Abstract:Unimolecular micelles have attracted intense interests as drug carriers for tumor chemotherapy owing to their superior stability in comparison with the self-assembled supramolecular ones. Among them, the dendritic polymers with the polar frameworks could favour the loading of chemotherapeutic drugs rather than the hyperbranched polymers via radical polymerization, by enhancing the interaction with drugs. While the tedious synthesis procedure for dendritic polymers could be simplified with the construction principle on urethane chemistry. Here, the PEGylated dendritic polyurethanes, Ph-DPUGly-PEG and Ph-DPUTEA-PEG, were designed with glycerol or triethanolamine as monomer, respectively. The effect of the molecular architecture of the Ph-DPU-PEGs unimolecular micelles on the controlled drug releasing performance was compared. It was found that the Ph-DPUTEA-PEG with tertiary amine as branching points could efficiently endow the pH-triggered drug release, due to its protonation.

Abstract:Unimolecular micelles have attracted intensely attention as drug delivery systems (DDSs) in tumor chemotherapy owing to their superior stability. However, their drug release performance is mainly determined by the hydrophilic-hydrophobic property of their cores, in which the chemotherapeutic drugs were loaded. Here, GSH-triggered disintegrable dendritic polyurethane was designed via a facile one-pot approach, with disulfide-containing diisocyanate. After PEGylation, it self-assembled into bioreducible unimolecular micelles with highly hydrophobic cores for chemotherapeutics-loading, showing a minimized premature drug leakage during blood circulation. After tumor cellular internalization, the GSH-triggered disintegration of the dendritic polyurethane-based cores in the unimolecular micelles occurred to release the loaded model drug (DOX), possessing tumor-specific chemotherapy with an enhanced tumor growth inhibition than free DOX.

Abstract:The synthesis of a new kind of amphiphilic poly(vinyl-co-ester)s via simple one-pot radical ring-opening polymerization (RROP) is reported. A series of biodegradable, biocompatible, and cross-linkable copolymers P(MDO-co-PEGMA-co-CMA) with different compositions were synthesized by copolymerization of 2-methylene-1,3-dioxepane (MDO), poly(ethylene glycol) methyl ether methacrylate (PEGMA) and 7-(2-methacryloyloxyethoxy)-4-methylcoumarin methacrylate (CMA). All the copolymers were well characterized by H-1 NMR, C-13 NMR, and gel permeation chromatography (GPC). The hydrolytic degradation and enzymatic degradation of the copolymers were investigated. The amphiphilic P(MDO-co-PEGMA-co-CMA) can self-assemble into micelles in aqueous solution. Polymeric micelles with different sizes were obtained through tuning the compositions of the copolymers, i.e., the hydrophilic/hydrophobic ratio. The size and morphology of the micelles were measured by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Because of the coumarin side groups in the micellar core, the micelles can be cross-linked upon exposure to long-wavelength UV light (lambda = 365 nm). These micelles also showed excellent biocompatibility by the in vitro cytotoxicity experiments. Furthermore, the biodegradable micelles were applied for the delivery of an anticancer drug doxorubicin (DOX). The cell viability of DOX-loading micelles compared to free DOX was discussed. The effects of cross-linking and the copolymer composition on the drug release behavior were well studied. The results indicate that this new kind of amphiphilic poly(vinyl-co-ester) s could serve as promising nanocarriers for the delivery of anticancer drugs.

Abstract:Nanoparticles are appealing drug delivery systems since they promise prolonged circulation time and predictable release behaviors. The current work reports a novel hyperbranched amphiphilic block copolymer synthesized using deactivation-enhanced atom transfer radical polymerization (DE-ATRP) for smart drug delivery. PEG2000-Br was applied as a macroinitiator to initiate acid-cleavable divinyl (ACD) monomers linked by acetal groups, formulating the hydrophilic (PEG) and hydrophobic (ACD) segments. The polymer appeared to self-assemble into micelles with diameters in the range of 70-100 nm, and was examined for the controlled release of doxorubicin (DOX). Results showed that DOX-loaded micelles (similar to 90 nm) could achieve drug loading as high as 8.2 wt%, with interesting pH-dependent release behaviors. Studies with flow cytometry (FCM) and confocal laser scanning microscopy (CLSM) showed that DOX-loaded micelles exhibited a high cellular uptake performance by HeLa cells, which indicated promising antitumor efficacy for such a drug delivery system. Additionally, such DOX-loaded micelles exhibited remarkable cytotoxicity against HeLa cells in a dose-and time-dependent manner due to the enhanced cell uptake behavior of micelles. These results indicated that the polymeric micelles might be used as a promising candidate for a pH-responsive drug delivery for cancer therapy.

