Abstract:Laser Doppler holography (LDH) is a full-field imaging technique that was recently used in the human eye to reveal blood flow contrasts in the retinal and choroidal vasculature non-invasively, and with high temporal resolution. We here demonstrate that the ability of LDH to perform quantitative flow measurements with high temporal resolution enables arteriovenous differentiation in the retina and choroid. In the retina, arteries and veins can be differentiated on the basis of their respective power Doppler waveforms. Choroidal arteries and veins can instead be discriminated by computing low and high frequency power Doppler images to reveal low and high blood flow images, respectively.

Abstract:Laser Doppler holography (LDH) is a full-field blood flow imaging technique able to reveal human retinal and choroidal blood flow with high temporal resolution. We here report on using LDH in the anterior segment of the eye without making changes to the instrument. Blood flow in the bulbar conjunctiva and episclera as well as in corneal neovascularization can be effectively imaged. We additionally demonstrate simultaneous holographic imaging of the anterior and posterior segments by simply adapting the numerical propagation distance to the plane of interest. We used this feature to track the movements of the retina and pupil with high temporal resolution. Finally, we show that the light backscattered by the retina can be used for retro-illumination of the anterior segment. Hence digital holography can reveal opacities caused by absorption or diffusion in the cornea and eye lens.
Retinal blood flow reversal quantitatively monitored in out-of-plane vessels with laser Doppler holography

Abstract:Laser Doppler holography is a planar blood flow imaging technique recently introduced in ophthalmology to image human retinal and choroidal blood flow non-invasively. Here we present a digital method based on the Doppler spectrum asymmetry that reveals the local direction of blood flow with respect to the optical axis in out-of-plane vessels. This directional information is overlaid on standard grayscale blood flow images to depict flow moving towards the camera in red and flow moving away from the camera in blue, as in ultrasound color Doppler imaging. We show that thanks to the strong contribution of backscattering to the Doppler spectrum in out-of-plane vessels, the local axial direction of blood flow can be revealed with a high temporal resolution, which enables us to evidence pathological blood flow reversals. We also demonstrate the use of optical Doppler spectrograms to quantitatively monitor retinal blood flow reversals.

Abstract:Laser Doppler holography was introduced as a full-field imaging technique to measure blood flow in the retina and choroid with an as yet unrivaled temporal resolution. We here investigate separating the different contributions to the power Doppler signal in order to isolate the flow waveforms of vessels in the posterior pole of the human eye. Distinct flow behaviors are found in retinal arteries and veins with seemingly interrelated waveforms. We demonstrate a full field mapping of the local resistivity index, and the possibility to perform unambiguous identification of retinal arteries and veins on the basis of their systolodiastolic variations. Finally we investigate the arterial flow waveforms in the retina and choroid and find synchronous and similar waveforms, although with a lower pulsatility in choroidal vessels. This work demonstrates the potential held by laser Doppler holography to study ocular hemodynamics in healthy and diseased eyes.

Abstract:Laser Doppler holography (LDH) is a full-field interferometric imaging technique recently applied in ophthalmology to measure blood flow, a parameter of high clinical interest. From the temporal fluctuations of digital holograms acquired at ultrafast frame rates, LDH reveals retinal and choroidal blood flow with a few milliseconds of temporal resolution. However, LDH experiences difficulties to detect slower blood flow as it requires to work with low Doppler frequency shifts which are corrupted by eye motion. We here demonstrate the use of a spatio-temporal decomposition adapted from Doppler ultrasound that provides a basis appropriate to the discrimination of blood flow from eye motion. A singular value decomposition (SVD) can be used as a simple, robust, and efficient way to separate the Doppler fluctuations of blood flow from those of strong spatial coherence such as eye motion. We show that the SVD outperforms the conventional Fourier based filter to reveal slower blood flow, and dramatically improves the ability of LDH to reveal vessels of smaller size or with a pathologically reduced blood flow.

