Abstract:The gene encoding a putative triphenylmethane reductase (TMR)-like protein derived from Geobacillus thermoglucosidasius C56-Y593 (named as GtAZR) was synthesized, heterologously expressed in Escherichia coli, and extensively characterized for the first time. The recombinant GtAZR displayed its maximum activity at pH 5.5 and 40 °C. GtAZR was stable at temperatures below 65 °C. It also exhibited a broad pH stability and retained more than 90 % of its initial activities in pH range of 4.5–10.5 after incubating in various buffers for 1 h. Moreover, GtAZR showed significant stability against metal ions and organic solvents. GtAZR displayed broad substrate spectrum toward both azo and triphenylmethane dyes. As a sequence and structural TMR-like protein, GtAZR was characterized as an azoreductase biochemically due to its high specificity for azo dye rather than triphenylmethane dye. Molecular docking and mutagenesis analysis revealed that amino acids Asp-79 and Thr-80 are responsible for its azoreductase activity, which eliminated the steric hindrance caused by His-77 and Tyr-78 at the correspond sites in other structural homologous triphenylmethane reductase. The robust stability and substrate promiscuity of GtAZR made it a promising candidate for practical removal of mixed dye wastewater.

Abstract:Azo dyes are toxic and carcinogenic and are often present in industrial effluents. In this research, azoreductase and glucose 1-dehydrogenase were coupled for both continuous generation of the cofactor NADH and azo dye removal. The results show that 85% maximum relative activity of azoreductase in an integrated enzyme system was obtained at the conditions: 1 U azoreductase:10 U glucose 1-dehydrogenase, 250 mM glucose, 1.0 mM NAD(+) and 150 mu M methyl red. Sensitivity analysis of the factors in the enzyme system affecting dye removal examined by an artificial neural network model shows that the relative importance of enzyme ratio between azoreductase and glucose 1-dehydrogenase was 22%, followed by dye concentration (27%), NAD(+) concentration (23%) and glucose concentration (22%), indicating none of the variables could be ignored in the enzyme system. Abstract:rAzoR2326, an azoreductase derived from Roseibium sp. H3510, functions as an FMN-dependent homodimer utilizing NADH as cofactor. It demonstrated maximum activity at 45 °C and retained moderate activity above 50 °C, exhibiting stability from pH 7-10. Evolution and structure guided rational design of wild-type rAzoR2326 (WT) efficiently yielded 6 single-point mutants with improved thermostability and activity from a 22-variant library. Further combinatorial mutation led to mutant M20 with substantially enhanced thermostability (15-fold longer half-life at 50 °C) and activity (3.24-fold higher kcat/Km). M20 exhibited superior catalytic properties for decolorizing Allura Red compared to WT. Specifically, its decolorization capacity at pH 10.0 was 4.26-fold higher than WT. Additionally, M20 demonstrated remarkable thermostability, retaining 76.83 % decolorization activity for Allura Red after 120 min at 50 °C, whereas WT nearly lost all catalytic activity under the same conditions. Molecular dynamics simulations revealed the structural changes in M20, such as improved hydrogen bonding and a new C-H···π interaction, led to a more compact and rigid enzyme structure. This resulted in a more stable FMN-binding pocket and substrate tunnel, thereby improving the catalytic stability and activity of M20. Given its enhanced dye decolorization ability and alkaline tolerance, M20 shows promise as a biocatalyst for treating azo dye effluents.

Abstract:Bio-degradation is the most affordable method of azo dye decontamination, while its drawbacks such as aromatic amines accumulation and low degradation efficiency must be overcome. In this study, a novel mechanism of azo dye degradation by a fungus was discovered. At a concentration of 400 mg/L, the decolorization efficiency of Acid Red 73 (AR73) by Aspergillus tabacinus LZ-M was 90.28%. Metabolite analysis and transcriptome sequencing analysis revealed a self-redox process of AR73 degradation, where the electrons generated in carbon oxidation were transferred to the reduction of -C-N = and –NN. The metabolites, 2-hydroxynaphthalene and N-phenylnitrous amide were mineralized into CO 2 through catechol pathway and a glycolytic process. Furthermore, the mineralization ratio of dye was computed to be 31.8% by the carbon balance and electron balance. By using comparative transcriptome, a novel decoloring enzyme Ord95 was discovered in unknown genes through gene cloning. It hydrolyzed AR73 into 2-hydroxynaphthalene and N-phenylnitrous amide, containing a glutathione S-transferase domain with three arginines as key active sites. Here the new mechanism of azo dye degradation was discovered with identification of a novel enzyme in Aspergillus tabacinus LZ-M.

