Abstract:Small molecules in plants - such as metabolites, phytohormones, reactive oxygen species (ROS), and inorganic ions - participate in the processes of plant growth and development, physiological metabolism, and stress response. Wearable electrochemical sensors, known for their fast response, high sensitivity, and minimal plant damage, serve as ideal tools for dynamically tracking these small molecules. Such sensors provide producers or agricultural researchers with noninvasive or minimally invasive means of obtaining plant signals. In this review we explore the applications of wearable electrochemical sensors in detecting plant small molecules, enabling scientific assessment of plant conditions, quantification of environmental stresses, and facilitation of plant health monitoring and disease prediction.

Abstract:Significance Feeding a population of 9 billion in 2050 coupled with the changing climate and environmental stresses motivate us to develop advances in plant science and technology. We present a high-throughput plant phenotyping platform for detection of abiotic stress. The proposed Raman spectroscopic technique for high-throughput stress phenotyping and early stress detection in vivo improves sensitivity with the ability to interrogate individual molecules simultaneously in plants. This technology holds promise for mobile automated systems and precision agriculture.

Abstract:Phytohormones, as crucial regulatory factors in plant growth and development, have garnered increased interest in improving crop stress resistance. It is essential to comprehend the distribution of phytohormones in plants to assess their health status and investigate their functions. This knowledge also serves as a guide for developing and using plant growth regulators. The advancement of fluorescent probe technology, along with the wide range of fluorophores and improvements in imaging methods, has made it a successful approach for monitoring phytohormones in plants. This technique has been confirmed to be effective in plants, particularly in detecting the response of fluorescent probes to phytohormones. In this Perspective, we highlight the utility of fluorescent probes in measuring and visualizing the distribution of phytohormones in plants under external stress. However, the visualization of phytohormones with high spatial resolution and the achievement of high biocompatibility in living plants have posed significant challenges for researchers. Nonetheless, there are also many untapped opportunities in this field. This paper seeks to delve into the potential for further discussion on the subject.

Abstract:Monitoring plant physiological information for gaining a comprehensive understanding of plant growth and stress responses contributes to safeguarding plant health. Light-emitting probes - in terms of small-molecule, nanomaterials-based, and genetically protein-based probes - can be introduced into plants through foliar and root treatment or genetic transformation. These probes offer exciting opportunities for sensitive and in situ monitoring of dynamic plant chemical information - for example, reactive oxygen species (ROS), calcium ions, phytohormones - with spatiotemporal resolution. In this review we explore the sensing mechanisms of these light-emitting probes and their applications in monitoring various chemical information in plants in situ. These probes can be used as part of a sentinel plant approach to provide stress warning in the field or to explore plant signaling pathways.
In vivo surface-enhanced Raman scattering nanosensor for the real-time monitoring of multiple stress signalling molecules in plants

Abstract:When under stress, plants release molecules to activate their defense system. Detecting these stress-related molecules offers the possibility to address stress conditions and prevent the development of diseases. However, detecting endogenous signalling molecules in living plants remains challenging due to low concentrations of these analytes and interference with other compounds; additionally, many methods currently used are invasive and labour-intensive. Here we show a non-destructive surface-enhanced Raman scattering (SERS)-based nanoprobe for the real-time detection of multiple stress-related endogenous molecules in living plants. The nanoprobe, which is placed in the intercellular space, is optically active in the near-infrared region (785 nm) to avoid interferences from plant autofluorescence. It consists of a Si nanosphere surrounded by a corrugated Ag shell modified by a water-soluble cationic polymer poly(diallyldimethylammonium chloride), which can interact with multiple plant signalling molecules. We measure a SERS enhancement factor of 2.9 × 107 and a signal-to-noise ratio of up to 64 with an acquisition time of ~100 ms. To show quantitative multiplex detection, we adopted a binding model to interpret the SERS intensities of two different analytes bound to the SERS hot spot of the nanoprobe. Under either abiotic or biotic stress, our optical nanosensors can successfully monitor salicylic acid, extracellular adenosine triphosphate, cruciferous phytoalexin and glutathione in Nasturtium officinale, Triticum aestivum L. and Hordeum vulgare L.-all stress-related molecules indicating the possible onset of a plant disease. We believe that plasmonic nanosensor platforms can enable the early diagnosis of stress, contributing to a timely disease management of plants.
Multicolor Fluorescence Imaging for the Early Detection of Salt Stress in Arabidopsis

