Abstract:Cu-II-catalysed diazo transfer and Cu-I-catalysed azide-alkyne 1,3-dipolar cycloaddition (CuAAC) "click chemistry" were used to synthesis C7N aminocyclitol-derivatised 1,2,3-triazoles. In the course of this work, the -N=N- moiety was transferred onto C7N aminocyclitols such as validamine, valienamine and valiolamine by employing imidazole-1-sulfonyl azide as the diazo transfer reagent with catalysis by Cu-II, Zn-II and Ni-II, in moderate to good yields. The obtained azidocyclitols were coupled with various terminal alkynes under modified Meldal's conditions with good to excellent yields. The stereo- and regiochemistry of the products were confirmed by 2D-NMR (NOESY and HMBC). One-pot syntheses of the corresponding 1,2,3-triazoles, as safer and more efficient procedures, were also investigated and gave moderate to good yields.

Abstract:Incorporation of a trifluoromethyl group with 1,2,3-triazoles motifs was described. We explored a click reaction approach for regioselective synthesis of 1-susbstituted-4-trifluoromethyl-1,2,3-triazoles in which 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) reacts with commercial 2-bromo-3,3,3-trifluoropropene (BTP) to form 3,3,3-trifloropropyne (TFP) in situ. Arising from merits associated with the availability and stability of BTP, and the high efficiencies of CuI/1,10-Phenanthroline (Phen)-catalyzed cycloaddition reactions of azides with alkynes, this readily performed click process takes place to form the target 1,2,3-triazoles in high yields, and with a wide azide substrate scope. The potential value of this protocol was demonstrated by its application to a gram-scale reaction.

Abstract:An air-stable supported Cu(I) catalyst, CuI@PS-Phen, was designed and synthesized. CuI@PS-Phen can efficiently catalyze the click polymerization of diynes a and diazides b to produce soluble and thermally stable polytriazoles with high molecular weights ( M w up to 30800), and low copper residue content (down to 190 ppm) in high yields (up to 94.2%) under mild reaction conditions without the exclusion of oxygen.

Abstract:The Cu(I)-catalyzed azide-alkyne click polymerization has become a powerful tool for the preparation of functional polytriazoles. However, copper residues in the products could harm the performance of optoelectronic devices and be cytotoxic in biological applications. The complete removal of these residues after reaction from the polymers is therefore necessary but difficult. The metal-free azide-alkyne click polymerization (MFCP) provides a solution to this problem. Currently, the MFCP is facilitated by activated alkynes, and no activated comonomer, i.e. azide, has been reported. In this paper, an activated azide, 4,4'-diazidoperfluorobenzophenone , was designed and synthesized. Compared to the ordinary azide, such a monomer can remarkably accelerate the MFCP with alkynes. Thus, a new MFCP of activated azide and alkyne was established, and polytriazoles with high regioregularity (fraction of 1,4-isomer higher than 82%) and satisfactory molecular weights (M-w up to 34 000) were produced in high yields (up to 95.1%).

Abstract:Click chemistry focuses on the development of highly selective reactions using simple precursors for the exquisite synthesis of molecules. Undisputedly, the Cu-I-catalyzed azide-alkyne cycloaddition (CuAAC) is one of the most valuable examples of click chemistry, but it suffers from some limitations as it requires additional reducing agents and ligands as well as cytotoxic copper. Here, we demonstrate a novel strategy for the azide-alkyne cycloaddition reaction that involves a photoredox electron-transfer radical mechanism instead of the traditional metal-catalyzed coordination process. This newly developed photocatalyzed azide-alkyne cycloaddition reaction can be performed under mild conditions at room temperature in the presence of air and visible light and shows good functional group tolerance, excellent atom economy, high yields of up to 99 %, and absolute regioselectivity, affording a variety of 1,4-disubstituted 1,2,3-triazole derivatives, including bioactive molecules and pharmaceuticals. The use of a recyclable photocatalyst, solar energy, and water as solvent makes this photocatalytic system sustainable and environmentally friendly. Moreover, the azide-alkyne cycloaddition reaction could be photocatalyzed in the presence of a metal-free catalyst with excellent regioselectivity, which represents an important development for click chemistry and should find versatile applications in organic synthesis, chemical biology, and materials science.