Decolorization and biodegradation of triphenylmethane dyes by a novel Rhodococcus qingshengii JB301 isolated from sawdust

Abstract:A sawdust sample collected from a wet environment was used to screen for crystal violet-decolorizing and methyl violet-decolorizing bacterial strains. A strain with degrading and decolorizing abilities was isolated and identified as Rhodococcus qingshengii JB301. The decolorization of crystal violet and methyl violet was improved by optimizing culture conditions. Based on liquid chromatography–mass spectrometry (LC-MS) analysis, similar degradation products of crystal violet and methyl violet were detected, while [N,N-dimethylaminophenyl] [aminophenyl] benzophenone or [N-methylaminophenyl] [N-methylaminophenyl] benzophenone was different from previous reports. A common degradation pathway was established to demonstrate the mechanism of crystal violet and methyl violet decolorization. It was found that lignin peroxidase activity was greatly enhanced after addition of crystal violet (by 514 %) and methyl violet (by 698 %) at 12 h, while NADH-DCIP (dichlorophenolindophenol) reductase activity was inhibited by the dyes. The growth of Triticum aestivum was inhibited by both dyes and the toxicity of the dyes was weakened after the degradation by R. qingshengii JB301.

Abstract:Azo dye has a massive marketplace in various industries, biological laboratories and clinics. A significant drawback is the excessive flush-off of azo dyes and their intermediate products are that they pollute the environment. Several environmental bacteria degrade azo dye under certain specific ecological conditions. Decolourisation by different bacteria occurs under aerobic, anoxic and anaerobic conditions. Bacterial genera such as Pseudomonas, Bacillus and Rhodococcus are used for the rapid decolourisation of azo dyes. Bacterial enzymes play an essential role in the degradation of azo dyes by cleavage of azo bonds (-N=N-). It is degraded by both oxidative and reductive enzymes including azo reductase, laccase, lignin peroxidase, NADH-DCIP reductase, malachite green reductase, polyphenol oxidase (tyrosinase), veratrol alcohol oxidase. This review describes the role and mechanism of these enzymes in the degradation of azo dyes.
Biodegradation of Methyl Red by Bacillus Sp. Strain UN2: Decolorization Capacity, Metabolites Characterization, and Enzyme Analysis.

Abstract:Azo dyes are recalcitrant and refractory pollutants that constitute a significant menace to the environment. The present study is focused on exploring the capability of Bacillus sp. strain UN2 for application in methyl red (MR) degradation. Effects of physicochemical parameters (pH of medium, temperature, initial concentration of dye, and composition of the medium) were studied in detail. The suitable pH and temperature range for MR degradation by strain UN2 were respectively 7.0–9.0 and 30–40 °C, and the optimal pH value and temperature were respectively 8.0 and 35 °C. Mg2+ and Mn2+ (1 mM) were found to significantly accelerate the MR removal rate, while the enhancement by either Fe3+ or Fe2+ was slight. Under the optimal degradation conditions, strain UN2 exhibited greater than 98 % degradation of the toxic azo dye MR (100 ppm) within 30 min. Analysis of samples from decolorized culture flasks confirmed biodegradation of MR into two prime metabolites: N,N′dimethyl-p-phenyle-nediamine and 2-aminobenzoic acid. A study of the enzymes responsible for the biodegradation of MR, in the control and cells obtained during (10 min) and after (30 min) degradation, showed a significant increase in the activities of azoreductase, laccase, and NADH-DCIP reductase. Furthermore, a phytotoxicity analysis demonstrated that the germination inhibition was almost eliminated for both the plants Triticum aestivum and Sorghum bicolor by MR metabolites at 100 mg/L concentration, yet the germination inhibition of parent dye was significant. Consequently, the high efficiency of MR degradation enables this strain to be a potential candidate for bioremediation of wastewater containing MR.
Bioinformatics analysis and molecular dynamics simulations of azoreductases (AzrBmH2) from Bacillus megaterium H2 for the decolorization of commercial dyes

