Abstract:BACKGROUND: Toxoplasmosis is caused by the Toxoplasma gondii parasite. The parasite is intracellular and can result in severe complications leading to death in immuno-deficient patients in particular. Diabetes is an important factor that increases susceptibility and risk of various infections in the host.OBJECTIVES: The present study focused on the serologic detection of Toxoplasma gondii infection in diabetic patients.MATERIALS AND METHODS: Through a case-control study, overall 184 serum samples including 91 from diabetic cases and 93 from healthy non-diabetic controls were investigated. Cases and controls were matched for age and gender. Serum samples were tested for sugar by an enzymatic method, and IgG antibodies were tested against Toxoplasma gondii by ELISA method.RESULTS: The prevalence of IgG antibodies against Toxoplasma gondii in diabetic patients and healthy controls were 60.43% and 38% respectively. Risk of toxoplasmosis infection in diabetic patients with was two folds higher than healthy controls (RR = 2.21, 95% CI; 1.6 - 3.7, P = 0.001).CONCLUSIONS: Diabetes may be caused by Toxoplasma gondii. Presence of T. gondii in the pancreas at the same time could directly undermines the pancreas cells. When β cells are destroyed, insulin secretion would then be affected. Probably the destruction of T. gondii affects nervous system and damages pancreatic cells leading to increased risk of diabetes.

Abstract:Abstract Background: Toxoplasma gondii is an opportunistic parasitic protozoan, which is a causative agent of serious complications such as abortion in pregnant women or fetal still birth. The aim of this study was evaluating the seroprevalence of toxoplasmosis infection in diabetic pregnant women from Sanandaj, Kurdistan, west of Iran. Methods: In this study 136 serum samples from diabetic pregnant women were collected from referred to the Toohid hospital diabetes center during June 2018 to October 2019 in Sanandaj, Kurdistan, west of Iran. IgM and IgG titers were evaluated by ELISA(enzyme-linked immunosorbent assay) method. The collected data were analyzed by SPSS 19 software and using Chi-Square and Fisher tests. Results: The ELISA method showed 35(25.7%) and 7(5.1%) of diabetic pregnant women were positive for IgG and IgM T.gondii antibodies respectively. There was statistically significant relationship between toxoplasma infection and education, gestational age and number of parity. Conclusion: The results of this study showed that public awareness and education about toxoplasmosis and its transmission routes before pregnancy may be effective in preventing congenital toxoplasmosis.

Abstract:To investigate anti-Toxoplasma gondii IgG and IgM antibodies in diabetic pregnant women in Ahvaz, southwest of Iran this experiment was performed. In current study the sera of 110 diabetic pregnant women as well as 110 non diabetic pregnant women referred to the hospitals affiliated with the Ahvaz Jundishapur University of Medical Sciences were assessed for anti-T. gondii IgG and IgM antibodies by ELISA and IFA methods. The ELISA assessments showed that 47 (42.7 %) and 3 (2.7 %) of diabetic women were positive for IgG and IgM antibodies, respectively. However, in the control group, 24 individuals (21.81 %) were positive for IgG antibody but no detection for IgM antibody. According to IFA method, 46 (41.8 %) and 3 (2.7 %) of diabetic women were positive for IgG and IgM antibodies, respectively, while in control group, 21 individuals (19.09 %) were positive for IgG antibody. In this method, IgM antibody was negative for all samples of control group (0 %). In both methods, the values obtained in the case group were significantly higher than those in the control group (p < 0.05). Prevalence of anti-Toxoplasma IgG and IgM antibodies in diabetic pregnant women was higher than that in non-diabetic pregnant women. It seems that screening tests for seeking patients and teaching the transmission routes should be considered as prenatal cares for diabetic women.

