Abstract:PURPOSE:To establish a rabbit dry eye model with topical medication of the ocular preparation preservative benzalkonium chloride (BAC). METHODS:Sixteen white rabbits were used. One eye of each rabbit was chosen randomly for topical administration of 0.1% BAC twice daily for 14 days. The other untreated eyes served as controls. Schirmer test, fluorescein, and rose bengal staining were performed before and after BAC treatment on days 3, 5, 7, and 14. Conjunctiva impression cytology specimens were collected on days 0, 7, and 14. The rabbits were killed after day 14. Immunofluorescence staining was performed to detect mucin-5 subtype AC (MUC5AC) on conjunctival cryosections. Cornea and conjunctiva structures were evaluated by light and electron microscopy. RESULTS:Compared with untreated controls, BAC-treated eyes showed significant decreases in Schirmer scores (P = 0.01) and increases in fluorescein scores (P < 0.001) on days 5, 7, and 14. A significant increase in rose bengal scores was noticed as early as day 3 (P = 0.001). Decreases in goblet cell density occurred on days 7 and 14 (P = 0.001). Decreased MUC5AC and histopathologic and ultrastructural disorders of the cornea and conjunctiva were also observed in the BAC group. CONCLUSIONS:These findings demonstrated that an ophthalmic preservative, benzalkonium chloride, induced a dry eye syndrome in rabbits with damage to the cornea and conjunctiva, decreased aqueous tear basal secretion, goblet cell loss, and MUC5AC deficiency. This rabbit model was consistent with human dry eye syndrome in both aqueous tear and mucin deficiency and may be appropriate for studying dry eye syndrome.

Abstract:Aqueous-deficient dry eye (ADDE) is a type of dry eye disease that can result in the reduction of tear secretion quantity and quality. Prolonged abnormal tear production can lead to a disturbance in the ocular surface environment, including corneal damage and inflammation. In severe cases, ADDE can cause vision loss or even blindness. Currently, dry eye treatment is limited to eye drops or physical therapy, which can only alleviate eye discomfort symptoms and cannot fundamentally cure dry eye syndrome. To restore the function of the lacrimal gland in dry eye, we have created an animal model of lacrimal gland dysfunction in rats induced by scopolamine. Through the comprehensive evaluation of the lacrimal gland, corneas, conjunctivas, and other factors, we aim to provide a full understanding of the pathological changes of ADDE. Compared with the current dry eye mouse model, this ADDE animal model includes a functional evaluation of the lacrimal gland, providing a better platform for studying lacrimal gland dysfunction in ADDE.

Evaluation of Establishing Rabbit Dry Eye Model by Cauterizing the Meibomian Gland Orifices

Abstract:Objective To evaluate the characteristic of rabbit's dry eye model by cauterizing the meibomian gland orifices and search new rabbit's dry eye model. Methods Sixteen New Zealand white rabbits were experimented.The meibomian gland orifices of one eye of every rabbit were cauterized,the other eye was used as control.Eyelid swelling,lid margin hyperemia,lid margin eschar,meibomian gland orifices,rose bengal stain,fluorescence stain,ferning test and SchirmerⅠtest(after anesthesia)were performed before operation and 1,2,4,7,14,21,28,35,42 days after operation(SchirmerⅠtest was not carried out at 1,2 days after operating).Conjunctival impression cytology test was done at 14,28 and 42 days before and after operation.At the 45th day,eyelid,conjunctiva,and cornea tissues biopsy was carried out under optical microscope.Results At the first day after operating,all cauterized eyes presented eyelid swelling,lid margin hyperemia,and eschar at the cautery site,which was reduced in 7 to 9 days.All meibomian gland orifices were closed all along.By the 4th week,abnormal rose bengal stain and fluorescence stain could be found in the experiment group(P0.05).By the sixth week,Ⅲ,Ⅳ grade fern increased in the experiment group,and the conjunctival goblet cell density was lower than the control group.Under optical microscope examination,some meibomian gland ducts was dilated in the experiment group.Conclusion Rabbit dry eye model by cauterizing the meibomian gland orifices is simple and practical.
Induction of Chorioretinopathy by Adrenaline Injection in Different Strains of Rabbits

