Abstract:Bacteriocins have been attracting a lot of interest as antibacterial agents in recent decades. Bacteriocins have long been used as food preservatives, but their potential as clinical antimicrobials and immunological modulators has recently received increased interest. Antibiotic-resistant bacterial infections have been identified as one major issue affecting worldwide public health. Bacteriocins may be able to address this widespread issue because of their broad- or narrow-spectrum abilities to attack antibiotic-resistant bacteria. New tactics are needed to combat antibiotic-resistant microorganisms. Selenium nanoparticles (SeNPs) are a subgroup of antimicrobial peptides that can be employed in addition to or instead of approved antibiotics. After the biosynthesis of SeNPs by partially purified crude bacteriocin with a broad spectrum from Staphylococcus warneri after screening methods, the SeNPs were characterized using multiple characterizing tests. We tested the toxicity of the biosynthesized nanoparticles and measured their antibacterial and antifungal capabilities of it. Finally, we used conventional PCR to detect the PhzM gene and RT‒PCR to estimate the effect of SeNPs on the gene expression of the pyocyanin PhzM gene. Our findings indicate that combining various antimicrobial agents with various modes of action resulted in increased potency and a wide range of inhibition, including in multidrug-resistant bacteria. In addition, the majority of the studies demonstrated significant antibacterial efficacy against the majority of Staphylococcus lentus , Staphylococcus warneri , Staphylococcus epidermidis , Staphylococcus aureus , and Staphylococcus haemolyticus isolates as well as Candida albicans , Candida guilliermondii , and Candida ciferrii . The gene expression of PhzM , the gene encoding pyocyanin that was present in ten of P . aeruginosa isolates, was evaluated. The expression of PhzM was significantly up- or down-regulated in the isolates treated with SeNPs compared to the untreated isolates, as revealed by real-time PCR. Further study is needed to critically assess the effectiveness of this alternative treatment in both ex vivo and in vivo settings to properly address the creation and development of safe and well-designed protocols.
