Abstract:Luminogens with aggregation-induced emission (AIEgens) characteristics have been well developed and applied in various areas such as bio-imaging, theranostics, organic photoelectronics and chemo/bio sensors. However, most of the reported AIEgens suffer from the disadvantages of complex organic synthesis and high cost, as well as being environmentally unfriendly and hard to degrade, which have largely limited their real applications. In this work, we discovered berberine chloride, a natural isoquinoline alkaloid isolated from Chinese herbal plants, as an unconventional rotor-free AIEgen with bright solid-state emission and water-soluble characteristics. Single crystal structure analysis and optical property, viscosity, and host-guest interaction studies suggested that intramolecular vibration and twisted intramolecular charge transfer were responsible for the AIE phenomenon of berberine chloride. Moreover, berberine chloride was biocompatible and could specifically target lipid droplets in a fluorescence turn-on and wash-free manner, demonstrating the great potential of natural products as promising AIE probes.

Abstract:Isocyanides have drawn increasing attention in biological applications due to their attractive properties and unique reactivities, which can undergo various reactions, such as multicomponent reactions, alpha-addition reactions, [4 + 1] cycloaddition reactions, and the reaction scope keeps expanding. In addition to acting as reactants for the preparation of structurally interesting and diverse N-heterocycles or peptidomimetics, this type of functionality may be a good choice in the labeling and modulation of biomolecules due to the high biocompatibility and small size to minimize modifications on the parent molecule. It has been demonstrated that isocyanides can participate in biomolecule labeling through three strategies, including the two-component bioorthogonal reaction, multicomponent reaction, and metal chelation. Among them, the isocyanide-tetrazine reaction has been better studied recently, augmenting the potency of isocyanide as a bioorthogonal handle. This review will focus on the recent progress in isocyanide chemistry for labeling of biomolecules. Meanwhile, methods to introduce isocyano groups into biomacromolecules are also described to facilitate wider applications of this unique functionality.

Abstract:Cancer is one of the most fatal diseases for decades. Aggregation‐induced emission luminogens (AIEgens) have been recently used as molecular imaging or therapeutic agents in cancers, due to the advantages of large Stokes shift, high quantum yield, great biocompatibility, and strong photostability. AIEgens can specifically target different types of cancer via diverse targeting strategies. AIEgen‐based fluorescence imaging, especially near‐infrared imaging, demonstrated deep penetration and suitable signal‐to‐noise ratio, which allows reliable in vivo cancer imaging. Combined with other imaging modalities, AIEgen‐based multimodal imaging could provide multidimensional cancer hallmarks from different perspectives. In addition, AIEgen‐based phototherapy can be used for photodynamic therapy and photothermal therapy, which facilitate ablation of cancer cells with good biosafety and high therapeutic effects in vivo. AIEgens nanoparticles fabricated with some specific chemicals, drugs, or siRNA, could display synergistic therapeutic effects for cancers. This paper comprehensively describes the current status and future perspectives of AIEgens, which have showed a great potential for the future preclinical and clinical translation on in vivo molecular imaging and theranostics in cancer.

Abstract:The development of high-performance theranostic platforms is always an intractable challenge in modern medicine research. Herein, new luminescent materials that possess the advantages of high sensitivity, high resolution, and deep tumor-penetrating imaging as well as multimodal synergistic therapeutic functions may be one of the best potential alternatives for efficient theranostics. In this work, a powerful Au(I)-containing aggregation-induce emission luminogen (AIEgen) with integrated multifunctions of bimodal imaging and multimodal combination therapy is developed. This simple small-molecule system can simultaneously realize high-contrast fluorescence imaging and computed tomography imaging. Additionally, it can act as an effective thioredoxin reductase inhibitor to exert inherent anticancer effect, and simultaneously work as an ideal radiosensitizer to produce efficient anticancer efficacy through a multimodal synergistic effect. Thus, this study highlights a new design guideline to endow AIE luminogens with extraordinary functions by Au(I) and opens a new avenue for the development of new luminescent theranostic systems.

