Abstract:A purified polysaccharide was acquired from a newly collected wild Morchella. The strain identification initially showed that the strain was Morchella sextelata. This study aimed to investigate the structural features and immunomodulating activities of the polysaccharide. Polysaccharide extracted from mycelia was purified by DEAE-cellulose chromatography and Sephadex G-25 size-exclusion chromatography in sequence. The main fraction of polysaccharide (MSP-II) was obtained during purification process. High Performance Liquid Chromatography (HPLC) analysis revealed that MSP-II was composed of Glc, Ara, Gal, Man, Rha, Fuc, GalUA and GluUA in ratio of 34.95:8.7:9.55:4.55:5.0:1.45:12.7:7.65. The structure of MSP-II was furtherly analyzed by FT-IR spectrum and 1H and 13C NMR spectroscopy, the results showed that MSP contained β-glycosidic bonds as well as α-glycosidic linkages. In vitro tests proved that MSP-II could not only promote the proliferation and phagocytosis of RAW264.7 cells, but also induce the section of nitric oxide (NO) of macrophages. Consequently, the polysaccharide has a potent immunomodulatory activity by stimulating macrophages and can be considered as a novel potential immunopotentiator in medical and food industries.
Effect of Surfactants on the Production of Polysaccharides from Schizophyllum Commune Through Submerged Fermentation.

Abstract:Schizophyllum commune (S. commune) polysaccharides are biomacromolecules with multiple biological activities and wide applications. In this study, polysaccharide production through submerged fermentation of S. commune using different surfactants was investigated. The addition of 1 g/L of polyoxyethylene sorbitan monooleate (Tween 80) at the beginning of the fermentation showed the best promotional effects on collective exopolysaccharide (EPS) production (which increased by 37.17%) while shortening the production cycle by 2 days. The monosaccharide composition of the EPS produced when the added Tween 80 was similar to that of the control; however, the molecular weight (Mw) was lower. Notably, the addition of Tween 80 significantly increased the ATP levels and the transcription levels of phosphoglucomutase and beta-glucan synthase genes in the polysaccharide synthesis pathway. The addition of Tween 80 reduced the pellet size of the mycelium compared to that of the control, but did not significantly change the microstructure of the mycelial cells. This study proposes an efficient strategy for the production of polysaccharides through submerged fermentation of S. commune, and elucidates the detailed mechanism of using Tween 80 as a fermentation stimulatory reagent.
Isolation, partial characterisation and immunomodulatory activities of polysaccharide from Morchella esculenta

Abstract:Background: Polysaccharides extracted from fungi or algae have shown a variety of medical activities, and the exploitation of polysaccharides for application in pharmacy has been very promising. In this study, the immunomodulatory properties of polysaccharides from Morchella esculenta were investigated to identify immunostimulatory activity and potentially contribute to the therapeutic potential of Morchella esculenta. Results: A water-soluble polysaccharide, MEP, was obtained from the fermentation broth of Morchella esculenta. Two fractions of this polysaccharide (MEP-I and MEP-II) were extracted and purified. High-performance gel permeation chromatography analysis showed the average molecular weights of two polysaccharides. Structural properties and compositions of these two fractions were examined by Fourier transform infrared and a high-performance liquid chromatography with an evaporative light scattering detector. Active experiments suggested that the MEP had typical immunostimulatory activity. Conclusion: These bioactivity tests showed that the polysaccharides from Morchella esculenta presented significant immune modulating activity, and this finding may offer the basis for the popular use of polysaccharides in functional foods or medicine.

Abstract:A novel polysaccharide (MSP-1) was isolated from the fruiting body of Morchella sextelata and purified using DEAE-52 and Sephadex G-75. The molecular weight of MSP-1 was 1.17 × 104 Da, as detected by HPLC analysis. The monosaccharide composition of MSP-1 was mannose and glucose at a ratio of 1.00: 1.25. Methylation and NMR results revealed that the backbone of MSP-1 was composed of →4)-β-D-Manp-(1→, →4)-β-D-Glcp-(1→, →4)-α-D-Glcp-(1→, and →4, 6)-α-D-Glcp-(1→. SEM images of MSP-1 presented a dense network structure with porous characterizations. The immunomodulatory activities of MSP-1 were evaluated using RAW264.7 cells, and the results showed that MSP-1 promoted proliferative and phagocytic activity and increased the production of NO, TNF-α and IL-6. These results indicated that MSP-1 exhibited significant immunomodulatory activities.

