Abstract:High intensity focused ultrasound (HIFU) has demonstrated its safety, efficacy and noninvasiveness in the ablation of solid tumor. However, its further application is limited by its inherent deficiencies, such as postoperative recurrence caused by incomplete ablation and excessive intensity affecting surrounding healthy tissues. Recent research has indicated that the integration of nanomaterials with HIFU exhibits a promising synergistic effect in tumor ablation. The concurrent utilization of nanomaterials with HIFU can help overcome the limitations of HIFU by improving targeting and ablation efficiency, expanding operation area, increasing operation accuracy, enhancing stability and bio-safety during the process. It also provides a platform for multi-therapy and multi-mode imaging guidance. The present review comprehensively expounds upon the synergistic mechanism between nanomaterials and HIFU, summarizes the research progress of nanomaterials as cavitation nuclei and drug carriers in combination with HIFU for tumor ablation. Furthermore, this review highlights the potential for further exploration in the development of novel nanomaterials that enhance the synergistic effect with HIFU on tumor ablation.

The problem that this paper attempts to solve is the application limitations of high - intensity focused ultrasound (HIFU) in tumor treatment. Although the HIFU technology has been widely used in the ablation treatment of solid tumors because of its non - invasiveness, safety and effectiveness, there are still some limitations in its clinical application, such as postoperative recurrence (caused by incomplete ablation), excessive intensity affecting surrounding healthy tissues, etc. These problems limit the further development and application of the HIFU technology. In order to solve the above problems, this paper explores the application of the combined use of nanomaterials and HIFU in anti - tumor treatment. By integrating nanomaterials, the inherent limitations of HIFU can be overcome, the targeting and ablation efficiency can be improved, the operating range can be expanded, the operating precision can be increased, and the stability and biosafety during the process can be improved. At the same time, this combination also provides a platform for multi - therapy and multi - mode imaging - guided. Specifically, the paper mainly focuses on the following aspects: 1. **Thermal effect**: Nanomaterials can enhance the thermal effect of HIFU by increasing the local thermal conductivity, thereby accelerating the tumor heating speed and reducing the damage to surrounding tissues. 2. **Mechanical effect**: By introducing exogenous cavitation nuclei (such as nanomaterials), the cavitation effect of HIFU can be enhanced and the tumor ablation efficiency can be improved. 3. **Image brightness enhancement**: Nanomaterials, as contrast agents, can improve the quality of ultrasonic imaging and the accuracy of diagnosis and treatment. 4. **Drug delivery**: Nanomaterials can be used as drug carriers to achieve the controlled release of drugs under the action of HIFU and improve the therapeutic effect of drugs. In conclusion, this paper aims to improve the efficacy and safety of HIFU in tumor treatment through the synergy between nanomaterials and HIFU, and promote the wider application of HIFU technology in the clinic.
