Abstract:Ultrafast real-time optical imaging is an indispensable tool for studying dynamical events such as shock waves, chemical dynamics in living cells, neural activity, laser surgery and microfluidics. However, conventional CCDs (charge-coupled devices) and their complementary metal-oxide-semiconductor (CMOS) counterparts are incapable of capturing fast dynamical processes with high sensitivity and resolution. This is due in part to a technological limitation-it takes time to read out the data from sensor arrays. Also, there is the fundamental compromise between sensitivity and frame rate; at high frame rates, fewer photons are collected during each frame-a problem that affects nearly all optical imaging systems. Here we report an imaging method that overcomes these limitations and offers frame rates that are at least 1,000 times faster than those of conventional CCDs. Our technique maps a two-dimensional (2D) image into a serial time-domain data stream and simultaneously amplifies the image in the optical domain. We capture an entire 2D image using a single-pixel photodetector and achieve a net image amplification of 25 dB (a factor of 316). This overcomes the compromise between sensitivity and frame rate without resorting to cooling and high-intensity illumination. As a proof of concept, we perform continuous real-time imaging at a frame speed of 163 ns (a frame rate of 6.1 MHz) and a shutter speed of 440 ps. We also demonstrate real-time imaging of microfluidic flow and phase-explosion effects that occur during laser ablation.

This work paves the way towards a fully-digital microscope allowing real-time, in-vivo, quantitative and deep inspection of tissues.

Abstract:SIGNIFICANCE: High-speed 3D imaging methods have been playing crucial roles in many biological discoveries.AIM: We present a hybrid light-field imaging system and image processing algorithm that can visualize high-speed biological events.APPROACH: The hybrid light-field imaging system uses the selective plane optical illumination, which simultaneously records a high-resolution 2D image and a low-resolution 4D light-field image. The high-resolution 4D light-field image is obtained by applying the hybrid algorithm derived from the deconvolution and phase retrieval methods.RESULTS: High-resolution 3D imaging at a speed of 100-s volumes per second over an imaging field of 250 × 250 × 80 μm3 in the x, y, and z axis, respectively, is achieved with a 2.5 times enhancement in lateral resolution over the entire imaging field compared with standard light-field systems. In comparison to the deconvolution algorithm, the hybrid algorithm addresses the artifact issue at the focal plane and reduces the computation time by a factor of 4.CONCLUSIONS: The new hybrid light-field imaging method realizes high-resolution and ultrafast 3D imaging with a compact setup and simple algorithm, which may help discover important applications in biophotonics to visualize high-speed biological events.

Abstract:Many problems in fluid mechanics require single-shot 3D measurements of fluid flows, but are limited by available techniques. Here, we design and build a novel flexible high-speed two-color scanning volumetric laser-induced fluorescence (H2C-SVLIF) technique. The technique is readily adaptable to a range of temporal and spatial resolutions, rendering it easily applicable to a wide spectrum of experiments. The core equipment consists of a single monochrome high-speed camera and a pair of ND: YAG lasers pulsing at different wavelengths. The use of a single camera for direct 3D imaging eliminates the need for complex volume reconstruction algorithms and easily allows for the correction of distortion defects. Motivated by the large data loads that result from high-speed imaging techniques, we develop a custom, open-source, software package, which allows for real time playback with correction of perspective defects while simultaneously overlaying arbitrary 3D data. The technique is capable of simultaneous measurement of 3D velocity fields and a secondary tracer in the flow. To showcase the flexibility and adaptability of our technique, we present a set of experiments: (1) the flow past a sphere, and (2) vortices embedded in laminar pipe flow. In the first experiment, two channel measurements are taken at a resolution of 512 × 512 × 512 with volume rates of 65.1 Hz. In the second experiment, a single-color SVLIF system is integrated on a moving stage, providing imaging at 1280 × 304 × 256 with volume rates of 34.8 Hz. Although this second experiment is only single channel, it uses identical software and much of the same hardware to demonstrate the extraction of multiple information channels from single channel volumetric images.

