Abstract:In this paper we present recent advances in Forster resonance energy transfer (FRET) sensing and bioimaging using nontoxic silicon quantum dots. (SiQDs) In our work, we prepare SiQDs-dye conjugates, with SiQDs serving as the donor which are covalently attached to organic dye acceptors via self-assembled monolayer linkers. Enzymatic cleavage of the peptide leads to changes in FRET response which was monitored using fluorescence lifetime imaging microscopy (FLIM-FRET). The combination of interfacial design and optical imaging presented in this work opens new opportunities for bio-applications using nontoxic silicon quantum dots.

Abstract:In this paper, bifunctional composite particles were synthesized by in situ grafting Au nanoparticles on the surface of SiO2/polyethyleneimine/Eu-polyoxometalates (SiO2/PEI/Eu-POM) spheres. It was found that the PEI layer on SiO2/PEI/Eu-POM spheres could act as a reductant for in situ reduction of metal ions to Au nanoparticles. The sizes of Au particles grafted on spheres could be tuned by adjusting the sizes of SiO2 spheres. The composite particles showed excellent red luminescence under UV light, which could be observed by naked eyes. In addition, they exhibited good catalytic activity in the oxidation of styrene and reduction of 4-nitrophenol. The smaller composite particles show the better overall performance in the catalytic reaction because of their lager active surface. The present strategy gives a promising way to prepare bifunctional composites for broad applications in catalysis and labeling. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:A seeded watermelon‐like mesoporous nanostructure (mSiO2@CdTe@SiO2, mSQS) composed of a novel dendritic mesoporous silica core, fluorescent CdTe quantum dots (QDs), and a protective solid silica shell is successfully fabricated by loading QDs into dendritic mesoporous silica nanoparticles through electrostatic interaction, and then coating with a solid silica shell by the modified Stöber method. The shell thickness of mSQS can be tuned from 0 to 32 nm as desired by controlling the reaction parameters, including the amount of silica precursor, tetraethyl orthosilicate, that is introduced, the solvent ratio (H2O:ethanol), and the amount of catalyst (NH3⋅H2O). These fluorescent mSiO2@QDs@SiO2 nanoparticles possess excellent stability and thickness‐dependent cytotoxicity, and are successfully applied to bioimaging.

Abstract:Au nanoparticle@SiO2@CdTe/CdS/ZnS quantum dot (QD) composite structures were synthesized by a liquid phase synthesis method. In this paper, three steps were adopted to gradually grow PVP-stabilized Au seeds from which PVP-stabilized Au nanoflower (NF) structures were successfully synthesized. For the sake of controlling the distance between Au NFs and QDs, silica was used as the shell material for coating Au NFs. The applications of this multi-functional nanoprobe in photothermal treatment and biolabeling were demonstrated on MCF-7 and MDA-MB-231 breast cancer cells labeled with Au NF@SiO2@QDs. The experimental results of viability staining indicate that Au NF@SiO2@QDs composites with an excitation threshold of the photothermal effect of only 1.0 W cm (2) exhibit excellent photothermal conversion efficiency owing to the large absorption cross sections of Au NFs. Compared with that of pure QDs, the fluorescence efficiency of Au NF@SiO2@QDs was increased by 40%, which could be attributed to localized surface plasmon enhanced dipole radiation. Fluorescence imaging results reveal that Au NF@SiO2@QDs targeted the membrane of cancer cells showing strong fluorescence. Therefore, it can be concluded that the composite structure combines the therapeutic and diagnostic modalities.
The Preparation and Properties of Polymer Beads and Quantum Dots Composites

Abstract:Quantum dots(QDs) were prepared in an organic system through wet chemistry method,which has the following virtues: the method is simple and new,and the cost of materials is low.Polymer beads with diameter of 60~70 nm and specific functional groups were synthesized with a particular seeded emulsion polymerization technique.The nano polymer beads have water-solubility and the functional groups being able to link with the biomolecules,and have more advantages than the micron beads.Moreover,QDs were embedded in the polymer beads with the specific functional groups through the dissolving and swelling method,which provides the condition for the conjunction of biomolecules and QDs as the fluorescent probes.The prepared samples are characterized with UV-Vis,PL,TEM,FTIR,CLSM and conductance titration etc.The results show that QDs are successfully embedded in polymer beads,which breaks the limitation of the conjunction of biomolecules and QDs only synthesized in aqueous system.