Methods: In this case-control study Toxoplasma gondii antibodies were tested in 91 cases as well as 90 controls from referred the Toohid hospital diabetes center using ELISA (enzyme-linked immunosorbent assay) method during June 2018 to October 2019 in Sanandaj, Kurdistan, west of Iran. The collected data were analyzed by SPSS 19 and using Chi-Square and Fisher tests. Results: The seropositivity for anti-Toxoplasma IgG in case group was 33% and 15.6% in the control group ( p = 0.006) showing a statistically significant difference between groups, while for anti-Toxoplasma IgM ( p = 0.31) showing no statistically significant differences between two groups. A significant relationship was observed between age ( p = 0.024) and abortion history ( p = 0.05) with anti-Toxoplasma IgG antibodies in cases and controls. Conclusion : The results of current study indicated statistically significant relationship between diabetes and toxoplasmosis. The abortion history and increase of age was found to be a major T. gondii infection risk factor.

Abstract:Background: Type 2 diabetes mellitus (T2DM) is a chronic disorder that constitutes a major health problem worldwide. Toxoplasma gondii is an intracellular parasite that may infect any nucleated cell. Toxoplasmosis is becoming a worldwide health threat, infecting 30–50% of the world’s human population. The studies that have been undertaken to investigate the link between T. gondii infection and diabetes have shown contradictory fi ndings. This research aimed to look at the possible link between T2DM and T. gondii infection. Methods and Subjects: The enzyme-linked immunosorbent assay (ELISA) approach was used to screen for T. gondii IgM and IgG antibodies in 69 patients with T2DM and 92 seemingly healthy persons as controls. Results: The results demonstrate that all participants were IgM negative, the percentage of T. gondii latent infection was (52.1%) among patients with T2DM and (31.5%) among non-diabetic individuals. The frequency of infection diff ers signifi cantly between diabetic and non-diabetic people. T. gondii infection was not linked to the studied risk factors. Conclusion: There is serological evidence of a link between T2D and T. gondii infection. Furthermore, Toxoplasmosis is a risk factor for type 2 diabetes.

Abstract:Objective: Toxoplasmosis is a parasitic infection caused by Toxoplasma gondii. Immunocompromised individuals and pregnant women are at risk, with the latter group being susceptible to miscarriages. This study aimed to determine the seropositivity of T. gondii antibodies and potential risk factors in pregnant women diagnosed with diabetes mellitus. Methods: The research was conducted at the Ankara City Hospital Perinatology Clinic between October 2021 and June 2022. The study included 277 pregnant women diagnosed with diabetes mellitus and 277 healthy pregnant women who had given birth. Retrospective analysis of anti-T. gondii immunoglobulin (Ig)G and IgM levels was performed for patients between January 2020 and February 2022. Participants were administered an informed consent form and a questionnaire. Data were analysed using SPSS 22. Results: Among pregnant women with diabetes, IgG seropositivity was 18.4%, IgM was 0.0%, and IgG+IgM was 0.0%. In healthy pregnant women, IgG seropositivity was 12.3%, IgM was 0.4%, and IgG+IgM was 0.4%. Overall, seropositivity rates were 15.3% for IgG, 0.2% for IgM, and 0.2% for IgG+IgM. The difference between the two groups was statistically significant (p<0.05). Among pregnant women with diabetes, there was a significant statistical difference (p<0.05) in anti-T. gondii IgG seropositivity related to education, employment status, number of pregnancies and live births, history of toxoplasmosis diagnosis in children, previous toxoplasmosis diagnosis, hygiene, nutrition, and social habits. Among healthy pregnant women, significant statistical differences were found (p<0.05) in IgG seropositivity related to age, income, education level, number of pregnancies and live births, previous toxoplasmosis diagnosis, hygiene, nutrition, and social habits. No invasive interventions were performed on infants born to seropositive mothers, and perinatal data were not available. Conclusion: The seroprevalence of toxoplasmosis in Ankara appears to be decreasing, but T. gondii infections continue to pose a public health concern and are significant in pregnant women with diabetes mellitus.

