Abstract:We designed and synthesized a novel near-infrared (NIR) mitochondria-targeted fluorescent probe RQNA for the specific detection of mitochondrial Cu2+ because mitochondria are important reservoirs of intracellular copper. For the preparation of this probe, a novel π-extended fluorescent xanthene dye RQN was firstly synthesized via an intramolecular nucleophilic substitution of aromatic hydrogen (SNArH) strategy. Then, probe RQNA was prepared by the reaction of RQN and hydrazine hydrate, followed by treatment with acetone. RQNA exhibited selectivity, sensitivity (22 nM), and fast response time (20 s) for the detection of Cu2+ via a specific Cu2+-triggered ring-opening and hydrolysis cascade reaction. RQNA is cell-membrane permeable and mitochondria-targetable, and can be used for monitoring mitochondrial Cu2+ in living cells.

Abstract:1,4-Dihydropyridines are a class of drugs with a wide range of biological activities and pharmacological effects. However, there are few reports on its optical activity, especially its application on fluorescent CN− probe. In this experiment, we designed and synthesized a fluorescent probe based on 1,4-dihydropyridines to detect CN−. Interestingly, the probe exhibited outstanding properties such as 100% water soluble, near infrared, ratiometric, fast response, high selectivity and anti-interference ability for other ions. The color change indicated that the probe can be used for naked eye identification. In particular, the probe showed a super large fluorescent emission peak shift (260 nm). In addition, the reaction mechanism of the probe has been studied by 1H NMR titration, high resolution mass spectrometry and theoretical calculations.

Abstract:A class of pyridazinone derivatives as near-infrared optical probes in fluorescence microscopy images was designed. The design strategy consisted of the stepwise extension and modification of pyridazinone by expansion of the electron-donating moiety to a larger π-conjugated system and anchoring a subcellular directing group such as triphenylphosphine or morpholine. All the desired products were successfully applied in cell imaging with high subcellular colocalization. Furthermore, these fluorescent probes showed excellent performance in mouse-brain imaging.

Abstract:A novel quinoline-fused Rhodamine dye is designed and synthesized to achieve near-infrared fluorescent emission by expanding the pi-conjugated system. Ratiometric fluorescent probe (RhQ) is further constructed by introducing the 3,5-ditrifluormethylthiophenol moiety into dye through a thioester bond to take advantage of spimcyclization open-ring states of Rhodamine platform. The probe exhibits remarkable dual-channel fluorescence response toward Cys/Hcy in aqueous solutions between near-infrared and visible emission, arising from the formation of closed spimlactam configuration disrupting molecular pi-conjugation system. RhQ is highly selective and sensitive to Cys/Hcy with detection limits of 0.12 mu M and 0.25 mu M for Cys and Hcy, respectively. Moreover, imaging in cellular experiment indicates that probe displays excellent mitochondrial targeting ability, lower cytotoxicity and ratiometric responses with two well-defined emissions to ensure exactly comparison and quantitative analysis of Cys/Hcy changes in the cell.

Abstract:Near-infrared (NIR) fluorophores, especially dicyano-based fluorophores and xanthene-based hemicyanines, have been put high expectation in bioimaging application due to their excellent optical properties. However, they suffer from inherent shortages such as short excitation/emission wavelength (less than 700 nm) or small Stokes shift (20-50 nm). Herein, we constructed a novel NIR dicyano-based fluorophore (DCO-HBTN). To our knowledge, it is the first reported dicyano-based fluorophore of which the excitation/emission wavelength is more than 650 nm and Stokes shift is more than 100 nm. To demonstrate the feasibility of our efforts, we developed two NIR fluorescent probes (Probe-Cys and Probe-H2S) based on the fluorophore, Probe-Cys displayed good selective and highly sensitive (LOD = 0.28 mu M) recognition of Cys over Hcy and GSH, which was used to visualize endogenous Cys in tumor tissue. Probe-H2S exhibited an. excellent specific and sensitive (LOD = 0.11 mu M) response to H2S, which was applied in monitoring H2S releasing from the prodrug in vitro and in vivo.