Abstract:Morchella esculenta is a precious edible and medicinal fungus rich in protein, polysaccharides, polyphenols, amino acids, triterpenes, and other active components. In this study, MS-1 was isolated from the fruiting body of M. esculenta. Through conducting single-factor experiments and the response surface analysis of the culture conditions, the optimal culture components of an M. esculenta fermentation broth for extracellular polysaccharide production were determined, namely, 3.7% glucose, 2% yeast extract, and 0.15% sodium chloride. The polysaccharides MSF and MSL were extracted from the fruiting body of M. esculenta and the fermentation broth, respectively, and analyzed with gel permeation chromatography (GPC), monosaccharide composition, scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), and in vivo and in vitro antioxidant and anti-inflammatory activity analyses. The research results show that the calculated MW of MSF is 2.34 × 105 Da, and the calculated MW of MSL is 1.40 × 105 Da. MSF is composed of three monosaccharides: D-galactose, D-glucose, and D-mannose (molar ratio of 4.34:90.22:5.45). MSL consists of five monosaccharides: D-arabinose, D-galactose, D-glucose, D-mannose, and glucuronic acid (molar ratio of 0.31:14.71:13.03:71.43:0.53). The in vitro antioxidant test results show that MSF and MSL both have significant antioxidant activities. Activity experiments on MSF and MSL in zebrafish showed that MSF and MSL have significant repair effects on the oxidative damage caused by metronidazole in zebrafish embryos, and there were significant changes in the transcriptional activity levels of the oxidative stress-related genes SOD, Keap1, and Nrf2. Therefore, the polysaccharides MSF and MSL from MS-1 can be used as important raw materials for functional foods and drugs.

Abstract:Morchella esculenta (L.) Pers., referred to as Morel, is a medicinal and edible homologous fungus, which contains many bioactive substances. In Morel, polysaccharides are the most abundant and have various bioactivities. In the present work, two novel polysaccharides, Se-MPS and MPS, were prepared and purified from selenium-enriched (Se-enriched) and common Morel mycelia, respectively, and their structural and immunomodulatory properties were evaluated. The results show that Se-enriched treatment significantly changed the polysaccharides' chemical composition, molecular weight, and sugar chain configuration. In addition, the Se-enriched treatment also improved the polysaccharides' fragmentation and thermal stability. Importantly, Se-enriched Morel polysaccharide (Se-MPS) could significantly enhance phagocytosis of RAW 264.7 macrophage cells and, remarkably, activate their immune response via activating the TLR4-TRAF6-MAPKs-NF-κB cascade signaling pathway, finally exerting an immunomodulatory function. Based on these findings, selenium-enriched Morel polysaccharide appears to have more potential for development and utilization in functional foods or medicines than ordinary Morel polysaccharide.

Abstract:Objective:to investigate the structure and in vitro immunomodulatory activity of polysaccharides from bamboo shavings (BSP).Methods:the structure was analyzed by the methods of methylation and nuclear magnetic resonance spectroscopy;the in vitro immunomodulatory activity was evaluated by the assays of mouse splenic lymphocyte proliferation,the phagocytosis of neutral red and NO secretion of RAW264.7 cell.Results:BSP was O-acetyl-(4-O-methylglucurono)-arabinoxylans consisting of a linear (l→4)-β-D-xylopyranosyl backbone decorated with branches at O-2 of 4-O-methylglucuronic acid units and acetyl groups or O-3 of α-L-arabinofuranosyl and acetyl groups.The structural element could be expressed as below:1,4-β-D-[2-O-Ac][3-O-Ac][4-O-Me-D-GlcpA-(1→2)][α-L-Araf-(1→3)]-xylp;BSP could evidently strengthen concanavalin A or lipopolysaccharide-induced splenic lymphocyte proliferation at 10-100μg/mL;BSP could significantly enhance the phagocytosis activity and NO production of RAW264.7 cell at 10-1 000 μg/mL in a dose-dependent manner.Conclusion:BSP exhibits excellent immunostimulatory activity in vitro,and can be considered as an ideal source of bio-active polysaccharides due to the abundance of its raw material.

