Abstract:In recent years, infection caused by drug-resistant bacteria has become a serious public health problem. The exploration of antibacterial therapies other than antibiotics has attracted more and more attention. Photothermal therapy (PTT) has become a promising antibacterial method due to its low invasiveness, low toxicity and avoidance of drug-resistant bacteria. However, when PTT is used alone, it requires a higher temperature to achieve a better antibacterial effect, which will not only kill bacteria, but also cause damage to normal tissues, even trigger new inflammation. Many reports have confirmed that a combination of other antibacterial methods with PTT could effectively reduce side effects on normal cells and enhance the therapeutic effect. In view of the rapid development of synergistic PTT in antibacterial therapy, this review mainly discusses and summarizes the advancements of several synergistic photothermal antibacterial methods within the last five years. In addition, the synergistic mechanism of antibacterial methods is also clarified. The remaining challenges and future opportunities in this field are also highlighted. We believe that this review will provide improved understanding of synergistic photothermal antibacterial therapy enabled by multifunctional nanomaterials and push nanoscience and nanotechnology one step at a time toward clinical applications.

Abstract:<p>With the prevalence of antibiotic-resistant bacteria, novel antibacterial strategies are urgently needed. In recent years, several antibiotics-independent physical approaches have attracted high attention and interests. Among those approaches, photothermal therapy (PTT), a novel non-invasive therapeutic technique, has exhibited great potentials in dealing with drug-resistant bacteria and bacterial biofilms. Photothermal agents (PTAs), which are either nanomaterials themselves or small molecules loaded in nanoparticles, are the essential element for PTT. How to deliver PTAs in a controlled manner is of great importance for high-efficiency and low-toxicity PTT. Therefore, a comprehensive understanding of various PTAs is required for the better application of PTT in antibacterial treatment. Herein, the physicochemical properties and antibacterial PTT of five types of PTAs are summarized. In addition, the PTT-involved multifunctional theranostics nanoplatforms and the potential approaches for reducing the side effects of PTT (such as targeted delivery and controlled release of PTAs) are also discussed.</p>

Abstract:Photothermal therapy has the advantages of non-invasiveness, low toxicity, simple operation, a broad spectrum of antibacterial ability, and non-proneness to developing drug resistance, which provide it with irreplaceable superiority in fighting against microbial infection. The effect of photothermal therapy is closely related to the choice of photothermal agent. Conjugated nanomaterials are potential candidates for photothermal agents because of their easy modification, excellent photothermal conversion efficiency, good photostability, and biodegradability. In this paper, the application of photothermal agents based on conjugated nanomaterials in photothermal antimicrobial treatment is reviewed, including conjugated small molecules, conjugated oligomers, conjugated polymers, and pseudo-conjugated polymers. At the same time, the application of conjugated nanomaterials in the combination of photothermal therapy (PTT) and photodynamic therapy (PDT) is briefly introduced. Finally, the research status, limitations, and prospects of photothermal therapy using conjugated nanomaterials as photothermal agents are discussed.

Abstract:Bacterial infection continues to be one of the biggest threats to human health. The therapeutic effect is significantly reduced as a result of the development of super-bacteria-resistant bacteria due to the overuse of antibiotics for conventional antimicrobial treatment. The emergence of drug-resistant bacteria makes it necessary to develop new antimicrobial treatments. Following the successful application of photothermal therapy in the field of oncology treatment, more and more researchers are applying it to antimicrobial treatments and achieving important results, especially in the treatment of drug-resistant bacterial infections. As a non-invasive anti-infection method, photothermal therapy has the advantages of broad-spectrum antimicrobial properties, a short treatment period, and low systemic impact. In the PTT, the antimicrobial efficacy strongly depends on the different laser properties and the choice of photothermal agents (PTAs). The purpose of this review is to discuss the most recent developments in photothermal nanomaterials that are antibacterial. We will delve into the antimicrobial principles and antimicrobial spectrum of different PTAs and provide a summary of current developments in synthesis, classification, structural features, physicochemical properties, and antibacterial performance research. The challenges and future prospects of enhanced antimicrobial performance and reduced cytotoxicity of PTAs are investigated.

