Abstract:The availability of non-invasive technologies, which can be used separately or in combination for obtaining chemical composition data and structural information of Cultural Heritage (CH) materials, is of prime importance for improving the understanding the environmental or ageing impact on monuments and artefacts and defining optimal strategies for their conservation. This paper overviews and assesses the potential of two emerging photonic technologies, the Non-linear Optical Microscopy (NLOM) and Photoacoustic (PA) imaging modalities, for a variety of diagnostic applications in preservation science. These techniques, which are well-established in biomedical research, during the last few years have been also investigated as non-invasive tools for the in-depth, high-resolution analysis of various CH objects, including paintings, documents and murals. We discuss on the applicability of these diagnostic optical methods to obtain precise stratigraphic information in artefacts, evaluating additionally the presence and the extent of potential morphological or chemical changes in several CH materials due to ageing. Furthermore, we demonstrate that the contrast complementarity of NLOM and PA imaging provides invaluable insights into the structural integrity of an artwork, which can be subsequently utilized for the early and accurate detection of depth degradation effects.

Anisotropic NMR data acquisition with a prototype 400 MHz cryogen-free NMR spectrometer

Abstract:High-temperature superconducting (HTS) materials have recently been incorporated into the construction of HTS cryogen-free magnets for nuclear magnetic resonance (NMR) spectroscopy. These HTS NMR spectrometers do not require liquid cryogens, thereby providing significant cost savings and facilitating easy integration into chemistry laboratories. However, the optimal performance of these HTS magnets against standard cryogen NMR magnets must be evaluated, especially with demanding modern NMR applications such as NMR in anisotropic media. The stability of the HTS magnets over time and their performance with complex pulse sequence experiments are the main unknown factors of this new technology. In this study, we evaluate the utility of our prototype 400 MHz cryogen-free power-driven HTS NMR spectrometer, installed in the fumehood of a chemistry laboratory, for stereochemical analysis of three commercial natural products (artemisinin, artemether, and dihydroartemisinin) via measurement of anisotropic NMR data, in particular, residual dipolar couplings. The accuracy of measurement of the anisotropic NMR data with the HTS magnet spectrometer is evaluated through the CASE-3D fitting protocol, as implemented in the Mestrenova-StereoFitter software program.
Advanced Ground Truth Multimodal Imaging Using Time Reversal (TR) Based Nonlinear Elastic Wave Spectroscopy (NEWS): Medical Imaging Trends Versus Non-destructive Testing Applications

Abstract:An innovative pragmatic approach is described using a symbiosis of modern signal processing techniques coming from nonlinear science and multiscale global analysis usually considered for the characterization of nanoscale systems with complex mesoscopic properties. The basis of this systemic approach comes from ultrasound imaging of the complexity with examples coming from the non-destructive testing (NDT) industry and from the medical harmonic imaging research. Modeling uses basic tools derived from the analysis of nonlinear dynamics, such as spectrum representations coming from harmonic analysis and multiscale analysis, in association with advanced signal processing such as similarity and invariance analysis based on group theory. Experimentation is based on multimodal imaging using time reversal (TR) based nonlinear elastic wave spectroscopy (NEWS). Practical implementation in the aeronautic industry and the biomedical imaging field is taken as a demonstration of feasibility of such advanced engineering TR-NEWS methods for imaging complex systems with an advanced ground truth approach including round robin tests. This new class of multiscale materials is studied with phenomenological approaches like the Preisach–Mayergoyz space (PM space), where some physical properties like hysteresis, end-point memory, and odd harmonic generations are extracted from the noise. As a consequence, this approach is proposed for including phenomenological tools to any organizations’ strategies in order to monitor the vulnerability in a societal emergency management context. These results could be applied for the organizational side of innovation, cognition, and practices for novelty and disruption in political organizations, social monitoring, and any system which will involve big data or artificial intelligence.
Sustainable and cost-effective MAS DNP-NMR at 30 K with cryogenic sample exchange