Biology-Oriented Design Strategies of AIE Theranostic Probes

Abstract:Theranostic probes integrate sensing, imaging, and therapy functions, which not only allow direct visualization of biochemical processes occurring in complex biological systems, but also offer facile therapeutic functions for various diseases. In this regard, organic fluorogens with aggregation-induced emission (AIEgens) are ideal materials to satisfy the needs of multifunctional theranostic probes. Under the guidance of specific biological principles, AIEgens can be deliberately engineered to have simultaneous optical and therapeutic functions, such as disease targeting, responsive drug release, and photosensitizing/photothermal capability, which could influence their interactions with biological microenvironments and further enable image-guided treatment of various diseases. In this perspective, we mainly focus on how new AIE multifunctional probes have been designed based on the latest advances in medicine and biology. We first summarize the biology-oriented design strategies for AIE probes for diagnosis and treatment of disease. Next, specific examples have been demonstrated to illustrate the probe design strategies. Finally, current challenges and future perspectives for development of AIE theranostic probes are presented in the conclusion section.

Abstract:Efficient theranostic systems can realize better outcomes in disease treatment because of precise diagnosis and the concomitant effective therapy. Aggregation-induced emission luminogens (AIEgens) are a unique type of organic emitters with intriguing photophysical properties in the aggregate state. Among the AIEgens studied for biomedical applications, so far, metal-based AIE systems have shown great potential in theranostics due to the enhanced multimodal bioimaging ability and therapeutic effect. This research field has been growing rapidly, and many rationally designed systems with promising activities to cancer and other diseases have been reported recently. In this review, we summarized the recent progress of metal-based AIE materials in bioimaging and biological theranostics, and deciphered the pertinent design strategies. We hope that this review can offer new insights into the development of this growing field.

Abstract:Aggregation-induced emission (AIE) refers to a unique photophysical phenomenon observed for some specific fluorophores, which are faintly or non-emissive in solution but become highly emissive in solid and aggregated states. In the cases of fluorescent probes, AIE active molecules possess some unparallel advantages over their classical counterparts. On one hand, it allows more AIE active molecules to bind to the target analyte, without worrying about the aggregation-induced fluorescence quenching; thus favors fluorescence detection. On the other hand, the feature of drastic enhancement in fluorescence intensity associating with an aggregation event can be used as a method of quantitative detection. In this paper, a series of representative AIE molecules are demonstrated; their applications as biological probes in sensing of protein, DNA, G4, and chiral amines are highlighted; their working principles and features of these molecules are described. Finally, a brief outlook on the design of new AIE-active molecules and the application of AIE-molecules in relevant fields is forwarded.

Abstract:The cell contains various organelles that perform different functions using different biological processes to form a complete organic architecture. Healthy cells and tumor cells often show clear differences in cellular homeostasis. Specific detection, imaging, and therapeutic strategies targeting organelles have been found to be effective for tumor diagnosis and therapy. The multiple functions of AIEgens make them attractive over other nanomaterials, and the application of AIEgens has increased dramatically in tumor-related nanomedicine, specifically, for the detection, bioimaging, and therapy of cancers. Here, recent advances in AIEgen applications targeting specific organelles are summarized. The ability of AIEgens to target specific organelles has enhanced various diagnostic and therapeutic applications of this technology, and this review will provide guidance for further exploration in AIEgen-related nanomedicine.

Abstract:Metabolic labeling based on bioorthogonal click chemistry has transformed the way living entities are manipulated in biological environments to enable more targeted, effective, and safer theranostic applications. Since the first demonstrations of this strategy, many other metabolic labeling approaches have been formulated to incorporate additional functionalities, such as those based on fluorogens with aggregation-induced emission characteristic (AIEgens), for light-up bioimaging and image-guided disease therapy. In this perspective, we discuss the emergence and development of AIEgen-based theranostic systems for bioorthogonal metabolic engineering and labeling. First, we highlight the fundamental principles guiding the design of AIEgen-based bioorthogonal metabolic labeling. Next, we describe the formulation and applications of different AIEgen-based synthetic and living theranostic systems for cancer and pathogenic bacterial imaging and ablation. We then provide our outlook on the current challenges, potential opportunities, and future research directions of this emerging field.