Abstract:The present study aimed to investigate the decolorization of various commercial dyes by azoreductases (AzrBmH21, AzrBmH22/3, and AzrBmH24/5) through bioinformatics means, comprising molecular docking, molecular dynamics simulation, and molecular mechanics Poisson–Boltzmann surface area (MM-PBSA). Therefore, four commercial dyes, namely acid orange 7, cresol red, methylene blue, and malachite green, were selected as potential targets for degradation by the above said azoreductases derived from Bacillus megaterium H2. The prediction of ligand binding or catalytic sites for AzrBmH21, AzrBmH22/3, and AzrBmH24/5 were performed using a machine learning algorithm based on the Prank Web and DeepSite chemoinformatic tool. This analysis revealed that several amino acids of AzrBmH2 interacted with the tested dyes, indicating the presence of distinct ligand-binding sites for AzrBmH2-dye complexes. Likewise, the binding affinity for AzrBmH21, AzrBmH22/3, and AzrBmH24/5 ranged from − 9.4 to − 5.5 kcal/mol, − 9.2 to − 5.4 kcal/mol, and − 9.0 to − 5.4 kcal/mol, respectively, with each complex stabilized at a minimum of 0–5 hydrogen bonds. MD simulations revealed stable AzrBmH2-dye complexes with RMSD and RMSF values ranging from 0.15 to 0.42 nm and 0.05 to 0.48 nm, respectively, with Rg values between 1.75 and 1.88 nm. MM-PBSA calculations indicated that the AzrBmH2–dye complexes, except for AzrBmH2–malachite green, exhibited the lowest binding energy (− 191.05 ± 7.08 to 314.19 ± 6.88 kcal/mol), with prevalent hydrophobic interactions (− 268.25 ± 12.25 to − 418.92 ± 29.45 kcal/mol) through van der Waals forces. Therefore, this study was able to highlight the potential role of enzymes, specifically azoreductases from Bacillus megaterium H2, in predicting the decolorization of commercial dyes. These findings could contribute to our understanding of the azoreductases' mechanisms in bioremediation and for biotechnological applications.

Abstract:Azo dyes are used as coloring agent in textile industries at larger scale. As a result, large quantity of dye-enriched waste water is generated which subsequently poses environmental problems. Biological tool involving bacteria having azoreductase enzyme has proved to be more effective and efficient in dye effluent treatment. Current work focuses on Staphylococcus caprae ( S. caprae ) for degradation and decolorization of Reactive Red-195 (RR-195) azo dye. For this purpose, factors such as pH, temperature, inoculums, carbon and nitrogen sources, and dye concentrations have been optimized for maximum decolorization and degradation. S. caprae (4 mg/mL) efficiently resulted into 90% decolorization of RR-195 dye under static condition at 100 μg/mL concentration, 30 °C and pH 7.0 at a 12-h contact period. FTIR analysis has revealed the formation of new functional groups in the treated dye such as O–H stretch at 3370 cm −1 , C-H band stretching at 2928 cm −1 , and new band at 1608 cm −1 which specify the degradation of aromatic ring, 1382 and 1118 cm −1 represents desulfonated peaks. Biodegraded metabolites of RR-195 dye such as phenol, 3, 5-di-tert-butylphenol, and phthalic acid have been identified respectively that find industrial applications. Phytotoxicity test has shown non-toxic effects of treated dye on germination of Vigna radiata and Triticum aestivum seeds. Further, antibiotic diffusion assay has confirmed the biosafety of S. caprae.

Case Application of a Combined Treating Process on Azo Dye Wastewater

Abstract:In this study, a combined treating process (neutralization, electrolysis, ozone oxidization, coagulation and cinder adsorption) was introduced to investigate the possibility of degradation of concentrated azo dye wastewater. The optimum operation parameters were obtained through small-scale bench tests and pilot-scale tests, and then were put into operation. The experimental results showed that the removal rates of chemical oxygen demand (COD) and chroma were all above 99.5%. After treatment, the COD of the outflow was lowered less than 100 mg/L, the chroma was reduced 50 times, and the pH was 6-9, with the outflow under the national standards for the wastewater discharge. The application of the combined treating process would be a good demonstration for the concentrated organic wastewater in dyestuff industry. Meanwhile, the sludge and the residue generated from each unit of the combined treating process could be processed for further resource utilization based on economic analysis; the treatment cost is 1.08 Yuan (RMB)/m3, so this application also captured considerable benefits in terms of environments and economics for protection of water resources effectively.