Abstract:Silicon quantum dots (SiQDs) have fully demonstrated their applicability in light of the fluorescence, the extension of their applications to other fields, especially considering their excellent biocompatibility, would be more appealing. Herein, a kind of versatile nitrogen-doped silicon quantum dots (N-SiQDs) was facilely synthesized via one-pot hydrothermal method with 3-aminopropyltrimethoxysilane (APTMS) and tetraethylpentylamine (TEPA) as sources. The N-SiQDs were used as a probe for the bacterial imaging owing to its good fluorescence property, stability and biocompatibility. Besides, thanks to N doping rendering the N-SiQDs stronger reducibility and Au affinity, the N-SiQDs displayed unique reduction capability, and were attempted as a reducing agent and stabilizer for the synthesis of the nanocomposite, i.e. N-SiQDs stabilized Au nanoparticles (N-SiQDs-AuNPs), under mild conditions. The N-SiQDs-AuNPs showed superior catalytic performance to citric-AuNPs due to the synergistical catalytic effect. In addition, the N-SiQDs exhibited good antibacterial properties against Gram-positive (S. aureus) and Gram-negative bacteria (E. coli) without obvious negative influence on the cells, particularly avoiding the use of any other external stimulation. This study may open a new avenue for using the SiQDs for synthesis of catalyst and other biomedicine applications beyond as the fluorescence probe.

Abstract:Magnetic and fluorescent bifunctional nanocomposites have great potential in biomolecule detection and biological imaging applications. So far, it remains a challenge to prepare bifunctional nanocomposites with high colloidal and fluorescent stability. To address this problem, we utilized a simple ring-opening reaction to conjugate Fe3O4 and CdZnSeS quantum dots (QDs). The surface amine groups of SiO2-coated Fe3O4 nanoparticles (Fe3O4@SiO2) opened the rings of the surface maleic anhydride groups of poly(styrene-co-maleic anhydride)-coated CdZnSeS QDs (QDs@PSMA), and then the resulting nanocomposite was functionalized with Jeffamine M-1000 polyetheramine (JMP) by a ring-opening reaction. The structures and properties of the bifunctional nanocomposites were fully characterized by transmission electron microscopy, dynamic light scattering, spectrofluorometry and vibrating sample magnetometry. The results indicated that the nanocomposites prepared by the conjugation method had dramatically higher quantum yields (QYs) than those prepared by the SiO2 co-encapsulation method. After introducing JMP, the nanocomposites exhibited high fluorescent and colloidal stability over a wide pH range (pH 2-13), a low level of protein adsorption in PBS with 10% fetal bovine serum at 37 degrees C, and a negligible level of nonspecific binding when incubated with HeLa cells. Sandwich fluoroimmunoassay results indicated that the nanocomposites can be successfully applied in a variety of diagnosis and bioimaging applications.

Abstract:Highly fluorescent SiO2@CdTe@SiO2 (SQS) nanoparticles as a new nanoprobe in biomedical imaging have been successfully synthesized by a novel approach. The size of SQS can be tuned from 39 to 76 nm by controlling reaction parameters, including the amount and the type of silica precursor, the ratio of silica precursor to ammonia solution, and the ratio of H2O to surfactant. The photo- and chemical stability, cytotoxicity and cell uptake of these nanoparticles were investigated. The results demonstrate that these properties are strong size-dependent, and reveal that the minimal shell thickness for effective protection of the embedded CdTe quantum dots is 6 nm. Our findings highlight the importance of controlling particle size and shell thickness during the preparation of fluorescent silica-QDs core-shell nanoparticles, and their potential in cell labeling and imaging.
A general route to efficient functionalization of silicon quantum dots for high-performance fluorescent probes.

Abstract:A general technique for efficient surface modification of silicon nanocrystals is highly desirable for the development of silicon quantum dots (SiQDs) as fluorescent probes for biological applications. Herein, a facile microwave-assisted hydrosilylation process for the preparation of stable SiQDs in a single step is presented. FTIR spectroscopy indicates that molecules with various terminal functionalities, such as alcohol, alkyl groups, and carboxylic acid, are grafted successfully onto the surface of silicon nanocrystals. The dispersibility of such SiQDs is clearly dependent on the terminal functional groups of the grafted molecules. In addition, the as-prepared SiQDs show excellent cell compatibility, photoluminescence properties, and stability, and their use as long-term intracellular fluorescent probes is also demonstrated. It is envisaged that this facile and effective method for the stabilization and functionalization of SiQDs with tailored wetting and chemical properties will enable wide application of SiQDs in a number of areas.

