Abstract:The development of thrombotic disease is one of the most common causes of morbidity and mortality in human. There are still disputes about the relationship between 4G/5G polymorphism of PAI-1 and plasma PAI-1 levels and thrombotic disorders.We analyzed the distribution of PAI-1 promoter 4G/5G polymorphism in Chinese and compared their effects on the transcriptional activity in vitro by using luciferase reporter vectors containing different PAI-1 promoters(4G or 5G). Methods: l.Genomic DNA was extracted from white blood cells from healthy blood donors (n=77) and PAI-1 4G/5G polymorphism was analyzed by allele-specific PCR. 2. The PAI-1 promoter sequence from -809 to +83 was amplified from individual homozygous for 5G insertion (I) or 4G deletion (D) allele and cloned into pGL3 vector. 3. The recombinant vectors were transfected into ECV304 and HepG2 cells,respectively.The transcriptional activities in basic state, and after chanllenges by thrombin or IL-1. were investigated. Results: l.The genotype frequency was 49.45% 44.1% 6.5% for 4G/4G 4G/5G 5G/5G genotypes.respectively, which differs from that reported in healthy Caucasians, with the 4G allele more frequently found in Chinese. 2.The 4G and 5G promoters were successfully cloned, and were identified by endonuclease cleavage analysis, and PCR, and DNA sequencing.3.Both of the two recombinants could be expressed in ECV304 and HepG2 cells; There were no significant difference in the basic transcriptional activities of the two promoters.4. Thrombin( 10-20 U/ml)had no effect on the transcriptional activities of the two promoters (4G and 5G) in ECV304 cells. IL-1(158 U/ml)increased the transcriptional activity of PAI-1-4G gene by 67% (p<0.05, n = 5) and had no effect on the transcription of PAI-1-5G. Conclusion: 1. The distribution of PAI-1 promoter 4G/5G polymorphism in Chinese differs from that in Caucasians, with the more 4G/4G genotype in Chinese. 2. The two promoters (PAI-1-4G and PAI-1-5G) cloned into luciferase reporter vector could be expressed in ECV304 and HepG2 cells, with more effective expression in HepG2 cell. 3. The basic transcriptional activities of the two promoters were almost the same and the response of PAI-1-4G promoter to IL-1 was higher than that of PAI-1-5G, suggesting that the 4G allele may be a possible responsive element to inflammatory stimuli.
