Abstract:Aza-BODIPY,developed from BODIPYs,have advantages of large molar absorption coefficients,narrow half peak widths,long fluorescence life,good optical stability and tunable fluorescence;hence they have been selected as excellent candidates for near infrared colorimetric and fluorescent probes. However,most of them are poor water solubility and the field of their application is limited. In this paper,the recent researches on Aza-BODIPY-based fluorescent probes were reviewed. Three methods of synthesis of Aza-BODIPYs are summarized briefly including the O'Shea method starting from chalcone,the Carreira method using 2,4-substituted pyrroles as starting materials and the Lukyanets method for fused-ring-Aza-BODIPYs. The sensing performances,mechanisms and practical applications of various probes for pH,H2O2,NH4+,F–,Hg2+,CN–,damtoxin and Cys are discussed. Although there are limited reports about Aza-BODIPY-based probes,they are expected to be developed in the biological and environment areas with the improvement of water solubility,the longer excitation and emission wavelengths in the near-infrared region and the increase of the type and range of identifying species by structural modifications.
Twisted Intramolecular Charge Transfer and Aggregation-Induced Emission of BODIPY Derivatives

Abstract:Boron dipyrromethene (BODIPY) derivatives 1 and 2 consisting of donor and acceptor units with dual photoresponses to solvent polarity and luminogen aggregation are developed through taking advantage of twisted intramolecular charge transfer (TICT) and aggregation-induced emission (ATE) processes. In nonpolar solvents, the locally excited (LE) states of the BODIPY luminogens emit intense green lights. Increasing solvent polarity brings the luminogens from the LE state to the TICT state, causing a large bathochromic shift in the emission color but a dramatic decrease in the emission efficiency. The red emission is greatly boosted by aggregate formation or ATE effect: addition of large amounts of water into the solutions of 1 and 2 in the polar solvents causes the luminogens to aggregate supramolecularly and to emit efficiently. The emission can be enhanced by increasing solvent viscosity and decreasing solution temperature, indicating that the ATE effect is caused by the restriction of the intramolecular rotations in the aggregates of the luminogens.

Abstract:On page 771, Bin Liu, Ben Zhong Tang, and co-workers report the synthesis of 2-(2,6-bis((E)-4-(phenyl(4′-(1,2,2-triphenylvinyl)-[1,1′-biphenyl]-4-yl)amino)styryl)-4Hpyran-4-ylidene)malononitrile (TPE-TPA-DCM), which has an aggregation-induced emission (AIE) feature. Further formulation of TPE-TPA-DCM using bovine serum albumin (BSA) as the polymer matrix allows for successful in vitro and in vivo farred/near-infrared (FR/NIR) bioimaging.

Abstract:A series of naphthalimide-tetrazines were developed as bioorthogonal fluorogenic probes, which could produce significant fluorescence enhancement, notable aggregation-induced emission (AIE) characters and multicolor emissions after bioorthogonal reaction with strained dienophiles. Manipulating the pi-bridge in the fluorophore skeleton allows fine-tuning of the emission wavelength and influences the AIE-active properties. With these probes, we succeeded in no-wash fluorogenic protein labeling and mitochondria-selective bioorthogonal imaging in live cells.

Abstract:Organic fluorescent probes are widely used in bioimaging and bioassays, but the notorious photobleaching hampers their applications. Encapsulation of organic dyes into nanoparticles (NPs) is an effective strategy to minimize photobleaching, but classical organic dye molecules tend to have their fluorescence quenched in aggregate states, which is termed aggregation-caused quenching (ACQ). Here we demonstrate our attempt to tackle this problem through the aggregation-induced emission (AIE) strategy. 3,4: 9,10-Tetracarboxylic perylene bisimide (PBI) is a well-known organic dye with a serious ACQ problem. By attaching two tetraphenylethene (TPE) moieties to the 1,7-positions, the ACQ-characteristic PBI-derivative was converted to an AIE-characteristic molecule. The obtained PBI derivative (BTPEPBI) exhibits several advantages over classical PBI derivatives, including pronounced fluorescence enhancement in aggregate state, red to near infrared emission, and facile fabrication into uniform NPs. Studies on the staining of MCF-7 breast cancer cells and in vivo imaging of a tumor-bearing mouse model with BTPEPBI-containing NPs reveal that they are effective fluorescent probes for cancer cell and in vivo tumor diagnosis with high specificity, high photostability and good fluorescence contrast.
Red/near-infrared (NIR) difluoroboron β-diketonate derivatives with reversible mechanochromism for cellular imaging