Abstract:Objective To establish a central serous chorioretinopathy (CSC) model on different strains of rabbits by intravenous injection of adrenaline,which may contribute to related researches of CSC.Methods This study was approved by Bioethics Committee of Fourth Military University and complied with Statement for the Use of Animals in Ophthalmic and Visual Research.Fundus fluorescein angiography (FFA) was initially performed on male New Zealand white rabbits (10),Belgium rabbits (5) and Chinchilla rabbits (10) to make sure that the retinas of subjects were normal.For the New Zealand white rabbits,adrenaline was injected via ear vein at a dose of 0.04 mg/kg once per day for the first 8 weeks and followed by a dose of 0.08 mg/kg for the next 4 weeks,while 0.04 mg/kg adrenaline was injected in the same way for 8 weeks in the Belgium rabbits and Chinchilla rabbits.FFA was performed every week after injection of adrenaline to evaluate the fluorescence leakage in ocular fundus.New Zealand white rabbits were sacrificed in 4 (3 rabbits),8 (3 rabbits) and 12 weeks (4 rabbits) after injection respectively,and Belgium rabbits and Chinchilla rabbits were sacrificed in the 8 weeks after injection.The eyeballs of the rabbits were enucleated to prepare the retinal sections for histopathological examination after hematoxylin-eosin staining.The results of FFA and retinal structure were compared among different strains of rabbits.Results No fluorescence leakage was found by FFA in ocular fundus,and the retinal structure was normal in all the 10 New Zealand white rabbits during the experiment.Fluorescence leakage was found by FFA in 2 Belgium rabbits at 1 week and 2 weeks after injection respectively,and retinal detachment and depigmentation of retinal pigment epithelium (RPE) with an enlarged intercellular space were shown by hematoxylin-eosin staining.For the Chinchilla rabbits,fluorescence leakages were found in 7 rabbits throughout the whole period of adrenaline administration.Circumscribed retinal detachment,depigmentation of RPE with enlarged intercellular space were also found in leakage lesions.Conclusions Repeated intravenous injection of adrenaline can induce CSC-like lesions in colored rabbits but not in albino rabbits.
Involvement of aberrant acinar cell proliferation in scopolamine-induced dry eye mice

Abstract:Dry eye is a multifactorial disease that causes dryness, inflammation and damage of ocular surface. Subcutaneous injection of the muscarinic cholinergic antagonist scopolamine under desiccating stress reduces tear production and induces dry eye symptoms in mice. However, the expression profile and pathogenic changes of the lacrimal gland remain incompletely understood. In the present study, we performed comparative transcriptomic analysis of lacrimal glands from the control and scopolamine-treated mice. Primary analysis identified 677 upregulated genes and 269 downregulated genes in the lacrimal gland of mice with scopolamine treatment. Unexpectedly, KEGG pathway and hierarchical clustering analysis showed the enrichment of "DNA replication" and "cell cycle" categories in the upregulated genes. Subsequently, we confirmed that the acinar cells were the major proliferating cells of lacrimal gland, which exhibited significant increasing of the proliferating cell nuclear antigen (PCNA) expression after scopolamine treatment, accompanied with the upregulation of DNA damage marker γ-H2AX. More importantly, both prophylactic and therapeutic administration of the cyclin-dependent kinase (CDK) inhibitor AT7519 rescued the tear reduction and alleviated dry eye severity in the scopolamine-treated mice, including corneal epithelial barrier function, lacrimal and corneal inflammation, and conjunctival goblet cell density. Therefore, we conclude that aberrant acinar cell proliferation is involved in the scopolamine-induced tear reduction and dry eye onset, which can be improved by AT7519 treatment.