Abstract:Salt stress is one of the abiotic factors that causes adverse effects in plants and there is an urgent need to detect salt stress in plants as early as possible. Multicolor fluorescence imaging, as a powerful tool in plant phenotyping, can provide information about primary and secondary metabolism in plants to detect the responses of the plants exposed to stress in the early stage. The purpose of this study was to evaluate the potential of multicolor fluorescence imaging’s application in the early detection of salt stress in plants. In this study, the measurements were conducted on Arabidopsis and the multicolor fluorescence images were acquired at 440, 520, 690, and 740 nm with a self-developed imaging system consisting of a UV light-emitting diode (LED) panel for an excitation at 365 nm, a charge coupled device (CCD) camera, interference filters, and a computer. We developed a classification method using the imaging analysis of multicolor fluorescence based on principal component analysis (PCA) and a support vector machine (SVM). The results showed that the four principal fluorescence feature combinations were the ideal indicators as the inputs of the SVM model, and the classification accuracies of the control and salt-stress treatment at 5 days and 9 days were 92.65% and 98.53%, respectively. The results indicated that multicolor fluorescence imaging combined with PCA and SVM could act as a tool for early detection in salt-stressed plants.

Abstract:Most agricultural land, as a result of climate change, experiences severe stress that significantly reduces agricultural yields. Crop sensing by imaging techniques allows early-stage detection of biotic or abiotic stress to avoid damage and significant yield losses. Among the top certified imaging techniques for plant stress detection is chlorophyll a fluorescence imaging, which can evaluate spatiotemporal leaf changes, permitting the pre-symptomatic monitoring of plant physiological status long before any visible symptoms develop, allowing for high-throughput assessment. Here, we review different examples of how chlorophyll a fluorescence imaging analysis can be used to evaluate biotic and abiotic stress. Chlorophyll a is able to detect biotic stress as early as 15 min after Spodoptera exigua feeding, or 30 min after Botrytis cinerea application on tomato plants, or on the onset of water-deficit stress, and thus has potential for early stress detection. Chlorophyll fluorescence (ChlF) analysis is a rapid, non-invasive, easy to perform, low-cost, and highly sensitive method that can estimate photosynthetic performance and detect the influence of diverse stresses on plants. In terms of ChlF parameters, the fraction of open photosystem II (PSII) reaction centers (qp) can be used for early stress detection, since it has been found in many recent studies to be the most accurate and appropriate indicator for ChlF-based screening of the impact of environmental stress on plants.

Abstract:Abstract Increased exposure to environmental stresses due to climate change have adversely affected plant growth and productivity. Upon stress, plants activate a signaling cascade, involving multiple molecules like H 2 O 2, and plant hormones such as salicylic acid (SA) leading to resistance or stress adaptation. However, the temporal ordering and composition of the resulting cascade remains largely unknown. In this study we developed a nanosensor for SA and multiplexed it with H 2 O 2 nanosensor for simultaneous monitoring of stress-induced H 2 O 2 and SA signals when Brassica rapa subsp. Chinensis (Pak choi) plants were subjected to distinct stress treatments, namely light, heat, pathogen stress and mechanical wounding. Nanosensors reported distinct dynamics and temporal wave characteristics of H 2 O 2 and SA generation for each stress. Based on these temporal insights, we have formulated a biochemical kinetic model that suggests the early H 2 O 2 waveform encodes information specific to each stress type. These results demonstrate that sensor multiplexing can reveal stress signaling mechanisms in plants, aiding in developing climate-resilient crops and pre-symptomatic stress diagnoses.