Abstract:Introduction: Toxoplasma gondii is an obligate intracellular parasite affecting a broad range of warm-blooded animals, including humans. Infection acquired during pregnancy can be transmitted to the fetus and leading to serious problems such as spontaneous abortion, stillbirth, or severe mental and/or physical handicaps in the child. The purpose of this study was to investigate the seroprevalence of Toxoplasma infection and related risk factors in pregnant woman. Methodology: The study enrolled 1200 serum samples of pregnant women from February-November 2017. Then the samples were tested for the presence of anti-T. gondii antibodies (Ab) using enzyme-linked immunosorbent assay. Results: Out of the 1200 samples, 381 (31.7%) and 41 (3.4%) subjects were positive for IgG and IgM Ab, respectively. Among the evaluated risk factors, the seroprevalence of Toxoplasma infection was not related to the occupation in a significant way. However significant relationship was observed with factors such as; contact with soil, cats, consumption of raw washed vegetables, and washed hands before meals. Conclusions: According to the results, more than two-thirds of pregnant women are susceptible to Toxoplasma infection, hence training health care programs should be provided to prevent infection.

Abstract:Toxoplasmosis ranks among the most prevalent parasitic diseases globally. It seems that chronic toxoplasmosis is associated with several neuropsychiatric and other harmful effects in infected people, therefore, there is a need to investigate the prevalence of toxoplasmosis across various world regions. In this study, we conducted a meticulous meta-analysis to estimate the seroprevalence of anti-Toxoplasma gondii IgG antibodies within the general population in Iran (GPI). International and national scientific databases for studies published between January 1, 2000, and September 30, 2023, were searched. Observational studies reporting anti-T. gondii IgG seroprevalence in the GPI was selected/included. The data were synthesized using a random-effects model to calculate with a 95% confidence interval (95% CI) the national and regional anti-T. gondii IgG seroprevalence rates in Iran. Additionally, subgroup analyses were conducted to investigate the frequency of exposition to T. gondii in different socio-demographic, climatic, and geographical scenarios. From 18661 identified studies, 327 were included in the present meta-analysis, encompassing 122,882 individuals across the 31 Iranian provinces. The pooled nationwide anti-T. gondii IgG seroprevalence among the GPI was determined to be 32.9% (95% CI: 30.9-35.1%). The highest anti-T. gondii IgG seroprevalence was observed in Mazandaran province (North of Iran) (61%), whereas the lowest was in Semnan province (12.5%).Anti-T. gondii IgG seroprevalence demonstrated a higher occurrence in provinces characterized by moderate temperatures of 16-21°C, high relative humidity, and annual precipitation. Additionally, a higher anti-T. gondii IgG seroprevalence was identified among individuals with a habit of consumption of undercooked meat, raw fruits or vegetables, and untreated water. Moreover, those reporting direct contact with cats, possessing a lower level of education, residing in rural areas, being engaged in farming occupations, or playing the role of housewives exhibited higher anti-T. gondii IgG seroprevalence figures.The anti-T. gondii IgG seroprevalence within GPI closely aligns with the estimated worldwide average exposition rates. This underscores the imperative for public health policymakers to prioritize educational efforts regarding toxoplasmosis transmission pathways and its link to harmful effects.

Abstract:Toxocariasis is one of the most important and widespread neglected tropical infectious diseases. We designed a cross-sectional study to assess the seroepidemiological aspects of toxocariasis among the general population in northern Iran. A total of 630 rural subjects were enrolled to participate in the study. The presence of anti-Toxocara immunoglobulin G (IgG) was tested using a commercially available enzyme-linked immunosorbent assay. A structured questionnaire was also used to evaluate the potential risk factors and related clinical signs/symptoms. The presence of anti-Toxocara IgG antibodies was detected in 148 of the 630 rural subjects (23.5% [95% confidence interval {CI} 21.8 to 25.1]). By multivariate analysis, age (odds ratio [OR] 2.89 [95% CI 1 to -8.3], p=0.04), eating improperly washed vegetables (OR 4.05 [95% CI 2.47 to 6.64], p<0.001), contact with dogs (OR 3.31 [95% CI 2.13 to 5.12], p<0.001) and exposure to soil (OR 3.56 [95% CI 2.13 to 5.13], p<0.001) were significantly associated with the seroprevalence of Toxocara. The clinical study also demonstrated that the seroprevalence of Toxocara infection was significantly associated with asthma (OR 3.78 [95% CI 1.63 to 8.75], p<0.001) and ophthalmic disorder (OR 1.83 [95% CI 1.04 to 3.21], p=0.034). The findings proved that residents of rural communities in tropical regions may be heavily exposed to Toxocara spp. We highly recommend more investigations in high-risk groups.