Abstract:The choroid is a highly vascularized tissues supplying the retinal pigment epithelium and photoreceptors. Its implication in retinal diseases is gaining increasing interest. However, investigating the anatomy and flow of the choroid remains challenging. Here we show that laser Doppler holography provides high contrast imaging of choroidal vessels in humans, with a spatial resolution comparable to state of the art indocyanine green angiography and optical coherence tomography. Additionally, laser Doppler holography contributes to sort out choroidal arteries and veins by using a power Doppler spectral analysis. We thus demonstrate the potential of laser Doppler holography to improve our understanding of the anatomy and flow of the choroidal vascular network.

Abstract:The eye offers a unique opportunity for non-invasive exploration of cardiovascular diseases. Optical angiography in the retina requires sensitive measurements, which hinders conventional full-field laser Doppler imaging schemes. To overcome this limitation, we used digital holography to perform laser Doppler perfusion imaging of the human retina in vivo with near-infrared light. Wideband measurements of the beat frequency spectrum of optical interferograms recorded with a 39 kHz CMOS camera are analyzed by short-time Fourier transformation. Power Doppler images and movies drawn from the zeroth moment of the power spectrum density reveal blood flows in retinal and choroidal vessels over 512 $\times$ 512 pixels covering 2.4 $\times$ 2.4 mm$^2$ on the retina with a 13 ms temporal resolution.

Abstract:This study presents a novel quantitative estimation method for total retinal arterial blood flow utilizing real-time Doppler holography at an unprecedented frame rate of 33,000 frames per second. This technique, leveraging high-speed digital holography, enables non-invasive angiographic imaging of the retina, providing detailed blood flow contrasts essential for assessing retinal health. The proposed quantitative analysis method consists of segmenting primary in-plane retinal arteries and calculating local blood velocity using Doppler frequency broadening. The analysis integrates a forward scattering model to achieve blood flow estimation. Our findings highlight the potential of Doppler holography as a powerful tool for diagnosing and monitoring the treatment of retinal vascular conditions, complementary to existing imaging methods.

Abstract:We report on local superficial blood flow monitoring in biological tissue from laser Doppler holographic imaging. In time averaging recording conditions, holography acts as a narrowband bandpass filter, which, combined with a frequency shifted reference beam, permits frequency selective imaging in the radiofrequency range. These Doppler images are acquired with an off axis Mach Zehnder interferometer. Microvascular hemodynamic components mapping is performed in the cerebral cortex of the mouse and the eye fundus of the rat with near-infrared laser light without any exogenous marker. These measures are made from a basic inverse method analysis of local first order optical fluctuation spectra at low radiofrequencies, from 0 Hz to 100 kHz. Local quadratic velocity is derived from Doppler broadenings induced by fluid flows, with elementary diffusing wave spectroscopy formalism in backscattering configuration. We demonstrate quadratic mean velocity assessment in the 0.1 to 10 millimeters per second range in vitro and imaging of superficial blood perfusion with a spatial resolution of about 10 micrometers in rodent models of cortical and retinal blood flow.

Abstract:We report on wide-field imaging of pulsatile motion induced by blood flow using heterodyne holographic interferometry on the thumb of a healthy volunteer, in real-time. Optical Doppler images were measured with green laser light by a frequency-shifted Mach-Zehnder interferometer in off-axis configuration. The recorded optical signal was linked to local instantaneous out-of-plane motion of the skin at velocities of a few hundreds of microns per second, and compared to blood pulse monitored by plethysmoraphy during an occlusion-reperfusion experiment.

Abstract:A comprehensive assessment of retinal health demands reliable and precise methods to measure localized blood perfusion. Despite considerable advancements in imaging techniques, such as indocyanine green and fluorescein angiography, along with optical coherence tomography angiography, their capacity to monitor blood flow dynamics across the cardiac cycle faces significant limitations. For more effective care of those with ocular conditions, innovating new approaches is paramount. Doppler holography, an emerging non-invasive optical imaging technique, meets this challenge by offering high temporal resolution imaging of retinal and choroidal blood flow. Now a burgeoning interdisciplinary research field, Doppler holography intertwines functional optical imaging system design, high-performance computing, and clinical investigation. Through collaborative efforts among universities, industry partners, and ophthalmic clinics, a network for its advancement and application is taking shape. This endeavor promises to propel the discovery of novel functional biomarkers, transforming the diagnosis and treatment of retinal diseases, refining disease severity categorization, and enhancing therapeutic monitoring-ultimately leading to improved healthcare outcomes.