Abstract:Plant responses to drought stress are complex due to various mechanisms of drought avoidance and tolerance to maintain growth. Traditional plant phenotyping methods are labor-intensive, time-consuming, and subjective. Plant phenotyping by integrating kinetic chlorophyll fluorescence with multicolor fluorescence imaging can acquire plant morphological, physiological, and pathological traits related to photosynthesis as well as its secondary metabolites, which will provide a new means to promote the progress of breeding for drought tolerant accessions and gain economic benefit for global agriculture production. Combination of kinetic chlorophyll fluorescence and multicolor fluorescence imaging proved to be efficient for the early detection of drought stress responses in the Arabidopsis ecotype Col-0 and one of its most affected mutants called reduced hyperosmolality-induced [Ca2+]i increase 1. Kinetic chlorophyll fluorescence curves were useful for understanding the drought tolerance mechanism of Arabidopsis. Conventional fluorescence parameters provided qualitative information related to drought stress responses in different genotypes, and the corresponding images showed spatial heterogeneities of drought stress responses within the leaf and the canopy levels. Fluorescence parameters selected by sequential forward selection presented high correlations with physiological traits but not morphological traits. The optimal fluorescence traits combined with the support vector machine resulted in good classification accuracies of 93.3 and 99.1% for classifying the control plants from the drought-stressed ones with 3 and 7 days treatments, respectively. The results demonstrated that the combination of kinetic chlorophyll fluorescence and multicolor fluorescence imaging with the machine learning technique was capable of providing comprehensive information of drought stress effects on the photosynthesis and the secondary metabolisms. It is a promising phenotyping technique that allows early detection of plant drought stress.

Abstract:Redox changes of pyridine nucleotides in cellular compartments are highly dynamic and their equilibria are under the influence of various reducing and oxidizing reactions. To obtain spatiotemporal data on pyridine nucleotides in living plant cells, typical biochemical approaches require cell destruction. To date, genetically encoded fluorescent biosensors are considered to be the best option to bridge the existing technology gap, as they provide a fast, accurate, and real-time readout. However, the existing pyridine nucleotides genetically encoded fluorescent biosensors are either sensitive to pH change or slow in dissociation rate. Herein, we employed the biosensors which generate readouts that are pH stable for in planta measurement of NADH/NAD(+) ratio and NADPH level. We generated transgenic Arabidopsis lines that express these biosensors in plastid stroma and cytosol of whole plants and pollen tubes under the control of CaMV 35S and LAT52 promoters, respectively. These transgenic biosensor lines allow us to monitor real-time dynamic changes in NADH/NAD(+) ratio and NADPH level in the plastids and cytosol of various plant tissues, including pollen tubes, root hairs, and mesophyll cells, using a variety of fluorescent instruments. We anticipate that these valuable transgenic lines may allow improvements in plant redox biology studies.
A SiPM-Enabled Portable Delayed Fluorescence Photon Counting Device: Climatic Plant Stress Biosensing