Abstract:Bacterial infection is still a ticklish clinical challenge even though some advanced antibacterial materials and techniques have been put forward. This work reports that rapid and effective antibacterial performance is achieved by the synergistic local photothermal and photodynamic therapy (PTDT). Within 10 min of light irradiation, both Escherichia coli and Staphylococcus aureus are almost completely eliminated by the action of photothermy (52.1 °C) and limited reactive oxygen species (ROS), the corresponding bacterial killing efficiencies are 99.91 and 99.97%, respectively, which are far higher than single modal therapy, i.e., photothermal therapy or photodynamic therapy with antibacterial efficacy of 50 or 70%, respectively. The mechanism is that bacterial membrane permeation is increased by PTDT because photothermy shows more severe impact only on E. coli by destroying the outmost bacterial panniculus, whereas the inner panniculus of the two kinds of bacteria is more sensitive to ROS. Hence, ROS penetrates the bacterial membrane more easily, and meanwhile, the proteins in the bacteria are severely lost after the bacterial membrane disruption, which leads to bacterial death. In vivo results reveal that rapid and effective sterilization is an important process to accelerate wound healing, and the traumas on the rats' backbones heal well within 12 days by PTDT. Furthermore, the PTDT is friendly to major organs of rats during the therapeutic process. Therefore, the synergistic therapy system can be a safe therapeutic system for clinical sterilization with great potential. More importantly, the antibacterial mechanism presented in this work has great guiding significance for the design of other advanced antibacterial systems and techniques.
A Universal Strategy to Enhance Photothermal Conversion Efficiency by Regulating the Molecular Aggregation States for Safe Photothermal Therapy of Bacterial Infections

Abstract:Photothermal therapy (PTT) has emerged as a promising approach for treating bacterial infections. However, achieving high photothermal conversion efficiency (PCE) of photothermal agents (PTAs) remains a challenge. Such a problem is usually compensated by the use of high-intensity laser, which inevitably causes tissue damage. Here, we present a universal strategy to enhance PCE by regulating the molecular aggregation states of PTAs within thermoresponsive nanogels. We demonstrate the effectiveness of this approach using aggregation-induced emission (AIE) and aggregation-caused quenching (ACQ) PTAs, showing significant enhancements in PCE without the need for intricate molecular modifications. Notably, the highest PCEs reached up to 80.9% and 64.4% for AIE-NG and ACQ-NG, respectively, which were nearly two-fold of their self-aggregate counteraparts. Moreover, we elucidate the mechanism underlying PCE enhancement, highlighting the role of strong intermolecular π−π interactions facilitated by nanogel-induced volume contraction. Furthermore, we validate the safety and efficacy of this strategy in in vitro and in vivo models of bacterial infections at safe laser power densities, demonstrating its potential for clinical translation. Our findings offer a straightforward, universal, and versatile method to improve PTT outcomes while minimizing cytotoxicity, paving the way for enhanced treatment of bacterial infections using safe PTT protocols.

Abstract:Abstract Photothermal therapy (PTT), which converts light energy to heat energy, has become a new research hotspot in cancer treatment. Although researchers have investigated various ways to improve the efficiency of tumor heat ablation to treat cancer, PTT may cause severe damage to normal tissue due to the systemic distribution of photothermal agents (PTAs) in the body and inaccurate laser exposure during treatment. To further improve the survival rate of cancer patients and reduce possible side effects on other parts of the body, it is still necessary to explore PTAs with high selectivity and precise treatment. In this review, we summarized strategies to improve the treatment selectivity of PTT, such as increasing the accumulation of PTAs at tumor sites and endowing PTAs with a self-regulating photothermal conversion function. The views and challenges of selective PTT were discussed, especially the prospects and challenges of their clinical applications.

Abstract:Multidrug-resistant (MDR) bacteria are rapidly emerging, coupled with the failure of current antibiotic therapy; thus, new alternatives for effectively treating infections caused by MDR bacteria are required. Hyperthermia-mediated photothermal therapy (PTT) and reactive oxygen species (ROS)-mediated photodynamic therapy (PDT) have attracted extensive attention as antibacterial therapies owing to advantages such as low invasiveness, low toxicity, and low likelihood of causing bacterial resistance. However, both strategies have notable drawbacks, including the high temperature requirements of PTT and the weak ability of PDT-derived ROS to penetrate target cells. To overcome these limitations, a combination of PTT and PDT has been used against MDR bacteria. In this review, we discuss the unique benefits and limitations of PTT and PDT against MDR bacteria. The mechanisms underlying the synergistic effects of the PTT–PDT combination are also discussed. Furthermore, we introduced advancements in antibacterial methods using nano-based PTT and PDT agents to treat infections caused by MDR bacteria. Finally, we highlight the existing challenges and future perspectives of synergistic PTT–PDT combination therapy against infections caused by MDR bacteria. We believe that this review will encourage synergistic PTT- and PDT-based antibacterial research and can be referenced for future clinical applications.