Acupuncture Alleviates Corneal Inflammation in New Zealand White Rabbits with Dry Eye Diseases by Regulating α7nAChR and NF-κB Signaling Pathway

Abstract:Purpose. The purpose of this study is to determine the mechanism of improvement in dry eye diseases (DEDs) treated by acupuncture. The inflammatory molecules and related pathways will be analyzed in our study. Methods. In order to establish the animal model for DEDs, healthy New Zealand white rabbits were treated with scopolamine (Scop) hydrobromide for 21 consecutive days. After 21 days, acupuncture, fluorometholone (Flu), and α7nAChR antagonist (α-BGT) treatments were performed, and the Scop injections were continued until day 35. The therapeutic effect of acupuncture on DED inflammation was evaluated by corneal fluorescence staining, tear film rupture time, tear flow measurement, in vivo confocal microscopy (IVCM), corneal histopathology, and cytokine protein chip technology. The influence of acupuncture on the corneal pathology and inflammatory factors ACh, α7nAChR, and NF-κB was detected by enzyme-linked immunosorbent assay (ELISA) and western blot. Results. Compared with the group Scop, acupuncture can significantly reduce corneal staining and increase the tear film rupture time and tear flow, which are accompanied by a decrease in corneal epithelial detachment and lymphocyte infiltration. Acupuncture can relieve the inflammation of corneal stroma and mitigate the expression of proinflammatory factors and chemokines. Acupuncture can upregulate the expression of ACh and α7nAChR and downregulate the expression of NF-κB. Conclusion. Our findings demonstrate that acupuncture can alleviate corneal inflammation in New Zealand white rabbits with DEDs via α7nAChR and NF-κB signaling pathway regulation. The expression indicates that α7nAChR/NF-κB signaling pathway may be active and that acupuncture is a potential therapeutic target for dry eye.

An Animal Study on the Effect of Topically Administered Ambroxol for Dry Eye

Abstract:Objective: To evaluate the effect of 0.2% ambroxol eye drop on tear secretion and corneal healing on a rabbit dry eye model, and to delineate potential underlying mechanisms. Materials and method: A mixed mechanism dry eye model was created using 12 healthy New Zealand rabbits by excision of the main lacrimal glands, Harderian gland and nictitating membrane. Establishment of the model was confirmed by the decrease of Schirmer I and increase of corneal fluorescein staining scores. Two weeks after model creation, the rabbits were randomly and evenly divided into NaCl, 0.1% sodium hyaluronate and 0.2% ambroxol groups. Each group was administered the respective eye drops 4 times a day for four weeks. The Schirmer I test and corneal fluorescein staining were performed at two and four weeks. After four weeks of treatment, the animals were sacrificed and the conjunctiva and eyelid specimens collected. Inflammatory factors IL-8, TNF-α, and goblet cell specific mucin MUC5AC were measured by ELISA while the lid meibomian gland was evaluated by oil red O staining. Results: Compared with the baseline, 2 weeks after the surgery, Schirmer I test value decreased significantly (20.35 ± 5.18 mm/5 min vs 13.95 ± 4.64 mm/5 min, p < 0.01), and the fluorescein staining score increased significantly (0.5 ± 0.6 vs 5.5 ± 1.4, p < 0.01). After four weeks of treatment, compared with the NaCl and sodium hyaluronate groups, tear secretion in ambroxol group increased significantly (p < 0.01), while the corneal fluorescence staining score decreased significantly (p < 0.01). In the conjunctival tissue, significant decrease was seen in TNF-α (p < 0.01) and IL-8 [p (unilateral) < 0.05] concentrations in ambroxol group, and significant increase in MUC5AC concentration (p < 0.01) in ambroxol group as well. The lipid content in the lid meibomian glands appeared increased after the administration of ambroxol. Conclusion: The present rabbit dry eye model study demonstrated potentials of topically administered 0.2% ambroxol in stimulating tear and mucin secretion, inhibiting ocular surface inflammation, promoting corneal healing, and possibly augmenting meibomian gland lipid production.
Therapeutic efficiency of amniotic extraction for dry eye rabbit model

