Abstract:
Treatment of oral pathogens is important for both oral and systemic health. The antimicrobial activity of chitosan (CS)-based scaffolds either loading antibiotics or compositing with other agents are well documented. However, the intrinsic antibacterial activity of CS scaffolds alone has never been reported. Herein, we fabricated the non-crosslinked CS scaffold and investigated its antibacterial activity against typical oral pathogens, Gram-negative Porphyromonas gingivalis and Gram-positive Streptococcus mutans. We found both pathogens were completely killed by 1 mg CS scaffolds at 6 h, due largely to the CS-induced time-dependent bacteria clustering. Interestingly, β-glycerophosphate crosslinked scaffolds showed no antibacterial activity. In conclusion, the bactericidal activity of CS scaffolds alone is reported for the first time. Together with the biodegradability, physical stability, biocompatibility and great antibacterial activity, the non-crosslinked CS scaffolds may have great potentials not only in treating oral diseases but also in wound healing and tissue engineering.

What problem does this paper attempt to address?
### The Problem the Paper Attempts to Solve This paper aims to explore whether chitosan (CS) scaffolds possess intrinsic antibacterial activity without the addition of any antibiotics or other active components and to verify their effectiveness against oral pathogens. #### Main Research Content: 1. **Preparation and Characterization of Non-Crosslinked Chitosan Scaffolds**: - Different weights of non-crosslinked chitosan scaffolds were prepared, and a porous structure was obtained through freeze-drying technology. - The performance of non-crosslinked chitosan scaffolds (CS scaffolds) was compared with crosslinked chitosan-β-glycerophosphate scaffolds (CS-GP scaffolds). 2. **Antibacterial Activity Evaluation**: - The antibacterial effect of chitosan scaffolds on Gram-negative Porphyromonas gingivalis and Gram-positive Streptococcus mutans was evaluated using the colony-forming unit (CFU) counting method. - It was found that non-crosslinked chitosan scaffolds almost completely killed these two pathogens within 6 hours, while the crosslinked CS-GP scaffolds had almost no antibacterial activity. 3. **Observation of Bacterial Morphological Changes**: - Scanning electron microscopy (SEM) was used to observe the morphological changes of bacteria after contact with chitosan scaffolds, revealing a time-dependent aggregation phenomenon of bacterial cells on the chitosan scaffolds. 4. **Protein Adsorption Capacity Evaluation**: - The protein adsorption capacity of chitosan scaffolds in BHI medium was quantified using the BCA method, showing that non-crosslinked chitosan scaffolds had a stronger protein adsorption capacity. 5. **Physical Stability and Biocompatibility Evaluation**: - The physical stability of chitosan scaffolds in BHI medium was assessed, and their biocompatibility with human gingival epithelial cells was verified through the CCK-8 assay. #### Research Conclusions: - Non-crosslinked chitosan scaffolds demonstrated significant antibacterial activity for the first time, effectively killing oral pathogens. - Chitosan scaffolds achieve antibacterial effects by inducing bacterial aggregation, and the positive charge of chitosan molecules in the non-crosslinked state is crucial for their antibacterial activity. - Due to their biodegradability, physical stability, biocompatibility, and excellent antibacterial activity, non-crosslinked chitosan scaffolds can be used not only for the treatment of oral diseases but also have broad application potential, such as in wound healing and tissue engineering.