Abstract:PURPOSE: A new noninvasive laser Doppler flowmetry (LDF) probe (one emitting fiber surrounded by a ring of eight collecting fibers, 1-mm interaxis distance) was tested for its sensitivity to assess the retinal/choroidal blood flow variations in response to hypercapnia, hyperoxia, diverse vasoactive agents and following retinal arteries photocoagulation in the rat.MATERIALS AND METHODS: After pupil dilation, a LDF probe was placed in contact to the cornea of anesthetized rats in the optic axis. Hypercapnia and hyperoxia were induced by inhalation of CO(2) (8% in medical air) and O(2) (100%) while pharmacological agents were injected intravitreously. The relative contribution of the choroidal circulation to the LDF signal was estimated after retinal artery occlusion by photocoagulation.RESULTS: Blood flow was significantly increased by hypercapnia (18%), adenosine (14%) and sodium nitroprusside (16%) as compared to baseline values while it was decreased by hyperoxia (-8%) and endothelin-1 (-11%). Photocoagulation of retinal arteries significantly decreased blood flow level (-45%).CONCLUSIONS: Although choroidal circulation most likely contributes to the LDF signal in this setting, the results demonstrate that LDF represents a suitable in vivo noninvasive technique to monitor online relative reactivity of retinal perfusion to metabolic or pharmacological challenge. This technique could be used for repeatedly assessing blood flow reactivity in rodent models of ocular diseases.

Abstract:Laser speckle contrast imaging has been widely used for quantifying blood perfusion. In this letter, we propose a rapid, wide-field laser speckle angiography technique for noninvasive imaging of retinal and choroidal vessels. The field of view (FOV) reaches similar to 50(circle) while the acquisition time was less than 1 s. Larger FOV (similar to 90(circle)) can be further improved by mosaicking. The results were comparatively studied with optical coherence tomography angiography and fundus fluorescence angiography. The low-cost technology can obtain wide-field fundus vascular and perfusion images less than 1 s. This technology is more suitable for optic disc and choroidal vasculature imaging. In addition, blood flow dynamic information such as pulsation can be provided by this technique.

Abstract:We describe the advantages of diffuse illumination in laser holography for ophthalmology. The presence of a diffusing element introduces an angular diversity of the optical radiation and reduces its spatial coherence, which spreads out the energy distribution of the illumination beam in the focal plane of the eyepiece. The field of view of digitally computed retinal images can easily be increased as the eyepiece can be moved closer to the cornea to obtain a Maxwellian view of the retina without compromising ocular safety. Compliance with American and European safety standards for ophthalmic devices is more easily obtained by preventing the presence of a laser hot spot observed in front of the cornea in the absence of a scattering element. Diffuse laser illumination does not introduce any adverse effects on digitally computed laser Doppler images.
High-resolution three-dimensional blood flow tomography in the subdiffuse regime using laser speckle contrast imaging