Abstract:A portable and sensitive time-resolved biosensor for capturing very low intensity light emission is a promising avenue to study plant delayed fluorescence. These weak emissions provide insight on plant health and can be useful in plant science as well as in the development of accurate feedback indicators for plant growth and yield in applications of agricultural crop cultivation. A field-based delayed fluorescence device is also desirable to enable monitoring of plant stress response to climate change. Among basic techniques for the detection of rapidly fluctuating low intensity light is photon counting. Despite its vast utility, photon counting techniques often relying on photomultiplier tube (PMT) technology, having restricted use in agricultural and environment measurements of plant stress outside of the laboratory setting, mainly due to the prohibitive cost of the equipment, high voltage nature, and the complexity of its operation. However, recent development of the new generation solid-state silicon photomultiplier (SiPM) single photon avalanche diode array has enabled the availability of high quantum efficiency, easy-to-operate, compact, photon counting systems which are not constrained to sophisticated laboratories, and are accessible owing to their low-cost. In this contribution, we have conceived, fabricated and validated a novel SiPM-based photon counting device with integrated plug-and-play excitation LED, all housed inside a miniaturized sample chamber to record weak delayed fluorescence lifetime response from plant leaves subjected to varying temperature condition and drought stress. Findings from our device show that delayed fluorescence reports on the inactivation to the plant's photosystem II function in response to unfavorable acute environmental heat and cold shock stress as well as chronic water deprivation. Results from our proof-of-concept miniaturized prototype demonstrate a new, simple and effective photon counting instrument is achieved, one which can be deployed in-field to rapidly and minimally invasively assess plant physiological growth and health based on rapid, ultra-weak delayed fluorescence measurements directly from a plant leaf.

Abstract:Photosynthesis is a pivotal process in plant physiology, and its regulation plays an important role in plant defense against biotic stress. Interactions with pathogens and pests often cause alterations in the metabolism of sugars and sink/source relationships. These changes can be part of the plant defense mechanisms to limit nutrient availability to the pathogens. In other cases, these alterations can be the result of pests manipulating the plant metabolism for their own benefit. The effects of biotic stress on plant physiology are typically heterogeneous, both spatially and temporarily. Chlorophyll fluorescence imaging is a powerful tool to mine the activity of photosynthesis at cellular, leaf, and whole-plant scale, allowing the phenotyping of plants. This review will recapitulate the responses of the photosynthetic machinery to biotic stress factors, from pathogens (viruses, bacteria, and fungi) to pests (herbivory) analyzed by chlorophyll fluorescence imaging both at the lab and field scale. Moreover, chlorophyll fluorescence imagers and alternative techniques to indirectly evaluate photosynthetic traits used at field scale are also revised.

Abstract:Globally, 91% of plant production encounters diverse environmental stresses that adversely affect their growth, leading to severe yield losses of 50–60%. In this case, monitoring the connection between the environment and plant health can balance population demands with environmental protection and resource distribution. Fluorescent chemosensors have shown great progress in monitoring the health and environment of plants due to their high sensitivity and biocompatibility. However, to date, no comprehensive analysis and systematic summary of fluorescent chemosensors used in monitoring the correlation between plant health and their environment have been reported. Thus, herein, we summarize the current fluorescent chemosensors ranging from their design strategies to applications in monitoring plant-environment interaction processes. First, we highlight the types of fluorescent chemosensors with design strategies to resolve the bottlenecks encountered in monitoring the health and living environment of plants. In addition, the applications of fluorescent small-molecule, nano and supramolecular chemosensors in the visualization of the health and living environment of plants are discussed. Finally, the major challenges and perspectives in this field are presented. This work will provide guidance for the design of efficient fluorescent chemosensors to monitor plant health, and then promote sustainable agricultural development.

Abstract:Determination of plant stresses such as infections by plant pathogens is currently dependent on time-consuming and complicated analytical technologies. Here, we report a leaf-attachable chemiresistive sensor array for real-time fingerprinting of volatile organic compounds (VOCs) that permits noninvasive and early diagnosis of plant diseases, such as late blight caused by Phytophthora infestans. The imperceptible sensor patch integrates an array of graphene-based sensing materials and flexible silver nanowire electrodes on a kirigami-inspired stretchable substrate, which can minimize strain interference. The sensor patch has been mounted on live tomato plants to profile key plant volatiles at low-ppm concentrations with fast response (<20 s). The multiplexed sensor array allows for accurate detection and classification of 13 individual plant volatiles with >97% classification accuracy. The wearable sensor patch was used to diagnose tomato late blight as early as 4 days post inoculation and abiotic stresses such as mechanical damage within 1 h.