Abstract:Combination chemotherapy has been recognized as a more powerful strategy for tumor treatment rather than the single chemotherapy. However, the interactive mechanism of the two hydrophobic chemotherapeutic drugs has not been explored by now. Aiming for a better synergistic effect, such interactive mechanism was investigated in the present work, by designing CPT@DOX-DPU TEA -PEG nanomedicine with encapsulated camptothecin (CPT) and conjugated doxorubicin (DOX). The synergistic controlled drug release effect was found for the two drugs loaded on the different sites of the dendritic polyurethane core. Synergism was achieved on the HepG2 cells with a combination index (CI) of 0.58 in the in vitro cellular experiments. The results demonstrated the promising application of the unimolecular micelles-based nanomedicine with independently loading of two hydrophobic chemotherapeutic drugs.

Abstract:A novel amphiphilic copolymer, poly (ethylene glycol)-graft-polyethyleneimine/amide (PEG-g-PEI/amide), is synthesized by grafting PEG and 1,2-cis-Cyclohexanedicarboxylic anhydride onto the PEI. PEGylated polymeric micelles can be assembled from the amphiphilic copolymers with well-defined nano-sizes, and anticancer drugs are successfully loaded into micelle core formed by the amide. The amides with neighboring carboxylic acid groups exhibit pH-dependent hydrolysis and can reversibly shield the cationic charge of amine groups on the PEI, giving the micelles a charge-conversion property from negative to positive in acidic tumor tissue environment. Meanwhile, the cleavage of amide bonds at acidic pH also results in the disassembly of the micelle and pH-responsive drug release. These micelles are promising drug delivery systems due to their smart properties: PEGylation, suitable size, charge-conversion, and simultaneous pH-sensitive drug release.

Abstract:Polyurethane micelles with disulfide linkage located at the interface of hydrophilic shell and hydrophobic core (PU-SS-I) have been shown enhanced drug release profiles. However, the payloads could not be released completely. The occurrence of aggregation of hydrophobic cores upon shedding hydrophilic PEG coronas was considered as the reason for the incomplete release. To verify the above hypothesis and to develop a new polyurethane based micelles with dual stimuli respond properties and controllable location of pH and reduction responsive groups in the PU main chains, a tertiary amine was incorporated into the hydrophobic core PU-SS-I, which resulted polyurethane with both reduction and pH sensitive properties (PU-SS-N). Biodegradable polyurethane with only disulfide linkages located between the hydrophilic PEG segment and the hydrophobic PCL segments (PU-SS-I) and polyurethane with only pH sensitive tertiary amine at the hydrophobic core (PU-N-C) were used as comparisons. Paclitaxel (PTX) was chosen as mode hydrophobic drug to evaluate the loading and redox triggered release profiles of the PU micelles. It was demonstrated that PU-SS-N micelles disassembled instantly at the presence of 10mM GSH and at an acidic environment (pH=5.5), which resulted the nearly complete release (~90%) of the payloads within 48h, while about ~70% PTX was released from PU-SS-I and PU-SS-N micelles at neutral environment (pH=7.4) with the presence of 10mM GSH. The rapid and complete redox and pH stimuli release properties of the PU-SS-N nanocarrier will be a promising anticancer drug delivery system to ensure sufficient drug concentration to kill the cancer cells and to prevent the emergency of MDR. The in vitro cytotoxicity and cell uptake of the PTX-loaded micelles was also assessed in H460 and HepG2 cells.