Abstract:Tyrosinase (TYR) detection will be crucial in the prevention and early diagnosis of some illnesses, including epilepsy, albinism, and melanoma, due to the close linkage of abnormal expression of TYR with the occurrence of these diseases. Therefore, how to obtain a fluorescent probe that has a high fluorescence quantum yield and high specificity for detecting TYR activity will be of significance. In this work, we improved the rhodamine structure by expanding the π-conjugation system and using asymmetric synthesis to provide a fluorescent probe with good optical properties for the specific detection of TYR. The fluorescence quantum yield of fluorophore released from the probe by the enzymatic reaction with TYR was as high as 0.22, which was significantly greater than that of other previously reported fluorescent probes. We also chose 3-hydroxybenzyl oxygen as the recognition group, which can successfully lessen the interference from reactive oxygen species (ROS). At the same time, the probe demonstrated great sensitivity in detecting TYR activity, and the detection limit of TYR for the probe was calculated to be 0.45 U/mL. In addition, the probe has currently been successfully utilized to evaluate TYR activity in cells. Therefore, this probe is anticipated to be crucial in identifying TYR activity in challenging settings.

Abstract:In this work, tetrahydro-acridine salt hybrid coumarin dyes was designed and developed as a Near-infrared (NIR) fluorescent probe CM for the detection and fluorescent bioimaging of Cys/Hcy. The activated chlorine atom in probe facilitates more efficient nucleophilic substitution reaction, resulting in fast responsive time (3 min toward 3 eq. Cys) and high sensitivity toward Cys with detection limit of 46 nM. More importantly, probe displays "Turn -on" near-infrared fluorescence emission (Emmax: 674 nm) with Large Strokes shift (about 200 nm) toward Cys/ Hcy. Furthermore, probe could be used as efficient tool for imaging and detection of Cys/Hcy in mitochondria of living cells and in vivo. This work innovatively proposed a new design strategy for NIR fluorescent probe platform for the detection and fluorescence imaging of bio-thiols.
An Activated Near-Infrared Fluorescent Probe with Large Stokes Shift for Discrimination of Bio-Thiols

Abstract:Bio-thiols such as cysteine (Cys), homocysteine (Hcy), and glutathione (GSH) play a crucial role in physiological processes. Distinguishing between Cys, Hcy, and GSH remains a challenge because of their similar structure and reactivity. Herein, we conceptualized and synthesized a near-infrared (NIR) fluorescent probe TAI based on acridine-conjugated indole salts. This innovative probe demonstrated effectiveness in sensitively distinguishing Cys, Hcy and GSH by NIR dual-channel and time-dependence fluorescence. Probe displays a significant turn-on NIR fluorescent response toward Cys at 654nm under 488nm excitation with Large Strokes shift (about 166nm) and high selectivity, but also shows turn-on NIR fluorescent response for Hcy/GSH at 695nm under 640nm excitation. More interestingly, the probe also exhibited an NIR fluorescence response at 654nm to Hcy and no fluorescence response to GSH when the response time is over 3h. The sensing mechanism was fully confirmed by NMR, HRMS spectra and DFT theoretical calculations. Moreover, successful intracellular imaging experiments in living cells suggest that the probe is suitable for discrimination imaging endogenous and exogenous bio-thiols. This work offers a promising near-infrared fluorescent imaging tool for the differential detection of bio-thiols.
A Coumarin-Indole-Based Near-Infrared Ratiometric Ph Probe for Intracellular Fluorescence Imaging