Abstract:Morchella esculenta L. is a rare and the most valuable edible mushrooms. The polysaccharides of M. esculenta L. boast high nutrition and health values. This paper describes microwave extraction of the polysaccharides of M. esculenta L. by ultrasonic microwave. The optimal extraction conditions, optimized by single factor experiments and response surface method, were determined as follows: the ultrasonic power is 250 W, the microwave power is 220 W, and the extraction time is 7 min with a yield of polysaccharides at 4.425%. The crude polysaccharides of M. esculenta L. at 4 mg/mL exhibited hydroxyl radical, 2-Diphenyl-1-picrylhydrazyl radical and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging rates at 76.70%, 60.73%, and 80.87%, respectively. Its Fe2+ chelating rates increased from 45.63% to 82.50%, as the polysaccharide concentration increased from 0.5 to 4.0 mg/mL.

Optimization of Submerged Culture for Exopolysaccharides Production by Morcella esculenta and its Antioxidant Activities In Vitro

Abstract:Morcella esculenta is an excellently edible and delicious morel mushroom found growing in China, Japan, and other Asian countries. Polysaccharides extracted from fungi have shown a variety of medical activities; however, the industrial large-scale production of Morcella esculenta can not be achieved now due to the natural conditions. So this paper is concerned with optimization of submerged culture conditions for mycelial growth and exopolysaccharides (EPS) production of Morcella esculenta. by airlift fermentation. And the antioxidant activities of EPS were revealed by various free radical scavenging assays in vitro. The optimal medium constituents were determined as follows: (g/L): glucose 20 g/L, peptone 4.5 g/L, KH2PO4 0.8 g/L, and MgSO4·7H2O 1.0 g/L. The optimum parameters of liquid culture were temperature 25 °C, cultivation time 5 d, rotary speed 140 rpm, and initial pH 6.0. This optimization strategy in shake flask culture leads to a mycelia yield of 4.93 ± 0.28 g/L, and EPS production of 1.65 ± 0.28 g/L, respectively. Under optimal culture conditions, the maximum mycelia and EPS concentration in a 7 L airlift tower loop reactor were 6.13 g/L and 2.12 g/L for the first report, which were considerably higher than those obtained in preliminary studies that indicated the EPS production was closely correlated to the mycelia growth by Morcella esculenta. Futuremore, the EPS demonstrated positively antioxidant potential on 1,1-diphenyl-2-picrylhydrazyl radical scavenging, reducing power, and hydroxyl radical scavenging. These findings may provide the basis for the popular use of EPS in functional food or medicine.
Improvement of Exo-Polysaccharides Production and Modeling Kinetics by Armillaria Luteo-Virens Sacc. in Submerged Cultivation

Abstract:Armillaria luteo-virens Sacc. is a special medicinal and edible mushroom in China, and its wild type has been investigated that it contains some functional components. The optimization of submerged culture conditions for mycelium growth and exo-polysaccharide (EPS) production in an edible mushroom A. luteo-virens Sacc. was studied in shake flasks and bioreactors with response surface methodology (RSM). A fractional factorial design (FFD) and central composite design (CCD) were applied to optimize the main factors that affect the fungal polysaccharides production in a submerged culture. Glucose is very important for EPS production by A. luteo-virens Sacc., and yeast extract powder also significantly affects fungal EPS production through fractional factorial experiment. The optimized cultivation medium constituents were derived from the predicted model using central composite design as follows: 31.26g/l glucose, 1.06g/l yeast extract powder, 1.00g/l K2HPO4, 0.50g/l KH2PO4, and 0.45g/l MgSO4·7H2O, pH unadjusted. The predicted maximal EPS concentration was 5.40g/l using the optimized culture medium compositions at 23°C for 5 days. Additionally, the submerged cultivation process was explained using the optimized cultivation medium. The cultivation kinetics model constructed was suitably depicting the culture process in bioreactor cultivation scale.

