SETD2-mediated H3K14 trimethylation promotes ATR activation and stalled replication fork restart in response to DNA replication stress

Abstract:Significance ATR is a central molecule involved in the DNA replication stress response and repair that ensures genome stability. Whether chromatin modifiers or chromatin modifications also regulate ATR activation, however, is unclear. We conclude that SETD2-mediated H3K14me3 recruits the RPA complex to chromatin and thus promotes ATR activation during conditions of replication stress. Using a series of biological and molecular approaches, we carefully delineated this new SETD2-H3K14me3-RPA-ATR axis that promotes ATR activation and improves cancer cell survival in response to DNA replication stress. Our data open up future opportunities to generate cancer therapeutics strategies based on this pathway.