Abstract:AIM: To investigate therapeutic efficiency of amniotic extraction on dry eye in rabbit model induced by topical benzalkonium chloride (BAC). <br> METHODS: Totally 26 rabbits (26 right eyes) with dry eye model were studied and divided into two groups:group A (control group with PBS eye drops, n = 13) and group B ( amniotic extraction group, n = 13). Another two rabbits were chosen as normal control. The SchirmerⅠ tests ( S Ⅰ t) and corneal fluorescein staining ( FL) were made, and the tear total protein content, amylase activity, lactoferrin, lysozyme contents, goblet cell density were performed in two groups before treatment and 1, 2, 4 and 8 wk after treatment. <br> RESULTS: There were significant differences in SIT, FL scores, lysozyme activity and goblet cell density among different groups at different time points (P<0. 05). But, there was no significant differences in SⅠt, FL scores, lysozyme activity and goblet cell density between two groups before treatment (P>0. 05). After 8wks' treatment with PBS, the mean differences of the group A showed great changes in SⅠt, lysozyme and goblet cell density compared with those before treatment ( P < 0. 05); but there was no significant differences in FL scores compared with those before treatment (P>0. 05). As for group B, 8wks after treatment, there were statistical changes in SⅠt, FL, lysozyme (P<0. 05); but there was no significant differences in goblet cell density compared with those before treatment ( P > 0. 05). It was evident that statistical differences were observed in S Ⅰ t, FL scores, lysozyme activity and goblet cell density between two groups at each time point (P<0. 05). However, there were no significant differences in total protein, lactoferrin, amylase activity at different time points (P>0. 05). Meanwhile there was no significant differences in total protein, lactoferrin, amylase activity between two groups before treatment ( P > 0. 05 ). But there were significant differences in total protein, lactoferrin, amylase activity between two groups after 4 and 8 wks'treatment (P<0. 05). <br> CONCLUSION: Amniotic extraction has significant therapeutic effect on the dry eye in rabbit model.
An Experimental Study of Amniotic Lacrimal Duct Stents in the Treatment of Perimenopausal Female Rabbits with Dry Eye

Abstract:The aim of the present study was to investigate the effects of an amniotic membrane lacrimal stent in the treatment of dry eye syndrome in a perimenopausal rabbit model. In total, 48 healthy female rabbits were randomly divided into four groups (12 rabbits/group): Group A, sham-operated group; group B, negative control group; group C, sham-implantation group; and group D, implantation with amniotic membrane lacrimal stents. A Schirmer I test (SIT), corneal fluorescein staining, optical coherence tomography angiography and corneal confocal microscopy were conducted and the biomechanical properties of the amniotic membrane were measured prior to and 2, 4 and 6 weeks after the operations. There were marked differences in the amount of tear secretion and the SIT following the operation compared with prior to the operation. The amount of tear secretion of group B and C was significantly reduced (F=8.894; P=0.0017). Compared with groups A, B and C, group D demonstrated a significant increase in the amount of tear secretion and a significant reduction in SIT (P<0.05). However, there was no statistical difference between the tear secretion of the A, B and C groups (P>0.05). The cornea epithelial of groups A and D was significantly thinner compared with groups B and C. The superior part [superior temporal (ST)5, superior (S)5, superior nasal (SN)5, ST6, S6 and SN6] was significantly thinner compared with the central part of the cornea epithelium. The corneal epithelia of all groups were thin in the center and thick at the edge. At 6 weeks after the operation, group D exhibited a markedly decreased number of anterior stromal luminescent cells, compared with groups A, B and C. Furthermore, the bending degree of corneal stromal neural trunks was significantly improved, and the density, branches and curvature of corneal epithelium sub-basal nerves ameliorated to a certain extent (P<0.05). Amniotic membrane lacrimal stents demonstrated specific therapeutic effects on dry eye in perimenopausal female rabbits.

