopy techniques like Phase contrast and Differential Interference Contrast, without modifying the microscope illumination unit.

Quantifying Light-assisted Collisions in Optical Tweezers Across the Hyperfine Spectrum

Abstract:We investigate the role of hyperfine structure in resonant-dipole interactions between two atoms co-trapped in an optical tweezer. Two-body loss rates from light-assisted collisions (LACs) are measured across the $^{87}$Rb hyperfine spectrum and connected to properties of molecular photoassociation potentials via a semi-classical model. To obtain our results, we introduce an imaging technique that leverages repulsive LACs to detect two atoms in a trap, thereby circumventing parity constraints in tweezers. Our findings offer key insights for exploiting hyperfine structure in laser-induced collisions to control cold atoms and molecules in a broad range of quantum science applications.

Abstract:A scheme that combines optoelectronic tweezers (OET) with spectroscopic analysis is presented. Referred to as spectral tweezers, the approach uses a single focused light beam that acts both as the trapping beam for OET and the probe beam for spectroscopy. Having simultaneous manipulation and spectral characterization ability, the method is used to isolate single micro-samples from clusters and perform spectral measurements. Experimental results show that a characteristic spectral signature can be obtained for a given sample. The proposed approach can be easily integrated into the optical setups used for conventional OETs with only a few additional optical components, making it a convenient tool for bio-analytical applications.

Abstract:Trapping of microspheres with a single focused laser beam is usually limited to materials with relative refractive indexes slightly larger than one. We show that directional light scattering can be employed to optically trap high-index materials. For this purpose, we propose a material platform to achieve zero backward scattering (ZBS), also known as the first Kerker condition, in a composite media containing spherical inclusions of silica embedded in a SiC microsphere. By tuning the volume filling fraction of inclusions and the microsphere radius, stable trapping can be achieved, provided that ZBS is combined with the condition for destructive interference between the fields reflected at the external and internal interfaces of the microsphere when located at the focal point. We show that our proposal also holds even in the presence of a significant amount of spherical aberration, which is a common condition in most optical tweezers setups. In this case, achieving ZBS is essential for trapping high-index materials.

Abstract:Optical tweezers (OT), or optical traps, are a device for manipulating microscopic objects through a focused laser beam. They are used in various fields of physical and biophysical chemistry to identify the interactions between individual molecules and measure single-molecule forces. In this work, we describe the development of a homemade optical tweezers device based on a cost-effective IR diode laser, the hardware, and, in particular, the software controlling it. It allows us to control the instrument, calibrate it, and record and process the measured data. It includes the user interface design, peripherals control, recording, A/D conversion of the detector signals, evaluation of the calibration constants, and visualization of the results. Particular stress is put on the signal filtration from noise, where several methods were tested. The calibration experiments indicate a good sensitivity of the instrument that is thus ready to be used for various single-molecule measurements.

Tuning the Optical Response of a Plasmonic T-shaped Dimer with Nanowire Loads for Improved SERS and Sensing Applications

Abstract:Plasmonic nanostructures have important applications for surface-enhanced Raman chips, optical sensors, perfect absorbers, and so on. In this paper, we theoretically investigate the optical properties of a T-shaped dimer (TD) nanostructure and a TD with nanowire loads (TD/NL) nanostructure. By depositing the NLs around the TD, the optical property of the TD is significantly modified, and the extinction spectrum of composite TD/NL appears the splitting resonance peak and shows the asymmetric characteristic of Fano resonance due to the plasmon hybridization. Besides, a stronger electric field enhancement can be obtained in the composite TD/NL nanostructure, and the calculated results show the field enhancement effect of the dark mode in designed TD/NL nanostructure is three times than that in the TD nanostructure. Furthermore, the Fano resonance in TD/NL nanostructure reveals the narrow spectral linewidth and high quality factor, which is advantageous to optical refractive index sensors, and the corresponding sensing sensitivity is as high as 1083 nm RIU−1.

