Abstract:Wavelength-controlled dynamic processes are mostly based on light-triggered isomerization or the cleavage/formation of molecular connections. Control over dynamic metathesis reactions by different light wavelengths, which would be useful in controllable dynamic chemistry, has rarely been studied. Taking advantage of the different bond energies of disulfide and diselenide bonds, we have developed a wavelength-driven exchange reaction between disulfides and diselenides, which underwent metathesis under UV light to produce Se-S bonds. When irradiated with visible light, the Se-S bonds were reversed back to those of the original reactants. The conversion of the exchange depends on the wavelength of the incident light. This light-driven metathesis chemistry was also applied to tune the mechanical properties of polymer materials. The visible-light-induced reverse reaction was compatible with reductant-catalyzed disulfide/diselenide metathesis, and could be utilized to develop a dissipative system with light as the energy input.

Abstract:Disulfide bonds, ubiquitous in natural and artificial systems, underpin protein folding, cellular redox homeostasis, materials science, and drug development. Despite numerous existing methods for disulfide synthesis, efficient and selective construction of asymmetric disulfides remains challenging. This review focuses on the latest advances in visible light photocatalytic and transition metal-catalyzed synthesis of disulfide compounds from 2021 to 2024, which have emerged as promising strategies offering milder and more sustainable reaction conditions. In the realm of visible light photocatalysis, we discuss novel methods such as photocatalytic decarboxylation, deoxygenative disulfidation of alcohols, and direct C-H disulfidation, which demonstrate broad substrate scope and excellent functional group tolerance under mild conditions. Regarding transition metal catalysis, we explore new reaction systems involving copper, nickel, palladium, chromium, molybdenum, and scandium, which achieve reductive coupling, selective oxidation, and novel insertion reactions, providing effective routes for constructing asymmetric disulfides and late-stage functionalization of complex molecules.

Abstract:Visible-light photoredox catalysis has been established as a popular and powerful tool for organic transformations owing to its inherent characterization of environmental friendliness and sustainability in the past decades. The thiol-ene/yne reactions, the direct hydrothiolation of alkenes/alkynes with thiols, represents one of the most efficient and atom-economic approaches for the carbon-sulfur bonds construction. In traditional methodologies, harsh conditions such as stoichiometric reagents or a specialized UV photo-apparatus were necessary suffering from various disadvantages. In particular, visible-light photoredox catalysis has also been demonstrated to be a greener and milder protocol for the thiol-ene/yne reactions in recent years. Additionally, unprecedented advancements have been achieved in this area during the past decade. In this review, we will summarize the recent advances in visible-light photoredox catalyzed thiol-ene/yne reactions from 2015 to 2021. Synthetic strategies, substrate scope, and proposed reaction pathways are mainly discussed.

Abstract:Visible light promoted difunctionalization of alkynes is reviewed. The difunctionalization reaction is achieved by different reagents. Radicals such as carbon (sp3), carbon (sp2), and other heteroatom (P, S, N, Se, O, and halide) radicals initiated by visible light can undergo radical addition to a carbon-carbon triple bond. Upon further transformation, the desired difunctionalized products are obtained. Some organometallic complexes can be activated by visible light; the difunctionalization of alkynes is catalyzed by these species. Other reagents like 1,3-dipole precursors could also react with alkynes to give difunctionalization products; here, the 1,3-dipole derivatives are obtained by visible light photocatalysis. So far, the strategy has been succeeded in the formation of C–C bonds and C–X bonds. Several valuable chemical skeletons have been constructed under mild conditions. However, high regio- and stereoselectivities in some direct difunctionalization methodologies are yet to be achieved.

