Abstract:Compared with the increasing and widespread bacterial resistance to clinical medicines and the urgent need for cures of intractable diseases, there is a dramatic decline in the numbers of drugs reaching the market or clinical trials. Accordingly, it has become imperative to discover more rational and efficient strategies to design and develop novel drugs. Structure-based drug design/discovery (SBDD) is one of the computer-aided methods, by which novel drugs are designed or discovered based on the knowledge of 3D structures of the relevant specific targets. During the past few decades, the great potentials and success of SBDD have been seen in the field of drug discovery. In this review, we present an overview of the key mechanisms of SBDD, the frequently used computer programs in SBDD and the reported successful cases. Finally, several typical design processes of lead components from SBDD are also highlighted in detail, such as the discovery of inhibitors of G protein-coupled receptors (GPCRs), antibacterial drugs, and anti-cancer drugs.

Abstract:Since the sulfur(VI) fluoride exchange reaction (SuFEx) was introduced by Sharpless and co-workers in 2014, this new-generation click chemistry has emerged as an efficient and reliable tool for creating modular intermolecular connections. Sulfonyl fluorides, one of the most important sulfur(VI) fluoride species, have attracted enormous attention in diverse fields, ranging from organic synthesis and material science, to chemical biology and drug discovery. This review aims to introduce seminal and recent progresses on the synthetic methods of sulfonyl fluorides, which include aromatic, aliphatic, alkenyl, and alkynyl sulfonyl fluorides. While not meant to be exhaustive, the purpose is to give a timely overview and insight in this field, and stimulate the development of more efficient synthetic methods of sulfonyl fluorides.

This short review provides a detailed introduction to the diverse applications of fluorosulfonyl-containing radical reagents in organic synthesis, such as C-H bond functionalizations, difunctionalization of olefins, and tandem reactions, highlighting their crucial role in synthesizing complex organic molecules.

Abstract:Four-membered heterocycles such as oxetanes and azetidines represent attractive and emergent design options in medicinal chemistry due to their small and polar nature, and potential to significantly impact physiochemical properties of drug molecules. The challenging preparation of these derivatives, especially in a divergent manner, has severely limited the combination of oxetanes with other medicinally and biologically important groups. Consequently, there is a substantial demand for mild and effective synthetic strategies to access new oxetane and azetidine derivatives and molecular scaffolds. Herein, we report the development and use of oxetane sulfonyl fluorides (OSF) and azetidine sulfonyl fluorides (ASF), which behave as precursors to carbocations in an unusual defluorosulfonylation reaction pathway (deFS). The small-ring sulfonyl fluorides are activated under mild thermal conditions (60 ºC), and the generated reactive intermediates couple with a broad range of nucleophiles. This reactivity enables the generation of new chemical motifs that make attractive design elements for drug discovery. Notably, oxetane and azetidine-heterocyclic, -sulfoximine and -phosphonate derivatives are prepared, several of which do not have comparable carbonyl analogues. Alternatively, a SuFEx pathway under anionic conditions accesses oxetane-sulfur (VI) derivatives. We demonstrate the synthetic utility of novel OSF and ASF reagents through the synthesis of 11 drug analogues, showcasing their potential for subsequent diversification and facile inclusion into medicinal chemistry programs. Moreover, we propose the application of the OSF and ASF reagents as linker motifs and demonstrate the incorporation of pendant groups suitable for common conjugation reactions. Productive deFS reactions with E3 ligase recruiters such as pomalidomide and related derivatives provide new degrader motifs, and potential PROTAC linkers.

Abstract:Background: The widespread application of fluorine in drug design benefits from distinctive properties. Incorporation of fluorine can positively modulate certain pharmacokinetics properties, including lipophilicity, eletrophilicity, metabolic stability, chemical stability, et al., which were of interest to medicinal chemists. Herein, fluorinated compounds, which have been designed and evaluated for anticancer, antimicrobial, anti-inflammatory and antivirus activity, et al. during the last 6 years (2010-2015) are summarized. Conclusion: The emphasis is to highlight the importance of fluorinated compounds and the changes in the properties of compounds when introducing fluorine atom.

This review focuses on the latest advancements in synthesizing these compounds using the radical sulfur dioxide insertion and fluorination (RSIF) strategy, underscoring the versatility and efficacy of the RSIF strategy in constructing sulfonyl fluoride compounds.

