Abstract:Cold plasma treatment is a new physical processing technology on crop seeds, the seeds after treatment will show a greater character. In order to ascertain the influence of cold plasma treatment on ear character and group quality of wheat, wheat seeds of Ningmai13, Huaimai22 and Huaimai33 were treated for 15 seconds by a cold plasma seed processing apparatus with the power of 80W, while the environmental medium was set to be air and the absolute pressure was set to be 60Pa. Then the seeds were cultivated in the field after placed for four days under the room temperature. At the harvest, the ear length, number of grain per ear and thousand-grain-weight of different varieties of wheat with different treatments were measured. After the harvest, volume-weight and grain protein content of wheat grains with different treatments were measured respectively according to GB-5498-1985 and GB-5009.5-2010. Results show that the ear length and number of grain per ear of fruiting wheat of Ningmai13, Huaimai22 and Huaimai33 treated by cold plasma are improved observably, the ear length increases by 10.2mm, 6.4mm, 3.2mm respectively, while number of grain per ear increases by 4.0, 3.3, 1.9 respectively. The influence of cold plasma treatment on thousand-grain-weight and grain protein content of fruiting wheat is different with varieties. The thousand-grain-weight and grain protein content of Huaimai22 increase by 0.93g, 1.017% respectively, but the effect on fruiting wheat of Ningmai13 and Huaimai33 is small. The difference of volume-weight of three varieties of fruiting wheat after treatment is indistinctive.

Abstract:Cold atmospheric pressure ambient air plasma generated by Diffuse Coplanar Surface Barrier Discharge (DCSBD) was investigated for inhibition of native microbiota and potentially dangerous pathogens (Aspergillus flavus, Alternaria alternata and Fusarium culmorum) on the maize surface. Moreover, the improvement of germination and growth parameters of maize seeds was evaluated. Maize (Zea mays L.; cv. Ronaldinio), one of the most important cultivated crops worldwide, was selected as the research material. Electrical measurements confirmed the high volume power density (80 W cm−3) of DCSBD plasma. Non-equilibrium plasma state evaluated using optical emission spectroscopy showed values of vibrational and rotational temperature (2700 ± 300) K and (370 ± 75) K, respectively. Changes on the plasma treated seeds surface were studied by water contact angle measurement, scanning electron microscope analysis and Fourier transform infrared spectroscopy. A complete devitalisation of native microbiota on the surface of seeds was observed after a short treatment time of 60 s (bacteria) and 180 s (filamentous fungi). The plasma treatment efficiency of artificially contaminated maize seeds was estimated as a reduction of 3.79 log (CFU/g) in F. culmorum after a 60-s plasma treatment, 4.21 log (CFU/g) in A. flavus and 3.22 log (CFU/g) in A. alternata after a 300-s plasma treatment. Moreover, the obtained results show an increase in wettability, resulting in a better water uptake and in an enhancement of growth parameters. The investigated DCSBD plasma source provides significant technical advantages and application potential for seed surface finishing without the use of hazardous chemicals.

Abstract:This work investigates the impact of a new geometry Atmospheric Cold-Plasma (ACP) system on mung bean (Vigna radiata) seed germination and seedling growth. Plasma is produced using a bipolar pulse power source, providing superior control of the generated species. A substantial increase in germination rates (86.67%–91.67%) and optimal seedling growth after 40 s of treatment is reported. The functional and morphological features have shown improvements. Plasma treatment significantly increases the specific surface area (7.695 m2/g) and total pore volume (0.007 cc/g) of the seeds compared to the control (2.214 m2/g and 0.001 cc/g, respectively), suggesting potential enhancements in water absorption and nutrient exchange. Radicle growth stimulation is also observed without alterations in the seed’s chemical structure. These findings highlight the potential of the ACP system for enhancing mung bean sprout germination and seedling development, which is helpful for agriculture applications.