Abstract:Near-infrared (NIR) fluorophores have promoted the development of materials for bioimaging, but traditional NIR dyes usually suffer from aggregation-caused quenching (ACQ), impeding their applications. Herein, we propose two difluoroboron β-diketonate complexes TBO and TBS, consisting a donor-acceptor (D-A) structure with triphenylamine (TPA) moiety as an electron donors and difluoroboron as well as furan or thiophene building block as an electron acceptor. The theoretical calculation and optical data shows that both of them have intramolecular charge transfer (ICT) characteristics. Such ICT characteristics endow them with both solvatochromism and dual-state emission (DSE) properties. In the solvent CH2Cl2, the emission wavelength of TBO ranges from 550 nm to 750 nm, with a low fluorescence quantum yield (Φ = 7.0 %). However, in the less polar solvent hexane, the emission wavelength blue-shifts, with an increased Φ reaching up to 18 %. Moreover, TBO and TBS exhibit mechanochromic characteristics and rare multi-channel fluorescence emission phenomena at solid-state. Their solid-state samples can emit fluorescence in four spectral bands with maximum emission wavelengths at 300 nm, 400 nm, 600 nm, and 770 nm under excitation at 240 nm. These unique optical properties are expected to be utilized for detecting polarity of system and deformation. Moreover, according to the results of cell imaging and flow cytometry, TBO molecular were easily internalized into Hela cells and distributed in the cytoplasm with strong red fluorescence. Therefore, this research inspires more insight into development of NIR luminogens for biomedical imaging.

Abstract:Fluorescent probes play a key role in modern biomedical research. As compared to inorganic quantum dots (QDs) composed with heavy metal elements, organic dye-based fluorescent nanoparticles have higher biocompatibility and are richer in variety. However, traditional organic fluorophores tend to quench fluorescence upon aggregation, which is known as aggregation-caused quenching (ACQ) effect that hinders the fabrication of highly emissive fluorescent nanoparticles. In this work, we demonstrate the synthesis of organic fluorescent dots with aggregation-induced emission (AIE) in far-red/near-infrared (FA/NIR) region. A conventional ACQ-characteristic fluorescent dye, 3,4:9,10-tetracarboxylic perylene bisimide (PBI), is converted into an AIE fluorogen through attaching two tetraphenylethylene (TPE) moieties. The fluorescent dots with surface folic acid groups are fabricated from PBI derivative (DTPEPBI), showing specific targeting effect to folate receptor-overexpressed cancer cells. In vivo studies also suggest that the folic acid-functionalized AIE dots preferentially accumulate in the tumor site through enhanced permeability and retention (EPR) effect and folate receptor-mediated active targeting effect. The low cytotoxicity, good FR/NIR contrast and excellent targeting ability in in vitro / in vivo imaging indicate that the AIE dots have great potentials in advanced bioimaging applications.

Abstract:BODIPY (4,4-difluoro-4-bora-3a,4a-diaza-s-indacene) is an emissive chromophore in solutions but suffers from fluorescence quenching when aggregated due to its flat molecular conformation and small Stokes shift. To create aggregate-state emissive BODIPY luminogens, tetraphenylethene (TPE), which is a popular luminogen with intriguing aggregation-induced emission (AIE) characteristic, is introduced as periphery to a methylated BODIPY core. Three TPE-BODIPY adducts are synthesized and characterized, and their photophysical properties and electronic structures are investigated. The incorporation of AIE-active TPE units alleviates aggregation-caused quenching of BODIPY core, furnishing emissive nanoparticles based on TPE-BODIPY adducts. Significantly, the two-photon absorption (TPA) and two-photon excited fluorescence (TPEF) properties are improved as more TPE units are attached. The luminogens with 3TPE units (3TPE-BODIPY) shows the strongest TPA and TPEF in the wavelength range of 750-830 nm, with cross-section values of 264 and 116 GM at 810 nm, respectively. Red emissive nanoparticles with a Stokes shift of 60 nm and a fluorescence quantum yield of 16% are attained by encapsulating 3TPE-BODIPY with 1,2-sistearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000]. The nanoparticles are biocompatible and function well in TPEF cellular imaging and mouse brain blood vascular visualization.
Tetraphenylethylene- and fluorene-functionalized near-infrared aza-BODIPY dyes for living cell imaging