Abstract:The past two decades have witnessed significant advances in the field of aggregation-induced emission (AIE). Luminescent materials with AIE characteristics (AIEgens) are of great importance because they can show enhanced and improved properties for different high-tech applications. Metal ions are vital for all living species. In the recent years, the AIE platform has been widely adopted in the design and synthesis of metal ion-responsive luminescent probes because of its excellent photophysical properties such as microenvironment sensitivity, high signal-to-noise ratio, high photostability, high emission efficiency in the aggregated state, and large Stokes' shift. In this review, we present an overview of the design principles of metal ion responsive AIEgens that can be generated using a modular approach. We have summarized the typical examples of metal ion responsive AIEgens, and their sensing mechanisms developed from 2005 to 2020.

Abstract:In recent years, aggregation-induced emission (AIE) materials have gained significant attention and have been developed for various applications in different fields including biomedical research, chemical analysis, optoelectronic devices, materials science, and nanotechnology. AIE is a unique luminescence phenomenon, and AIEgens are fluorescent moieties with relatively twisted structures that can overcome the aggregation-caused quenching (ACQ) effect. Additionally, AIEgens offer advantages such as non-washing properties, deep tissue penetration, minimal damage to biological structures, high signal-to-noise ratio, and excellent photostability. Fluorescent probes with AIE characteristics exhibit high sensitivity, short response time, simple operation, real-time detection capability, high selectivity, and excellent biocompatibility. As a result, they have been widely applied in cellular imaging, luminescent sensing, detection of physiological abnormalities in the human body, as well as early diagnosis and treatment of diseases. This review provides a comprehensive summary and discussion of the progress over the past four years regarding the detection of metal ions, small chemical molecules, biomacromolecules, microbes, and cells based on AIE materials, along with discussing their potential applications and future development prospects.

Abstract:Organometallic d6, d8 and d10 metal complexes are known for their interesting photophysical and photochemical properties. This allows many of them to develop into luminescent probes for biosensing and imaging. In addition, some of the complexes have been found to show promising anticancer properties, with different molecular targets from the classical cisplatin and platinum-based chemotherapeutic agents, illustrating the possibility of using them to develop new therapy for cancers. Notably, there are examples of luminescent metal complexes showing good therapeutic values. They have been employed as theranostic agents which enable diagnosis and therapy simultaneously. This review highlights key features of organometallic d6, d8 and d10 metal complexes reported in the past 10 years and their applications in biosensing, imaging, cancer treatment and theranosis. We also discuss the limitations found in these complexes and provide insights into the future development of organometallic metal complexes for biomedical applications.

Abstract:It is challenging to achieve precise control on the properties of organic π‐functional materials to widen their practical applications. On the other hand, the study of aggregation‐induced emission luminogens (AIEgens) helps achieve such goals because of inherent relationships between their luminescence behaviors and conformational variations that allow for the visual monitoring of the changes in the material properties. Inspired by this, in this work, three AIE isomers are fabricated in structures consisting of tetraphenylpyrazine and triphenylethene units with para‐, meta‐, and ortho‐position linkages, respectively. The isomerism effect brings about significantly decreased luminescence efficiency, subtly blueshifted emission, basically reduced AIE effect but boosted porosity in the aggregate state as the conformation of AIEgens evolves from an extended to a folded one. Based on the distinct properties, their respective use in blue organic light‐emitting diodes, nanofluorescent probes, and molecule‐capturing porous crystals are investigated. This work not only achieves precise property control by using the isomerism effect of AIEgens but also provides useful information on the future design of π‐conjugated materials with advanced functionalities.