These compounds were prepared by the reaction of 4-propargyl substituted 5-iodo-6-methyl-2(methylthio) pyrimidine with aryl and benzyl azides via copper-catalyzed azide-alkyne cycloaddition reactions in the presence of sodium ascorbate, as a reducing agent. Furthermore, the synthesized compounds were screened against the two bacterial strains pseudomonas aeruginosa and saphylococcus aureus .

Abstract:The copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is regarded as a prime example of "click chemistry", but the asymmetric click cycloaddition of internal alkynes still remains challenging. A new asymmetric Rh-catalyzed click cycloaddition of N-alkynylindoles with azides was developed, providing atroposelective access to C-N axially chiral triazolyl indoles, a new type of heterobiaryl, with excellent yields and enantioselectivity. This asymmetric approach is efficient, mild, robust and atom-economic, and features very broad substrate scope with easily available Tol-BINAP ligands.

Abstract:The synthesis of 1-monosubstituted aryl 1,2,3-triazoles was achieved using calcium carbide as a source of acetylene. The copper-catalyzed 1,3-dipolar cycloaddition reactions were carried out without nitrogen protection and in a MeCN-H2O mixture. The yields ranged from moderate to excellent. The reaction conditions were found to be successful for aryl azide reactants, including analogues with various functionalities.

Abstract:1,2,3-triazole is a vital structural motif of various drugs and therapeutic leads, as well as a linker for bioconjugation and molecular recognition. Cu-catalysed click cycloaddition of azides with terminal alkynes (CuAAc) is an important reaction to construct the triazole core. In recent years, various decarboxylative click strategies utilizing alkynoic acids as stable surrogates for low boiling or gaseous alkynes have been developed. For instance, propiolic acid, which is easy to transport, is a safe alternative for flammable gaseous acetylene. In this review article, we have covered the recent development in the decarboxylative click cycloaddition of alkynoic acids with azides leading to the synthesis of diversely substituted triazoles, including monosubstituted, 1,4-disubstituted and fully substituted 1,2,3-triazoles. Various aspects such as mechanistic insights and optimization conditions/role of catalyst are highlighted.

Abstract:Chiral 1,2,3-triazoles are highly attractive motifs in various fields. However, achieving catalytic asymmetric click reactions of azides and alkynes for chiral triazole synthesis remains a significant challenge, mainly due to the limited catalytic systems and substrate scope. Herein, we report an enantioselective azidation/click cascade reaction of N-propargyl-β-ketoamides with a readily available and potent azido transfer reagent via copper catalysis, which affords a variety of chiral 1,2,3-triazoles with up to 99% yield and 95% ee under mild conditions. Notably, chiral 1,5-disubstituted triazoles that have not been accessed by previous asymmetric click reactions are also prepared with good functional group tolerance.

Abstract:An effective strategy for constructing substituted 1-benzyl-1 H -1,2,3-triazoles was developed through click reactions of benzylic halides with sodium azide and calcium carbide as sources of nitrogen and acetylene, respectively. Abstract:Carbonyl-containing 1,4,5-trisubstituted- and 1,4-disubstituted-1,2,3-triazoles are well-known for their wide range of applications in pharmaceutical and medicinal chemistry, but their high-yielding metal-free synthesis has always remained challenging, as no comprehensive protocol has been outlined to date. Owing to their structural and medicinal importance, herein, we synthesized various carbonyl-containing 1,4,5-trisubstituted- and 1,4-disubstituted-1,2,3-triazoles and unsymmetrical 4,5'-bitriazoles with high yields and chemo-/regioselectivity from the library of 2,4-diketoesters and azides in a sequential one-pot manner through the combination of organocatalytic enolization, in situ [3 + 2]-cycloaddition, and hydrolysis reactions. The commercial availability of the starting materials/catalysts, diverse substrate scope, performance in a one-pot manner, chemo-/regioselectivity of organo-click reaction, quick synthesis of unsymmetrical 4,5'-bitriazoles, a large number of synthetic applications, and numerous medicinal applications of carbonyl-containing 1,2,3-triazoles are the key attractions of this metal-free organo-click work.