Abstract:Azo dyes, as the most widely used synthetic dyes, are considered to be one of the culprits of water resources and environmental pollution. Anoxybacillus sp. PDR2 is a thermophilic bacterium with the ability to degrade azo dyes, whose genome contains two genes encoding azoreductases (named AzoPDR2-1 and AzoPDR2-2). In this study, through response surface methodology (RSM), when the initial pH, inoculation volume and Mg 2+ addition amount were 7.18, 10.72% and 0.1 g/L respectively, the decolorization rate of methyl red (MR) (200 mg/L) could reach its maximum (98.8%). The metabolites after biodegradation were detected by UV–Vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), and liquid chromatography mass spectrometry (LC-MS/MS), indicating that MR was successfully decomposed into 4-aminobenzoic acid and other small substrates. In homologous modeling, it was found that both azoreductases were flavin-dependent azoreductases, and belonged to the α/β structure, using the Rossmann fold. In their docking results with the cofactor flavin mononucleotide (FMN), FMN bound to the surface of the protein dimer. Nicotinamide adenine dinucleotide (NADH) was superimposed on the plane of the pyrazine ring between FMN and the activity pocket of protein. Besides, both azoreductase complexes (azoreductase-FMN-NADH) exhibited a substrate preference for MR. Asn104 and Tyr74 played an important role in the combination of the azoreductase AzoPDR2-1 complex and the azoreductase AzoPDR2-2 complex with MR, respectively. This provided assistance for studying the mechanism of azoreductase biodegradation of azo dyes in thermophilic bacteria.

Abstract:Azo dyes are significant organic pollutants known for their adverse effects on humans and aquatic life. In this study, anthraquinone-2-sulfonate (AQS) immobilized on biochar (BC) was employed as a novel carrier in up-flow anaerobic fixed-bed reactors to induce specific biofilm formation and promote the biotransformation efficiency of azo dyes. Novel carrier–packed reactor 1 (R1) and BC-packed reactor 2 (R2) were used to treat red reactive 2 (RR2) under continuous operation for 175 days. The decolorization rates of R1 and R2 were 96–83% and 91–73%, respectively. The physicochemical characteristics and extracellular polymeric substances (EPS) of the biofilm revealed a more stable structure in R1. Furthermore, the microbial community in R1 interacted more closely with each other and contained more keystone genera. Overall, this study provides a feasible method for improving the biotransformation of azo dyes, thus providing support for practical applications in wastewater treatment projects.

Biosurfactant production from Pseudomonas taiwanensis L1011 and its application in accelerating the chemical and biological decolorization of azo dyes

Abstract:Dye dispersion and the interaction efficiency between azoreductases and dye molecules are rate-limiting steps for the decolorization of azo dyes. In this study, a biosurfactant-producing strain, Pseudomonas taiwanensis L1011, was isolated from crude oil. To increase the yield of the biosurfactant BS-L1011 from P. taiwanensis L1011, culture conditions were optimized including temperature, initial pH, carbon source, nitrogen source and C/N ratio. A maximum yield of 1.12g/L of BS-L1011 was obtained using D-mannitol as carbon source and yeast extract/urea as compound nitrogen source with C/N ratio of 10/4, pH 7.0 and 28°C. BS-L1011 exhibited a low critical micelle concentration (CMC) of 10.5mg/L and was able to reduce the surface tension of water to 25.8±0.1 mN/m. BS-L1011 was stable over a wide range of temperatures, pH values and salt concentrations. The biosurfactant is reported for the first time to accelerate chemical decolorization of Congo red by sodium hypochlorite, and biological decolorization of Amaranth by Bacillus circulans BWL1061, thus showing a potential in the treatment of dyeing wastewater.
Exclusive Extracellular Bioreduction of Methyl Orange by Azo Reductase-Free Geobacter Sulfurreducens.