Abstract:Triblock copolymer PEG-block-poly(ortho ester urethane)-block-PEG (PEG-POEU-PEG) was conveniently prepared via polycondensation reaction between the ortho ester-containing diol and hexamethylene diisocyanate (HDI) at varying reaction time followed by termination with polyethylene glycol monomethyl ether (mPEG). PEG-POEU-PEG could further self-assemble into nano-scaled micelles. Ortho ester degradation in these micelles was significantly accelerated at pH 5.0 due to its pH sensitivity. In vitro drug release profiles showed that only a small amount of the loaded DOX was released in PBS solution at pH 7.4, while DOX could be quickly released at pH 5.0. MTT assay reveals that the micelles were completely nontoxic to cultured cells. Moreover, DOX-loaded micelles exhibited efficient antitumor efficiency against cancer cells. Therefore, the micelles may be potential drug carriers in cancer therapy.

Abstract:The development of intelligent biomaterials system that can efficiently accumulate at the tumor site and release drug in a controlled way is very important for cancer chemotherapy. PEG is widely selected as hydrophilic shell to acquire prolonged circulation time and enhanced accumulation at tumor site, but it also restrains the cellular transport and uptake and leads to insufficient therapeutic efficacy. In this work, a PEG-detachable pH-responsive polymer form micelles from copolymer cholesterol grafted poly(ethylene glycol) methyl ether-Dlabile-poly(β-amino ester)-Dlabile-poly(ethylene glycol) methyl ether (MPEG-Dlabile-PAE-g-Chol) is developed to overcome aforementioned challenges based on pH value changes among normal physiological, extracellular (pHe) and intracellular (pHi) environment. PEGylated doxorubicin (DOX)-loaded polymeric micelles (DOX-PMs) can accumulate at the tumor site via enhanced permeability and retention (EPR) effect, and PEG shell is detachable induced by cleavage of pHe-labile linker between PEG segment and the main chain. Meanwhile, the pHi-sensitive poly(β-amino ester) (PAE) segment is protonated and has high positive charge. The detachment of PEG and protonation of PAE facilitate cellular uptake of DOX- polymeric micelles (PMs) by negatively charged tumor cells, along with the escape from endo/lysosome due to "proton-sponge" effect. The DOX molecules are controlled release from the carries at specific pH values. The results demonstrate that DOX-PMs have capability of show high therapeutic efficacy and negligible cytotoxicity compared with free DOX in vitro and in vivo. Overall, we anticipate that this PEG-detachable and tumor-acidity-responsive polymeric micelle can mediate effective and biocompatible drug delivery "on-demand" with clinical application potential.

Abstract:An amphiphilic metallo-supramolecular poly(propylene glycol)-block-poly(ethylene glycol) block copolymer containing a bis(2,2':6',2"-terpyridine) ruthenium (II) complex as a supramolecular connection between the two constituting blocks was used to prepare stable aqueous micelles which displayed a high intensity focused ultrasound (HIFU) triggered release behavior. By adjusting the HIFU time and intensity, the novel modality of HIFU triggered release allows for fine-tuning of the release kinetics of the encapsulants from the micelles in a remote and controlled way. Nuclear magnetic resonance spectroscopy, ultraviolet spectroscopy, and matrix-assisted laser desorption/ionization-time of flight mass spectrometry confirmed that the degradation of the micelles was due to the cleavage of the ether bond connected to the pyridine ring. This well controlled HIFU-copolymer micelle drug delivery system has considerable potential in targeted therapy.

Abstract:The hydrophobic block of polymeric micelles formed by amphiphilic copolymers has no direct therapeutical effect, and the metabolites of these hydrophobic segments might lead to some unexpected side effects. Here the hydrophobic core of polymeric micelles is replaced by highly water-insoluble drugs themselves, forming a new micellar drug delivery system. By grafting hydrophobic drugs of paclitaxel (PTX) onto the surface of hydrophilic hyperbranched poly(ether-ester) (HPEE), we constructed an amphiphilic copolymer (HPEE-PTX). HPEE-PTX could self-assemble into micellar nanoparticles in aqueous solution with tunable drug contents from 4.1 to 10.7%. Moreover, the hydrolysis of HPEE-PTX in serum resulted in the cumulative release of PTX. In vivo evaluation indicated that the dosage toleration of PTX in mice had been improved greatly and HPEE-PTX micellar nanoparticles could be used as an efficient prodrug with satisfactory therapeutical effect. We believe that most of the lipophilic drugs could improve their characters through this strategy.