Abstract:We report here instrumental developments to achieve sustainable, cost-effective cryogenic Helium sample spinning in order to conduct dynamic nuclear polarisation (DNP) and solid-state NMR (ssNMR) at ultra-low temperatures (<30 K). More specifically, we describe an efficient closed-loop helium system composed of a powerful heat exchanger (95% efficient), a single cryocooler, and a single helium compressor to power the sample spinning and cooling. The system is integrated with a newly designed triple-channel NMR probe that minimizes thermal losses without compromising the radio frequency (RF) performance and spinning stability (±0.05%). The probe is equipped with an innovative cryogenic sample exchange system that allows swapping samples in minutes without introducing impurities in the closeloop system. We report that significant gain in sensitivity can be obtained at 30-40 K on large micro-crystalline molecules with unfavorable relaxation timescales, making them difficult or impossible to polarize at 100 K. We also report rotor-synchronized 2D experiments to demonstrate the stability of the system.
Methodological Developments for Metabolic NMR Spectroscopy from Cultured Cells to Tissue Extracts: Achievements, Progress and Pitfalls

Abstract:This is a broad overview and critical review of a particular group of closely related ex vivo and in vitro metabolic NMR spectroscopic methods. The scope of interest comprises studies of cultured cells and excised tissue, either intact or after physicochemical extraction of metabolites. Our detailed discussion includes pitfalls that have led to erroneous statements in the published literature, some of which may cause serious problems in metabolic and biological interpretation of results. To cover a wide range of work from relevant research areas, we consider not only the most recent achievements in the field, but also techniques that proved to be valid and successful in the past, although they may not have generated a very significant number of papers more recently. Thus, this comparative review also aims at providing background information useful for judiciously choosing between the metabolic ex vivo/in vitro NMR methods presented. Finally, the methods of interest are discussed in the context of, and in relation to, other metabolic analysis protocols such as HR-MAS and cell perfusion NMR, as well as the mass spectrometry approach.

Abstract:Modern life science research is a collaborative effort. Few research groups can single‐handedly support the necessary equipment, expertise and personnel needed for the ever‐expanding portfolio of technologies that are required across multiple disciplines in today's life science endeavours. Thus, research institutes are increasingly setting up scientific core facilities to provide access and specialised support for cutting‐edge technologies. Maintaining the momentum needed to carry out leading research while ensuring high‐quality daily operations is an ongoing challenge, regardless of the resources allocated to establish such facilities. Here, we outline and discuss the range of activities required to keep things running once a scientific imaging core facility has been established. These include managing a wide range of equipment and users, handling repairs and service contracts, planning for equipment upgrades, renewals, or decommissioning, and continuously upskilling while balancing innovation and consolidation.

Nutation-Based Longitudinal Sensing Protocols for High-Field NMR With Nitrogen-Vacancy Centers in Diamond

Abstract:Nitrogen vacancy (NV) centers in diamond enable nuclear magnetic resonance (NMR) spectroscopy of samples at the nano- and micron scales. However, at typical tesla-scale NMR magnetic field strengths, NV-NMR protocols become difficult to implement due to the challenge of driving fast NV pulse sequences sensitive to nuclear Larmor frequencies above a few megahertz. We perform simulations and theoretical analysis of the experimental viability of NV-NMR at tesla-scale magnetic fields using a new measurement protocol called DRACAERIS (Double Rewound ACquisition Amplitude Encoded Radio Induced Signal). DRACAERIS detects the NMR sample's longitudinal magnetization at a much lower driven Rabi frequency, more suitable technically for NV detection. We discuss how pulse errors, finite pulse lengths, and nuclear spin-spin couplings affect the resulting NMR spectra. We find that DRACAERIS is less susceptible to pulse imperfections and off-resonance effects than previous protocols for longitudinal magnetization detection. We also identify reasonable parameters for experimental implementation.

Abstract:Nuclear Magnetic Resonance (NMR) is a non-contact, powerful structure-elucidation technique for biochemical analysis. NMR spectroscopy is used extensively in a variety of life science applications including drug discovery. However, existing NMR technology is limited in that it cannot run a large number of experiments simultaneously in one unit. Recent advances in micro-fabrication technologies have attracted the attention of researchers to overcome these limitations and significantly accelerate the drug discovery process by developing the next generation of high-throughput NMR spectrometers using Complementary Metal Oxide Semiconductor (CMOS). In this paper, we examine this paradigm shift and explore new design strategies for the development of the next generation of high-throughput NMR spectrometers using CMOS technology. A CMOS NMR system consists of an array of high sensitivity micro-coils integrated with interfacing radio-frequency circuits on the same chip. Herein, we first discuss the key challenges and recent advances in the field of CMOS NMR technology, and then a new design strategy is put forward for the design and implementation of highly sensitive and high-throughput CMOS NMR spectrometers. We thereafter discuss the functionality and applicability of the proposed techniques by demonstrating the results. For microelectronic researchers starting to work in the field of CMOS NMR technology, this paper serves as a tutorial with comprehensive review of state-of-the-art technologies and their performance levels. Based on these levels, the CMOS NMR approach offers unique advantages for high resolution, time-sensitive and high-throughput bimolecular analysis required in a variety of life science applications including drug discovery.