Abstract:Cold atmospheric pressure plasma (CAP) has attracted increased interest in recent years for possible biomedical, environmental and agricultural applications. A wide range of cold plasma treatment effects is observed in agricultural applications, like effects on the seed germination and seedling growth, but more systematic investigations are needed. The aim of this study was to identify the most appropriate combinations of the plasma source and duration of treatment positively affecting seed germination. In addition, the effect of cold plasma on the seedling growth and osmotic stress tolerance was studied. The seeds of three Bulgarian durum wheat cultivars were treated with cold plasma in twelve variants. The results obtained were processed statistically via two-way ANOVA. The treatment of seeds with a plasma torch for 20 s and the treatment with underwater diaphragm discharge for 5 min when the seeds were placed in both cameras in two different positions (relative to the electrodes between which the plasma is supplied, "+" and "−") have the greatest positive effect on the all traits related to germination. The analysis of variance reveals that the variation in germination energy, shoot length and root length after the cold plasma treatment of seeds is mainly due to the interaction between the genotype and treatment variant and to a small degree due to the genotype. The treatment of seeds with cold plasma improves the osmoregulation ability of cells and therefore increases the drought resistance of genotypes.

Abstract:Effects of a cold atmospheric pressure plasma (CAPP) treatment on the germination, production of biomass, vigor of seedlings, uptake of water of wheat seeds (Triticum aestivum L. cv. Eva) were investigated. The CAPP treatment influence on the inactivation of microorganisms occurring on the surface of wheat seeds was investigated also. The so-called Diffuse Coplanar Surface Barrier Discharge generating a cold plasma in ambient air with high power volume density of some 100 W/cm3 was used for the treatment of seeds at exposure times in the range of 10–600 s. The optical emission spectroscopy and the electrical measurements were used for estimation of CAPP parameters. The obtained results indicate that the germination rate, dry weight and vigor of seedlings significantly increased for plasma treatment from 20 to 50 s. The plasma treatment of seeds led to an extensive increase in wettability and faster germination comparing with the untreated seeds. The growth inhibition effect of CAPP on the surface microflora of wheat seeds increased with the increase of the treatment time. The efficiency of the treatment of wheat seeds artificially contaminated with pure cultures of filamentous fungi decreased in the following order: Fusarium nivale > F. culmorum > Trichothecium roseum > Aspergillus flavus > A. clavatus.

Abstract:The present study was designed to highlight the effects of cold plasma (10 kV) treatment and priming with 2 mM salicylic acid (SA) and their combination (10 kV of plasma + 2 mM SA) on the physiological parameters and metabolism of two cultivars of Oryza sativa, i.e., Zhu Liang You 06 (ZY) and Qian You No. 1 (QY), under salinity stress (150 mM NaCl) and normal growth condition (0 mM NaCl). Seed germination and seedling growth were enhanced by SA priming and cold plasma treatment either alone or in combination under salinity stress. Photosynthetic pigments, photosynthetic gas exchange, and chlorophyll fluorescence were improved by cold plasma treatment and SA priming under salinity stress as compared to the untreated seeds. The activities of antioxidant enzymes were significantly improved by the combination of SA priming and cold plasma treatment in both cultivars under salinity stress. There were rapid changes in the cellular content of sodium (Na+) and calcium (Ca+), where the plants grown under saline conditions accumulate more Na+ and less Ca+ contents resulting in ionic imbalances. Interestingly, cold plasma and SA treatments diminished this action by reducing Na+ accumulation and increasing K+ and Ca+ contents in the plant cell under salinity stress. The activities of enzymes involved in secondary metabolism assimilation were up-regulated with cold plasma and SA priming either alone or combination under salinity stress. An increase in reactive oxygen species (ROS) accumulation and malondialdehyde (MDA) content was also observed under salinity stress condition. On contrast, seed treated with SA and plasma alone or combined resulted in a significant decrease in ROS and MDA contents under salinity stress. Our results indicated that SA priming and cold plasma treatment either alone or combined improved plant uptake of nutrients in both cultivars under stress conditions. The ultrastructural changes were observed to be more prominent in ZY than QY cultivar. Plants without SA priming or cold plasma treatments have a big vacuole due to the movement of ions into the vacuole directly from the apoplast into the vacuole through membrane vesiculation leading to membrane destabilization. However, SA priming and cold plasma treatment alone or combined helped the plants to recover their cell turgidity under salinity stress.