Abstract:Azo dyes are a class of recalcitrant organic pollutants causing severe environmental pollution. For their biodecolorization, the azo reductase system was considered as the major molecular basis in bacteria. However, the intracellular localization of azo reductase limits their function for efficient azo dye decolorization. This limitation may be circumvented by electrochemically active bacteria (EAB) which is capable of extracellular respiration. To verify the essential role of extracellular respiration in azo dye decolorization, Geobacter sulfurreducens PCA, a model EAB, was used for the bioreduction of methyl orange (MO), a typical azo dye. G. sulfurreducens PCA efficiently reduced MO into amines. Kinetic results showed that G. sulfurreducens PCA had the highest decolorization efficiency among the currently known MO reducing bacteria. Electrons from acetate oxidization by this strain were transferred by the respiratory chain to MO. The mass and electron balances, fluorescent probing and proteinase K treatment experimental results indicate that the biodecolorization of MO by G. sulfurreducens PCA is an exclusive extracellular process. OmcB, OmcC and OmcE were identified as the key outer-membrane proteins for the extracellular MO reduction. This work deepens our understanding of EAB physiology and is useful for the decontamination of environments polluted with azo dyes. The contribution of extracellular respiration to pollutants reduction will broaden the environmental applications of EAB.

Abstract:The environmental pollution caused by azo dyes at high temperatures has become an urgent problem. However, little attention has been paid to decolorizing azo dyes by thermophilic consortiums. In this study, a thermophilic bacterial consortium (BCGR-T) mainly composed of two genera, namely, Caldibacillus (70.90%) and Aeribacillus (17.63%) was first enriched, which can decolorize Brilliant Crocein GR (BCGR) at high temperatures (50–75 °C), pH values of 6∼8, dye concentrations (100–400 mg/L) and salinities (1–5%, w/v). The enzyme activity results showed that the azoreductase activity was nearly 8.8 times that of the control (p < 0.01), and the intracellular lignin peroxidase was also highly expressed with enzyme activity of 5.64 U (min −1 mg −1 protein) (p < 0.05), indicated that both azoreductase and intracellular lignin peroxidase played an important part in the decolorization process. Furthermore, seven new intermediate metabolic products, including aniline, phthalic acid, 2-carboxy benzaldehyde, phenylacetic acid, benzoic acid, toluene, and 4-methyl-hexanoic acid, were identified. In addition, functional genes related with the azo dye decolorization, such as those encoding the azoreductase, laccase, FMN reductase, NADPH-/NADH-quinone oxidoreductases and NADPH-/NADH dehydrogenases, catechol dioxygenase, homogentisate 1,2-dioxygenase, protocatechuate 3,4-dioxygenase, gentisate 1,2-dioxygenase, azobenzene reductase, naphthalene 1,2-dioxygenase, benzoate/toluate 1,2-dioxygenase, and anthranilate 1,2-dioxygenase and so on were found in the metagenome of the consortium BCGR-T. Finally, a new decolorization pathway of the thermophilic consortium BCGR-T was proposed. In addition, the phototoxicity of BCGR decreased after decolorization. Overall, the thermophilic consortium BCGR-T could be a promising candidate in the treatment of high concentration azo dye wastewater at high temperatures.

Abstract:<p>Azo dyes have caused serious water pollution which significantly threaten human health. In this work, the MnBi powders were successfully fabricated and used to degrade the azo dye. The efficiency, thermodynamics and mechanism of the degradation of the azo dyes were systematically investigated. It was found that the MnBi alloy powder has an efficacious degrading effect on the azo dyes and the ball milled MnBi powder can effectively improve the degradation rate and efficiency. The high degradation efficiency of MnBi alloy powders could be mainly attributed to the Mn elements. In addition, with the increase of reaction temperature, the degradation reaction type of the azo dyes changed from second-order to first-order reactions. The effect of different reaction concentration and temperature on degradation efficiency were explained according to the kinetics and thermodynamics analysis of the degradation reaction. Therefore, this work provided a new material for the effective degradation of azo dyes.</p>

