Abstract:The experiment of exo-ecosystem and the exploration of extraterrestrial habitability aims to explore the adaptation of terrestrial life in space conditions for the manned space program and the future interstellar migration, which shows great scientific significance and public interests. By our knowledge the early life on Earth, archaea and extremophile have the ability to adapt to extreme environmental conditions and can potentially habitat in extraterrestrial environments. Here we proposed a design and framework for the experiment on exo-ecosystem and extraterrestrial habitability. The conceptual approach involves building an ecosystem based on archaea and extremophiles in a simulated extraterrestrial environment, with a focus on assessing the exobiological potential and adaptability of terrestrial life forms in such conditions through controlled experiments. Specifically, we introduce the Chinese Exo-Ecosystem Space Experiment (CHEESE), which investigates the survivability and potential for sustained growth, reproduction, and ecological interactions of methanogens under simulated Mars and Moon environments using the China Space Station (CSS) as a platform. We highlight that the space station provides unique yet relatively comprehensive conditions for simulating extraterrestrial environments. In conclusion, space experiments involving exo-ecosystems could pave the way for long-term human habitation in space, ensuring our ability to sustain colonies and settlements beyond Earth while minimizing our ecological impact on celestial bodies.

Abstract:Space life science is a discipline dedicated to investigating the origin,evolution,and distri-bution of life under space environmental conditions,along with the phenomena and principles governing life.This field delves into the mechanisms of life's origin and early evolution from both theoretical and experimental perspectives,while also exploring fundamental scientific inquiries such as the habitability and forms of extraterrestrial life.With the implementation of major scientific projects such as the Chi-nese space station,the Chang'E lunar exploration program,and the Tianwen series,research on space life in China has witnessed rapid development.This paper provides an overview of the domestic and in-ternational trends in space life,identifies key areas for future development of space life science in China,optimizes disciplinary layouts,and aims to promote high-quality advancement in space life research.

Abstract:Unprecedented experimental conditions were provided for research in space biology following the completion of the Chinese Space Station. The next decade is predicted to witness considerable developments in this subject. Space cell culture is a crucial experimental technique in space biology. The Cell Tissue Culture Experiment Module (CTCEM) aboard the space station's Biotechnology Experiment Rack is customized equipment designed for the microgravity environment in space. It provides suitable culture conditions for cell growth, including temperature and CO2 concentration control, automatic liquid exchange, and automated observation via visible light microscopy, fluorescence microscopy, and laser confocal microscopy. The Tianzhou-5 Launch Cell Life Support Module (LCLSM) was developed to meet the requirements for transporting samples for space station cell experiments. This device can provide the required temperature, CO2 concentration, and nutrient solution replacement for cell experiment sample transportation. It also stores cells during ground transportation, launches, and in-orbit flights to ensure that they arrive at a space station with good physiological conditions. This article describes space cell bioreactors, the detailed functions and usage methods of CTCEM and LCLSM, and discusses the entire process of transporting cells to the space station and conducting space cell culture experiments through the TZ5 mission.

Abstract:An innovative Chinese space mission, the Earth 2.0 (ET) mission, is being developed to combine the transit and microlensing method together to search for Earth-sized exoplanets in the Galaxy, including the most precious ones-Earth 2.0s, i.e., habitable Earth-sized (0.8-1.25 Earth radii) planets orbiting solar type stars, cold and free-floating low-mass planets. ET's 6 transit telescopes will monitor a FoV of 500 square degrees (covering the Kepler field) continuously for at least four years and generate a huge database containing high-cadence and ultra-high photometry precision light curves of 1.2 million FGKM dwarfs. With such a high value database in hand, many unsolved issues in the exoplanet field and even stellar sciences will be well addressed. Besides looking for Earth 2.0s and constraining its occurrence rate, ET will be dedicated to map a much wider radius-period diagram of terrestrial-like exoplanets than ever and reveal how it depends on the stellar properties and environments. With the 4-yr legacy data of Kepler, ET will observe some planet systems for up to 8 years and catch additional components in a multi-planet system, e.g. cold Giant, cold sub-Earths, exomoons, exorings and even exocomets. Are exomoons and exocomets common in a planet system? What's the favorite number of planets in a multi-planet system? What's the most common orbital configuration of planet systems? With these new data, ET will deepen our understandings on how unique our Solar system is and how do multi-planet systems evolve. In addition to exoplanet sciences, ET's time series data will also benefit the studies in asteroseismology, archeology in the Galaxy, time-domain astrophysics and black hole science.