Abstract:A central quest in optics is to rapidly extract quantitative information from a sample. Existing topographical imaging tools allow non-contact and three-dimensional measurements at the micro and nanoscales and are essential in applications including precision engineering and optical quality control. However, these techniques involve acquiring a focal stack of images, a time-consuming process that prevents measurement of moving samples. Here, we propose a method for increasing the speed of topographic imaging by orders of magnitude. Our approach involves collecting a reduced set of images, each integrated during the full focal scan, whilst the illumination is synchronously modulated during exposure. By properly designing the modulation sequence for each image, unambiguous reconstruction of the object height map is achieved using far fewer images than conventional methods. We describe the theoretical foundations of our technique, characterise its performance, and demonstrate sub-micrometric topographic imaging over 100 μm range of static and dynamic systems at rates as high as 67 topographies per second, limited by the camera frame rate. The high speed of the technique and its ease of implementation could enable a paradigm shift in optical metrology, allowing the real-time characterisation of large or rapidly moving samples.

Abstract:Optical imaging through complex scattering media is one of the major technical challenges with important applications in many research fields, ranging from biomedical imaging, astronomical telescopy, and spatially multiplex optical communications. Although various approaches for imaging though turbid layer have been recently proposed, they had been limited to two-dimensional imaging. Here we propose and experimentally demonstrate an approach for three-dimensional single-shot imaging of objects hidden behind an opaque scattering layer. We demonstrate that under suitable conditions, it is possible to perform the 3D imaging to reconstruct the complex amplitude of objects situated at different depths.

Abstract:Optical clearing is a versatile approach to improve imaging quality and depth of optical microscopy by reducing scattered light. However, conventional optical clearing methods are restricted in the efficiency-first applications due to unsatisfied time consumption, irreversible tissue deformation, and fluorescence quenching. Here, we developed an ultrafast optical clearing method (FOCM) with simple protocols and common reagents to overcome these limitations. The results show that FOCM can rapidly clarify 300-µm-thick brain slices within 2 min. Besides, the tissue linear expansion can be well controlled by only a 2.12% increase, meanwhile the fluorescence signals of GFP can be preserved up to 86% even after 11 d. By using FOCM, we successfully built the detailed 3D nerve cells model and showed the connection between neuron, astrocyte, and blood vessel. When applied to 3D imaging analysis, we found that the foot shock and morphine stimulation induced distinct c-fos pattern in the paraventricular nucleus of the hypothalamus (PVH). Therefore, FOCM has the potential to be a widely used sample mounting media for biological optical imaging.

Abstract:Many phenomena of interest in nature and industry occur rapidly and are difficult and cost-prohibitive to visualize properly without specialized cameras. Here we describe in detail the Virtual Frame Technique (VFT), a simple, useful, and accessible form of compressed sensing that increases the frame acquisition rate of any camera by several orders of magnitude by leveraging its dynamic range. VFT is a powerful tool for capturing rapid phenomenon where the dynamics facilitate a transition between two states, and are thus binary. The advantages of VFT are demonstrated by examining such dynamics in five physical processes at unprecedented rates and spatial resolution: fracture of an elastic solid, wetting of a solid surface, rapid fingerprint reading, peeling of adhesive tape, and impact of an elastic hemisphere on a hard surface. We show that the performance of the VFT exceeds that of any commercial high speed camera not only in rate of imaging but also in field of view, achieving a 65MHz frame rate at 4MPx resolution. Finally, we discuss the performance of the VFT with several commercially available conventional and high-speed cameras. In principle, modern cell phones can achieve imaging rates of over a million frames per second using the VFT.