Abstract:Objective To explore the effects of amniotic lacrimal duct stenting on the prevention of dry eye in castrated rabbits.Methods Thirtysix healthy male rabbits were selected,the third eyelid were cut off and antiinfection treatment were given,which were randomly divided into 3 groups (12 cases in each group),the castrated male rabbits models were made.Among them,group A was negative control group,group B was dry eye model group,group C was group of lacrimal amniotic membrane group.At 2 weeks before implantation of amniotic lacrimal duct stent,2 weeks,4 weeks and 6 weeks after implantation,the fluorescent (FL) examination,Western blot,Schirmer I examination,immunofluorescence staining and corneal confocal microscopy were performed.Results The levels of tear secretion and FL in the three groups among different time points were significantly different (F=7.126,P =0.009;F =9.658,P =0.016),and there were significant differences among three groups (F =12.582,P =0.005;F =13.187,P =0.013).The tendency of tear secretion and FL in the three groups were also significantly changed (F =8.531,P =0.007;F =10.652,P =0.019).The epithelial basal cells at 6 weeks after implantation in three groups were 3811 ±414,3820 ± 314,2789 ± 353,and the density of inflammatory cells was 266 ±28,266 ± 29,67 ± 13,there were significant differences among three groups (F =13.442,P =0.012;F =9.231,P =0.021).The K1 6 staining in the duct epithelium were negative,and the expression of α-SMA in the lacrimal duct tissue of group A,B and C was not changed at all time points after implantation of amniotic lacrimal stent,and there was no significant difference (F =14.681,P =0.002).Conclusion The amniotic lacrimal stent implantation has certain effect on the prevention of dry eye in rabbit.
[Effect of androgen replacement therapy on mixed dry eye in a rabbit model]

Abstract:Objective: To explore the effect of androgen replacement therapy in a rabbit dry eye model characterized by androgen deficiency and meibomian gland dysfunction (MGD). Methods: Thirty 6-month-old male Chinchilla rabbits were randomly divided into the treatment group, model group and control group, with 10 rabbits in each group. In the treatment and model groups, 2/3 of the meibomian gland openings were closed by cauterization with electric coagulation pen, and bilateral testes were removed. One gram gel containing 1% testosterone was applied for 28 days on the skin of rabbits in the treatment group since day 28 after the surgery. The model group and control group received transdermal petrolatum instead. Tear secretion, tear breakup time (TBUT), corneal fluorescein staining, and serum free testosterone level were monitored throughout the study period. The globes and eyelids were collected for hematoxylin-eosin staining and periodic acid-Schiff staining. Conjunctival tissue was tested for the expression of miRNA-744-5p and interleukin-6. Meibum was collected for fatty acid analysis. Results: Animals presented with typical dry eye signs and androgen deficiency. After 28-day androgen replacement therapy, compared with the model group, the treatment group had a significantly higher tear secretion rate [(14.7±5.2) vs (10.3±3.6) mm, P=0.001], higher TBUT [(15.0±4.2) vs (10.2±3.6) s, P=0.003], lower fluorescein staining score [0 (0, 1) vs 2 (1, 4), P<0.001], and higher goblet cell density at conjunctival fornix (27.2±7.6 vs 10.7±4.8, P<0.001). Additionally, compared with the model group, the conjunctiva of the treatment group expressed a significantly lower level of miRNA-744-5p (1.67±0.24 vs 2.63±0.58, P<0.001) and interleukin-6 [2.38 (1.84, 4.61) vs 4.82 (3.99, 6.36), P=0.022]. Meanwhile, the treatment group showed significantly increased level of 16∶1, Δ9 fatty acid [(10.31±1.00)% vs (3.87±0.45)%, P<0.001] and iso-18∶0 fatty acid [(7.09±0.93)% vs (2.44±0.70)%, P<0.001], but decreased level of iso-26∶0 fatty acid [(5.72±1.07)% vs (8.02±0.65)%, P<0.001] in the meibum compared with the model group. Conclusion: Androgen replacement therapy can alleviate dry eye signs in rabbits presented with combined androgen deficiency and MGD.
The application of Mulberry eye drops on xerophthalmia in perimenopausal female rabbit model