Abstract:The objective of this study was to determine the influence of low-temperature plasma (LTP) on seed surface modification, water uptake by seeds, seed germination and vigor of seedlings, as well as changes in the content of endogenous hormones in pea, (Pisum sativum L. var. Prophet). The study’s authors used diffuse coplanar surface barrier discharge as the source of LTP in various duration times of treatment (from 60 to 600 s). The SEM analysis showed that LTP induced significant changes on the seeds’ surface, which was related to water permeability into the seeds. LTP increased the germination percentage of pea seeds as well as the growth parameters (root and shoot length, dry weight), and the vigor of seedlings and the effects of LTP also depended on exposure time. The LTP-pretreatment produced changes in endogenous hormones (auxins and cytokinins and their catabolites and conjugates), which correlated with increased growth of the pea seedlings. The results suggested an interaction among the modification of seed structure demonstrated by LTP in the induction of faster germination and hormonal activities related to plant signaling and development during the early growth of pea seedlings.

Abstract:Cold plasma technologies are an efficient approach to improve the germination properties of seeds, especially in a stacking configuration within a dielectric barrier device. In such dry atmospheric plasma priming process, we show that a He-N2 plasma treatment of 20 min can reduce the median germination time of lentil seeds from 1420 min to 1145 minutes, ie a gain in vigor of 275 min (or +19.4 %). Considering that this result depends on the plasma-seed interaction and therefore on the contact surfaces between the seeds and the plasma, a topographic modeling of a 100 seeds-stack is performed in the dielectric barrier device. This model drives to the distinction between the seed-seed contact surfaces (276 contacts standing for a total area of 230.6 mm2) and the seed-wall contact surfaces (134 contacts standing for a total area of 105.9 mm2). After a single plasma treatment, the outer envelope of each seed is 92% exposed to plasma: a value high enough to support the relevance of the plasma process but which also opens the way to process optimizations. In this outlook, we propose to replace the single 20-minute plasma treatment by a "plasma sequence", ie a succession of shorter plasma treatments whose total duration remains 20 min. Between two successive plasma treatments, the seeds can follow either a trapping procedure (seeds in same positions and orientations) or a randomizing procedure (seeds in same positions but reoriented) or a stirring procedure (seeds vigorously shaken). A sequence of 10 plasma treatments (upon 2 min) separated by short stirring procedures leads to a gain in vigor as high as 405 min vs 275 min for a single plasma treatment of 20 min. We propose to understand these results by correlating the gain in vigor with the water uptake of the seeds and the wettability state of their coating.
Effects of Non-Thermal Plasma Treatment on Seed Germination and Early Growth of Leguminous Plants—A Review

Abstract:The legumes (Fabaceae family) are the second most important agricultural crop, both in terms of harvested area and total production. They are an important source of vegetable proteins and oils for human consumption. Non-thermal plasma (NTP) treatment is a new and effective method in surface microbial inactivation and seed stimulation useable in the agricultural and food industries. This review summarizes current information about characteristics of legume seeds and adult plants after NTP treatment in relation to the seed germination and seedling initial growth, surface microbial decontamination, seed wettability and metabolic activity in different plant growth stages. The information about 19 plant species in relation to the NTP treatment is summarized. Some important plant species as soybean (Glycine max), bean (Phaseolus vulgaris), mung bean (Vigna radiata), black gram (V. mungo), pea (Pisum sativum), lentil (Lens culinaris), peanut (Arachis hypogaea), alfalfa (Medicago sativa), and chickpea (Cicer aruetinum) are discussed. Likevise, some less common plant species i.g. blue lupine (Lupinus angustifolius), Egyptian clover (Trifolium alexandrinum), fenugreek (Trigonella foenum-graecum), and mimosa (Mimosa pudica, M. caesalpiniafolia) are mentioned too. Possible promising trends in the use of plasma as a seed pre-packaging technique, a reduction in phytotoxic diseases transmitted by seeds and the effect on reducing dormancy of hard seeds are also pointed out.