Abstract:The Earth 2.0 (ET) space mission has entered its phase B study in China. It seeks to understand how frequently habitable Earth-like planets orbit solar-type stars (Earth 2.0s), the formation and evolution of terrestrial-like planets, and the origin of free-floating planets. The final design of ET includes six 28 cm diameter transit telescope systems, each with a field of view of 550 square degrees, and one 35 cm diameter microlensing telescope with a field of view of 4 square degrees. In transit mode, ET will continuously monitor over 2 million FGKM dwarfs in the original Kepler field and its neighboring fields for four years. Simultaneously, in microlensing mode, it will observe over 30 million I < 20.5 stars in the Galactic bulge direction. Simulations indicate that ET mission could identify approximately 40,000 new planets, including about 4,000 terrestrial-like planets across a wide range of orbital periods and in the interstellar space, ~1000 microlensing planets, ~10 Earth 2.0s and around 25 free-floating Earth mass planets. Coordinated observations with ground-based KMTNet telescopes will enable the measurement of masses for ~300 microlensing planets, helping determine the mass distribution functions of free-floating planets and cold planets. ET will operate from the Earth-Sun L2 halo orbit with a designed lifetime exceeding 4 years. The phase B study involves detailed design and engineering development of the transit and microlensing telescopes. Updates on this mission study are reported.
Survival of soil microbial community exposed to hyper-gravity conditions

Abstract:Abstract Earth is the cradle of mankind, but it is impossible for human beings to live in the cradle forever. Sending soil microbial spores through space to foreign planets will be a likely initial process in planet colonization. Periods of hyper-gravity are likely to be a challenge for the candidate microorganisms during their interstellar transportation, raising questions about their survival rates and community-level responses. To address these questions, the impacts of hyper-gravity on soil microbial community composition and activity were tested by applying 1×g or 2500×g centrifugal force to soil for 6 days. The results indicated an increased diversity and absolute abundance of soil total bacterial community and a relatively stable active bacterial community under hyper-gravity condition. Besides, hyper-gravity had no observable effect on the relative abundance of soil microorganisms. These results suggest that soil microorganisms could survive during short periods of hyper-gravity. Our findings represent the first step towards a better understanding of the potential for survival of soil microbiomes during space travel and provide a basis for further interstellar soil research.

Abstract:A space mission called “Earth 2.0 (ET)” is being developed in China to address a few of fundamental questions in the exoplanet field: How frequently habitable Earth-like planets orbit solar type stars (Earth 2.0s)? How do terrestrial planets form and evolve? Where did floating planets come from? ET consists of six 30 cm diameter transit telescope systems with each field of view of 500 square degrees and one 35 cm diameter microlensing telescope with a field of view of 4 square degrees. The ET transit mode will monitor ~1.2M FGKM dwarfs in the original Kepler field and its neighboring fields continuously for four years while the microlensing mode monitors over 30M I< 20.6 stars in the Galactic bulge direction. ET will merge its photometry data with that from Kepler to increase the time baseline to 8 years. This enhances the transit signal-to-noise ratio, reduce false positives, and greatly increases the chance to discover Earth 2.0s. Simulations show that ET transit telescopes will be able to identify ~17 Earth 2.0s, about 4,900 Earth-sized terrestrial planets and about 29,000 new planets. In addition, ET will detect about 2,000 transit-timingvariation (TTV) planets and 700 of them will have mass and eccentricity measurements. The ET microlensing telescope will be able to identify over 1,000 microlensing planets. With simultaneous observations with the ground-based KMTNet telescopes, ET will be able to measure masses of over 300 microlensing planets and determine the mass distribution functions of free-floating planets and cold planets. ET will be operated at the Earth-Sun L2 orbit with a designed lifetime longer than 4 years.
Microbiome in a ground-based analog cabin of China Space Station during a 50-day human occupation