Abstract:One of the major pursuits of biomedical science is to develop advanced strategies for theranostics, which is expected to be an effective approach for achieving the transition from conventional medicine to precision medicine. Supramolecular assembly can serve as a powerful tool in the development of nanotheranostics with accurate imaging of tumors and real‐time monitoring of the therapeutic process upon the incorporation of aggregation‐induced emission (AIE) ability. AIE luminogens (AIEgens) will not only enable fluorescence imaging but will also aid in improving the efficacy of therapies. Furthermore, the fluorescent signals and therapeutic performance of these nanomaterials can be manipulated precisely owing to the reversible and stimuli‐responsive characteristics of the supramolecular systems. Inspired by rapid advances in this field, recent research conducted on nanotheranostics with the AIE effect based on supramolecular assembly is summarized. Here, three representative strategies for supramolecular nanomaterials are presented as follows: a) supramolecular self‐assembly of AIEgens, b) the loading of AIEgens within nanocarriers with supramolecular assembly, and c) supramolecular macrocycle‐guided assembly via host–guest interactions. Meanwhile, the diverse applications of such nanomaterials in diagnostics and therapeutics have also been discussed in detail. Finally, the challenges of this field are listed in this review.

Abstract:The integration of aggregation-induced emission luminogens (AIEgens) and inorganic constituents to generate multifunctional nanocomposites has attracted much attention because it couples the bright aggregate-state fluorescence of AIEgens with the diverse imaging modalities of inorganic constituents. Herein, a facile and universal strategy to prepare metal-phenolic-network (MPN)-coated AIE dots in a high encapsulation efficiency is reported. Through precise control on the nucleation of AIEgens and deposition of MPNs in tetrahydrofuran/water mixtures, termed as coacervation, core-shell MPN-coated AIE dots with bright emission are assembled in a one-pot fashion. The optical properties of MPN-coated AIE dots can be readily tuned by varying the incorporated AIEgens. Different metal ions, such as Fe3+ , Ti4+ , Cu2+ , Ni2+ , can be introduced to the nanoparticles. The MPN-coated AIE dots with a red-emissive AIEgen core are successfully used to perform magnetic resonance/fluorescence dual-modality imaging in a tumor-bearing mouse model and blood flow visualization in a zebrafish larva. It is believed that the present study provides a tailor-made nanoplatform to meet the individual needs of in vivo bioimaging.

Abstract:Multi-functional fluorescent probes have attracted more interest recently. Herein, two simple fluorescent probes (FAS and DPAS) from AIEgens with the ESIPT process of salicylaldehyde Schiff-base structures are developed. The aggregates of FAS and DPAS exhibit good selectivity and sensitivity towards Cu2+ in aqueous media without any water-miscible organic solutions as buffers. Their larger association constants and appropriate hydrophilic and hydrophobic properties play key roles in achieving lower detection limits. Based on their fluorescence on/off processes in the presence or absence of Hg2+, the composite probes are further prepared.

Abstract:Aggregation-induced emission (AIE) is a unique phenomenon observed in various materials such as organic luminophores, carbon dots (CDs), organic-inorganic nanocomposites, fluorescent dye molecules, and nanoparticles (NPs). These AIE-active materials, or AIEgens, are ideal for balancing multifunctional phototheranostics and energy dissipation. AIE properties can manifest in organic fluorescent probes, rendering them effective for cancer treatment due to their ability to penetrate deeply and provide high therapeutic efficacy. This efficacy is attributed to their high photobleaching thresholds, ability to induce Stokes shifts, and capacity to activate fluorophores. Therefore, the development of innovative AIE-based materials for disease diagnosis and treatment, particularly for cancer, is both important and promising. Recent years have seen successful demonstrations of nanoparticles with AIE properties being used for photodynamic therapy (PDT) and multimodal imaging of tumor cells. These fluorophores have been shown to impact mitochondria and lysosomes, generate reactive oxygen species (ROS), activate the immune system, load and release drugs, and ultimately induce apoptosis in tumor cells. In this review, we examine previous studies on the manufacturing methods and effects of AIEgens on cancer cells, with a theranostic strategy of simultaneous treatment and imaging. We also investigate the factors affecting drug delivery on different cancer cells, including internal stimuli such as pH, ROS, enzymes, and external stimuli like near-infrared (NIR) light and ultrasound waves.