Abstract:Engineering hotspots is of crucial importance in many applications including energy harvesting, nano-lasers, subwavelength imaging, and biomedical sensing. Surface-enhanced Raman scattering spectroscopy is a key technique to identify analytes that would otherwise be difficult to diagnose. In standard systems, hotspots are realised with nanostructures made by acute tips or narrow gaps. Owing to the low probability for molecules to reach such tiny active regions, high sensitivity is always accompanied by a large preparation time for analyte accumulation which hinders the time response. Inspired by transformation optics, we introduce an approach based on warped spaces to manipulate hotspots, resulting in broadband enhancements in both the magnitude and volume. Experiments for single molecule detection with a fast soaking time are realised in conjunction with broadband response and uniformity. Such engineering could provide a new design platform for a rich manifold of devices, which can benefit from broadband and huge field enhancements.

Abstract:Tip-enhanced Raman spectroscopy (TERS) exhibits new selection rule and sub- nanometer spatial resolution, which is attributed to the plasmonic near-field confinement. Despite recent advances in simulations of TERS spectra under highly confined fields, a simply physical mechanism has remained elusive. In this work we show that single-molecule TERS images can be explained by local sub-molecular density changes induced by the confined near-field during the Raman process. The local sub-molecular density changes determine the spatial resolution in TERS and the gradient-based selection rule. Using this approach we find that the four-fold symmetry of meso-tetrakis(3,5- di-tert-butylphenyl)porphyrin (H2TBPP) TERS images observed in experiments arises from the combination of degenerate normal modes localized in the functional side groups rather than the porphyrin ring as previously considered. As an illustration of the potential of the method, we demonstrate how this new theory can be applied to microscopic structure characterization.
A Micro-Raman Spectroscopy Combing with Optical Tweezers

Abstract:A hybrid "optical tweezers-micro Raman spectroscopy" technique is introduced in this letter. Applying this system, the micro-Raman spectrums of rat erythrocyte and saccharomyces cerevisiae in vivo were obtained. The results show that this technique can obviously improve signal-to-noise ratio compared with conventional approaches.Besides, the damage from lasers on the cells were analyzed. It was revealed that the damage of cell from intense IR laser were much lower than that from the weak Ar~+ laser. Finally, the system was also applied in carbon nano-tube (CNT) analysis and Raman spectra from CNT were obtained.

Abstract:We have constructed a counterpropagating optical tweezers setup embedded in a Sagnac interferometer in order to increase the sensitivity of position tracking for particles in the geometrical optics regime. Enhanced position determination using a Sagnac interferometer has previously been described theoretically by Taylor et al. [Journal of Optics 13, 044014 (2011)] for Rayleigh-regime particles trapped in an antinode of a standing wave. We have extended their theory to a case of arbitrarily-sized particles trapped with orthogonally-polarized counterpropagating beams. The working distance of the setup was sufficiently long to optically induce particle oscillations orthogonally to the axis of the tweezers with an auxiliary laser beam. Using these oscillations as a reference, we have experimentally shown that Sagnac-enhanced back focal plane interferometry is capable of providing an improvement of more than 5 times in the signal-to-background ratio, corresponding to a more than 30-fold improvement of the signal-to-noise ratio. The experimental results obtained are consistent with our theoretical predictions. In the experimental setup, we used a method of optical levitator-assisted liquid droplet delivery in air based on commercial inkjet technology, with a novel method to precisely control the size of droplets.

Abstract:A tip-enhanced Raman spectroscopy (TERS) based on plasmonic lens (PL) excitation is proposed in this work. A PL expected to realize a strong longitudinal electric field focus is designed. The focusing performance of the PL is calculated via finite-difference time-domain (FDTD) simulation and experimentally detected by a scattering-type scanning near-field optical microscope. The PL is introduced to a TERS system as a focusing device. Experimental results with carbon nanotube samples indicate that the Raman scatting signal is significantly enhanced. It proves experimentally that the combination of a PL focused excitation field with a metallic tip in a TERS system is a promising method.