Abstract:Four near-infrared (NIR) boron-azadipyrromethene (aza-BODIPY) dyes 5a-d bearing tetraphenylethylene- and fluorenyl substituents at the 1,7-position or 3,5-position were synthesized and characterized by H-1 NMR, C-13 NMR and HRMS. The single crystal structure of compound 5c was studied by X-ray crystallography. UV/vis and fluorescence spectroscopic studies of the aza-BODIPY dyes 5a-d indicated that the absorption and emission maxima of these dyes were bathochromically shifted up to 48 nm in comparison with that of the 1,3,5,7-tetraphenyl aza-BODIPY. Moreover, fluorescence quantum yields up to 0.45 were observed for the new dyes. The redox properties of these dyes were investigated by cyclic voltammetry. The observed optical and electrochemical properties of these dyes were further elucidated by the DFT calculation of the frontier molecular orbitals. Owing to their outstanding fluorescence properties, these dyes can be applied as fluorescence imaging probes for living cells.

Abstract:Three novel donor-pi-acceptor two-photon absorption compounds (1PZPy, 2PZIm, 3CZPy) bearing the 10-butyl-10H-phenothiazine (9-butyl-9H-carbazole) donor, the pyridinium (benzimidazolium) acceptor, and the 2,5-divinylthiophene pi-bridge were synthesized and fully characterized by H-1 NMR, C-13 NMR, FT-IR, and HRMS. The linear and nonlinear photophysical properties were systematically investigated. Their absorption properties show a strong solvent dependence, while the emission properties are nearly independent of solvent polarity. All of them possess large Stokes shifts (Delta lambda=149-190 nm in H2O). 1PZPy and 3CZPy exhibit red fluorescence emission centered at about 635 and 660 nm, respectively. The two-photon absorption cross-sections measured by the open aperture Z-scan technique are determined to be 486 (1PZPy), 601 (2PZIm), and 753 GM (3CZPy) in DMF. The density functional theory calculations were further carried out to reveal their electronic structures. All the target compounds are verified to have low cytotoxicity in the working solution and good capability for one- and two-photon excitation fluorescence imaging, suggesting their potential application in bioimaging. Moreover, they show the organelle targeting ability in living cells with the high Pearson's coefficients above 0.94 for the endoplasmic reticulum.

Abstract:The development of biological fluorescent probes is of great significance to the field of cancer bio-imaging. However, most current probes within the bulky hydrophobic group have limited application in aqueous medium and restricted imaging under physiological conditions. Herein, we proposed two efficient molecules to study their physical properties and imaging work, and the absorption and fluorescence intensity were collected with varying ions attending in aqueous medium. We enhance the water solubility through the quaternization reaction and form a balance between hydrophilic and hydrophobicity with dipyrrome-theneboron difluoride (BODIPY) fluorophore. We introduced pyridine and dimethylaminopyridine (DMAP) by quaternization and connected the BODIPY fluorophore by ethylenediamine. The final synthesized probes have achieved ideal affinity with HeLa cells (human cervical carcinoma cell line) in live-cell imaging which could be observed by Confocal Microscope. The probes also have a good affinity with subcutaneous tumor cells in mice in in vivo imaging, which may make them candidates as oncology imaging probes.