Abstract:Well-defined amphiphilic poly(ethylene glycol) and poly(dehydroabietic ethyl methacrylate) block copolymers (PEG-b-PDAEMA) were prepared by atom transfer radical polymerization. The methacrylate block contains a characteristic hydrophobic, biocompatible and economical dehydroabietic moiety. PEG-b-PDAEMA block copolymer micellar nanoparticles loaded with piperlongumine (PLGM) were successfully prepared by a nanoprecipitation method. In vitro and in vivo behaviors of these nanoparticles were thoroughly examined by a set of characterization techniques. Confocal laser scanning microscopy study revealed that these nanoparticles could be well taken up by cancer cells. In vivo near-infrared fluorescence imaging showed that the PLGM-loaded nanoparticles effectively targeted the tumor site by the enhanced permeability and retention (EPR) effect in H22 tumor-bearing mice. The in vivo antitumor examination found that PLGM-loaded nanoparticles exhibited superior efficacy in impeding the tumor growth compared to the commercially available Taxol (R) and free PLGM formulation. The changes in body weights and blood biochemical profiles were also compared to investigate the safety of PLGM and PEG-b-PDAEMA nanoparticle drug delivery system.
Leakage-free DOX/PEGylated Chitosan Micelles Fabricated Via Facile One-Step Assembly for Tumor Intracellular Ph-Triggered Release.

Abstract:A general strategy was developed to fabricate the chitosan-based micelles as smart drug delivery system (DDS) for anti-cancer drug doxorubicin (DOX), via modulating the feeding ratio of PEG (PEG2000-CHO) and molecular weight of chitosan. Typically, benefiting from the solubility of chitosan in media with different pH values as an "off-on" switch, the DOX/CS100K-PEG2 micelles possess spherical core with a narrow dispersed diameter of 64nm and regular shell with a thickness of 14nm in pH 7.4 PBS. Interestingly, the DOX/CS-PEG micelles exhibited drug leakage-free behavior in a physiological environment, with a cumulative release ratio of only 2.32% under pH 7.4 PBS, while achieving rapid drug release and a cumulative release ratio of 72.76% in the intracellular microenvironment. Moreover, the micelles transformed into the water-soluble CS100K-PEG2 after the pH-triggered intracellular release of DOX, and it makes them easy to be biodegraded and eliminated by metabolic system. As revealed by MTT assays and CLSM images, DOX could be efficiently and rapidly transported into cell nuclei of tumor cells and showed significant cell inhibition. The simple fabrication and excellent properties make them intelligent and favorable on-demand DDS in cancer treatment.

Abstract:A series of novel amphiphilic triblock copolymers of poly(ethyl ethylene phosphate) and poly(-caprolactone) (PEEP-PCL-PEEP) with various PEEP and PCL block lengths were synthesized and characterized. These triblock copolymers formed micelles composed of a hydrophobic core of poly(-caprolactone) (PCL) and a hydrophilic shell of poly(ethyl ethylene phosphate) (PEEP) in aqueous solution. The micelle morphology was spherical, determined by transmission electron microscopy. It was found that the size and critical micelle concentration values of the micelles depended on both hydrophobic PCL block length and PEEP hydrophilic block length. The in vitro degradation characteristics of the triblock copolymers were investigated in micellar form, showing that these copolymers were completely biodegradable under enzymatic catalysis of Pseudomonas lipase and phosphodiesterase I. These triblock copolymers were used for paclitaxel (PTX) encapsulation to demonstrate the potential in drug delivery. PTX was successfully loaded into the micelles, and the in vitro release profile was found to be correlative to the polymer composition. These biodegradable triblock copolymer micelles are potential as novel carriers for hydrophobic drug delivery.