Abstract:Photothermal therapy (PTT) has been regarded as a promising strategy considering its advantages of high inherent specificity and a lower invasive burden. Since the photothermal killing of cells/bacteria showed different patterns of death depending on the varying temperature in PTT, the temperature change of PTT is vital to cell/tissue response in scientific research and clinical application. On one hand, mild PTT has received substantial attention in the treatment of cancer and soft/hard tissue repair. On the other hand, the high temperature induced by PTT is capable of antibacterial capacity, which is better than conventional antibiotic therapy with drug resistance. Herein, we summarize the recent developments in the application of temperature‐dependent photothermal biomaterials, mainly covering the temperature ranges of 40–42°C, 43–50°C, and over 50°C. We highlight the biological mechanism of PTT and the latest progress in the treatment of different diseases. Finally, we conclude by discussing the challenges and perspectives of biomaterials in addressing temperature‐orchestrated PTT. Given a deep understanding of the interaction between temperature and biology, rationally designed biomaterials with sophisticated photothermal responsiveness will benefit the outcomes of personalized PTT toward various diseases. Photothermal therapy can be used for the treatment of tumors, tissue defects, and anti‐infection, depending on the effects at varying temperatures. In this review, we summarize the recent developments in the application of temperature‐dependent photothermal biomaterials, including the temperature ranges of 40–42°C, 43–50°C, and over 50°C. A NIR-II emissive polymer AIEgen for imaging-guided photothermal elimination of bacterial infection

Abstract:The development of antibiotics resistance has made multidrug-resistant (MDR) bacterial infection one of the most serious global health issues. Photothermal therapy (PTT) is an emerging therapeutic mode which can be applied to bacterial infection without inducing resistance. Moreover, enhanced therapeutic efficacy and less tissue damage can be realized with NIR-II fluorescence imaging (FLI) guided PTT. Herein, a polymeric luminogen with aggregation-induced emission (AIEgens) characteristics, poly(dithieno[3,2-b:2′,3′-d]pyrrole-benzo[1,2-c:4,5-c']bis([1,2,5]thiadiazole)) (PDTPTBT), was synthesized and used as a photothermal agent for PTT of bacterial infections. PDTPTBT was encapsulated into liposomes (L-PDTPTBT) for improved water dispersibility. Upon 808 nm NIR irradiation, L-PDTPTBT can eliminate multiple bacteria including the Gram-positive methicillin-resistant Staphylococcus aureus and Enterococcus faecalis, the Gram-negative Escherichia coli and Pseudomonas aeruginosa. Serious damage of bacterial membrane and leakage of cytoplasm is observed after photothermal treatment using L-PDTPTBT. The potential of the formulation has been demonstrated in two infected animal models: (i) a subcutaneous abscess model and (ii) a diabetic skin infection model. In the diabetic skin infection model, the death of mice is largely suppressed and the wounds can heal more quickly with treatment of L-PDTPTBT under NIR irradiation. The excellent photothermal bactericidal ability and low cytotoxicity make L-PDTPTBT potential candidate for treating MDR bacterial infections in the future.

Abstract:In the past few decades, great progress has been made in the field of nanomaterials against bacterial infection. However, with the widespread emergence of drug-resistant bacteria, people try their best to explore and develop new antibacterial strategies to fight bacteria without obtaining or increasing drug resistance. Recently, multi-mode synergistic therapy has been considered as an effective scheme for the treatment of bacterial infections, especially the combination of photothermal therapy (PTT) and photodynamic therapy (PDT) with controllable, non-invasive, small side effects and broad-spectrum antibacterial characteristics. It can not only improve the efficiency of antibiotics, but also do not promote antibiotic resistance. Therefore, multifunctional nanomaterials which combine the advantages of PTT and PDT are more and more used in the treatment of bacterial infections. However, there is still a lack of a comprehensive review of the synergistic effect of PTT and PDT in anti-infection. This review first focuses on the synthesis of synergistic photothermal/photodynamic nanomaterials and discusses the ways and challenges of photothermal/photodynamic synergism, as well as the future research direction of photothermal/photodynamic synergistic antibacterial nanomaterials.

Abstract:The construction of efficient photothermal antibacterial platform is a promising strategy for the treatment of drug-resistant bacterial infections. Herein, through the introduction of excited-state intramolecular proton transfer to promote the photothermal effect, N-(2,4-Dihydroxybenzylidene)-4-aminophenol (DOA)-polyvinyl alcohol (PVA) systems (DPVA) can reach 55°C within 10 s under irradiation. It shows superior antibacterial behavior against drug-resistant bacterial and therapeutic effect on infected skin wounds with only 100 s of irradiation, much faster than those of reported photothermal materials (5~10 mins). This work provides a convenient approach to fabricate broad-spectrum antibacterial wound dressing for treating bacteria-infected wounds, greatly contributing to the design and applications of photothermal antibacterial platform.