Abstract:Introduction: Historic data, often in analog format, is a valuable resource for assessing effects of directional changes in climate and climatic variability. However, historic data can be difficult to locate, interpret, and reformat into a useful state. Methods: Teams of scientists, librarians, archivists, and data managers at four US institutions have undertaken various projects to gather, describe, and in some cases, transform historic data. They have also surveyed researchers who either possess historic data or have used it in their work. Results: Historic data projects involved locating data, writing data descriptions, and connecting with individuals who had knowledge about the data's collection. The surveys and interviews found that researchers valued historic data and were worried that it was at risk of loss. They noted the lack of best practices. Discussion: Each project attempting to rescue or enhance access to historic data has a unique path but being guided by FAIR principles should be at the core whether or not the end result is machine-readable data. Working with a team incorporating librarians, archivists, and data managers can aid individual researchers' in producing accessible, and reusable datasets. There is much work to be done in raising awareness about the value of historic data but motivating factors for doing so include its usefulness in environmental research and other disciplines and its risk of loss as researchers retire and are unsure of how to save historic data, both in analog and electronic formats.

Abstract:Science, especially large-scale basic research, is a collaborative endeavor, often drawing on the skills of people from a wide variety of disciplines. These people include not just scientists, but also administrators, engineers, and many others. Fermilab, a Department of Energy National Laboratory and the United States' premier particle physics laboratory, exemplifies this kind of research; many of its high-energy physics experiments involve hundreds of collaborators from all over the world. The Fermilab Archives seeks to document the history of the lab and the unique scientific research its staff and visitors perform. Adequately documenting the lab's work often requires us to go far beyond things like the writings and correspondence of scientists to also capture the administrative and social histories of the experiments and the context in which they were performed. At Fermilab, we have sought to capture these elements of the lab's activities through an oral history program that focuses on support staff as well as physicists and collection development choices that recognize the importance of records documenting the cultural life of the lab. These materials are not merely supplementary, but rather essential documentation of the many types of labor that go into the planning and execution of an experiment or the construction of an accelerator and the context in which this work is performed.

Abstract:This Faraday Discussion explored the field of NMR crystallography, and considered recent developments in experimental and theoretical approaches, new advances in machine learning and in the generation and handling of large amounts of data. Applications to a wide range of disordered, amorphous and dynamic systems demonstrated the range and quality of information available from this approach and the challenges that are faced in exploiting automation and developing best practice. In these closing remarks I will reflect on the discussions on the current state of the art, questions about what we want from these studies, how accurate we need results to be, how we best generate models for complex materials and what machine learning approaches can offer. These remarks close with thoughts about the future direction of the field, who will be carrying out this type of research, how they might be doing it and what their focus will be, along with likely possible challenges and opportunities.

Abstract:Since commencing operation in 2014, the Dingo neutron imaging facility has expanded capabilities of investigating materials of cultural heritage significance at the Australian Nuclear Science and Technology Organisation. This paper offers an overview of the diverse range of artefacts investigated on the beamline during the last decade. A selection of case studies established through national and international collaborations showcases how neutron imaging can provide university and museum users with a powerful analytical tool for non-invasive real-space evaluation of a variety of ancient artefacts with properties complementary to other nuclear methods.

Abstract:State-of-the-art NMR spectrometers using superconducting magnets have enabled, with their ultrafine spectral resolution, the determination of the structure of large molecules such as proteins, which is one of the most profound applications of modern NMR spectroscopy. Many chemical and biotechnological applications, however, involve only small-to-medium size molecules, for which the ultrafine resolution of the bulky, expensive, and high-maintenance NMR spectrometers is not required. For these applications, there is a critical need for portable, affordable, and low-maintenance NMR spectrometers to enable in-field, on-demand, or online applications (e.g., quality control, chemical reaction monitoring) and co-use of NMR with other analytical methods (e.g., chromatography, electrophoresis). As a critical step toward NMR spectrometer miniaturization, small permanent magnets with high field homogeneity have been developed. In contrast, NMR spectrometer electronics capable of modern multidimensional spectroscopy have thus far remained bulky. Complementing the magnet miniaturization, here we integrate the NMR spectrometer electronics into 4-mm(2) silicon chips. Furthermore, we perform various multidimensional NMR spectroscopies by operating these spectrometer electronics chips together with a compact permanent magnet. This combination of the spectrometer-electronics-on-a-chip with a permanent magnet represents a useful step toward miniaturization of the overall NMR spectrometer into a portable platform.