Abstract:New pH-activatable ratiometric fluorescent probes have been developed that have a coumarin structure attached to benzothiazole or indole units with one or two conjugated bonds. They have optical responses in acidic conditions. Probes with benzothiazole (1) and indole (2, 3) units have opposite responses to changes in pH, in terms of shifts in absorption and emission. Quantum chemical calculations with the B3LYP exchange functional provide insights into the differences in optical properties between 1 and 2/3. Probe 3 (7-diethylamino-3-[4-(3,3-dimethyl-3H-indol-2-yl)-buta-1,3dienyl]- chromen-2-one) displayed a significant 122 nm red shift of the emission maximum; the new peak is located in the near-infrared region. In vitro assays demonstrate that probe 3 can be used in quantitative fluorescence imaging for the detection of intracellular pH distributions. In addition to the large Stokes shift, it has rapid responsivity, excellent cell permeability, and high selectivity. This study reveals that the conjugation of coumarin with benzothiazole or indole to form an intramolecular charge transfer (ICT) platform is an effective method to construct new fluorescent pH probes for acidic microenvironments.
A unique approach toward near-infrared fluorescent probes for bioimaging with remarkably enhanced contrast

Abstract:Near-infrared (NIR) fluorescent probes are attractive molecular tools for bioimaging because of their low autofluorescence interference, deep tissue penetration, and minimal damage to sample. However, most previously reported NIR probes exhibit small Stokes shift, typically less than 30 nm, and low fluorescence quantum yield, strictly limited contrast and spatial resolution for bioimaging. Herein, by expanding the π-conjugated system of rhodamine B, while, at the same time, keeping its rigid and planar structure, we reported an efficient NIR dye, HN7, with large stokes shift of 73 nm and fluorescence quantum yield as high as 0.72 in ethanol, values superior to those of such traditional cyanine NIR dyes as Cy5. Using HN7, living cells, tissues and mice were imaged, and the results showed significantly enhanced contrast, improved spatial resolution, and satisfactory tissue imaging depth when compared to Cy5. Moreover, the nonfluorescent spirocyclic structure of rhodamine B is an inherent component of HN7; therefore, our strategy provided a universal platform for the design of efficient NIR turn-on bioimaging probes for various targets. As a proof-of-concept, two different NIR probes, HN7-N2 and HN7-S for NO and Hg2+, respectively, were designed, synthesized, and successfully applied for the imaging of NO and Hg2+ in living cells, tissues and mice, respectively, demonstrating the potential bioimaging applications of the new probes. In sum, this new type of dye may present new avenues for the development of efficient NIR fluorescent probes for contrast-enhanced imaging in biological applications.

Abstract:Near-infrared (NIR) fluorescent probes have been considered as powerful tools for exploring bioactive species and biological processes in living systems by virtue of the substantial merits of the NIR window (such as minimal photo-toxicity, deep tissue penetration, and low background interference). However, the development of effective NIR fluorescent probes for biological application is still facing great challenges due to some weakness (for example, pH sensitivity, poor chemical and photo-stability) of mainly current NIR fluorophores. Herein, we synthesized a novel fluorine atom substituted NIR chromenylium-cyanine fluorophore, termed CCF-OH, with an optically tunable hydroxyl group. Compound CCF-OH possess several optical advantages, including high chemical stability, high photo-stability, and high fluorescence quantum yield (Phi = 0.47). Significantly, a low pK(a) value (6.3) of CCF-OH was obtained, making its fluorescence signal steady around the pH ranging from 6.8 to 8.1. To demonstrate the practicability of CCF-OH, a mitochondria-targeted near-infrared fluorescent probe, N-Cys, was developed by masking the hydroxyl group of CCF-OH with an acrylate group as the recognition unit for cysteine (Cys). N-Cys responded to Cys with an obvious ratiometric signal. Furthermore, N-Cys has also been successfully applied for ratiometric NIR imaging of Cys in living cells, suggesting the great potential of the new NIR fluorophore, CCF-OH, for practical applications in living systems. Abstract:A novel near-infrared fluorescent probe TM2 based on a rhodamine-bearing framework was disclosed with a large Stokes shift (100 nm). TM2 exhibits highly selective recognition for Cu2+ in EtOH/H2O (1 : 1, v/v) solution with a low detection limit (2.3 μM) and a wide detection range (0-50 μM). Detection of Cu2+ is undisturbed at physiological pH levels of 5-9. This recognition mechanism is attributed to the formation of a 1 : 1 complex between TM2 and Cu2+, validated by Job's plot, 1H NMR titration, and LC-MS experiments. Moreover, the successful fluorescence imaging of Cu2+ both in vitro and in vivo was also accomplished.