Abstract:The dynamics of thiol/disulfide exchange is essential to many cellular processes, such as oxidative folding in the endoplasmic reticulum. This group of reactions has usually been examined by bulk techniques, which are intrinsically limited when studying out-of-equilibrium reactions, characterizing transient intermediates, or inspecting the directionality of the reaction (regioselectivity). In this work, we present a single-molecule approach that allows a regiospecific description of the reaction pathways available for the reduction of a protein with two disulfide bonds. Two disulfides were introduced in the model protein I27; I272S-S. Single polyproteins based on I272S-S were mechanically extended using an atomic force microscope working in force-clamp mode and in the presence of reducing agents. In our experiments, the disulfides limit the mechanical extension of I272S-S, and the reduction events are detected as step-wise increases in the length of the polyprotein. Importantly, the magnitude of such steps depends on the number of amino acids sequestered by the disulfide being reduced, which allowed us to unambiguously assign specific steps to the cleavage of specific disulfide bonds. Cysteine residues resulting from the reduction of the first disulfide are excellent nucleophiles able to attack the remaining disulfide. This generates two additional intermediates and a total of four distinct reaction pathways. All of them can be unequivocally discriminated by our method. The frequency of appearance of the different pathways depends on the concentration of the reducing agent, as a consequence of the competition between inter- and intramolecular reactions. We observed that thiol/disulfide exchange reactions are highly regiospecific even within an unfolded polypeptide. Our results provide new insights into physiologically relevant reactions at an unprecedented level of detail.

Abstract:A metal‐free and NBS‐mediated direct thiol‐disulfide exchange reaction at room temperature is developed. A variety of unsymmetrical disulfides have been prepared with good to excellent yields. Efficient late‐stage modification of amino acids and drugs demonstrates the utility of this strategy. In addition, easy to operate, broad substrate scope and good functional group tolerance further demonstrate that this reaction provides an essential complementary route to access unsymmetrical disulfides.

Abstract:Herein, we present an efficient synthesis method utilizing Bi quantum dots (QDs) as catalysts for the photochemical oxidative coupling of thiols to disulfides under mild conditions. The protocol offers low catalyst loading, simplicity in reaction setup as well as high efficiency. Aromatic thiols and aliphatic thiols are successfully transformed into disulfides with good to quantitative yields, demonstrating tolerance to various substituents and heterocycles. Additionally, the protocol extends to the synthesis of asymmetric disulfides and allows for gram-scale synthesis. An efficient synthesis method utilizing Bi quantum dots (QDs) as catalysts for the photochemical oxidative coupling of thiols to disulfides under mild conditions is reported. The protocol offers low catalyst loading with high efficiency. Symmetric and asymmetric disulfides have been successfully obtained which allows for gram-scale synthesis.

Abstract:Dichalcogenides (disulfides and diselenides), as reactants for organic transformations, are important and widely used because of their potential to react with nucleophiles, electrophilic reagents, and radical precursors. In recent years, in combination with photochemical technology, the application of dichalcogenides as stable radical reagents has opened up a new route to the synthesis of various sulfur- and selenium-containing compounds. In this paper, synthetic strategies for disulfides and diselenides and their applications with photochemical technology are reviewed: (i) Cyclization of dichalcogenides with alkenes and alkynes; (ii) direct selenylation/sulfuration of C−H/C−C/C−N bonds; (iii) visible-light-enabled seleno- and sulfur-bifunctionalization of alkenes/alkynes; and (iv) Direct construction of the C(sp)–S bond. In addition, the scopes, limitations, and mechanisms of some reactions are also described.

Abstract:This article investigates a copper-catalyzed alkynyl disulfuration reaction of aryne formed in situ to synthesize o‐alkynyl aryl disulfides. The transformation proceeds through the in situ generation of aryne, followed by alkynylation under copper catalysis and disulfuration with disulfide reagent, which results in the formation of one C-C bond and one C-SS bond in one-pot under mild conditions. The reaction above can be readily employed to facilitate the introduction of a disulfide group and aryl alkynes into an aryne, and thus provide a modular approach to unsymmetrical disulfides.

Abstract:Organosulfur compounds, especially sulfur ylides and sulfonium salts, offer extraordinary potential in organic chemistry due to their distinctive structural properties and reactivities. In the past five years, tremendous progress of sulfur ylide and sulfonium salt chemistry has been made with the advent of photoredox catalysis. Increasing unprecedented reactivities and reactions of organosulfur compounds have been unearthed, which are distinguished from their conventional ionic reactivity. This review highlights the recent advances in alkylation procedures with sulfur ylides, sulfoxonium ylides, sulfonium salts and thianthrenium salts driven by photocatalysis—with emphases on reactions, mechanisms, and applications—aiming to provide guidance for the potential of sulfur-containing species in a wide range of reactions and to inspire further progress in organosulfur chemistry.