Abstract:In the last two decades, click chemistry has become a modular synthetic procedure to assemble new molecular structures. It is a powerful methodology that relies on the construction of carbon-heteroatom bonds of a variety of reactants. In modern synthetic chemistry, it has been applied in a large number of applications, from pharmaceutical to material science. The copper-catalyzed 1,2,3-triazole preparation, reacting organic azides with alkynes, has become the star of click chemistry due to its reliability and biocompatibility. As a consequence, this reaction has found many applications in diverse areas such as bioconjugation, material science, and drug discovery. In order to understand the effect of copper catalysts and ligands in click chemistry, it is important to understand the structural and mechanistic aspects involved. In this review, several physicochemical aspects of click chemistry are discussed. First, the mechanisms and intermediates involved in the preparation of 1,2,3-triazoles. Second, the different types of copper catalysts are used to perform the reaction regioselectively. The last section shows the structure and characteristics of effective ligands utilized to improve click chemistry under different experimental conditions.

Abstract:A Cu (I)-catalyzed three-component click reaction of alkyl bromide, terminal alkyne and sodium azide was developed in micro flow reactor. Sodium ascorbate was added as additive to prevent the oxidative coupling of desired product with terminal alkyne. Under optimized condition, various 1,4-disubstituted-1,2,3-triazoles were obtained in 85-95% yield with exclusive 1,4-regioisomeric selectivity. The present smooth procedure greatly accelerated the reaction due to the excellent mixing and mass transfer in micro flow system.

Abstract:Cu(I)-catalyzed alkyne–azide ‘click’ cycloaddition (CuAAC) is an important ‘‘click chemistry’’ reaction that is widely known in materials science, chemical biology, and pharmaceutical chemistry. The CuAAC reaction of terminal alkynes affords an efficient and mild production of triazolic 1,4-disubstituted compounds. In this work, a green and valuable method was introduced for the synthesis of the category of different new 1,4-disubstituted 1,2,3-triazole swapped with a 2-amino-4,8-dihydropyrano[3,2-b]pyran-3-cyano moiety. These triazolic derivatives were produced by treatment of various 2-amino-6-(azidomethyl)-4,8-dihydropyrano[3,2-b]pyran-3-carbonitriles with phenylacetylene in the presence of CuI as a catalyst with excellent yields (because CuAAC is selective to 1,4-disubstituted triazole derivatives) in a green solvent (ethanol/water). All structures were evaluated by 13C, 1H NMR, and FT-IR spectroscopy and a compound was analyzed by crystallography (X-ray) technique.

Abstract:A facile, high-yielding and straightforward methodology for the copper-catalyzed synthesis of 1,4-disubstituted 1,2,3-triazoles in water, using in situ prepared copper nanoparticles (NPs) on modified poly(styrene-co-maleic anhydride) [SMA] catalyst, is reported. The polymer support was easily prepared from the reaction of SMA with 4-aminopyridine and subsequently underwent reaction with CuI NPs. The catalyst was applied for the preparation of triazoles under air, followed by chromatographic separation of the products. The polymer-supported catalyst not only showed high catalytic activity but also showed high 1,4-regioselectivity for the [3 + 2] Huisgen cycloaddition in water as solvent. The products were obtained in good to excellent yields in all cases. The catalyst can be used without pre-activation and reloaded for at least five runs without significant decrease in its activity. The catalyst was characterized by SEM, energy dispersive spectroscopy analysis of X-rays, and inductively coupled plasma.

Abstract:Enzyme-catalyzed novel protocols for the regioselective construction of fully substituted 1,2,3-triazoles by employing 2-azido-1,3,5-triazine (ADT) as a 1,3-dipole for the cycloaddition reaction with the activated alkene in an aqueous medium have been developed. Various 4-heterosubstituted-1,2,3-triazoles were readily assembled in good to excellent yields with high regioselectivity. This reaction also features wide substrate scope, strong functional group tolerance, gram-scale synthesis, and an environmentally friendly process.

Abstract:Introduction: An effective Cu-complex, [Cu(NH3)4SO4 • H2O] was prepared conveniently from the inexpensive and easily available starting reagents in a simple route. Materials and methods: Excellent reactivity of the catalyst was observed towards two competent clickcycloadditions: (a) oxidative cycloaddition of azides with electron-poor olefins and (b) one-pot cycloaddition of alkynes with boronic acid and sodium azide under "click-appropriate" conditions. Results: No external oxidant, short reaction time, high product yield, wide substrate scope, and aqueous solvent media make the azide-olefin cycloaddition approach a greener route in contrast to the reported methods. Conclusion: The newly developed mild, green, and rapid three-component strategy shows product diversity with superb yields at room temperature by reducing the synthetic process time and using only 1 mol % of the synthesized copper complex.