Abstract:The reduction of the azo dye model compounds Reactive Red 2 (RR2) and Reactive Orange 14 (RO14) by mesophilic (30 degrees C) and thermophilic (55 degrees C) anaerobic consortia was studied in batch assays. The contribution of fermentative and methanogenic microorganisms in both temperatures was evaluated in the presence of the fermentative substrate glucose and the methanogenic substrates acetate, H2/CO2, methanol, and formate. Additionally, the effect of the redox mediator riboflavin on electron shuttling was assessed. We concluded that the application of thermophilic anaerobic treatment is an interesting option for the reductive decolorization of azo dyes compared to mesophilic conditions. The use of high temperature may decrease or even take the place of the need for continuous redox mediator dosage in bioreactors, contrarily to the evident effect of those compounds on dye reduction under mesophilic conditions. Both fermenters and methanogens may play an important role during reductive decolorization of dyes, in which mediators are important not only for allowing the different microbes to participate more effectively in this complex reductive biochemistry but also for assisting in the competition for electrons between dyes and other organic and inorganic electron acceptors.

Abstract:The accelerating effect of quinones has been studied in the bio-decolorization processes, but there are no literatures about the incorporation bio-treatment technology of the bromoamine acid (BA) wastewater and azo dyes wastewaters under high-salt conditions (NaCl, 15%, w/w). Here we described the BA wastewater as a redox mediator in the bio-decolorization of azo dye wastewaters. Decolorization of azo dyes was carried out experimentally using the salt-tolerant bacteria under the BA wastewater and high-salt conditions. The BA wastewater used as a redox mediator was able to increase the decolorization rate of wastewater containing azo dyes. The effects of various operating conditions such as dissolved oxygen, temperature, and pH on microbial decolorization were investigated experimentally. At the same time, BA was tested to assess the effects on the change of the Oxidation–Reduction Potential (ORP) values during the decolorization processes. The experiments explored a great improvement of the redox mediator application and the new bio-treatment concept.
Sequential electrochemical oxidation and bio-treatment of the azo dye congo red and textile effluent

Abstract:Textile effluent is often difficult to manage as it contains a high concentration of toxic and recalcitrant synthetic dyes. In this study, congo Red and textile effluent were treated by electrochemical oxidation using RuO2-IrO2 coated titanium electrode as an anode followed by biodecolorization using Pseudomonas stutzeri MN1 and Acinetobacter baumannii MN3. Effluent pre-treatment is often necessary to minimize the inhibitory effects of textile dyes on dye degrading bacterial during bio-treatment. The pre-treatment of Congo Red by electrochemical oxidation for 10 min resulted in a decolorization rate of 98% at a pH, NaCl concentration, and current density of 7, 2 g L-1, and 20 mA cm-2. Subsequent bio-treatment of the pretreated Congo Red enhanced the biodegradation to 93%. The COD removal efficiency in real textile effluent following electrochemical pretreatment and biological treatment using bacterial consortium were 3.8% and 93%, respectively. Therefore, integrating electrochemical oxidation and microbial consortia offers an effective and environmentally friendly approach for treating complex industrial effluents.

Abstract:Azo dyes impact the environment and deserve attention due to their widespread use in textile and tanning industries and challenging degradation. The high temperature, pH, and salinity used in these industries render industrial effluent decolorization and detoxification a challenging process. An enrichment technique was employed to screen for cost-effective biodegraders of Direct Red 81 (DR81) as a model for diazo dye recalcitrant to degradation. Our results showed that three mixed bacterial cultures achieved ≥80% decolorization within 8 h of 40 mg/L dye in a minimal salt medium with 0.1% yeast extract (MSM-Y) and real wastewater. Moreover, these mixed cultures showed ≥70% decolorization within 24 h when challenged with dye up to 600 mg/L in real wastewater and tolerated temperatures up to 60 °C, pH 10, and 5% salinity in MSM-Y. Azoreductase was the main contributor to DR81 decolorization based on crude oxidative and reductive enzymatic activity of cell-free supernatants and was stable at a wide range of pH and temperatures. Molecular identification of azoreductase genes suggested multiple AzoR genes per mixed culture with a possible novel azoreductase gene. Metabolite analysis using hyphenated techniques suggested two reductive pathways for DR81 biodegradation involving symmetric and asymmetric azo-bond cleavage. The DR81 metabolites were non-toxic to Artemia salina nauplii and Lepidium sativum seeds. This study provided evidence for DR81 degradation using robust stress-tolerant mixed cultures with potential use in azo dye wastewater treatment.