Abstract:Chlorophyll fluorescence is interesting for phenotyping applications as it is rich in biological information and can be measured remotely and non-destructively. There are several techniques for measuring and analysing this signal. However, the standard methods use rather extreme conditions, e.g., saturating light and dark adaption, which are difficult to accommodate in the field or in a greenhouse and, hence, limit their use for high-throughput phenotyping. In this article, we use a different approach, extracting plant health information from the dynamics of the chlorophyll fluorescence induced by a weak light excitation and no dark adaption, to classify plants as healthy or unhealthy. To evaluate the method, we scanned over a number of species (lettuce, lemon balm, tomato, basil, and strawberries) exposed to either abiotic stress (drought and salt) or biotic stress factors (root infection using Pythium ultimum and leaf infection using Powdery mildew Podosphaera aphanis). Our conclusions are that, for abiotic stress, the proposed method was very successful, while, for powdery mildew, a method with spatial resolution would be desirable due to the nature of the infection, i.e., point-wise spread. Pythium infection on the roots is not visually detectable in the same way as powdery mildew; however, it affects the whole plant, making the method an interesting option for Pythium detection. However, further research is necessary to determine the limit of infection needed to detect the stress with the proposed method.

Abstract:The field of plant phenotype is used to analyze the shape and physiological characteristics of crops in multiple dimensions. Imaging, using non-destructive optical characteristics of plants, analyzes growth characteristics through spectral data. Among these, fluorescence imaging technology is a method of evaluating the physiological characteristics of crops by inducing plant excitation using a specific light source. Through this, we investigate how fluorescence imaging responds sensitively to environmental stress in garlic and can provide important information on future stress management. In this study, near UV LED (405 nm) was used to induce the fluorescence phenomenon of garlic, and fluorescence images were obtained to classify and evaluate crops exposed to abiotic environmental stress. Physiological characteristics related to environmental stress were developed from fluorescence sample images using the Chlorophyll ratio method, and classification performance was evaluated by developing a classification model based on partial least squares discrimination analysis from the image spectrum for stress identification. The environmental stress classification performance identified from the Chlorophyll ratio was 14.9% in F673/F717, 25.6% in F685/F730, and 0.209% in F690/F735. The spectrum-developed PLS-DA showed classification accuracy of 39.6%, 56.2% and 70.7% in Smoothing, MSV, and SNV, respectively. Spectrum pretreatment-based PLS-DA showed higher discrimination performance than the existing image-based Chlorophyll ratio.

Abstract:Plant stress detection is considered one of the most critical areas for the improvement of crop yield in the compelling worldwide scenario, dictated by both the climate change and the geopolitical consequences of the Covid-19 epidemics. A complicated interconnection of biotic and abiotic stressors affect plant growth, including water, salt, temperature, light exposure, nutrients availability, agrochemicals, air and soil pollutants, pests and diseases. In facing this extended panorama, the technology choice is manifold. On the one hand, quantitative methods, such as metabolomics, provide very sensitive indicators of most of the stressors, with the drawback of a disruptive approach, which prevents follow up and dynamical studies. On the other hand qualitative methods, such as fluorescence, thermography and VIS/NIR reflectance, provide a non-disruptive view of the action of the stressors in plants, even across large fields, with the drawback of a poor accuracy. When looking at the spatial scale, the effect of stress may imply modifications from DNA level (nanometers) up to cell (micrometers), full plant (millimeters to meters), and entire field (kilometers). While quantitative techniques are sensitive to the smallest scales, only qualitative approaches can be used for the larger ones. Emerging technologies from nuclear and medical physics, such as computed tomography, magnetic resonance imaging and positron emission tomography, are expected to bridge the gap of quantitative non-disruptive morphologic and functional measurements at larger scale. In this review we analyze the landscape of the different technologies nowadays available, showing the benefits of each approach in plant stress detection, with a particular focus on the gaps, which will be filled in the nearby future by the emerging nuclear physics approaches to agriculture.