Abstract:Maize grains (Zea mays convar. Indentata Sturt.) were treated with non-thermal plasma, where Gliding Arc plasma discharge at an atmospheric pressure was used (working gas: Air; time duration: 0 s, 180 s, 300 s, 600 s). The experiment was conducted at a temperature of 18 °C, light/dark 12/12 h, and a light intensity of 100 μmol/m2s. Seed germination, seedling growth, photosynthetic parameters, and hormone (abscisic acid, jasmonic acid, salicylic acid, indole-3-acetic acid, and cytokinin) contents were measured. The highest stimulation of seed germination (to 141%), root length (to 221%), shoot length (to 298%), and root weight (to 122%) in comparison with the control was recorded after Gliding Arc plasma treatment for 600 s. The photochemical and non-photochemical Chl fluorescence parameters were not significantly affected by Gliding Arc plasma treatment. In contrast, hormonal pools in maize were significantly affected. The short-term plasma treatment (180 s) was associated with a decrease in the stress hormones abscisic acid, salicylic acid, jasmonic acid, and jasmonate isoleucine, while indole-3-acetic acid and cytokinin precursors were elevated. Longer-term treatment (300 s, 600 s) had an opposite effect—an elevation of abscisic acid, jasmonic acid, and jasmonate isoleucine as well as active cytokinins. The content of auxin decreased. Gliding plasma treatment may significantly affect maize physiology, dependent on the treatment duration.

Abstract:Low-temperature plasma treatment of wheat seeds was performed by a plasma jet and dielectric barrier discharge, both at atmospheric pressure. The influence of the plasma treatment on the growth parameters such as root and shoot dry weight and length as well as on the mean germination time was investigated. Stimulation of germination and an increase of the mean germination time were achieved for the shortest plasma exposure time. The ability of water imbibition by wheat seeds was found to increase by 20–30%. These phenomena were explained by the enhancement of hydrophilicity of the seed surface, which resulted in the reduction of the apparent contact angle. The positive effect of the plasma treatment was also achieved for biomass parameters such as average length, dry weights of roots and sprouts, and other. Thus, the plasma treatment can prove to be an effective alternative to traditional pre-sowing seed treatments used in agriculture.

Abstract:Sustaining the quality of seeds is a major task in attempting to supply nutrition to the growing world population. In this study, the seeds of Cicer arietinum were exposed to cold atmospheric plasma (CAP). A significant reduction of the natural microbiota attached to the seed surface was observed for increasing CAP treatment times—2 and 5 min were sufficient to achieve a 1 and 2 log reductions, respectively. Furthermore a 1 min CAP treatment showed a strongly improved seed germination (89.2 %), speed of germination (7.1 ± 0.1 seeds/day), and increased seed vigor, beside a decrease in the mean germination time (2.7 days) compared with controls. The roughness profile of the seed cotyledon was altered significantly, only in case of longer treatment times from 5 min. These results suggest that CAP technology has the potentiality to reduce health risks associated with contaminated seeds, while improving food quality.

Abstract:Meadow restoration and creation projects have faced a lack of local seed diversity due to the limited availability of seed sources. Non-thermal plasma technologies are being developed for agriculture and do not cause damage to heat-sensitive biological systems. This technology has shown the potential to improve agronomic seed quality by enhancing germination and promoting plant growth. However, there is almost no information about the effect of non-thermal plasma pretreatment on the seedlings' emergence of wild plant species. Therefore, this study aimed to evaluate the effect of non-thermal plasma on the emergence of 17 plant seeds originating from local meadows in Lithuania and compare it with the cold stratification pretreatment. The results obtained indicate that there were differences in emergence parameters among the species. However, NTP did not show statistically significant differences from the control. Non-thermal plasma improved the kinetic parameters of emergence for a few specific species' seeds, such as Anthyllis vulneraria and Prunella grandiflora, while the cold stratification pretreatment enhanced emergence for a broader range of plants. Significant differences were observed between non-thermal plasma and stratification pretreatment, as well as between the control and stratification groups. Both methods also had a negative impact.