Abstract:Quantum dot-doped mesoporous microbeads (QDMMs) are encapsulated with silica shells for enhanced chemical stability. The results show that a micro-emulsion procedure is highly efficient in coating QDMMs with polyvinyl alcohol (PVA), which is important in the subsequent deposition of a silica shell. Incorporation of fluorescent silane precursors allows direct observation of silica shells by fluorescence microscopy. The resulting silica coated QDMMs (QDMM@SiO(2)) exhibit remarkable stability against solvent-induced QD leaching and chemical-induced fluorescence quenching compared with uncoated QDMMs. Further development of this technology such as optimization of silica shell thickness, surface modification with non-fouling polymers, and conjugation with biomolecular probes will enable clinical translation of the optical barcoding technology for highly multiplexed detection and screening of genes and proteins.

One-Pot Synthesis of Silicon Quantum Dots-Based Fluorescent Nanomaterial and Its Application

Abstract:Conventionally obtained silicon quantum dots (Si QDs) generally suffer from the disadvantages of a cumbersome preparation process, large fluctuation in the quality of Si QDs, poor water solubility, and aggregation-caused quenching (ACQ) phenomenon. Here we report a facile one-pot strategy to synthesize a novel Si QDs-based fluorescent nanomaterial in which Si QDs are confined into dendritic mesoporous silica, named as SiQDs@DMSNs. The prepared SiQDs@DMSNs, with adjustable particle sizes ranging from 140 to 300 nm, emit blue fluorescence around 410 nm upon excitation by ultraviolet light at a wavelength of 300 nm. It is found that the addition of sodium salicylate (NaSAL) plays a crucial role in the in situ generation of Si QDs. The obtained SiQDs@DMSNs exhibit excellent fluorescence intensity, water solubility, and stability, facilitating easy surface modification, without being limited by the ACQ phenomenon. It is expected to be widely used in many fields such as biosensors, nanomedicines, in vivo imaging, fingerprint identification, and anticounterfeiting labels.
Controllable synthesis, characterization and optical properties of colloidal PbS/gelatin core-shell nanocrystals

Abstract:Native quantum dots (QDs) made up of semiconductor nanocrystals (NCs) are toxic in nature but due to their excellent optical properties, they have proven themselves to be an attractive choice in biological labeling and targeting. In order to improve the general biocompatibility of lead sulfide (PbS) NCs, we present a new and simple procedure for preparing PbS/gelatin core-shell nanoparticles cross-linked with glutaraldehyde (GA) molecules. The phase composition, morphology, luminescence and in vitro photostability of the samples were characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR), transmission electron microscope (TEM) and fluorescence spectroscopy, respectively. The XRD analysis showed that the PbS NCs were of the cubic structure, the mean crystallite size was calculated to be 13.5 nm and the calculated lattice constant using Bragg's equation was 0.5950 nm, which was very close to its value in the standard card (JCPDS No. 5-592). In vitro test revealed that compared with bare PbS NCs, the photostability of the core-shell nanostructure remarkably improved. In addition, possible formation mechanisms of the PbS/gelatin nanoparticles were discussed in detail. Consequently, the advantages of high stability as well as high fluorescent intensity and biocompatibility make the core-shell nanoparticles promising candidates for in vivo biological targeting applications.

Abstract:High-quality cadmium selenide (CdSe) quantum dots (QDs) with good thermal stability were prepared by using liquid paraffin as a solvent and oleic acid as a ligand under relative low temperatures. Moreover, the as-prepared CdSe quantum dots were embedded into the porous polystyrene (PS) beads to fabricate optically encoded beads, which suggests their promising application in multiplexed bioassays.