Abstract:Dead-corner areas in space station that untouched by the clean-up campaign often experience microorganisms outbreaks, but the microbiome of these areas has never been studied. In this study, the microbiome in a ground-based analog ``Tianhe'' core module of China Space Station was first investigated during a 50-day three-crew occupation. Dead-corner areas were receiving attention by adopting a new sampling method. Results indicate that the astronauts occupation did not affect the dominant bacteria community, but affected a small proportion. Due to the frequent activity of astronauts in the work and sleep areas, the biomarkers in these two areas are common human skin surface and gut microorganisms, respectively. For areas that astronaut rarely visits, the biomarkers in which are common environmental microbial groups. Fluorescence counting showed that 70.12-84.78% of bacteria were alive, with a quantity of 104-105 cells/100 cm2. With the occupation time extension, the number of microorganisms increased. At the same sampling time, there was no significant bioburden difference in various locations. The cultivable bioburden ranged from 101 to 104 colony forming unit (CFU)/100 cm2, which are the following eight genera Penicillium, Microsphaeropsis, Stachybotrys, Humicola, Cladosporium, Bacillus, Planomicrobium, and Acinetobacter. Chryseomicrobium genus may be a key focus for future microbial prevention and control work.

Abstract:The Earth 2.0 (ET) mission is a Chinese next-generation space mission to detect thousands of Earth-sized terrestrial planets, including habitable Earth-like planets orbiting solar type stars (Earth 2.0s), cold low-mass planets, and freefloating planets. To meet the scientific goals, the ET spacecraft will carry six 30 cm diameter transit telescopes with each field of view of 500 square degrees, and one 35 cm diameter microlensing telescope with a field of view of 4 square degrees, monitor ~1.2M FGKM dwarfs in the original Kepler field and its neighboring fields continuously while monitoring over 30M stars in the Galactic bulge direction. The high precision transit observations require high photometry precision and pointing stability, which is the key drive for the ET spacecraft design. In this paper, details of the overall mission modeling and analysis will be presented. The spacecraft orbit, pointing strategy, stability requirements are presented, as well as the space-ground communication analysis. The ET spacecraft adopts an ultra-high photometry precision & high stable platform, largely inherited from other space science missions. The preliminary design of spacecraft which meets mission requirements is introduced, including the spacecraft overall configuration, observation modes, avionics architecture and development plan, which pays great attention to the pointing stability and huge volume science telemetry download.
The Tianlin Mission: a 6m UV/Opt/IR space telescope to explore the habitable worlds and the universe

Abstract:[Abridged] It is expected that the ongoing and future space-borne planet survey missions including TESS, PLATO, and Earth 2.0 will detect thousands of small to medium-sized planets via the transit technique, including over a hundred habitable terrestrial rocky planets. To conduct a detailed study of these terrestrial planets, particularly the cool ones with wide orbits, the exoplanet community has proposed various follow-up missions. The currently proposed ESA mission ARIEL is capable of characterization of planets down to warm super-Earths mainly using transmission spectroscopy. The NASA 6m UV/Opt/NIR mission proposed in the Astro2020 Decadal Survey may further tackle down to habitable rocky planets, and is expected to launch around 2045. In the meanwhile, China is funding a concept study of a 6-m class space telescope named Tianlin (A UV/Opt/NIR Large Aperture Space Telescope) that aims to start its operation within the next 10-15 years and last for 5+ years. Tianlin will be primarily aimed to the discovery and characterization of rocky planets in the habitable zones (HZ) around nearby stars and to search for potential biosignatures mainly using the direct imaging method. Transmission and emission spectroscopy at moderate to high resolution will be carried out as well on a population of exoplanets to strengthen the understanding of the formation and evolution of exoplanets. It will also carry out in-depth studies of the cosmic web and early galaxies, and constrain the nature of the dark matter and dark energy. We describe briefly the primary scientific motivations and main technical considerations based on our preliminary simulation results. We find that a monolithic off-axis space telescope with a primary mirror diameter larger than 6m equipped with a high contrast chronograph can identify water in the atmosphere of a habitable-zone Earth-like planet around a Sun-like star.

