Abstract:Mercury (Hg) biomagnification occurs in many ecosystems, resulting in a greater potential for toxicological effects in higher-level trophic feeders.However, Hg transport pathways through different food-web channels are not well known, particularly in high-latitude systems affected by the atmospheric Hg deposition associated with snow and ice.Here, we report on stable carbon and nitrogen isotope ratios, and Hg concentrations, determined for 26, late 19th and early 20th century, polar bear (Ursus maritimus) hair specimens, collected from catalogued museum collections.These data elucidate relationships between the high-latitude marine food-web structure and Hg concentrations in polar bears.The carbon isotope compositions of polar bear hairs suggest that polar bears derive nutrition from coupled food-web channels, based in pelagic and sympagic primary producers, whereas the nitrogen isotope compositions indicate that polar bears occupy > fourth-level trophic positions.Our results show a positive correlation between polar bear hair Hg concentrations and d 15 N. Interpretation of the stable isotope data in combination with Hg concentrations tentatively suggests that polar bears participating in predominantly pelagic food webs exhibit higher mercury concentrations than polar bears participating in predominantly sympagic food webs.

In the last 25 years there has been an important increase in the amount of data collected from animal-mounted sensors (bio-probes) which are often used to study the animals' behaviour or environment. We focus here on an example of the latter, where the interest is in sea surface temperature (SST), and measurements are taken from sensors mounted on elephant seals in the southern Indian Ocean. We show that standard geostatistical models may not be reliable for this type of data, due to the possibility that the regions visited by the animals may depend on the SST. This phenomenon is know in the literature as preferential sampling, and, if ignored, it may affect the resulting spatial predictions and parameter estimates. Research on this topic has been mostly restricted to stationary sampling locations such as monitoring sites. The main contribution of this manuscript is to extend this methodology to observations obtained by devices that move through the region of interest, as is the case with the tagged seals. More specifically, we propose a flexible framework for inference on preferentially sampled fields where the process that generates the sampling locations is stochastic and moving over time through a two-dimensional space. Our simulation studies confirm that predictions obtained from the preferential sampling model are more reliable when this phenomenon is present, and they compare very well to the standard ones when there is no preferential sampling. Finally, we note that the conclusions of our analysis of the SST data can change considerably when we incorporate preferential sampling in the model.

Abstract:Biological responses to climate and environmental changes in remote polar regions are of increasing interest in global change research. Terrestrial and marine polar ecosystems have suffered from impacts of both rapid climate change and intense human activities, and large fluctuations in the population sizes of seabirds, seals, and Antarctic krill have been observed in the past decades. To understand the mechanisms driving these regime shifts in polar ecosystems, it is important to first distinguish the influences of natural forcing from anthropogenic activities. Therefore, investigations of past changes of polar ecosystems prior to human contact are relevant for placing recent human-induced changes within a long-term historical context. Here we focus our review on the fossil, sub-fossil, archaeological, and biogeochemical remains of marine vertebrates in polar sediments. These remains include well-preserved tissues such as bones, hairs and feathers, and biogeochemical markers and other proxy indicators, including deposits of guano and excrement, which can accumulate in lake and terrestrial sediments over thousands of years. Analyses of these remains have provided insight into both natural and anthropogenic impacts on marine vertebrates over millennia and have helped identify the causal agents for these impacts. Furthermore, land-based seabirds and marine mammals have been shown to play an important role as bio-vectors in polar environments as they transport significant amounts of nutrients and anthropogenic contaminants between ocean and terrestrial ecosystems.

Abstract:The Arctic faces increasing exposure to environmental chemicals such as metals, posing health risks to humans and wildlife. Biomonitoring of polar bears (Ursus maritimus) can be used to quantify chemicals in the environment and in traditional foods consumed by the Inuit. However, typically, these samples are collected through invasive or terminal methods. The biomonitoring of feces could be a useful alternative to the current metal monitoring method within the Arctic. Here, we aim to 1) quantify the relationship between concentrations of metals in the feces and tissues (muscle, liver, and fat) of polar bears using predictive modeling, 2) develop an easy-to-use conversion tool for use in community-based monitoring programs to non-invasively estimate contaminant concentrations in polar bears tissues and 3) demonstrate the application of these models by examining potential exposure risk for humans from consumption of polar bear muscle. Fecal, muscle, liver, and fat samples were harvested from 49 polar bears through a community-based monitoring program. The samples were analyzed for 32 metals. Exploratory analysis indicated that mean metal concentrations generally did not vary by age or sex, and many of the metals measured in feces were positively correlated with the internal tissue concentration. We developed predictive linear regression models between internal (muscle, liver, fat) and external (feces) metal concentrations and further explored the mercury and methylmercury relationships for utility risk screening. Using the cross-validated regression coefficients, we developed a conversion tool that contributes to the One Health approach by understanding the interrelated health of humans, wildlife, and the environment in the Arctic. The findings support using feces as a biomonitoring tool for assessing contaminants in polar bears. Further research is needed to validate the developed models for other regions in the Arctic and assess the impact of environmental weathering on fecal metal concentrations.

Abstract:Climate changes cause a dramatical increase in the ice-free season in the Arctic, forcing polar bears ashore, closer to human settlements associated with new and non-natural food objects. Such a diet may crucially transform the intestinal microbiome and metabolism of polar bears. The aim of this study was to characterize changes in the gut bacterial and fungal communities resulting from the transition to anthropogenic food objects by the means of 16S and ITS metabarcoding. Thus, rectal samples from 16 wild polar bears from the Kara-Barents subpopulation were studied. Human waste consuming resulted in a significant increase in the relative abundance of fermentative bacteria (Lactobacillaceae, Leuconostocaceae, and Streptococcaceae) and a decrease in proteolytic Enterobacteriaceae. However, the alpha-diversity parameters remained similar. Also, for the first time, the composition of the fungal community of the polar bear intestine was determined. Diet change is associated with the displacement of eurybiontic fungi (Thelebolus, Dipodascus, Candida (sake), and Geotrichum) by opportunistic Candida (tropicalis), Kazachstania, and Trichosporon. Feeding on human waste does not cause any signs of dysbiosis and probably leads to adaptive changes in the bacterial microbiome. However, the emergence of fungal facultative pathogens increases the risk of infections.