Abstract:Modified fluorescent nanoparticles continue to emerge as promising candidates for drug delivery, bioimaging, and labeling tools for various biomedical applications. The ability of nanomaterials to fluorescently label cells allow for the enhanced detection and understanding of diseases. Silica nanoparticles have a variety of unique properties that can be harnessed for many different applications, causing their increased popularity. In combination with an organic dye, fluorescent nanoparticles demonstrate a vast range of advantageous properties including long photostability, surface modification, and signal amplification, thus allowing ease of manipulation to best suit bioimaging purposes. In this study, the Stöber method with tetraethyl orthosilicate (TEOS) and a fluorescent dye sulfo-Cy5-amine was used to synthesize fluorescent silica nanoparticles. The fluorescence spectra, zeta potential, quantum yield, cytotoxicity, and photostability were evaluated. The increased intracellular uptake and photostability of the dye-silica nanoparticles show their potential for bioimaging.

Abstract:We report a novel kind of oxidized silicon nanospheres (O-SiNSs), which simultaneously possess excellent aqueous dispersibility, high photoluminescent quantum yield (PLQY), ultra photostability, wide pH stability, and favorable biocompatibility. Significantly, the PLQY of the O-SiNSs is as high as 25%, and is stable under intense UV irradiation and in acidic-to-basic environments covering the pH range 2-12. To our best knowledge, it is the first example of water-dispersed silicon nanoparticles which possess both high PLQY and robust pH stability suitable for broad utility in bioapplications. Furthermore, the O-SiNSs are readily conjugated with antibody, and the resultant O-SiNSs/antibody bioconjugates are successfully applied in immunofluorescent cell imaging. The results show that the highly luminescent and stable O-SiNSs/antibody bioconjugates are promising fluorescent probes for wide-ranging bioapplications, such as long-term and real-time cellular labeling.

Abstract:Silica coating is an effective approach for rendering luminescent quantum dots (QDs) with water dispersibility and biocompatibility. However, it is still challenging to prepare silica-coated QDs (QD@SiO2) with high emission efficiency, small size and great stability in favor for bioapplication. Herein, we reported a modified Stober method for silica coating of aqueously-prepared CdTe/CdS QDs. With the coexistence of Cd2+ and thioglycolic acid (TGA), a thin silica shell was formed around QDs by the hydrolysis of tetraethyl orthosilicate (TEOS). The resultant QD@SiO2 with a small size (similar to 5 nm in diameter) exhibits significantly higher emission efficiencies than that of the initial QDs. Also, QD@SiO2 has extraordinary photo and colloidal stability (pH range of 5-13,4.0 M NaCl solution). Protected by the silica shell, the cytotoxicity of QDs could be reduced. Moreover, the QD@SiO2 conjugated with folic acid (FA) presents high specific binding toward receptor-positive HeLa cells over receptor-negative A549 cells. (C) 2014 Elsevier Ltd. All rights reserved.
An Efficient Method for Preparing High-Performance Multifunctional Polymer Beads Simultaneously Incorporated with Magnetic Nanoparticles and Quantum Dots

Abstract:An efficient modified method combining conventional swelling method with high-temperature swelling method was applied to prepare high-stable multifunctional MNPs-QD-encoded polystyrene beads simultaneously with large encoding capacity and fast separation. In this paper, the results demonstrate that encapsulating QDs into pre-prepared MNPs-beads is the best sequence, which keeps high brightness and fast separation. The MNPs-QD-encoded microcarriers designed by our proposed method exhibit excellent performance for magnetic manipulation and optical encoding. What is more, they also have better physical and chemical stabilities. Compared with beads prepared via conventional swelling method, the leakage of nanoparticles from the designed MNPs-QD-encoded poly (styrene-co-ethylene glycol dimethacrylate-co-methacrylic acid) beads (PSEMBs) induced by organic solvents (e. g. cyclohexane) is less than 6%; meanwhile, fluorescence intensity of MNPs-QDs-encoded PSEMBs fluctuates slightly more in a wide range of pH 2-12 buffers. In addition, immunoassay performance for human IgG detections indicates that carboxyl groups on fluorescent microsphere surface facilitate efficient attachment of biomolecule and therefore they can be further applied to fast separation and multiplexed biomolecular assays.

The thermal stability and the effects of the nanocrystal (NC) purification on the optical properties of the as-prepared CdSe QDs were also studied for their further bioapplication. The results demonstrate that the CdSe QDs have good thermal stability and a high photoluminescence quantum yield (PLQY) after NC purification, which is very important for their bioapplication. Moreover, optically encoded microbeads have been successfully prepared by directly embedding the as-prepared oil soluble CdSe QDs into the carboxyl group-capped porous polystyrene (PS) beads through our route, which suggests their promising application in multiplexed bioassays.