Abstract:The diblock copolymer, PEG-b-PMEA, was synthesized by reversible-addition fragmentation chain transfer polymerization (RAFT). The PMEA block contained a polymethacrylamide backbone and twin ortho ester rings in the side-chains. At neutral pH, PEG-b-PMEA self-assembled to form stable micelles. At pH 5, the twin ortho ester rings were quickly hydrolyzed to completion in 12h, and releasing nearly 70% of the encapsulated Nile Red dye. The PEG-b-PMEA micelles were completely nontoxic to cultured cells as determined by the MTT assay. Paclitaxel (PTX)-loaded micelles showed toxicity toward lung cancer cells comparable to that of the free PTX at equivalent doses. These results suggest that the PEG-b-PMEA micelles could be useful nano-carriers for pH-responsive delivery of poorly soluble anticancer drugs.
Synthesis of Branched Cyclic Copolymer for pH-responsive Drug Delivery System

Abstract:A branched cyclic amphiphilic block copolymer ( PEG-b-PACDs ) was synthesized via a deactivation-enhanced atom transfer radical polymerization ( DE-ATRP) with brominated polyethylene glycol (PEG2KBr) and di(2-acry-loyloxy ethoxy)-[4-methoxy-phenyl]methane (ACD) as initiator and hydrophobic monomer, respectively. The blank micelles were spherical in shape with a particle size of 70 nm, while the particle size of doxorubicin ( DOX)-loaded micelles were about 90 nm. The encapsulation efficiencies of DOX-loaded micelles were 48. 7%. The DOX-loaded PEG-b-PACDs shows obvious pH-responsive drug release property under acidic condition.
Supramolecular Copolymer Micelles Based on the Complementary Multiple Hydrogen Bonds of Nucleobases for Drug Delivery.

Abstract:Novel supramolecular copolymer micelles with stimuli-responsive abilities were successfully prepared through the complementary multiple hydrogen bonds of nucleobases and then applied for rapid intracellular release of drugs. First, both adenine-terminated poly(ε-caprolactone) (PCL-A) and uracil-terminated poly(ethylene glycol) (PEG-U) were synthesized. The supramolecular amphiphilic block copolymers (PCL-A:U-PEG) were formed based on multiple hydrogen bonding interactions between PCL-A and PEG-U. The micelles self-assembled from PCL-A:U-PEG were sufficiently stable in water but prone to fast aggregation in acidic condition due to the dynamic and sensitive nature of noncovalent interactions. The low cytotoxicity of supramolecular copolymer micelles was confirmed by MTT assay against NIH/3T3 normal cells. As a hydrophobic anticancer model drug, doxorubicin (DOX) was encapsulated into these supramolecular copolymer micelles. In vitro release studies demonstrated that the release of DOX from micelles was significantly faster at mildly acid pH of 5.0 compared to physiological pH. MTT assay against HeLa cancer cells showed DOX-loaded micelles had high anticancer efficacy. Hence, these supramolecular copolymer micelles based on the complementary multiple hydrogen bonds of nucleobases are very promising candidates for rapid controlled release of drugs.
Novel Biodegradable Polylactide/poly(ethylene glycol) Micelles Prepared by Direct Dissolution Method for Controlled Delivery of Anticancer Drugs

Abstract:Purpose The aim of this study is to develop novel polylactide/poly(ethylene glycol) (PLA/PEG) micelles as carrier of hydrophobic drug (paclitaxel) by direct dissolution method without using any organic solvents. The in vitro and in vivo release properties were studied in comparison with micelles prepared by dialysis. Methods Drug encapsulation efficiency (EE) and loading content (LC) of the micelles were evaluated by high-performance liquid chromatography. Micelle diameters and structures were determined by dynamic light scattering and transmission electron microscopy. In vitro release was performed in phosphate-buffered saline (pH 7.4) at 37°C, and in vivo experiments were realized in lung cancer-bearing mice. Results Similar EE and LC values were obtained for micelles by direct dissolution method and those by dialysis. L- and D-PLA/PEG mixed micelles present higher drug encapsulation ability than separate micelles due to stereocomplexation. Micelle diameters are enlarged by drug-loading. Faster drug release was obtained for micelles by direct dissolution than those by dialysis. Compared with current clinical formulation and micelles by dialysis, paclitaxel-loaded micelles by direct dissolution showed the highest antitumor ability. Conclusion The L- and D-PLA/PEG mixed micelles by direct dissolution method present many advantages such as easy formulation and absence of toxic organic solvents, which shows great potential as carrier of hydrophobic drugs.