Abstract:Extensive investigations in outer space have revealed not only how life adapts to the space environment, but also that interesting biophysical phenomena occur. These phenomena affect human health and other life forms (animals, plants, bacteria, and fungi), and to ensure the safety of future human space exploration need to be further investigated. This calls for joint research efforts between biologists and physicists, as these phenomena present cross-disciplinary barriers. Various national organizations provide useful forums for bridging this gap. Additional discussion avenues and database resources are helpful for facilitating the interdisciplinary investigations of these phenomena. In this paper, we present the newly established Space Life Investigation Database (SpaceLID, https://bidd.group/spacelid/ ) which provides information about biophysical phenomena occurring in space. Examples obtained using the database are given while discussing the underlying causes of these phenomena and their implications for the physiology and health of life in space.

Abstract:In the era of deep space exploration, extremophile research represents a key area of research w.r.t space survival. This review thus delves into the intriguing realm of 'Space and Astro Microbiology', providing insights into microbial survival, resilience, and behavioral adaptations in space-like environments. This discussion encompasses the modified behavior of extremophilic microorganisms, influencing virulence, stress resistance, and gene expression. It then shifts to recent studies on the International Space Station and simulated microgravity, revealing microbial responses that impact drug susceptibility, antibiotic resistance, and its commercial implications. The review then transitions into Astro microbiology, exploring the possibilities of interplanetary transit, lithopanspermia, and terraforming. Debates on life's origin and recent Martian meteorite discoveries are noted. We also discuss Proactive Inoculation Protocols for selecting adaptable microorganisms as terraforming pioneers. The discussion concludes with a note on microbes' role as bioengineers in bioregenerative life support systems, in recycling organic waste for sustainable space travel; and in promoting optimal plant growth to prepare Martian and lunar basalt. This piece emphasizes the transformative impact of microbes on the future of space exploration.

Abstract:With the construction of the International Space Station, humans have been continuously living and working in space for 22 years. Microbial studies in space and other extreme environments on Earth have shown the ability for bacteria and fungi to adapt and change compared to "normal" conditions. Some of these changes, like biofilm formation, can impact astronaut health and spacecraft integrity in a negative way, while others, such as a propensity for plastic degradation, can promote self-sufficiency and sustainability in space. With the next era of space exploration upon us, which will see crewed missions to the Moon and Mars in the next 10 years, incorporating microbiology research into planning, decision-making, and mission design will be paramount to ensuring success of these long-duration missions. These can include astronaut microbiome studies to protect against infections, immune system dysfunction and bone deterioration, or biological in situ resource utilization (bISRU) studies that incorporate microbes to act as radiation shields, create electricity and establish robust plant habitats for fresh food and recycling of waste. In this review, information will be presented on the beneficial use of microbes in bioregenerative life support systems, their applicability to bISRU, and their capability to be genetically engineered for biotechnological space applications. In addition, we discuss the negative effect microbes and microbial communities may have on long-duration space travel and provide mitigation strategies to reduce their impact. Utilizing the benefits of microbes, while understanding their limitations, will help us explore deeper into space and develop sustainable human habitats on the Moon, Mars and beyond.

Abstract:Giant planets like Jupiter and Saturn, play important roles in the formation and habitability of Earth-like planets. The detection of solar system analogs that have multiple cold giant planets is essential for our understanding of planet habitability and planet formation. Although transit surveys such as Kepler and TESS have discovered thousands of exoplanets, these missions are not sensitive to long period planets due to their limited observation baseline. The Tianyu project, comprising two 1-meter telescopes (Tianyu-I and II), is designed to detect transiting cold giant planets in order to find solar system analogs. Featuring a large field of view and equipped with a high-speed CMOS camera, Tianyu-I will perform a high-precision photometric survey of about 100 million stars, measuring light curves at hour-long cadence. The candidates found by Tianyu-I will be confirmed by Tianyu-II and other surveys and follow-up facilities through multi-band photometry, spectroscopy, and high resolution imaging. Tianyu telescopes will be situated at an elevation about 4000 meters in Lenghu, China. With a photometric precision of 1% for stars with V < 18 mag, Tianyu is expected to find more than 300 transiting exoplanets, including about 12 cold giant planets, over five years. A five-year survey of Tianyu would discover 1-2 solar system analogs. Moreover, Tianyu is also designed for non-exoplanetary exploration, incorporating multiple survey modes covering timescales from sub-seconds to months, with a particular emphasis on events occurring within the sub-second to hour range. It excels in observing areas such as infant supernovae, rare variable stars and binaries, tidal disruption events, Be stars, cometary activities, and interstellar objects. These discoveries not only enhance our comprehension of the universe but also offer compelling opportunities for public engagement in scientific exploration.