Abstract:Significance: Visualizing high-resolution hemodynamics in cerebral tissue over a large field of view (FOV), provides important information in studying disease states affecting the brain. Current state-of-the-art optical blood flow imaging techniques either lack spatial resolution or are too slow to provide high temporal resolution reconstruction of flow map over a large FOV. Aim: We present a high spatial resolution computational optical imaging technique based on principles of laser speckle contrast imaging (LSCI) for reconstructing the blood flow maps in complex tissue over a large FOV provided that the three-dimensional (3D) vascular structure is known or assumed. Approach: Our proposed method uses a perturbation Monte Carlo simulation of the high-resolution 3D geometry for both accurately deriving the speckle contrast forward model and calculating the Jacobian matrix used in our reconstruction algorithm to achieve high resolution. Given the convex nature of our highly nonlinear problem, we implemented a mini-batch gradient descent with an adaptive learning rate optimization method to iteratively reconstruct the blood flow map. Specifically, we implemented advanced optimization techniques combined with efficient parallelization and vectorization of the forward and derivative calculations to make reconstruction of the blood flow map feasible with reconstruction times on the order of tens of minutes. Results: We tested our reconstruction algorithm through simulation of both a flow phantom model as well as an anatomically correct murine cerebral tissue and vasculature captured via two-photon microscopy. Additionally, we performed a noise study, examining the robustness of our inverse model in presence of 0.1% and 1% additive noise. In all cases, the blood flow reconstruction error was <2 % for most of the vasculature, except for the peripheral vasculature which suffered from insufficient photon sampling. Descending vasculature and deeper structures showed slightly higher sensitivity to noise compared with vasculature with a horizontal orientation at the more superficial layers. Our results show high-resolution reconstruction of the blood flow map in tissue down to 500 μm and beyond. Conclusions: We have demonstrated a high-resolution computational imaging technique for visualizing blood flow map in complex tissue over a large FOV. Once a high-resolution structural image is captured, our reconstruction algorithm only requires a few LSCI images captured through a camera to reconstruct the blood flow map computationally at a high resolution. We note that the combination of high temporal and spatial resolution of our reconstruction algorithm makes the solution well-suited for applications involving fast monitoring of flow dynamics over a large FOV, such as in functional neural imaging.
Ultrafast imaging of laser-induced modifications in planar targets via single-shot in-line holography

Abstract:We present a holographic single-shot coherent diffractive imaging method based on in-line holography that allows the ultrafast characterization of 2D transmission maps of semi-transparent planar targets such as foils in amplitude and phase. Holographic information is obtained from the interference of the transmitted primary beam with the fields scattered from the modified or unknown target regions. A specialized iterative phase retrieval is used to incorporate the holographic nature of the approach and to accelerate and improve convergence. The achievable quality and reproducibility of the reconstructed transmission maps as well as optimal setup parameters are investigated using realistic pre-characterized reference targets. We used non-circular laser-induced hole structures in 30 nm thin gold foils that represent the final state of a laser modification and show that the far field error of the reconstructed diffraction images can be used to estimate and optimize the reconstruction quality in the object plane in order to obtain accurate and reproducible transmission maps. Our results mark the important first step towards the full spatio-temporal analysis of all stages of laser material modification or laser ablation in two-color pump probe experiments, including

Abstract:A portable choroidal laser Doppler flowmeter (LDF) with enhanced sensitivity based on a scattering plate is developed. The portable LDF is weighted 2 kg operated at center wavelength of 780 nm, leading to a better penetration into the eye fundus in contrast to the previous LDF operated at center wavelength of 670 nm. Enhancement of number of detected photons that undergo Doppler scattering and improved measured speed of choroidal blood flow are achieved with the use of a scattering plate positioned in front of the eye. The mechanism of detection and sensitivity enhancement is theoretically analyzed. Evaluation of system performance is done by in vivo measurements on ten volunteers. The results demonstrate that an increased percentage of backscattering light at high Doppler shift frequency is collected due to utilization of the scattering plate. However, this kind of light detection influences spatial resolution of the system and decreases the total signal measured. The proposed method for detection and sensitivity enhancement might be useful in a case where the perception of very slight alternation of blood flow is pursued and the spatial resolution is not as critical as that in a choroidal vascular bed. Abstract:High-speed, phase contrast retinal and blood flow imaging using an adaptive optics partially confocal multi-line ophthalmosocope (AO-pcMLO) is described. It allows for simultaneous confocal and phase contrast imaging with various directional multi-line illumination by using a single 2D camera and a digital micromirror device (DMD). Both vertical and horizontal line illumination directions were tested, for photoreceptor and vascular imaging. The phase contrast imaging provided improved visualization of retinal structures such as cone inner segments, vessel walls and red blood cells with images being acquired at frame rates up to 500 Hz. Blood flow velocities of small vessels (<40 μ m in diameter) were measured using kymographs for capillaries and cross-correlation between subsequent images for arterioles or venules. Cardiac-related pulsatile patterns were observed with normal resting heart-beat rate, and instantaneous blood flow velocities from 0.7 to 20 mm/s were measured.