Abstract:Background: Toxoplasma gondii is a pathogenic protozoan that causes toxoplasmosis and spreads worldwide. Objectives: This study aimed to investigate the prevalence of toxoplasmosis by serological and molecular methods in working children and a control group in Tehran. Methods: The study participants comprised 460 children aged 7 - 14 years, including 278 working children and 182 age-matched controls. Blood samples were collected, and a serological test was performed to evaluate IgM and IgG antibodies against T. gondii. Peripheral blood mononuclear cells (PBMCs) were isolated from the blood specimens by gradient centrifugation method. Real-time polymerase chain reaction (PCR) was performed using primer B1 on PBMC samples in children’s blood to determine the status of Toxoplasma infection. Results: Seroprevalence of IgG and IgM antibodies against T. gondii was 24.8% and 0.7%, respectively, in working children; however, in the control group, 12.1% and 2.2% had IgG and IgM antibodies against T. gondii, respectively. The mean IgG titer was 160 ± 86.39 IU/mL and 69.36 ± 88 IU/mL for working children and the control group, respectively (P < 0.0001); however, the mean IgM titer was 4.65 ± 3.04 IU/mL and 3.85 ± 4 IU/mL for working children and control group, respectively (P = 0.8187). Real-time PCR results indicated two (0.7%) positive cases among working children and three (1.65%) samples in the control group. The present study showed a significant difference between working children and the control group regarding the frequency of IgG antibodies (P = 0.0012). However, there was no significant difference in the frequency of IgM antibodies in the two mentioned groups. Conclusions: Seroprevalence of IgG antibody against T. gondii was more in working children than in the control group in Tehran. This investigation revealed a significant difference in frequency and titer of IgG antibodies between working children and the control group. More exposure to the soil and contaminated hands before drinking water or food may be considered factors in the development of toxoplasmosis infection in these children.

Abstract:BACKGROUND: Toxoplasmosis is the most common disease in humans and animals (zoonosis) caused by the protozoan parasite Toxoplasma gondii. The disease is usually appeared as asymptomatic in immunocompromised individuals but its most common symptom is lymphadenopathy. Shortly before or during the first trimester of pregnancy, this disease can be transferred to the fetus and cause serious infection in the fetus. In late pregnancy (third trimester), the complications of this infection is very low or unsigned. Due to the absence of non-specific clinical symptoms or slight infection in pregnant women, prenatal diagnosis is often impossible.OBJECTIVES: Since no research compared these two methods, we decided to compare these methods and determine which method works better for diagnosis of toxoplasmosis.PATIENTS AND METHODS: In this study, 50 pregnant women who referred to the Chalus Health Center laboratory were included and the blood samples were tested for presence of IgG and IgM antibodies of Toxoplasma gondii by both ELISA and Chemiluminescence methods.RESULTS: Of the 50 samples tested by the ELISA method, 26 samples (52%) were positive for IgG . No samples were positive for IgM. Of the 50 samples tested by the Chemiluminescence method, 28 samples (56%) were positive for IgG. No samples were positive for IgM.CONCLUSIONS: A significant relationship between the age of the youngest child and the infection rate was seen. No significant correlation between age, number of individuals in the household, number of children, location, type of construction, consumption of greens, the way of greens and meat consumption, drug use, history of stillbirth and infection levels was seen.
Serological survey of Toxoplasma gondii infection in a breeding kennel with reproductive disorders

