Abstract:Natural bioactive compounds are valuable resources for drug discovery due to their diverse and unique structures. However, these compounds often lack optimal drug-like properties. Therefore, structural optimization is a crucial step in the drug development process. By employing medicinal chemistry principles, targeted molecular operations can be applied to natural products while considering their size and complexity. Various strategies, including structural fragmentation, elimination of redundant atoms or groups, and exploration of structure-activity relationships, are utilized. Furthermore, improvements in physicochemical properties, chemical and metabolic stability, biophysical properties, and pharmacokinetic properties are sought after. This article provides a concise analysis of the process of modifying a few marketed drugs as illustrative examples.

What problem does this paper attempt to address?
The problem this paper attempts to address is the insufficient biological properties of natural bioactive compounds when developed as drugs. Specifically, these compounds typically have the following limitations: 1. **Low activity**: Many natural bioactive compounds have low activity, making them insufficient to be used directly as drugs. 2. **Limited specificity**: These compounds may have weak binding ability to specific targets, leading to suboptimal drug efficacy. 3. **Significant toxicity**: Some natural compounds have high toxicity, limiting their safe use. 4. **Unfavorable pharmacokinetic properties**: Natural compounds may exhibit poor characteristics in absorption, distribution, metabolism, and excretion (ADME) processes in the body, affecting their efficacy. Therefore, the focus of the paper is on optimizing the structure of these natural bioactive compounds through medicinal chemistry strategies to enhance their biological activity, selectivity, safety, and pharmacokinetic properties, thereby making them effective drug candidates. Specific strategies include: - **Structural fragmentation**: Breaking down complex natural products into simpler fragments to facilitate further chemical synthesis and optimization. - **Elimination of redundant atoms or groups**: Removing redundant atoms or groups that do not participate in target binding to improve the physicochemical properties and pharmacokinetic characteristics of the compounds. - **Exploration of structure-activity relationships**: Systematically modifying the structure of compounds to study their impact on biological activity, thereby finding the optimal structure. Through these strategies, researchers hope to develop new drugs with higher efficacy and better safety profiles.