Abstract:The work presents a new non‐invasive way to image dynamic events occurring deep within rapidly decorrelating, diffusely scattering media. By combining a novel single‐photon avalanche diode array with a custom‐designed fiber bundle probe and a new data‐driven approach, the work demonstrates the ability to reconstruct images of transient dynamic events beneath decorrelating tissue phantoms. Noninvasive optical imaging through dynamic scattering media has numerous important biomedical applications but still remains a challenging task. While standard diffuse imaging methods measure optical absorption or fluorescent emission, it is also well‐established that the temporal correlation of scattered coherent light diffuses through tissue much like optical intensity. Few works to date, however, have aimed to experimentally measure and process such temporal correlation data to demonstrate deep‐tissue video reconstruction of decorrelation dynamics. In this work, a single‐photon avalanche diode array camera is utilized to simultaneously monitor the temporal dynamics of speckle fluctuations at the single‐photon level from 12 different phantom tissue surface locations delivered via a customized fiber bundle array. Then a deep neural network is applied to convert the acquired single‐photon measurements into video of scattering dynamics beneath rapidly decorrelating tissue phantoms. The ability to reconstruct images of transient (0.1–0.4 s) dynamic events occurring up to 8 mm beneath a decorrelating tissue phantom with millimeter‐scale resolution is demonstrated, and it is highlighted how the model can flexibly extend to monitor flow speed within buried phantom vessels.

Abstract:Light transport has been analyzed extensively, in both the primal domain and the frequency domain. Frequency analyses often provide intuition regarding effects introduced by light propagation and interaction with optical elements; such analyses encourage optimal designs of computational cameras that efficiently capture tailored visual information. However, previous analyses have relied on instantaneous propagation of light, so that the measurement of the time dynamics of light–scene interaction, and any resulting information transfer, is precluded. In this paper, we relax the common assumption that the speed of light is infinite. We analyze free space light propagation in the frequency domain considering spatial, temporal, and angular light variation. Using this analysis, we derive analytic expressions for information transfer between these dimensions and show how this transfer can be exploited for designing a new lensless imaging system. With our frequency analysis, we also derive performance bounds for the proposed computational camera architecture and provide a mathematical framework that will also be useful for future ultra-fast computational imaging systems.
Single picture single photon single pixel 3D imaging through unknown thick scattering medium

Abstract:Imaging through thick scattering media presents significant challenges, particularly for three-dimensional (3D) applications. This manuscript demonstrates a novel scheme for single-image-enabled 3D imaging through such media, treating the scattering medium as a lens. This approach captures a comprehensive image containing information about objects hidden at various depths. By leveraging depth from focus and the reduced thickness of the scattering medium for single-pixel imaging, the proposed method ensures robust 3D imaging capabilities. We develop both traditional metric-based and deep learning-based methods to extract depth information for each pixel, allowing us to explore the locations of both positive and negative objects, whether shallow or deep. Remarkably, this scheme enables the simultaneous 3D reconstruction of targets concealed within the scattering medium. Specifically, we successfully reconstructed targets buried at depths of 5 mm and 30 mm within a total medium thickness of 60 mm. Additionally, we can effectively distinguish targets at three different depths. Notably, this scheme requires no prior knowledge of the scattering medium, no invasive procedures, reference measurements, or calibration.

Fourier synthesis optical diffraction tomography for kilohertz rate volumetric imaging

Abstract:Many biological and soft matter processes occur at high speeds in complex 3D environments, and developing imaging techniques capable of elucidating their dynamics is an outstanding experimental challenge. Here, we introduce Fourier Synthesis Optical Diffraction Tomography (FS-ODT), a novel approach for high-speed quantitative phase imaging capable of recording the 3D refractive index at kilohertz rates. FS-ODT introduces new pattern generation and inverse computational strategies that multiplex tens of illumination angles in a single tomogram, dramatically increasing the volumetric imaging rate. We validate FS-ODT performance by imaging samples of known composition and accurately recovering the refractive index for increasing pattern complexity. We further demonstrate the capabilities of FS-ODT for probing complex systems by studying the hindered diffusion of colloids in solution and the motility of single-cellular bacterial swimmers. We believe that FS-ODT is a promising approach for unlocking challenging imaging regimes in biophysics and soft matter that have been little explored, including understanding the physical interactions of colloids and microswimmers with their viscous 3D environment and the interplay between these stimuli and the molecular response of biological systems.