Abstract:Sulfur(VI) Fluoride Exchange (SuFEx) is a new family of click chemistry transformations which relies on readily available materials to produce compounds bearing the S-VI-F motif. The potential of SuFEx in drug discovery has just started to be explored. We report the first method of SuFEx chemistry for the conversion of phenolic compounds to their respective arylfluorosulfate derivatives in situ in 96-well plates. This method is compatible with automated synthesis and screening to quickly assess the biological activities of the in situ generated, crude products. Using this method, we perform late-stage functionalization of a panel of known anticancer drugs to generate the corresponding arylfluorosulfates. These in situ generated arylfluorosulfates are directly tested in a cancer-cell growth inhibition assay in parallel with their phenolic precursors. We discover three arylfluorosulfates that exhibit improved anticancer cell proliferation activities compared to their phenol precursors. Among these three compounds, the fluorosulfate derivative of Fulvestrant possesses significantly enhanced activity to down-regulate estrogen receptor (ER) expression in ER+ breast cancer cell line MCF-7 and the fluorosulfate derivative of Combretastatin A4-a general anticancer drug currently being evaluated under clinical trials-exhibits a 70-fold increase in potency in the drug resistant colon cancer cell line HT-29.

Abstract:Since their introduction as a new strategy for synthesizing diverse chemotypes, sulfur(VI)–fluoride exchange (SuFEx) transformations have found applications ranging from polymer chemistry and covalent probe development to bioconjugation tools and chemistries for the synthesis of compound libraries. The collection of SuFEx reactions has expanded significantly since their introduction as a concept, comprising functionalities with varying reactivities towards different nucleophiles; thus, enabling the generation of a wide array of sulfur-containing functional groups for the linkage of structural elements in diverse chemotypes. In this review, we focus on the most recent developments in the use of SuFEx chemistry as a means for the preparation of compound libraries for biological screening as well as the introduction of SuFEx hubs into various biomolecules.

Abstract:Vinyl sulfones, with their exceptional chemical properties, are known as the "chameleons" of organic synthesis and are widely used in the preparation of various sulfur-containing structures. However, their most alluring feature lies in their biological activity. The vinyl sulfone skeleton is ubiquitous in natural products and drug molecules and boasts a unique molecular structure and drug activity when compared to conventional drug molecules. As a result, vinyl sulfones have been extensively studied, playing a critical role in organic synthesis and pharmaceutical chemistry. In this review, we present a comprehensive analysis of the recent applications of vinyl sulfone structures in drug design, biology, and chemical synthesis. Furthermore, we explore the prospects of vinyl sulfones in diverse fields, offering insight into their potential future applications.

Abstract:The introduction of trifluoromethylsulfinyl group into the required target molecules has become an indispensable strategy for pharmaceutical chemists to improve the success of new drug discovery. Compared with the indirect methods, direct trifluoromethylsulfinylation by employing available trifluoromethylsulfinylating reagents is an important and elegant strategy for the rapid assembly of valuable CF3S(O)‐derived organic molecules. This review focuses on the recent synthetic approaches and future perspectives of trifluoromethylsulfinyl compounds based on the indirect and direct methods in this area. These transformations proceed effectively with high yields and good functional group compatibility under mild conditions.

Abstract:Sulfonyl fluorides have gained significant importance due to their classification as a click reaction and therefore have seen increased use in drug discovery and biochemistry. Their use, however, is complicated by the methods by which they are synthesized and their general synthetic instability. This results in sulfonyl fluorides being introduced late in a synthetic route with minimal structural diversity. Masking the reactivity of a sulfonyl fluoride by protecting the parent sulfinate is one method to ameliorate these issues. This study outlines discovery and selection of sulfinate protecting groups (SPGs) based on their overall stability, ease of synthesis, and simple deprotection conditions. This includes the discovery of two novel, photolabile sulfinate protecting groups (SPGs), para¬-methoxybenzyl Rongalite and ortho-nitrobenzyl Rongalite that can be directly converted to the sulfonyl fluoride using light and selectfluor. Along with known SPG, 2-trimethylsilylethyl sulfone (SES), all three SPGs were found to possess broad stability when exposed to numerous common synthetic conditions and are easily coupled to aryl halides from their sulfinate salt precursor. Overall, having access to a wide range of stable, easily functionalized SPGs will aid in increasing the structural diversity of sulfonyl fluorides.

Abstract:Sulfonyl drugs are widely used to treat various types of diseases, in which sultones have several desirable properties that have led to their increased use, including improving its simple and effective synthetic methods and/or potentially enhancing potency of drugs. This review focuses on the recent progress in typical synthesis of sultones and the latest developments in various therapeutic applications, including carbonic anhydrase inhibitory activity, HIV-I and HIV-II inhibitory activity, human cytomegalovirus inhibitory activity, and cholinesterase inhibitory activity and their structure–activity relationship. We hope that this review will encourage medicinal chemists to consider the potential benefits to incorporating sultone scaffold into future drug discovery.