Abstract:AIM: To investigate the therapeutic efficiency of Mulberry eye drops on dry eye in perimenopausal female rabbit model. METHODS: Totally 36 female New Zealand white rabbits were selected and operated with bilateral ovariectomy and fed for 60d postoperatively to make perimenopausal female rabbit dry eye model(36 eyes, all right eyes). And 24 of them were randomly divided into control group and experimental group(12 for each): the control group used PBS, the experimental group used Mulberry eye drops, the other 12 did not use any eye drops. The Schirmer I tests(SⅠt)and corneal fluorescein(FL)were made, and the tear total protein content, amylase activity, lactoferrin and lysozyme contents and confocal scanning microscopy were performed in two groups before treatment and 2, 4, 6wk after treatment. RESULTS: There was no significant difference in SⅠt,FL scores,total proteins,lysozyme,lactoferrin contents and amylase activity between two groups at pre-therapy(all P>0.05).After 2, 4 and 6wk of treatment, there were significant changes in SⅠt and FL scores in the two groups(P<0.05). There were no significant changes in lysozyme content, lactoferrin, amylase activity and total tear protein in the experimental group compared with those before treatment(all P>0.05), while there were significant changes in lysozyme content, lactoferrin, amylase activity and total tear protein in the control group(all P<0.05). At 2, 4 and 6wk after treatment, there were differences in the scores of SⅠt and FL, lysozyme content, lactoferrin, amylase activity and total protein content in tear between the two groups(P<0.05). At 6 wk after treatment, the mean number of corneal basal cells and inflammatory cells in control group were 4436±289mm2 and 321±91mm2,respectively,which in experimental group were 3219±223mm2 and 36±11mm2, respectively,there were statistical differences between two groups( all P<0.05). After 6wk treatment, there were no change of corneal nerve bending and less density in control group while the nerve fiber bending and density decreased significantly in experimental group,there were statistical differences between two groups(all P<0.05).CONCLUSION: Mulberry eye drops has significantly therapy effects on the dry eye induced by decreased androgen and it has a definite clinical application value.
Mucoadhesive Bletilla striata Polysaccharide-Based Artificial Tears to Relieve Symptoms and Inflammation in Rabbit with Dry Eyes Syndrome

Abstract:Dry eye syndrome (DES) is a multifactorial disorder of the ocular surface affecting many people all over the world. However, there have been many therapeutic advancements for the treatment of DES, substantial long-term treatment remains a challenge. Natural plant-based polysaccharides have gained much importance in the field of tissue engineering for their excellent biocompatibility and unique physical properties. In this study, polysaccharides from a Chinese ground orchid, Bletilla striata, were successfully extracted and incorporated into the artificial tears for DES treatment due to its anti-inflammatory and mucoadhesive properties. The examination for physical properties such as refractive index, pH, viscosity and osmolality of the Bletilla striata polysaccharide (BSP) artificial tears fabricated in this study showed that it was in close association with that of the natural human tears. The reactive oxygen species (ROS) level and inflammatory gene expression tested in human corneal epithelium cells (HCECs) indicated that the low BSP concentrations (0.01–0.1% v/v) could effectively reduce inflammatory cytokines (TNF, IL8) and ROS levels in HCECs, respectively. Longer retention of the BSP-formulated artificial tears on the ocular surface is due to the mucoadhesive nature of BSP allowing lasting lubrication. Additionally, a rabbit’s DES model was created to evaluate the effect of BSP for treating dry eye. Schirmer test results exhibited the effectiveness of 0.1% (v/v) BSP-containing artificial tears in enhancing the tear volume in DES rabbits. This work combines the effectiveness of artificial tears and anti-inflammatory herb extract (BSP) to moisturize ocular surface and to relieve the inflammatory condition in DES rabbit, which further shows great potential of BSP in treating ocular surface diseases like DES in clinics in the future.