Abstract:Introduction: Toxoplasma gondii (T. gondii) infection in dogs is distributed globally. Given the potential impact of T. gondii on human reproductive health and its zoonotic potential, focusing on infected dogs with breeding disorders may provide valuable new insights into the role of toxoplasmosis on reproductive disorders. This study aimed to elucidate the potential role of T. gondii on reproductive disorders by conducting a serosurvey on a kennel with breeding disorders in Kerman, Iran. Materials and methods: Purebred dogs with American Kennel Club-certified Pedigrees were randomly selected in both genders from a breeding kennel with a history of reproductive problems such as neonatal deaths, abortion stillbirths, and male infertility. Blood samples were collected from selected dogs, and serosurvey was done with a Toxo-Screen kit using the modified agglutination test (MAT). Findings: From 46 selected breeding dogs, 20 (43.5 %) were positive in the Toxoplasma MAT test, consisting of 24 samples (52.17 %) with different reproductive disorders and 22 (47.83 %) healthy dogs. However, the presence of reproductive disorder had no significant relationship with seropositivity (p = 0.067). Seroprevalence of toxoplasmosis had no significant relationship with sex (p = 0.28) and age (p = 0.18) in the studied population. Conclusion: Screening with the standard MAT method reduces the risk of transmitting toxoplasmosis by preventing the entry of infected dogs into the kennel, identifying the infected cases to start specific treatments, optimizing the proper nutritional conditions, and ensuring the hygiene of the environment. Fresh/refrigerated raw meat, unpasteurized milk feeding, rodent infestation of grass-covered dog runs, and the main dry food storage facilities were the primary risk factors in kennels.

Abstract:Toxoplasmosis is a frequent infection among the human population. The infection can cause devastating complications for the fetus during pregnancy. The present study aimed to determine the serological and molecular prevalence of the infection and molecular characterization of Toxoplasma gondii isolates among pregnant women referred to Kowsar Hospital, Urmia, Iran. In a cross-sectional study, 340 blood samples were collected from pregnant women referred to Kowsar Hospital, Urmia, Iran from May to July 2022. Anti- T. gondii IgG and IgM seropositivity were determined by enzyme-linked immunosorbent assay. PCR was carried out by targeting the GRA6 gene of the parasite on all patients' buffy coats. Anti- T. gondii IgG and IgM antibodies were positive in two (0.6%) women, and 101 (29.7%) women had anti- T. gondii IgG and 70.3% were seronegative. PCR was positive in two IgM-positive women, and both isolates belonged to T. gondii carrying the GRA6 allele of lineage I. The risk of infection was significantly higher in women who had constant contact with cats and soil, and who were residents of rural areas. The two IgM-positive women were asymptomatic regarding acute toxoplasmosis. According to the results of the present study, the prevalence of toxoplasmosis in pregnant women in Urmia is similar to its prevalence in other areas in northwestern Iran, and despite the low prevalence of acute infection, it should not be ignored.

Abstract:Gestational diabetes (GDM), the onset of any degree of glucose intolerance during pregnancy, increases a wide range of adverse health outcomes for both the mother and the fetus. The aim of the present study was to evaluate the association of Toxoplasma gondii infection with GDM in a case-control study with regard to the levels of leptin and tumor necrosis factor alpha (TNF-α) as two inflammatory biomarkers. Fifty-one pregnant diabetic cases and 109 controls were selected from a prenatal care clinic of a general hospital in Shiraz, southern Iran during July-November 2020. Cases and controls were similar in age, gestational age and number of parturitions. The presence of IgG antibodies against T. gondii , and serum concentrations of leptin and TNF-α were determined by ELISA. Anti-Toxoplasma antibodies were detected in 25 subjects (15.6%, 95% CI: 9.9 - 21.3). Nine (18%) diabetic cases were infected with Toxoplasma compared to 16 (15%) healthy controls (P=0.63). Level of leptin was higher (P=0.07) while TNF-α was lower in diabetic cases compared to healthy controls (P=0.08). When subjects were classified according to the combination of GDM and T. gondii , leptin was significantly lower in healthy (non-diabetic, non-infected) subjects compared to diabetics (P = 0.026), and TNF-α was higher in healthy subjects compared to Toxoplasma-infected diabetics (P = 0.032). These findings can be interpreted as both comorbidities being individually associated with increasing serum leptin and decreasing TNF-α concentrations, with modifying effects on each other. The present study opens a new perspective on GDM and its complex pathophysiological mechanism. Future research in this area is needed to better understand the underlying pathway for the development of GDM and the role of T. gondii and inflammatory biomarkers.

