Abstract:Maintaining translation fidelity is a critical step within the process of gene expression. It requires the involvement of numerous regulatory elements to ensure the synthesis of functional proteins. The efficient termination of protein synthesis can play a crucial role in preserving this fidelity. Here, we report on investigating a protein of unknown function, YNR069C (also known as BSC5), for its activity in the process of translation. We observed a significant increase in the bypass of premature stop codons upon the deletion of YNR069C. Interestingly, the genomic arrangement of this ORF suggests a compatible mode of expression reliant on translational readthrough, incorporating the neighboring open reading frame. We also showed that the deletion of YNR069C results in an increase in the rate of translation. Based on our results, we propose that YNR069C may play a role in translation fidelity, impacting the overall quantity and quality of translation. Our genetic interaction analysis supports our hypothesis, associating the role of YNR069C to the regulation of protein synthesis.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the role of the gene YNR069C (also known as BSC5) with unknown function in *Saccharomyces cerevisiae* in the process of protein synthesis. Specifically, the researchers focused on the possible role that the YNR069C gene may play in the translation termination process, especially how it affects the readthrough of Premature Termination Codons (PTCs) and the translation rate. ### Main research questions: 1. **The role of YNR069C in translation termination**: By deleting the YNR069C gene, the researchers observed that its deletion led to an increase in read - through at premature termination codons, indicating that YNR069C may be involved in the translation termination process. 2. **The influence of YNR069C on the translation rate**: Further research found that the deletion of YNR069C also led to an increase in the translation rate, suggesting that YNR069C may play a balancing role between maintaining the accuracy and speed of translation. 3. **Function verification of YNR069C**: To verify these observations, the researchers conducted large - scale screening experiments, using β - galactosidase reporter gene constructs with different premature termination codons, as well as quantitative β - galactosidase activity assays and qRT - PCR analysis to confirm changes in translation activities. 4. **Genetic interaction analysis**: Through Synthetic Genetic Array (SGA) analysis, the researchers explored the genetic interactions between YNR069C and other genes, further supporting the role of YNR069C in translation regulation. ### Research background: - **Translation termination**: Translation termination occurs when the ribosome A - site encounters one of the three stop codons (UAA, UAG, UGA) and requires the participation of release factors eRF1 and eRF3 to complete. - **Read - through of premature termination codons**: When a nearly matching tRNA binds to the stop codon instead of eRF1, it will cause the translation to continue until the next in - frame stop codon. This phenomenon is called read - through. - **Characteristics of YNR069C**: YNR069C encodes a protein with unknown function, but previous studies have shown that it may carry a functional premature termination codon and has a high read - through rate. ### Research significance: - **Maintenance of translation fidelity**: Understanding the role of YNR069C in the translation process helps to reveal how cells control gene expression, especially in terms of translation fidelity. - **Potential therapeutic applications**: The research results may provide clues for the development of new treatment methods, for example, using the translation read - through mechanism to treat some genetic diseases caused by premature termination codons. In conclusion, this paper aims to comprehensively analyze the specific function and mechanism of YNR069C in yeast protein synthesis through various experimental methods.