Effect of fibrin glue amniotic rod on treatment of dry eye in models of castrated rabbit

Abstract:Objective To investigate the effect of fibrin glue amniotic rod on prevention and treatment of dry eye in models of castrated rabbit. Methods A total of 60 castrated male rabbits (60 right eyes) were randomly divided into: group A (sham group), group B (sham plus fibrin glue amniotic rod for 5 seconds), group C (sham plus fibrin glue amniotic rod for 6 weeks), group D (dry eye), group E (dry eye plus fibrin glue amniotic rod for 5 seconds) and group F (dry eye plus fibrin glue amniotic rod for 6 weeks). Animals were observed for 6 weeks after surgery. Schirmer I test (SIT), Corneal fluorescein (FL), Immunofluorescent staining on cornea and Western blot analysis of K16 expression were performed at baseline, 2 weeks, 4 weeks and 6 weeks after treatment. Results As for group C, SIT showed beneficial effect of fibrin glue amniotic rod sensitively on the rates at each recording time after treatment when compared with those in the baseline ( P<0.05). Group C had a significant improvement in terms of SIT in comparison with group A and B at various time points ( P<0.05). Group C had a significant improvement in terms of FL in comparison with group A and B at the time point of 4 weeks and 6 weeks after treatment ( P< 0.05). Group F experienced a dramatic amelioration of SIT and FL at the time point of 2 weeks, 4 weeks, and 6 weeks after treatment ( P<0.05). Epithelial basal cells and inflammatory cells were significantly decreased in group C and F ( P< 0.05), Immunofluorescent staining and Western blot showed downregulated expression of K16 in lacrimal mucosa in group C and F when compared with other groups ( P<0.05). Conclusion Fibrin glue amniotic rod can prevent and treat dry eye in models of castrated rabbit.

Abstract:Objective To investigate the experimental effect of mistletoe eye drops in prevention of xerophthalmia of castrated male rabbit. Method Thirty-six healthy male rabbits were chosen. The castrated dry eye models were established in 24 male rabbits and randomly divided into 2 groups, i.e. group A (mistletoe eye drop group) and group B (normal saline group). In the remaining 12 rabbits (12 eyes, group C) the scrotum was cut and the testes were saved. Schirmer test (SIT), corneal fluorescein (FL), tear proteins, and corneal condition under confocal microscope were checked before treatment and 2, 4 and 8 weeks after treatment. Results After treatment for 2, 4 or 8 weeks, SIT and FL were slightly changed in the group A without statistical differences ( p> 0.05), while SIT and FL were significantly worsened than those before treatment in the group B ( p<0.05). In the group A, tear protein, lactoferrin, lysozyme and amylase activity 2 weeks after treatment had statistical differences from those before treatment ( p<0.05). In the group B tear total protein content, lactoferrin, lysozyme and amylase activity at all time points significantly decreased compared with those before treatment ( p< 0.05). Observation of the base layer of corneal epithelium in the castrated male rabbits under corneal confocal microscope revealed that corneal inflammatory cells and epithelial basal cells were reduced in the group A. Conclusions Mistletoe eye drops can effectively reduce the signs and symptoms of dry eyes of the castrated male rabbits and have a certain preventive effect.

Abstract:Research on the effect of acupuncture for tear secretion and morphological changes of the lacrimal gland. Acupuncture therapy was given on the rabbits of New Zealand origin. The needles were inserted into the following acupoints around the right eye: Extra 1 (Taiyang), BL 2 ( Zanzhu) and SJ 23 (Sizhukong). The Schirmer Test I (S1T, The Schirmer test is probably the most commonly performed method of measuring aqueous tear production) values pre and post acupuncture therapy were blindly recorded. The lacrimal glands of the normal lacrimal gland group were stained by Hematoxylin and Eosin (HE) for light microscope examination and observed by transmission electron microscope. The S1T value was about sixty percent higher after acupuncture than before. Meanwhile changes also took place in the morphology of lacrimal gland tissu, indicated an active glandular function of synthesis and secretion. Acupuncture can increase lacrimal secretion by stimulating the rabbit lacrimal glandular function of synthesis and secretion.