Abstract:Toxoplasma gondii is a neurotropic protozoan parasite that affects neuronal processing in the brain. This study aimed to investigate the prevalence of T. gondii infection in psychiatric disorder patients. We also investigated the potential association between sociodemographic, clinical manifestation, and behaviour of Toxoplasma -seropositive patients with psychiatric disorders. Commercial ELISAs (IgG, IgM, and IgG avidity) using serum and PCR using buffy coat were performed on samples from 54 individuals in each of the following groups: patients diagnosed with depressive disorder, bipolar disorder, and schizophrenia, as well as psychiatrically healthy subjects (control group). They were recruited from the Hospital Universiti Sains Malaysia in Kelantan, Malaysia. Of 54 patients with depressive disorder, 24/54 (44.4%) were seropositive for IgG, and four (16.7%) were IgG+/IgM+. Among the latter, a high avidity index indicating a past infection was observed in half of the samples (50.0%), and the other half (50.0%) showed a low avidity index, indicating a possible recent infection. Meanwhile, 30/54 (55.6%) patients with bipolar disorder were seropositive for IgG+, five (16.7%) were IgG+/IgM+, and four of them had a high avidity index, and one had a low avidity index. Patients with schizophrenia showed 29/54 (53.7%) seropositive for IgG, two of them (6.9%) were IgG+/IgM+; one of latter had a high avidity index, and one had a low avidity index. Of 54 people in the control group, 37.0% (20/54) were seropositive for T. gondii IgG antibodies. However, no significant difference was observed in seroprevalence between the control group and each patient group. No PCR-positive results were documented. A Chi-Square and multiple logistic regression showed that age ( p =0.031), close contact with cats/pets ( p =0.033) and contact with soil ( p =0.012) were significantly associated with Toxoplasma seropositivity in patients with psychiatric disorders. Additional research is needed to elucidate the causal relationships and underlying mechanisms.

Abstract:Introduction/Aims: Toxoplasmosis modifies various hormones and cytokines in the infected hosts which may result in several disorders. This study was conducted to assess testosterone, DHEA, and prolactin concentration levels among Toxoplasma gondii infected and uninfected infertile couples. Methods: Blood samples were collected and sera were separated. The sera were analyzed for the detection of anti-Toxoplasma (IgG& IgM) antibodies using commercial ELISA kits. The level of DHEA was measured by ELISA and the levels of testosterone and prolactin were evaluated by enzyme-linked fluorescent assay (ELFA, VIDAS). Results: The overall seroprevalence of toxoplasmosis was 58.0% (218/376). Among women, 56.9% (107/188) and 6.5% (7/107) were positive for anti-T. gondii antibodies IgG and IgM, respectively. The IgG and IgM were detected in the sera of 111/188 (59.0%) and 9/111 (8.1%) in male subjects, respectively. A positive association was observed between T. gondii infection and the upper and lower ranges of the normal value of testosterone in males (x 2 = 6.8, p = 0.033) but not in females (x 2 = 0.62, p = 0.99). A positive correlation was seen between toxoplasmosis and the upper and lower ranges of the normal value of prolactin in females (x 2 = 6.5, p = 0.039) but not in male cases (x 2 = 1.06, p = 0.59). Conclusion: Our results demonstrated no statistically significant differences between the level of sexual hormones between Toxoplasma-infected individuals and Toxoplasma-free subjects suffering from infertility. These findings suggest that further studies should be performed on infertile cases with large sample sizes in a case-control format.