Abstract:NMR spectroscopy is an indispensable method of analysis in chemistry, which until recently suffered from high demands for space, high costs for acquisition and maintenance, and operational complexity. This has changed with the introduction of compact NMR spectrometers suitable for small-molecule analysis on the chemical workbench. These spectrometers contain permanent magnets giving rise to proton NMR frequencies between 40 and 80 MHz. The enabling technology is to make small permanent magnets with homogeneous fields. Tabletop instruments with inhomogeneous fields have been in use for over 40 years for characterizing food and hydrogen-containing materials by relaxation and diffusion measurements. Related NMR instruments measure these parameters in the stray field outside the magnet. They are used to inspect the borehole walls of oil wells and to test objects nondestructively. The state-of-the-art of NMR spectroscopy, imaging and relaxometry with compact instruments is reviewed.

Abstract:Nuclear magnetic resonance (NMR) spectroscopy is an indispensable technique in the organic chemistry laboratory for elucidation of molecular structure. Miniaturization of NMR technology has attracted attention and acceptance due to a considerable reduction in the cost to acquire and maintain the instrument. The advent of low-field, low-cost, and low-maintenance tabletop NMR technology has provided academic institutions with limited resources the option of hands-on NMR experimentation for undergraduates. Abstract:Progress evolves. Like mortality, it has a unique timeline. New technology can enhance previous methodology and bring new insight to research; new interpretations build on the shoulders of what has already been learned or divined. In all matters pertaining to science and research, we move onwards and upwards and the arrow of time is always assumed to be pointing forwards. Nevertheless, there are occasions when critical information can only be gleaned from the past, when the inclusion of heritage data is pivotal to a research problem. Modern data management is not on our side in this respect, and the matter is growing more serious daily. In this chapter, we (a) examine the present state of affairs regarding those heritage data which deserve and need far more attention than they have been afforded until now, (b) outline some of the science that can only be made possible by including past observations along with new ones, and (c) discuss how we might focus a global effort on this problem before it is too late.
Complex mixtures by NMR and complex NMR for mixtures: experimental and publication challenges

Abstract:Untargeted strategies have changed the rules of the game in complex mixture analysis, introducing an amazing potential for medical and biological applications that is just starting to be tapped. But with great power come great challenges; although untargeted mixture analysis opens the road for many exciting possibilities, the road is still full of perils. On the one hand, this article highlights some of the difficulties that need to be sorted for mixture analysis by NMR to fulfill its potential, along with insight on how they may be managed. Highlighted key points include the need for 'computer friendly' solutions for sharing data, experimental design and algorithm to facilitate the steady growth of knowledge and modeling ability in the field, and the need for large-scale studies to improve confidence in newly identified biomarkers. On the other hand, the second part of this article presents some breakthroughs in NMR experiments that, when combined, may modify the landscape of mixture analysis. The mission of the INFN-Cultural Heritage Network: the multifaceted example of the Macro-XRF scanner experience

Abstract:Abstract The mission of the Cultural Heritage Network of the Italian Institute of Nuclear Physics (INFN-CHNet) is presented here through a “virtuous” example: the activity related to the Macro-X-Ray Fluorescence (MA-XRF) scanner. The main focus of INFN-CHNet is the collaboration and sharing of knowledge among the network members, to better address the issues of Cultural Heritage operators, while the fields of activity are research (instrumental development and data management), analysis of cultural objects, education of young scholars and professionals, and dissemination to the general public. In this context, the MA-XRF scanner exemplifies the objectives of INFN-CHNet, being the first instrument developed, tested and optimised jointly by several INFN-CHNet partners. It was designed to be an open (free access to information on processes, components and materials), low-cost system, based on sustainability and interoperability, and modular. All these features allow for easy replication of instruments in any node of the network or reuse of system/subsystem also in different projects. Moreover, for the MA-XRF scanner, the whole path from data acquisition to their storage, management and visualisation/reuse has been addressed. The activities related to the scanner clearly demonstrate the effectiveness of the approach of CHNet in all the issues that arise in conservation and study of CH.