Abstract:Understanding behavioral responses of species to environmental change is critical to forecasting population-level effects. Although climate change is significantly impacting species' distributions, few studies have examined associated changes in behavior. Polar bear (Ursus maritimus) subpopulations have varied in their near-term responses to sea ice decline. We examined behavioral responses of two adjacent subpopulations to changes in habitat availability during the annual sea ice minimum using activity data. Location and activity sensor data collected from 1989 to 2014 for 202 adult female polar bears in the Southern Beaufort Sea (SB) and Chukchi Sea (CS) subpopulations were used to compare activity in three habitat types varying in prey availability: (1) land; (2) ice over shallow, biologically productive waters; and (3) ice over deeper, less productive waters. Bears varied activity across and within habitats with the highest activity at 50-75% sea ice concentration over shallow waters. On land, SB bears exhibited variable but relatively high activity associated with the use of subsistence-harvested bowhead whale carcasses, whereas CS bears exhibited low activity consistent with minimal feeding. Both subpopulations had fewer observations in their preferred shallow-water sea ice habitats in recent years, corresponding with declines in availability of this substrate. The substantially higher use of marginal habitats by SB bears is an additional mechanism potentially explaining why this subpopulation has experienced negative effects of sea ice loss compared to the still-productive CS subpopulation. Variability in activity among, and within, habitats suggests that bears alter their behavior in response to habitat conditions, presumably in an attempt to balance prey availability with energy costs.

Abstract:Understanding an animal's metabolic rate and thermal history is pivotal for ecological research. Recent studies have proposed the use of stable carbon and oxygen isotopes (δ 13 C and δ 18 O) in biogenic carbonates as proxies of metabolic rate and experienced temperature, respectively, to overcome the challenges of directly measuring these data in the field. Our study represents the first experimental investigation to develop δ 13 C and δ 18 O proxies in octopus. Octopus berrima hatchlings were raised in captivity, at varying water temperatures, for up to 110 days. O. berrima statoliths were then subsequently analysed for δ 13 C and δ 18 O values. The proportion of metabolically derived carbon, or respired carbon (C resp ), increased as the octopus grew (slope= 0.076, R 2 = 0.72), suggesting an influence of somatic growth rate and body mass on δ 13 C values. Additionally, we identified an inverse correlation between δ 18 O values and environmental temperature (slope= -0.163, R 2 = 0.91), which was subsequently used to develop a thermal reconstruction model. Our experiment aids in interpreting stable isotopic values in statoliths and their application as temperature and metabolic proxies in wild-caught octopus. Such proxies will increase our monitoring capabilities of these ecologically and commercially significant cephalopods and contribute to their conservation and effective management.

Abstract:Ecological understanding of the role of consumer-resource interactions in natural food webs is limited by the difficulty of accurately and efficiently determining the complex variety of food types animals have eaten in the field. We developed a method based on DNA metabarcoding multiplexing and next-generation sequencing to uncover different taxonomic groups of organisms from complex diet samples. We validated this approach on 91 faeces of a large omnivorous mammal, the brown bear, using DNA metabarcoding markers targeting the plant, vertebrate and invertebrate components of the diet. We included internal controls in the experiments and performed PCR replication for accuracy validation in postsequencing data analysis. Using our multiplexing strategy, we significantly simplified the experimental procedure and accurately and concurrently identified different prey DNA corresponding to the targeted taxonomic groups, with ≥ 60% of taxa of all diet components identified to genus/species level. The systematic application of internal controls and replication was a useful and simple way to evaluate the performance of our experimental procedure, standardize the selection of sequence filtering parameters for each marker data and validate the accuracy of the results. Our general approach can be adapted to the analysis of dietary samples of various predator species in different ecosystems, for a number of conservation and ecological applications entailing large-scale population level diet assessment through cost-effective screening of multiple DNA metabarcodes, and the detection of fine dietary variation among samples or individuals and of rare food items.

Abstract:We assessed the potential for plastic ingestion in polar bears (Ursus maritimus (Phipps (1774)) using fecal analysis. Two methods studies ensured our protocols could effectively recover and identify plastics in feces. First, microplastics (film, foam, or fragments) were intentionally introduced into a model organic matrix. Recovery rates (mean ± standard deviation) averaged 95.8 ± 14.7% (n = 18) and were significantly affected by microplastic morphology, but not digestion status. Second, microplastics of three polymers were intentionally introduced to polar bear feces. Recovery rates averaged 79.3 ± 21.6% (n = 8), and Raman microscopy successfully identified all polymers in 87.5% of samples. The main study then investigated the presence of microplastics in hunter-collected polar bear feces in the Canadian Arctic. Feces from colons of hunted bears (n = 15) and field scat (n = 15) were collected from 30 bears through collaboration with Inuit communities. Polypropylene, polyethylene, and/or polyethylene terephthalate were detected in feces from eight bears. Concentrations of microplastics in feces were, on average, less than 1 particle/g dry weight feces and at or near detection limits. Overall, this work suggests microplastic ingestion by Canadian polar bears may be low, and demonstrates the utility of fecal sampling for community-based monitoring programs.