Abstract:The dynamic disulfide linkage plays a vital role in various biological processes as well as drugs and biomaterials. The conversion of thiols to their corresponding disulfides is a hallmark of sulfur chemistry, but notoriously difficult to control. Achieving optimal reactivity and selectivity continues to pose significant challenges. Here, we describe a click chemistry for disulfide formation from thiols in both batch and flow-mode using SO2F2, which display exceptional selectivity toward disulfide formation through an effective nucleophilic substitution cascade. This reaction's unique characteristics satisfy the stringent click-criteria with its high thermodynamic driving force, straightforward conditions, wide scope, quantitative yields, exceptional chemoselectivity, and non-chromatographic purification process. The modular synthesis of symmetrical, unsymmetrical, cyclic and polydisulfides is demonstrated, along with the formation of disulfide cross-linked hydrogels.

Abstract:We demonstrated herein a versatile protocol for visible-light-induced sulphide anion-catalysed decarboxylative cross-couplings. An array of primary, secondary, tertiary and amino acid-derived redox-active esters (RAEs) were all amenable substrates to undergo radical decarboxylation to be coupled with a diverse range of nucleophiles, creating challenging C(sp3)-C(sp3) and C(sp3)-C(sp2) bonds with high efficiency. We note that Katritzky’s salt and Togni’s reagent were also suitable substrates, affording deaminative alkylation and trifluoromethylation products in high yields. Density functional theory calculations and mechanism experiments indicate that a charge-transfer complex was formed between sulphide anions and RAEs in the amide solvent. Additionally, a one-pot two-step telescoped procedure and continuous-flow process further increase the synthetic utility of this catalytic system.

Abstract:Herein, we report the α‐C–H acyloxylation of sulfides and disulfides enabled by hypervalent iodine(III) reagents via photoredox catalysis. The methoxylbenziodoxole derivatives serve as the hydrogen atom transfer agent, the mild oxidant, and the acyloxylation source in the reaction. External nucleophiles can also be introduced to the α‐C–H of sulfides. The reaction applies to various aryl and alkyl sulfides including methionine peptide derivatives. Disulfides are applicable for the first time with excellent chemoselectivity and functional group compatibility.

Abstract:Herein, a facile and efficient thiolation method is presented, involving Ni-catalyzed cross-coupling of readily available disulfides with diverse bromides, promoted by ball milling. This Ni-catalyzed modular approach demonstrates excellent functional group tolerance and a broad substrate scope, including acidic molecules. Furthermore, the plausible mechanism was extensively investigated and proposed, incorporating multiple valence states of Ni to complete the catalytic cycle. Moreover, green factors were compared with conventional solution methods. The practicality of this transformation is thus demonstrated for the synthesis of sulfur-containing organic compounds, highlighting its high efficiency and eco-friendliness.

Abstract:Dynamic covalent bonds are extensively employed in dynamic combinatorial chemistry. The metathesis reaction of disulfide bonds is widely used, but requires catalysis or irradiation with ultraviolet (UV) light. It was found that diselenide bonds are dynamic covalent bonds and undergo dynamic exchange reactions under mild conditions for diselenide metathesis. This reaction is induced by irradiation with visible light and stops in the dark. The exchange is assumed to proceed through a radical mechanism, and experiments with 2,2,6,6-tetramethylpiperidin-1-yloxyl (TEMPO) support this assumption. Furthermore, the reaction can be conducted in different solvents, including protic solvents. Diselenide metathesis can also be used to synthesize diselenide-containing asymmetric block copolymers. This work thus entails the use of diselenide bonds as dynamic covalent bonds, the development of a dynamic exchange reaction under mild conditions, and an extension of selenium-related dynamic chemistry.