An AIE-based ratiometric fluorescent probe for highly selective detection of H2S in plant stress responses

Abstract:Hydrogen sulfide (H2S) has emerged as a crucial signaling molecule in plant stress responses, playing a significant role in regulating various physiological and biochemical processes. In this study, we report an aggregation-induced emission (AIE)-based ratiometric fluorescent probe TPN-H2S for the highly selective detection of H2S in plant tissues. The probe exhibited excellent sensitivity and selectivity towards H2S over other analytes, enabling real-time monitoring of H2S dynamics in living cell. Furthermore, the AIE-based ratiometric probe TPN-H2S allowed for accurate quantification of H2S levels, providing valuable insights into the spatiotemporal distribution of Cys metabolism produces H2S. Importantly, the physiological pathways and signaling mechanisms of H2S production of was investigated in plant tissues under Cr and nano-plastics stress. Utilizing a high-throughput screening approach, we identified exogenous substances such as calcium chloride (CaCl2) and abscisic acid (ABA) that could induce higher level of H2S production during the stress response in plants. Overall, those findings demonstrate the potential of the AIE-based ratiometric fluorescent probe TPN-H2S as a powerful tool for unraveling the role of H2S in plant stress responses and pave the way for further exploration of H2S-mediated signaling pathways in plants.

Abstract:Environmental stresses on plants are among the major limitations to crop productivity. Plants are naturally equipped with a set of acclimatory mechanisms enabling them to withstand stresses without irreversible damage. Here, we overhauled the approach to non-invasive gauging of the response of maize to combined drought and high-light stresses. This was done to leverage the advantages of a recently developed framework based on using the reflected signal absorption coefficient modality. We document the deployment of two responses, one based on non-photochemical quenching (NPQ) of the absorbed light energy and the second on the adjustment of leaf photosynthetically active radiation (PAR) interception. The NPQ-based response, implemented by the xanthophyll cycle and inferred from changes in the reflection of light in the blue-green region of the spectrum, engaged at the beginning of the stress but quickly reached a plateau. We demonstrate that altering leaf absorption in PAR is a fundamental plant response mechanism to diverse stresses. This response is quantified by the PAR absorption coefficient retrieved from the non-destructively measured reflectance. A profound decrease in the PAR absorption coefficient in the whole PAR region was shown to be a reliable measure of the degree of stress regardless of its cause. The effects of superficial shading and leaf position were readily detectable by the leaf PAR absorption coefficient. We consider this report a proof of concept for quantifying plant stress and monitoring the acclimation state via the absorption coefficient in the PAR region through non-destructive, remotely sensed techniques.

Abstract:Revolutionary new technologies, namely in the areas of DNA sequencing and molecular imaging, continue to impact new discoveries in plant science and beyond. For decades we have been able to determine properties of enzymes, receptors and transporters in vitro or in heterologous systems, and more recently been able to analyze their regulation at the transcriptional level, to use GFP reporters for obtaining insights into cellular and subcellular localization, and tp measure ion and metabolite levels with unprecedented precision using mass spectrometry. However, we lack key information on the location and dynamics of the substrates of enzymes, receptors and transporters, and on the regulation of these proteins in their cellular environment. Such information can now be obtained by transitioning from in vitro to in vivo biochemistry using biosensors. Genetically encoded fluorescent protein-based sensors for ion and metabolite dynamics provide highly resolved spatial and temporal information, and are complemented by sensors for pH, redox, voltage, and tension. They serve as powerful tools for identifying missing processes (e.g., glucose transport across ER membranes), components (e.g., SWEET sugar transporters for cellular sugar efflux), and signaling networks (e.g., from systematic screening of mutants that affect sugar transport or cytosolic and vacuolar pH). Combined with the knowledge of properties of enzymes and transporters and their interactions with the regulatory machinery, biosensors promise to be key diagnostic tools for systems and synthetic biology.