Abstract:In this study, a new near-infrared (NIR) fluorescent turn-on probe featuring a large Stokes shift (198 nm) was developed for the detection of biothiols. The probe was based on a dicyanoisophorone derivative serving as the fluorophore and a 2,4-dinitrobenzenesulfonyl (DNBS) group functioning as both a recognition site and a fluorescence quencher. In the absence of biothiols, the fluorescence of the probe was low due to the photoinduced electron transfer (PET) effect between the fluorophore and DNBS. Upon the presence of biothiols, the DNBS group underwent a nucleophilic aromatic substitution reaction with the sulfhydryl group of biothiols, leading to the release of the fluorophore and a notable emission peak at 668 nm. This developed probe exhibited exceptional selectivity and sensitivity to biothiols in solution, with an impressive detection limit of 28 nM for cysteine (Cys), 22 nM for homocysteine (Hcy), and 24 nM for glutathione (GSH). Furthermore, the probe demonstrated its applicability by successfully visualizing both endogenous and exogenous biothiols in living systems.

Abstract:Developing fluoran dyes as fluorescent probes is interesting because it is of great significance to extend their applications from the printing industry to other fields such as fluorescence analysis and medicine. We here present a new quinoline-fluoran hybrid dye QF, which delivers fluorescent signals in water (pH: 4.0 - 9.0) with the maximum absorption and emission centered at 514 nm and 597 nm, respectively. In addition, a large Stokes shift (83 nm) can be observed when compared to that of rhodamine B. QF is further employed for the preparation of a new fluorescent probe QFH, which exhibits high selectivity towards copper ions with the limit of detection down to 1.54 nM via a Cu2+-triggered spirolactam ring opening and hydrolysis process. Significantly, QFH can be applied for the detection of copper ions at physiological pH conditions (pH: 6.0 - 9.0) and shows low cell cytotoxicity, which allows it to be a candidate for tracing Cu2+ by cell imaging.

The probe exhibited good response fluorescent characteristic, and cells experiments showed the probe had high affinity and without apparent cytotoxicity. Fluorescent image experiments in living L929 cells, MCF-7 cells, HepG2 cells, further demonstrated its potential applications in biological systems. Abstract:Cytochrome P450 1B1 (CYP1B1) is induced during the early stage of cancer and is universally overexpressed in tumors. Thus, it was considered as a potential biomarker for the monitoring of cancer. In this study, we designed and synthesized CYP1B1-targeted near-infrared (NIR) fluorescence molecular probes based on the latest reported open conformation of the CYP1B1-alpha-naphthoflavone (ANF) complex. According to the architecture of the open channel, we introduced linkers and a Cy5.5 fragment at the 5 ' position of ANF derivatives with strong CYP1B1 inhibitory activity to obtain probes 19-21. Then, in vitro cell-based studies showed that the probes could be enriched in tumor cells by binding to CYP1B1. During in vivo and ex vivo imaging in a xenograft mouse model, probe 19 with the best binding affinity was proven to be able to identify tumor sites in both fluorescence imaging and photoacoustic imaging modes.

Abstract:IR-789, a novel near-infrared fluorescent probe, was designed, synthesized, and applied to living cells. The probe exhibited better response fluorescence characteristics than the only FDA-approved agent, indocyanine green. Cell experiments showed that the probe had high affinity and without apparent cytotoxicity. Abstract:A novel near-infrared fluorescent light- up probe with a tumor-homing pentapeptide, CREKA (Cys-Arg-Glu-Lys-Ala), specifically binds to fibrin-fibronectin complexes was rationally designed and developed for biomedical imaging. Its superior practical applications in tumor imaging and drug-induced liver injury detection are well demonstrated for the first time.