Abstract:Bacterial infection remains one of the most dangerous threats to human health due to the increasing cases of bacterial resistance, which is caused by the extensive use of current antibiotics. Photothermal therapy (PTT) is similar to photodynamic therapy (PDT), but PTT can generate heat energy under the excitation of light of specific wavelength, resulting in overheating and damage to target cells or sites. Polydopamine (PDA) has been proved to show plenty of advantages, such as simple preparation, good photothermal conversion effects, high biocompatibility, and easy functionalization and adhesion. Taking these advantages, dopamine is widely used to synthesize the PDA nanosystem with excellent photothermal effects, good biocompatibility, and high drug loading ability, which therefore play more and more important roles for anticancer and antibacterial treatment. PDA nanosystem-mediated PTT has been reported to induce significant tumor inhibition, as well as bacterial killings due to PTT-induced hyperthermia. Moreover, combined with other cancer or bacterial inhibition strategies, PDA nanosystem-mediated PTT can achieve more effective tumor and bacterial inhibitions. In this review, we summarized the progress of preparation methods for the PDA nanosystem, followed by advances of their biological functions and mechanisms for PTT uses, especially in the field of antibacterial treatments. We also provided advances on how to combine PDA nanosystem-mediated PTT with other antibacterial methods for synergistic bacterial killings. Moreover, we further provide some prospects of PDA nanosystem-mediated PTT against intracellular bacteria, which might be helpful to facilitate their future research progress for antibacterial therapy.

Abstract:Photothermal therapy (PTT) has received increasing attention due to its high selectivity, non-invasiveness, negligible drug resistance, oxygen independence, and minimal side effects. In PTT, photothermal agents (PTAs) absorb light energy and generate heat to kill cancer cells. Although various PTAs with strong near-infrared (NIR) light absorption and high photothermal conversion efficiency have been developed, their clinical uses in PTT still face with challenges. For example, maintaining a temperature of above 50°C for several minutes is needed to effectively ablate tumors, but this high temperature may destroy the surrounding normal tissues. Moreover, this rapid heating can induce an immune response that increases the cell tolerance to heat, which can weaken the therapeutic effects of PTT. In this review, we mainly focus on two approaches that have been developed to overcome the limitations of PTT, including inhibiting the expression of heat shock protein (Hsp) and regulating the metabolic processes of cells to develop low temperature PTT (LTPTT). We then discuss the existing challenges in clinical uses of LTPTT for treating tumors, and finally introduce future perspectives on the design of more effective therapeutic agents.

Abstract:Antibiotics are currently the main strategy to treat bacterial infections, but they can cause antimicrobial resistance. Thus, it is urgent to solve this problem. The emergence of photothermal therapy provides a new opportunity for the prevention and control of bacterial infection. In recent years, photothermal agents have been widely used in infection control and wound healing due to their strong antibacterial properties and low drug resistance. Photothermal agents (PTAs) are nanomaterials themselves, or small molecules loaded in nanoparticles, and are the basic elements of PPT. In this review, we discuss the characteristics of wound dressings in skin wound healing, types and main functions of antibacterial photothermal therapy (PTA), and the fabrication and application of wound dressings. Abstract:Pathogens on wounds and infected tissues or pathogens in drinking water or public facilities have been doing great harm in human life. Because of booming drug resistance and superbacteria, the abuse or excessive use of antibiotics during systemic treatment has caused a global antibiotic crisis. However, it usually takes a long time to develop antibiotics. In recent years, photocatalytic antibacterial agents have no drug resistance and side-effects due to their rapid and efficient bactericidal efficacy. They are becoming one of the most hopeful substitutions to antibiotics for dealing with the bacterial diseases and water pollution caused by certain pathogens. Photocatalysis has unique advantages in the field of antibacterials, and its controllability plays an irreplaceable role. This review focuses on the mechanism of photocatalysis, which involves representative photocatalytic semiconductors (metal oxides, metal sulfides, carbon nitride, heterojunction composite materials) and organics (organic polymers and organic small molecules-aggregation induced emission) as well as their photocatalytic antibacterial mechanism. In this paper, we summarize the photocatalytic antibacterial mechanisms by the numbers and current developing of photocatalytic antimicrobial materials applications. Current difficulties and expectations for the future in these fields are presented to stimulate the developing of material manufacturing technologies and their industrialization to combat bacterial infections. In addition, potential application limitations and future research potential are highlighted.
