Abstract:Malaria is one of the oldest and deadliest parasitic diseases globally, and it still poses a global health issue today because of the emergence of new strains resistant to the first-line treatment (artemisinin-based combination therapy). The development of new, effective antimalarial drugs to eradicate this lethal disease has never been more important. Gold complexes have a long history in the field of medicinal chemistry, especially because of their therapeutic activity against rheumatoid arthritis and cancer. Their more recently discovered antimalarial activities are also promising and triggered further research. This review presents an overview of the bioactive gold(I)/(III) compounds as antimalarial agents. We will discuss the synergistic effects of the gold-based drugs and the effect of ligands such as phosphine, S-donors and NHC. Studies providing insight into the possible mechanisms of action against essential parasite targets and validation attempts are also presented.