Abstract:Toxoplasmosis is a parasitic zoonotic disease caused by Toxoplasma gondii , a common protozoan in the Apicomplexa phylum. Several studies in Iran have demonstrated the presence of the parasite in various hosts, but no data on T. gondii genotyping in HIV patients in Khuzestan, Southwest Iran, is available. One hundred of blood samples from AIDS patients were collected and tested by real-time PCR High Resolution Melting analyses for T.gondii detection and genotyping. T. gondii was discovered in 8 out of 100 (8%) AIDS patients with dominant Type I. This study suggest that HRM method demonstrated excellent discriminating ability for T. gondii , and AIDS patients should be tested for Toxoplasma detection and genotyping to prevent parasite pathogenicity.
Seroprevalence of Toxoplasma gondii and Associated Risk Factors among Pregnant Women in Algeria

Abstract:Toxoplasma gondii is an obligate intracellular protozoan parasite affecting all warm-blooded vertebrates, including humans. Infections in humans can lead to severe clinical manifestations in pregnant women and immunocompromised patients. The aim of the present study was to assess seroprevalence of T. gondii infection and to identify the associated risk factors among pregnant women from southern Algeria. A cross-sectional study was conducted from March 2021 to May 2022 among 1,345 pregnant women. A structured questionnaire was used to obtain information on risk factors associated with infection. Participants were screened for anti T. gondii IgG and IgM antibodies using the enzyme-linked fluorescent assay. The overall IgG and IgM seroprevalence was 13.6% and 0.89%, respectively. A significant association was found between seroprevalence of anti T. gondii IgG and history of spontaneous abortion ( P = 0.016). Moreover, an increasing trend of seroprevalence was detected in the autumnal season ( P = 0.030), and eating raw or undercooked meat was found to be significantly associated with anti– T. gondii IgM seropositivity ( P = 0.002). Multivariate analysis showed that women who had experienced spontaneous abortion, regularly ate undercooked meat, and used bottled water in summer were more likely to contract infection with T. gondii . The majority (86.4%) of the studied pregnant women were serologically negative for toxoplasmosis and hence were susceptible to primary acute infection during pregnancy and possible fetal anomalies. Therefore, health education and awareness of the disease and its transmission to women, especially during pregnancy, is imperative.

Abstract:Background: Toxoplasmosis is a parasitic disease caused by compilation protozoan agent Toxoplasma gondii, leading to significant financial and quality-adjusted life-year losses. Overcooked or raw meat consumption has been a considerable transmission route. The present study was conducted to determine the seropositivity rate of T. gondii in sheep and goats by serological and molecular tests and genotyping of obtained isolates in northeast Iran. Methods: Blood and tissue samples (diaphragm, heart) of 296 animals (including 168 sheep and 128 goats) were collected from the slaughterhouse in Quchan Country from august 2016 to April 2017. Modified agglutination test (MAT) and the PCR method performed to detect parasite DNA on tissues.PCR-RFLP method of GRA6 gene was used to determine the genotype of T. gondii. In addition, sequencing analysis was performed to evaluate the Toxoplasma type strains. Results: Serum positive for MAT results were found in 27.4% of sheep and 23.4% of goats. Positive PCR of B1 gene results in diaphragm and heart tissues of sheep and goats was 47.8% and 26.1%, 40% and 23.3%, respectively. PCR of GRA6 gene results were positive in 10 samples that RFLP technique results using MseІ enzyme revealed genotype І. Sequencing and phylogenetic analysis revealed DNA of all samples was closely related to Toxoplasma type І. Conclusion: Concerning the high seropositivity rate of toxoplasmosis, undertaking an appropriate preventive program for reducing the prevalence of T. gondii infection by raw or undercooked meat consumption of livestock is recommended. Our study supports the notion that these animals' consumption of raw and undercooked meat can be a probable source of human toxoplasmosis.