Abstract:BIOMEX (BIOlogy and Mars EXperiment) is an ESA/Roscosmos space exposure experiment housed within the exposure facility EXPOSE-R2 outside the Zvezda module on the International Space Station (ISS). The design of the multiuser facility supports-among others-the BIOMEX investigations into the stability and level of degradation of space-exposed biosignatures such as pigments, secondary metabolites, and cell surfaces in contact with a terrestrial and Mars analog mineral environment. In parallel, analysis on the viability of the investigated organisms has provided relevant data for evaluation of the habitability of Mars, for the limits of life, and for the likelihood of an interplanetary transfer of life (theory of lithopanspermia). In this project, lichens, archaea, bacteria, cyanobacteria, snow/permafrost algae, meristematic black fungi, and bryophytes from alpine and polar habitats were embedded, grown, and cultured on a mixture of martian and lunar regolith analogs or other terrestrial minerals. The organisms and regolith analogs and terrestrial mineral mixtures were then exposed to space and to simulated Mars-like conditions by way of the EXPOSE-R2 facility. In this special issue, we present the first set of data obtained in reference to our investigation into the habitability of Mars and limits of life. This project was initiated and implemented by the BIOMEX group, an international and interdisciplinary consortium of 30 institutes in 12 countries on 3 continents. Preflight tests for sample selection, results from ground-based simulation experiments, and the space experiments themselves are presented and include a complete overview of the scientific processes required for this space experiment and postflight analysis. The presented BIOMEX concept could be scaled up to future exposure experiments on the Moon and will serve as a pretest in low Earth orbit.

Abstract:Biological experiments performed in space crafts like space stations, space shuttles, and recoverable satellites has enabled extensive spaceflight life investigations (SLIs). In particular, SLIs have revealed distinguished space effects on microbial growth, survival, metabolite production, biofilm formation, virulence development and drug resistant mutations. These provide unique perspectives to ground-based microbiology and new opportunities for industrial pharmaceutical and metabolite productions. SLIs are with specialized experimental setups, analysis methods and research outcomes, which can be accessed by established databases National Aeronautics and Space Administration (NASA) Life Science Data Archive, Erasmus Experiment Archive, and NASA GeneLab. The increasing research across diverse fields may be better facilitated by databases of convenient search facilities and categorized presentation of comprehensive contents. We therefore developed the Space Life Investigation Database (SpaceLID) , which collected SLIs from published academic papers. Currently, this database provides detailed menu search facilities and categorized contents about the studied phenomena, materials, experimental procedures, analysis methods, and research outcomes of 448 SLIs of 90 species (microbial, plant, animal, human), 81 foods and 106 pharmaceuticals, including 232 SLIs not covered by the established databases. The potential applications of SpaceLID are illustrated by the examples of published experimental design and bioinformatic analysis of spaceflight microbial phenomena.

Abstract:Arguably one of the most important discoveries in the history of human being is the detection of a planet orbiting a main sequence star beyond the solar system.By the time Science celebrated its 125th anniversary,～150 exoplanets have been discovered.However,there was no known habitable exoplanet.Neither was it clear whether there is habitable environment in the solar system beyond the Earth.Thus the possibility to "listen" to radio signals emitted by extraterrestrial intelligence was a focus of the 1 1th science question "Are we alone in the universe?".We equally care about the 56th question "Is/Was there life elsewhere in the solar system?".In this essay we discuss the following 4 aspects:(1) non-carbon-based life;(2) habitability of solar system planets/moons;(3) habitability of exoplanets;(4)prospects of detecting habitable environments and extraterrestrial life.

Abstract:The search for life on planets outside our solar system has largely been the province of the astrophysics community until recently. A major development since the NASA Astrobiology Strategy 2015 document (AS15) has been the integration of other NASA science disciplines (planetary science, heliophysics, Earth science) with ongoing exoplanet research in astrophysics. The NASA Nexus for Exoplanet System Science (NExSS) provides a forum for scientists to collaborate across disciplines to accelerate progress in the search for life elsewhere. Here we describe recent developments in these other disciplines, with a focus on exoplanet properties and environments, and the prospects for future progress that will be achieved by integrating emerging knowledge from astrophysics with insights from these fields.