Abstract:Three-dimensional single-pixel imaging (3D SPI) has become an attractive imaging modality for both biomedical research and optical sensing. 3D-SPI techniques generally depend on time-of-flight or stereovision principle to extract depth information from backscattered light. However, existing implementations for these two optical schemes are limited to surface mapping of 3D objects at depth resolutions, at best, at the millimeter level. Here, we report 3D light-field illumination single-pixel microscopy (3D-LFI-SPM) that enables volumetric imaging of microscopic objects with a near-diffraction-limit 3D optical resolution. Aimed at 3D space reconstruction, 3D-LFI-SPM optically samples the 3D Fourier spectrum by combining 3D structured light-field illumination with single-element intensity detection. We build a 3D-LFI-SPM prototype that provides an imaging volume of ∼390 × 390 × 3,800 μm3 and achieves 2.7-μm lateral resolution and better than 37-μm axial resolution. Its capability of 3D visualization of label-free optical absorption contrast is demonstrated by imaging single algal cells in vivo. Our approach opens broad perspectives for 3D SPI with potential applications in various fields, such as biomedical functional imaging.

Abstract:Here we report a method for visualization of volumetric structural information of live biological samples with no exogenous contrast agents. The process is made possible through a technique that involves generation, synthesis and analysis of three-dimensional (3D) Fourier components of light diffracted by the sample. This leads to the direct recovery of quantitative cellular morphology with no iterative procedures for reduced computational complexity. Combing with the fact that the technique is easily adaptive to any imaging platform and requires minimum sample preparation, our proposed method is particularly promising for observing fast, volumetric and dynamic events previously only accessible through staining methods.

Abstract:We report the experimental demonstration of deeply subwavelength far-field optical imaging of unlabelled samples with resolution better than $\lambda/20$. We beat the ~$\lambda$/2 diffraction limit of conventional optical microscopy several times over by recording the intensity pattern of coherent light scattered from the object into the far-field. We retrieve information about the object with a deep learning neural network trained on scattering events from a large set of known objects.

Abstract:Abstract Single-photon lidar has emerged as a prime candidate technology for depth imaging through challenging environments. Until now, a major limitation has been the significant amount of time required for the analysis of the recorded data. Here we show a new computational framework for real-time three-dimensional (3D) scene reconstruction from single-photon data. By combining statistical models with highly scalable computational tools from the computer graphics community, we demonstrate 3D reconstruction of complex outdoor scenes with processing times of the order of 20 ms, where the lidar data was acquired in broad daylight from distances up to 320 metres. The proposed method can handle an unknown number of surfaces in each pixel, allowing for target detection and imaging through cluttered scenes. This enables robust, real-time target reconstruction of complex moving scenes, paving the way for single-photon lidar at video rates for practical 3D imaging applications.

Abstract:Ultrafast imaging is a powerful tool for studying space-time dynamics in photonic material, plasma physics, living cells, and neural activity. Pushing the imaging speed to the quantum limit could reveal extraordinary scenes about the questionable quantization of life and intelligence, or the wave-particle duality of light. However, previous designs of ultrafast photography are intrinsically limited by framing speed. Here, we introduce a new technique based on a multiple non-collinear optical parametric amplifier principle (MOPA), which readily push the frame rate into the area of ten trillion frames per second with higher spatial resolution than 30 line pairs per millimeter. The MOPA imaging is applied to record the femtosecond early evolution of laser-induced plasma grating in air for the first time. Our approach avoids the intrinsic limitations of previous methods, thus can be potentially optimized for higher speed and resolution, opening the way of approaching quantum limits to test the fundamentals.

Abstract:As light travels through a disordered medium such as biological tissues, it undergoes multiple scattering events. This phenomenon is detrimental to in-depth optical microscopy, as it causes a drastic degradation of contrast, resolution and brightness of the resulting image beyond a few scattering mean free paths. However, the information about the inner reflectivity of the sample is not lost; only scrambled. To recover this information, a matrix approach of optical imaging can be fruitful. Here, we report on a de-scanned measurement of a high-dimension reflection matrix R via low coherence interferometry. Then, we show how a set of independent focusing laws can be extracted for each medium voxel through an iterative multi-scale analysis of wave distortions contained in R. It enables an optimal and local compensation of forward multiple scattering paths and provides a three-dimensional confocal image of the sample as the latter one had become digitally transparent. The proof-of-concept experiment is performed on a human opaque cornea and an extension of the penetration depth by a factor five is demonstrated compared to the state-of-the-art.