Abstract:Introduction: In vivo screening studies, in which the efficacy of dozens of drugs is tested to select several applicants for further study of their safety in humans, are the main stage in the study of the pharmacodynamics of promising antiglaucoma drugs. This imposes a number of specific requirements both on experimental models of glaucoma and on laboratory animals used in the experiment. Materials and Methods: 32 male rabbits of the Soviet Сhinchilla breed, 6 male albino rabbits weighing 3-3.5 kg, and 20 outbred white rats weighing 220-250 g were used in total in experiments to reproduce the glaucoma process. All manipulations on the rabbit eye were performed by an ophthalmologist under general anesthesia with telazol. Triamcinolone (vitreous injection) was used to simulate glaucoma in rabbits, lauromacrogol 400 or fine kaolin (anterior chamber injection) was used to simulate glaucoma in rabbits; adrenaline hydrochloride (intraperitoneal administration) was used to simulate glaucoma in rats. Results and Discussion: Double intravitreal administration of a suspension of triamcinolone at a dose of 4 mg was the most attractive model in terms of the technique of reproducing the pathology and the results obtained in modeling glaucoma in rabbits. However, this model did not produce a stable increase in intraocular pressure (IOP). Doubling the dose of triamcinolone and replacing chinchilla rabbits with albinos did not lead to a positive result. The introduction of the venous sclerosing drug lauromacrogol 400 into the anterior chamber of the eye proved to be ineffective either. The introduction of finely dispersed kaolin into the anterior chamber of the eye of rabbits led to a persistent increase in IOP. The intraperitoneal administration of epinephrine hydrochloride to rats according to the described method gave no stable results. The increase in IOP became stable only after a significant increase in the dose of adrenaline. Conclusion: The conducted studies of four models of glaucoma and their three modifications in animals made it possible to select two of them, which contributed to a stable and fairly long-term increase in IOP in rabbits (introduction of finely dispersed kaolin into the anterior chamber of the eye) and rats (adrenaline-induced model).
Establishment of Dry Eye Model by Alkali Burns in Conjunctiva

Abstract:Objective To establish by treatment a new animal model of dry eye.Methods Dry eye model was established with 1.0 mmol/L NaOH solutions in both eyes of ten New Zealand white rabbits. Another ten rabbits were used as control. Examinations of dry eye perormed periodically. Conjunctival impression cytology and immunohistochemistry were performed.Results The level of congestion and secretion in conjunctiva, fluorescence stain and rose Bengal stain rose most significantly the 1st day and gradually decreased; Tear break-up time declined most distinctly on 7th day and was subsequently slowly prolonged, with statistical difference compared with the control(P0.05), Schirmer I test was not statistically different compared with the control(P0.05); the density of goblet cells and quantity of MUC5AC were statistically different during 4 weeks compared with those of the control(P0.05).Conclusion Alkali burn in conjunctiva is a simple and stable dry eye model.
Developing a model for aqueous deficient dry eye secondary to periglandular cicatrizing conjunctivitis

Abstract:Purpose: The current study used various techniques to develop a rabbit animal model of lacrimal gland damage caused by scarring conjunctivitis in the periglandular area. Methods: Left eyes of New Zealand white rabbits were injected with 0.1 ml of 1M NaOH subconjunctivally around superior and inferior lacrimal gland orifices (Group 1, n = 4), touched with 1M NaOH for 100 s to the superior and inferior fornices with conjunctival denuding (Group 2; n = 4), and electrocauterization to the ductal opening area (Group 3; n = 4). The ocular surface staining, Schirmer I, lacrimal gland, and conjunctival changes were observed at baseline,1, 4, 8, and 12 weeks. The degree of glandular inflammation, conjunctival fibrosis (Masson Trichrome), and goblet cell density (PAS) were also assessed. Results: At 12 weeks, the lacrimal glands of group 1 rabbits with periglandular injection showed severe inflammation with mean four foci/10HPF and a significant mean reduction in the Schirmer values by 7.6 mm (P = 0.007). Lacrimal glands had diffuse acinar atrophy, loss of myoepithelial cells, and ductular dilatation. The overlying conjunctiva showed fibrosis, goblet cell loss, and corneal vascularization in the inferotemporal quadrant. No lacrimal gland or ocular surface changes were observed in groups 2 and 3 at 12 weeks, except for localized subconjunctival fibrosis. Conclusion: Periglandular injection of 0.1 ml of 1M NaOH induced extensive lacrimal gland damage with reduced secretion and scarring in the subconjunctival plane compared to direct cauterization or direct NaOH contact to the ductal orifices of the rabbit lacrimal gland.