Abstract:Multi‐resonant thermally activated delayed fluorescence (MR‐TADF) emitters have attracted strong interest for use in electroluminescent devices due to their high photoluminescence quantum yield and narrowband emission. These properties, along with red emission, are also crucial for bioimaging probes. In this work, two red‐emissive MR‐TADF emitters are designed and characterized and then used for the first time as imaging probes via encapsulation in glassy organic dots. Multi‐resonant thermally activated delayed fluorescence (MR‐TADF) emitters have attracted strong interest for organic electroluminescent devices due to their high photoluminescence quantum yield (ΦPL) and narrowband emission. These properties are also crucial for high‐performance biological probes, especially red emitters. Orange and red MR‐TADF emitters, PhDPA‐DiKTa and MeODPA‐DiKTa, are designed by decorating the DiKTa core with di([1,1′‐biphenyl]‐4‐yl)amine and bis(4‐methoxyphenyl)amine, respectively. As 5 wt.% doped films in 1,3‐di(9H‐carbazol‐9‐yl)benzene (mCP), PhDPA‐DiKTa emits at λPL of 617 nm, while MeODPA‐DiKTa emits at λPL of 655 nm. Both show delayed fluorescence, with delayed lifetimes, τd, of 658.4 and 249.2 μs, respectively. Water‐dispersible glassy organic dots (g‐Odots) based on these materials are prepared by encapsulating them and mCP host into an amphiphilic DSPE‐PEG2k polymer. Both families of g‐Odots showed a deeper red emission and enhanced ΦPL compared to the corresponding 5 wt.% doped films in mCP (λPL = 618 nm, ΦPL = 77% for PhDPA‐DiKTa g‐Odots, λPL = 663 nm, ΦPL = 38% for MeODPA‐DiKTa g‐Odots). The TADF character of the emitters is conserved in the g‐ODots, with τd of 203.9 μs for PhDPA‐DiKTa g‐Odots and 131.6 μs for MeODPA‐DiKTa g‐Odots. These MR‐TADF g‐Odots are successfully demonstrated as biological imaging probes of HeLa cells.

Abstract:In this work, a series of new diketopyrrolopyrrole-based dyes have been synthesized by connecting tetraphenylethene to the diketopyrrolopyrrole core. All the four compounds exhibit good aggregation induced emission property with nonemissive in the solution but strong red fluorescence in the aggregate or solid state. Also, these new dyes exhibit large two-photon absorption cross sections (a), in which the a data measured by the open aperture Z-scan technique are determined to be 150, 300, 1140 and 1016 GM for four dyes, respectively. In addition, compound with stilbene and four tetraphenylethene units (DPP-TPE-3) was used as the luminogen and encapsulated into nanoparticles for cell imaging and two-photon excited fluorescence blood vessels imaging. The result indicates that it can be used as the effective fluorescence probe for bioimaging and has great potential for bioapplications. (C) 2016 Elsevier Ltd. All rights reserved.

Abstract:Small-molecule fluorophores enable the observation of biomolecules in their native context with fluorescence microscopy. Specific labeling via bio-orthogonal tetrazine chemistry combines minimal label size with rapid labeling kinetics. At the same time, fluorogenic tetrazine–dye conjugates exhibit efficient quenching of dyes prior to target binding. However, live-cell compatible long-wavelength fluorophores with strong fluorogenicity have been difficult to realize. Here, we report close proximity tetrazine–dye conjugates with minimal distance between tetrazine and the fluorophore. Two synthetic routes give access to a series of cell-permeable and -impermeable dyes including highly fluorogenic far-red emitting derivatives with electron exchange as the dominant excited-state quenching mechanism. We demonstrate their potential for live-cell imaging in combination with unnatural amino acids, wash-free multicolor and super-resolution STED, and SOFI imaging. These dyes pave the way for advanced fluorescence imaging of biomolecules with minimal label size.This work presents probes that turn fluorescent upon specific chemical reactions in biological environments, which are highly beneficial for signal improvement in fluorescence microscopy.This article has not yet been cited by other publications.

Abstract:A series of BODIPY probes with a wide emission range were prepared via aminoacylation at the <i>meso</i>-position. Functional moieties were also introduced to induce bathochromic shifts in emission, improve water solubility, increase Stokes shifts, and construct bioorthogonal turn-on probes. The developed analogues were successfully used in live-cell imaging, suggesting that the described strategy can be used to prepare probes with improved bioimaging potential.

Abstract:Red/near-infrared dyes are highly demanded for biological applications but most of them are far from satisfactory. In this work, a series of red/nearinfrared fluorophores based on electron-withdrawing benzo[1,2-b:4,5-b']dithiophene 1,1,5,5-tetraoxide (BDTO) are synthesized and characterized. They possess both aggregation-induced emission, and hybridized local and charge-transfer characteristics. Crystallographic, spectroscopic, electrochemical and computational results reveal that the oxidation of benzo[1,2-b: 4,5-b']dithiophene to BDTO can endow the fluorophores with greatly red-shifted emission, enhanced emission efficiency, reduced energy levels, enlarged two-photon absorption cross section, and increased reactive oxygen species generation efficiency. The nanoparticles fabricated with a near-infrared fluorophore TPA-BDTO show high photostability and biocompatibility with good performance in targeted photodynamic ablation of cancer cells and two-photon fluorescence imaging of intravital mouse brain vasculature.