Abstract:Our previous investigations have successfully identified the repeating structural units of EPS53, an exopolysaccharide derived from Streptococcus thermophilus XJ53 fermented milk, and substantiated its potential immunomodulatory properties. The present study further elucidated the structural characteristics of EPS53 and investigated the underlying mechanisms governing its in vitro immunoreactivity as well as its in vivo immunoreactivity. The results obtained from multi-detector high performance gel filtration chromatography revealed that EPS53 adopted a rigid rod conformation in aqueous solution, with the weight-average molecular weight of 1464 kDa, the number-average molecular weight of 694 kDa, and the polydispersity index of 2.11. Congo red experiment confirmed the absence of a triple helix conformation. Scanning electron microscopy showed that EPS53 displayed a three-dimensional fibrous structure covered with flakes. The in vitro findings indicated that EPS53 enhanced phagocytosis ability, reactive oxygen species (ROS) production, and cytokine levels of macrophages via the TLR4-mediated NF-κB/MAPK signaling pathways as confirmed by immunofluorescence staining experiments, inhibition blocking experiments, and Western blot assay. Additionally, the in vivo experiments demonstrated that EPS53 significantly increased macrophage and neutrophil number while enhancing NO and ROS levels in zebrafish larvae; thus, providing further evidence for the immunomodulatory efficacy of EPS53.

Abstract:In our continuous exploration for bioactive polysaccharides, a novel polysaccharide FMP-2 was isolated and purified from the fruiting bodies of Morchella esculenta by alkali-assisted extraction. FMP-2 had an average molecular weight of 1.09 × 10<sup>6</sup> Da and contained mannose, glucuronic acid, glucose, galactose, and arabinose in a molar ratio of 4.10:0.22:1.00:5.75:0.44. The backbone of FMP-2 mainly consisted of 1,2-α-D-Galp, 1,6-α-D-Galp, and 1,4-α-D-Manp, with branches of 1,4,6-α-D-Manp and 1,2,6-α-D-Galp. FMP-2 can stimulate phagocytosis and promote the secretion of NO, ROS, and cytokines like IL-6, IL-1β, and TNF-α in RAW264.7 cells ranging from 25 to 400 μg/mL. FMP-2 had great repairing effect on the immune injury of zebrafish induced by chloramphenicol. The phagocytosis ability of zebrafish macrophages and the proliferation of neutrophils can be greatly enhanced by polysaccharide FMP-2 with concentrations from 50 to 200 μg/mL. These findings suggest that FMP-2 might be used as a potential immunomodulator in the food and pharmaceutical industries.

Abstract:Mucor sp. has a wide range of applications in the food fermentation industry. In this study, a novel exopolysaccharide, labeled MSEPS, was separated from Mucor sp. fermentation broth through ethanol precipitation and was purified by ion-exchange chromatography, as well as gel filtration column chromatography. MSEPS was composed mostly of mannose, galactose, fucose, arabinose, and glucose with a molar ratio of 0.466:0.169:0.139:0.126:0.015 and had a molecular weight of 7.78 × 104 Da. The analysis of methylation and nuclear magnetic resonance results indicated that MSEPS mainly consisted of a backbone of →3,6)-α-d-Manp-(1→3,6)-β-d-Galp-(1→, with substitution at O-3 of →6)-α-d-Manp-(1→ and →6)-β-d-Galp-(1→ by terminal α-l-Araf residues. MTT assays showed that MSEPS was nontoxic in normal cells (HK-2 cells) and inhibited the proliferation of carcinoma cells (SGC-7901 cells). Additionally, morphological analysis and flow cytometry experiments indicated that MSEPS promoted SGC-7901 cell death via apoptosis. Therefore, MSEPS from Mucor sp. can be developed as a potential antitumor agent.

