Abstract:Driven by the continuous demand for optically active fluoro-containing molecules from the chemical society, the field of fluorochemistry has emerged as a hot topic recently. While protocols focusing on catalytic asymmetric fluorination, (di)fluoroalkylation, and trifluoromethylation have been well developed, elegant strategies for the synthesis of enantioenriched monofluoro- and gem-difluoroalkenes remain highly sought after. As two vital subunits of organofluorides, both monofluoro- and gem-difluoroalkenes are pervasive within many biologically active molecules. They serve as the mimics of alkenyl, enol, and peptide bonds and the carbonyl group, respectively, which may enhance the bioactivities of the parent molecules. In addition, they are active handles for the construction of other fluorinated molecules. In this review, insightful discussions on the latest achievements of the catalytic asymmetric strategies associated with the facile synthesis of both monofluoro- and gem-difluoroalkenes are provided. The related mechanism considerations, applications in streamlining syntheses, limitations, and the opportunities of the state-of-the-art methodologies will be highlighted. We hope that this will trigger novel ideas, helping in pushing current frontiers further.

Abstract:gem-Difluoroalkyl groups, characterized as a gem-difluoromethylene group substituted by a hydrogen atom or an alkyl or other functional group, are not only valuable for the modulation of the properties of organic compounds, but are also useful for the synthesis of various fluorine-containing compounds. Currently, the catalytic asymmetric construction of stereogenic carbon centers that feature a gem-difluoroalkyl group is in its infancy. This Focus Review summarizes the latest achievements and discusses the difficulties and challenges in this field.

Abstract:As a special class of alkyne, gem-difluoromethylene alkynes exhibit a variety of fascinating property due to the presence of the gem-difluoro substitution. This substitution highlights the distinctive fluorine effects in influencing the chemoselectivity of reactions. As a result, chemical scientists have shown great interest and enthusiasm in investigating their reactions. In this review, we briefly summarize recent advances in transition-metal-catalysed reactions of gem-difluoromethylene alkynes with multiple reaction pathways. The mechanistic studies and challenges will be highlighted. The purpose of this review is to provide illustrations of elegant gem-difluoromethylene alkynes, and thereby elicit further interest among synthetic chemists in creating innovative transformation of gem-difluoromethylene alkynes.

The highly stereoselective synthesis of this privileged structural motif has attracted great synthetic attention. This review summarizes recent progresses in highly stereoselective synthesis of monofluoroalkenes from aldehydes, ketones and diazo compounds, other substrates such as alkynes, alkenyl metallic species and alkenes are also included. The advantages and disadvantages of different methods are discussed.

Abstract:We report a simple and economical method to synthesize monofluoroalkenes via the electrochemical hydrodefluorination of gem-difluoroalkenes. This reaction proceeds efficiently at room temperature, eliminating the requirement for a costly transition metal catalyst, ligand, and external reducing agent. The monofluoroalkene products can be obtained in medium to good yields and up to 99:1 E/Z selectivity. The reaction is easily scalable to gram scale.

Abstract:Chiral organofluorine compounds featuring a monofluoromethyl (CH 2 F)-substituted stereocenter are often encountered in a number of drugs and bioactive molecules. Consequently, the development of catalytic asymmetric methods for the enantioselective construction of CH 2 F-substituted stereocenters has received great progress over the past two decades, and a variety of enantioselective transformations have been accordingly established. According to the kinds of fluorinated reagents or substrates employed, these protocols can be divided into the following major categories: i) enantioselective ring opening of epoxides or azetidinium salts by fluoride anion; ii) asymmetric monofluoromethylation with 1-fluorobis(phenylsulfonyl)methane; iii) asymmetric fluorocyclization of functionalized alkenes with Selectfluor; iv) asymmetric transformations involving α-CH 2 F ketones, α-CH 2 F alkenes, or other CH 2 F-containing substrates. This feature article aims to summarize these recent advances, discuss the possible reaction mechanisms, advantages and limitations of each protocol and their applications. The synthetic opportunities still open for further development are illustrated as well. This review article would inspire for the researchers engaged in asymmetric catalysis, organofluorine chemistry, and medicinal chemistry.

Abstract:The gem-difluoroalkenes and related compounds have gained much attention from the organic synthetic community due to their widespread applications as versatile fluorinated building blocks for the synthesis of pharmaceuticals, agrochemicals and functional materials. In the past two decades, significant progress has been made to the development of efficient methods for the construction of gem-difluoroalkenes and useful reactions involving the cleavage of C-F bond in gem-difluoroalkenes. Abstract:The chiral fluorene fragment constitutes one of the most important scaffolds frequently found in numerous bioactive molecules, ligands, and advanced functional materials due to its unique physical properties. In this context, various transition metal‐ and organo‐catalyzed asymmetric reactions have been developed to prepare chiral fluorenes with high optical purity. In the present mini‐review, we summarize fifteen years of achievements in the catalytic asymmetric synthesis of chiral fluorenes and their derivatives. Particular attention is given to the strategies used and mechanistic aspects of some significant reactions. Moreover, the cutting‐edge developments in the catalytic asymmetric synthesis of silafluorenes are also highlighted.