Abstract:In this work, we investigated the impacts of atmospheric pressure dielectric barrier discharge (DBD), i.e., plasma treatment, on pearl millet seeds germination and plant growth. The effect of plasma discharge on water activation, by introducing the reactive species, was explored. We evidenced that about 30 min plasma treated pearl millet seeds exhibited 20% higher germination rate than the control seed watered with tap water. The HR-SEM study revealed that the plasma treatment increased the roughness and FTIR study showed that new oxygen functional groups were introduced on the seed surface. Moreover, it was observed that the water contact angle decreased for plasma treated seeds (50%) and the water uptake also increased considerably as compared to control seeds. These findings indicate that the seed surface has turned more hydrophilic after plasma treatment. A cylindrical double dielectric barrier discharge (D-DBD) reactor was employed for water activation, and 30 min of treatment under air has decreased the pH of deionized water from 7.4 to 4.5 and produced about 1.78 ppm of nitrate (NO 3 − ) and 4.2 ppm of hydrogen peroxide (H 2 O 2 ). Interestingly, the plasma activated water (PAW) improved the pearl millet seed germination by 30% (after 24 h of sowing) and plant growth as compared to tap water and deionized water. Remarkably, when PAW and plasma-treated seeds were combined, a beneficial impact in seed germination (95 ± 2%) and seedling growth have been evidenced owing to synergistic effect. We evidenced that among the long-lived species in PAW, NO 3 − enhanced the seed germination and plant growth under similar conditions. Abstract:The impacts of a low temperature plasma treatment on wheat seed germination and field growth were investigated using a plasma system about the first generation (T0), the second generation (T1) and the third generation (T2). Untreated wheat seeds were set as the control (CK). Plasma treatments of 80 W and 100 W were employed during the experiments. Germination characteristics, seedling growth parameters, biological characters and yield components were measured following the plasma treatments. The wheat seed germination index, plant height, tiller, growth and grain number obviously changed significantly after the low-temperature plasma treatment. Furthermore, the moderate-intensity plasma had an active impact on wheat seed germination and growth. The results showed that the theoretical yield of T0 significantly increased by 8.92% and 8.14%, while the actual yield increased by 8.12% and 6.10% compared with untreated. The theoretical yield of T1 increased by 5.69% and 6.62%, while it increased by 3.70% and 0.45% for T2. The actual yield of T1 increased by 5.75% and 4.94%, while it increased by 3.81% and 0.82% for T2. Plasma treatment exhibited the following trend of increasing: T0 > T1 > T2 > CK. We conclude that the effects of low-temperature plasma on the biological characteristics and yield components of wheat seeds exhibited a consistent trend from T0 to T1 to T2.

Abstract:Low temperature atmospheric pressure plasma has emerged as a very powerful technology to improve agricultural and food industries. Recently, various types of atmospheric pressure plasmas have applied to seed germination and plant growth. In this study, the barley (Hordeum vulgare) seeds have treated using a surface dielectric barrier discharge to investigate the biological effects on the seed germination, plant growth and functional metabolites. It was observed that the seed surface was cracked and eroded after plasma treatment, while the surface of non‐treated seed was intact. The growth of plasma treated barley was increased by at the lowest 15% and at the highest 110% depending on plasma treatment duration, respectively. GABA content of plasma treated Saechal barley was slightly increased under no germination process, while DPPH activity was decreased at the same condition. Plasma may affect the biological effects on seed germination, plant growth, and changing of secondary metabolites through physical and chemical interactions, but it needs further experimental investigations and verified data to clarify their mechanisms. Abstract:Among the innovative technologies being elaborated for sustainable agriculture, one of the most rapidly developing fields relies on the positive effects of non-thermal plasma (NTP) treatment on the agronomic performance of plants. A large number of recent publications have indicated that NTP effects are far more persistent and complex than it was supposed before. Knowledge of the molecular basis and the resulting outcomes of seed treatment with NTP is rapidly accumulating and requires to be analyzed and presented in a systematic way. This review focuses on the biochemical and physiological processes in seeds and plants affected by seed treatment with NTP and the resulting impact on plant metabolism, growth, adaptability and productivity. Wide-scale changes evolving at the epigenomic, transcriptomic, proteomic and metabolic levels are triggered by seed irradiation with NTP and contribute to changes in germination, early seedling growth, phytohormone amounts, metabolic and defense enzyme activity, secondary metabolism, photosynthesis, adaptability to biotic and abiotic stress, microbiome composition, and increased plant fitness, productivity and growth on a longer time scale. This review highlights the importance of these novel findings, as well as unresolved issues that remain to be investigated.