Abstract:Two polysaccharides, DGS1 and DGS2, were obtained by solid-state fermentation (SSF) of Fusarium solani DO7, an endophytic fungus isolated from the orchid Dendrobium officinale. Structural characterizations revealed that DGS1 consisted of arabinose, glucose, mannose and galactose with a molar ratio of 2.9:13.4:3.0:1, respectively, and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp, (1 → 2,6)-Manp and (1 → 6)-Galp glycosidic linkages, while DGS2 was composed of arabinose, glucose, mannose and galactose in a molar ratio of 3.5:8.1:2.1:1, respectively, and contained (1 → 5)-Araf, (1 → 4)-Glcp, (1 → 6)-Glcp, (1 → 3)-Manp and (1 → 6)-Galp glycosidic linkages. Neither polysaccharide was toxic to human embryonic kidney cells or mouse RAW 264.7 macrophage cells. An immunomodulatory activity assay indicated that both polysaccharides could significantly enhance the levels of TNF-α, IL-6 and NO by activating TNF-α, IL-6 and iNOs gene expression, respectively, especially DGS2. Interestingly, DGS2 also possessed relatively high antioxidant activity. These results illustrate that, due to its cost-effectiveness and environmentally friendly features, SSF has significant potential as a commercially competitive source of natural products, including fungal polysaccharides, with immunomodulatory activity.
Extraction optimization, structural characterization of soluble dietary fiber from Morchella importuna, and its in vitro fermentation impact on gut microbiota and short-chain fatty acids

Abstract:Soluble dietary fiber was extracted from spent Morchella importuna by optimizing the extraction conditions using the response surface method. Response surface analysis results indicated that the extraction yield of MI-SDF reached up to 15.38 ± 0.16% under the optimized conditions with optimal lye concentration of 2.9%, extraction temperature of 71°C and extraction time of 2 h. The results of scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffraction analyses suggested that M . importuna -derived soluble dietary fiber (MI-SDF) exhibited the typical porous fiber structures, polysaccharide functional groups, and cellulose crystal structure. And MI-SDF was applicable to food processing at <250°C. During in vitro fecal fermentation, MI-SDF significantly increased the abundance and diversity of beneficial bacteria and decreased those of harmful bacteria. The levels of total and individual short-chain fatty acids were significantly increased. This study will provide valuable information for further investigation of MI-SDF. GRAPHICAL

Abstract:This study investigated the purification and characterization of a new immunomodulatory GlcNAc-containing polysaccharide (MIPB70-1) from Morchella importuna with molecular weights of 20.6 kDa. Structural analysis indicated that MIPB70-1 was composed of GlcNAc:Gal:Glc:Man with molar ratios of 1.00:7.16:5.54:5.61, and its primary structure was characterized as a repeating unit consisting of →6)-α-D-Glcp-(1→, α-D-GlcpNAc-(1→, α-D-Galp-(1→, β-D-Glcp-(1→, →6)-α-D-Manp-(1→, →4)-α-D-GlcpNAc-(1→, →4)-β-D-Glcp-(1→, →3,6)-α-D-Manp-(1→, →2)-α-D-Galp-(1→, →2,3,6)-α-D-Manp-(1→. Immunological assays indicated that MIPB70-1 enhanced the phagocytic function and promoted the secretion of nitric oxide (NO) as well as cytokines through targeting Toll-like receptor 4 (TLR4) on macrophage membrane and activating the downstream signaling pathways in RAW 264.7 cells. MIPB70-1 regulated mouse immunity to counteract the immune damage caused by the chemotherapy drug cyclophosphamide (CTX) in vivo. Furthermore, MIPB70-1 enhanced the anti-tumor activity of doxorubicin (DOX) and inhibited the growth of tumors, by immunomodulation in the orthotopic murine model of 4T1 breast cancer. These results demonstrate the potential of this GlcNAc-containing polysaccharide as an immune enhancer.

Abstract:A polysaccharide isolated from spores of the fungus, Ganoderma lucidum, was found to be a complex glucan. On the basis of compositional and methylation analyses, periodate oxidation, Smith degradation, 1D and 2D NMR, and ESIMS experiments of the native polysaccharide and its degraded products, the polysaccharide was shown to have a backbone of beta-(1-->3)-linked D-glucopyranosyl residues, with branches of mono-, di- and oligosaccharide side chains substituting at the C-6 of the glucosyl residues in the main chain. Conformational analysis in aqueous solution and immunological activities of the native and degraded glucans were also investigated. The results suggested that the degree of substitution on the main chain and the length of side chains may be very important factors in determining the conformation and the biological activities of beta-(1-->3)-linked glucans.