Abstract:Herein, a photocatalytic system for defluorinative [4+2] annulation of gem‐difluoroalkenes with unactivated alkenes is realized. This protocol was compatible with a broad scope of unactivated alkenes, affording the diverse monofluorocyclohexene products in moderate to excellent yields. Furthermore, the value of this method was demonstrated by the gram‐scale synthesis, late‐stage functionalization and synthesis of various valuable and complex monofluorocyclohexene derivatives.

Abstract:Organofluorines have a broad range of industrial applications, such as pharmaceuticals, liquid crystal displays (LCDs), solar cells, textiles, and construction coatings, and are used in peptidomimetics, surfactants, refrigerants, anesthetics, and agrochemicals. Among them are versatile monofluoroalkenes that play a crucial role in medicinal and synthetic chemistry. The synthetic strategies for this class of molecules are limited, and prior efforts frequently suffered from poor atom- and step-economies. As a surrogate pathway for traditional cross-coupling transformations, transition metal (TM)-catalyzed C-H direct α-fluoroalkenylation overcomes these obstacles and provides straightforward techniques to access monofluoroalkenes. Nevertheless, substrate scope is still a challenge for catalysis, where gem-bromofluoroalkene synthons are applicable with electronically biased substrates such as azoles, while gem-difluoroalkene-based strategies are limited to substrates containing N-based directing groups. Herein, we review the cutting-edge fluoroalkenylation research for direct synthesis of monofluoroalkenes achieved during the last decade (2013-2023). This review is divided into two main parts: the first part discusses TM-catalyzed direct α-fluoroalkenylation via the merging of C-H activation and C(sp2)-Br cleavage strategies using gem-bromofluoroalkenes, and the second part describes the same reaction, albeit with C(sp2)-F cleavage of highly explored gem-difluoroolefins. Our review surveys all previously reported monofluoroalkenes in this research area, including their preparation techniques, stereoselectivity, and yield percentages. Furthermore, optimal conditions, reactant scope, mechanistic investigations, synthetic applications, benefits, and drawbacks of each presented methodology are critically discussed.

Abstract:An efficient decarboxylative/defluorinative alkylation for synthesizing gem‐difluoroalkenes is described, providing a general method for installation of the challenging alkyl fragments containing α‐electron‐withdrawing groups into α‐trifluoromethyl alkenes. Mechanistic studies suggest that this process involves an SN2′‐type synthetic route in the absence of transition‐metal catalysts or photocatalysis. Moreover, this protocol can easily be scaled up, and successfully applied to the modification of biologically active molecules, thus complementing methodologies that give access to structurally versatile gem‐difluoroalkenes.

Abstract:The judicious incorporation of a fluoroalkyl moiety often brings about beneficial effects on the properties of bioactive molecules. Consequently, efficient methods for selective fluoroalkylation are much sought after in drug discovery. Despite significant achievements in trifluoromethylation, selective mono- and difluoroalkylation is still undeveloped. Catalytic functionalization of fluorinated silyl enol ethers (FSEEs) emerges as a fruitful approach for the diversity-oriented synthesis of value-added α-mono or difluoroalkylated ketones. In this feature article, we detail our efforts in developing catalytic selective mono- and difluoroalkylation reactions using FSEEs. Specifically, we highlight our findings such as activating FSEEs by amines for catalytic enantioselective synthesis, taking advantage of the often observed high activity of FSEEs over the non-fluorinated analogues for reaction development, and the influence of C-FH-X interactions on reactivity and selectivity.

Abstract:The activation and functionalization of C-F bonds for the construction of C-C and C-X bonds have drawn significant research attention in recent years. However, the chemo-and stereoselective control of dual C-F bond functionalization of gem-difluoroalkenes remains a formidable challenge. Herein, a Ni-catalyzed reductive cross-coupling of gem-difluoroalkenes with alkenyl-electrophiles and D2O that allowed the generation of C(sp2)-C(sp2) bonds and C(sp2)-D bonds in one pot by successive defluorination is described. This methodology offers facile access to various deuterated 1,3-dienes with broad functional group compatibility and (E)-selectivity under mild conditions. Preliminary mechanistic studies indicate that alpha-alkenyl-substituted monofluoroalkenes might be intermediates in this protocol.

Abstract:Abstract A robust and practical difluoroalkylation synthon, α,α‐difluoroenol species, which generated in situ from trifluoromethyl diazo compounds and water in the presence of dirhodium complex, is disclosed. As compared to the presynthesized difluoroenoxysilane and in situ formed difluoroenolate under basic conditions, this difluoroenol intermediate displayed versatile reactivity, resulting in dramatically improved enantioselectivity under mild conditions. As demonstrated in catalytic asymmetric aldol reaction and Mannich reactions with ketones or imines in the presence of chiral organocatalysts, quinine‐derived urea, and chiral phosphoric acid (CPA), respectively, this relay catalysis strategy provides an effective platform for applying asymmetric fluorination chemistry. Moreover, this method features a novel 1,2‐difunctionalization process via installation of a carbonyl motif and an alkyl group on two vicinal carbons, which is a complementary protocol to the metal carbene gem‐difunctionalization reaction.