Comparative study on physicochemical properties and hypoglycemic activities of intracellular and extracellular polysaccharides from submerged fermentation of Morchella esculenta

Abstract:The structural characteristic, physicochemical properties and structure-hypoglycemic activity relationship of intracellular (IPS) and extracellular (EPS) from submerged fermentation of Morchella esculenta were systematically compared and assessed. Both IPS and EPS were neutral, with a triple-helical conformation, and composed of galactose, glucose and mannose monosaccharides in different molar ratios. The molecular weight and particle size of IPS were higher than those of EPS. FTIR and SEM showed that the main functional group absorption peak intensity, glycosidic bond type and surface morphology of the two polysaccharides differed. Analysis of rheological and thermal properties revealed that the viscosity of IPS was higher than that of EPS, while thermal stability of EPS was greater than that of IPS. Hypoglycemic activity analysis in vitro showed that both IPS and EPS were non-competitive inhibitors of α-amylase and α-glucosidase. EPS showed strong digestive enzyme inhibitory activity due to its higher sulphate content and molar ratio of galactose, lower Mw and particle size. Meanwhile, with its higher Mw and apparent viscosity, IPS showed stronger glucose adsorption capacity and glucose diffusion retardation. These results indicate that IPS and EPS differed considerably in structure and physicochemical properties, which ultimately led to differences in hypoglycemic activity. These results not only suggested that IPS and EPS has the potential to be functional foods or hypoglycemic drugs, but also provided a new target for the prevention and treatment of diabetes with natural polysaccharides.

Abstract:Sanghuang as a medicinal fungus in China has a history of more than 2000 years, and is known as the "forest gold". Most notably, the polysaccharides of Sanghuangporus sp. have attracted widespread attention due to their significant bioactivity in recent years. At present, extensive studies are being carried out on the extraction methods, structural characterization, and activity evaluation of polysaccharides. Here, we aimed to evaluate the structure and bioactivity of LEPS-1, an exopolysaccharide derived from the S. sanghuang JM-1 strain. The structure was elucidated by chromatography/spectral methods and hydrolyzation, and the solubility, the antioxidant activity, hypoglycemic activity and immunomodulatory activity were investigated. Results showed that LEPS-1 contained a →2)-α-Manp(1→6)-α-Galp(1→[2)-α-Manp(1→]n→2,6)-α-Manp(1→6,2)-α-Manp(1→3)-α-Manp(1→ backbone substituted at the O-6 and O-2 positions with side chains. These two branching fragments were β-Manp(1→. The molecular weight of LEPS-1 is 36.131 kDa. The results of biological activity analysis suggested that LEPS-1 was easily soluble in water, with reducing capability and DPPH radical scavenging capability. Furthermore, the IC50 values of LEPS-1 against α-amylase and α-glucosidase were 0.96 mg/mL and 1.92 mg/mL. LEPS-1 stimulated RAW264.7 cells to release NO, TNF-α and IL-6 with no cytotoxicity, showing potent potential for immunomodulatory activity. These findings describe a potential natural exopolysaccharide with medicinal value and a basis for the development of S. sanghuang exopolysaccharides.

Structure Identification of Two Polysaccharides from Morchella sextelata with Antioxidant Activity

Abstract:Mushrooms of the Morchella genus exhibit a variety of biological activities. Two polysaccharides (MSP1-1, 389.0 kDa; MSP1-2, 23.4 kDa) were isolated from Morchella sextelata by subcritical water extraction and column chromatography fractionation. Methylation and nuclear magnetic resonance analysis determined MSP1-1 as a glucan with a backbone of (1→4)-α-D-glucan branched at O-6, and MSP1-2 as a galactomannan with coextracted α-glucan. Light scattering analysis and transmission electron microscopy revealed that MSP1-1 possessed a random coil chain and that MSP1-2 had a network chain. This is the first time that a network structure has been observed in a polysaccharide from M. sextelata. Despite the differences in their chemical structures and conformations, both MSP1-1 and MSP1-2 possessed good thermal stability and showed antioxidant activity. This study provides fundamental data on the structure–activity relationships of M. sextelata polysaccharides.
