Abstract:Nanomaterial mediated hyperthermia, or termed nano-hyperthermia, is emerging as a very promising approach to innovate conventional cancer thermal therapy. The current industry is witnessing a rapid growth in the numbers of filed and issued patents in this area. Various types of nanomaterials, such as gold nanoparticles, magnetic nanoparticles, carbon nanotubes, and multifunctional particles etc., are being invented as heating enhancers, image contrast agents and drug delivery carriers etc. Meanwhile, tremendous efforts are also made to improve the heating and energy administration modalities and devices in nano-hyperthermia by taking full use of the many unique merits offered by the nanotechnologies and new heating strategies. This article is dedicated to present a comprehensive analysis on the latest advancement made towards finding new materials and devices for nano-hyperthermia. Abstract:Magnetic nanoparticles (MNPs) have evolved tremendously during recent years, in part due to the rapid expansion of nanotechnology and to their active magnetic core with a high surface-to-volume ratio, while their surface functionalization opened the door to a plethora of drug, gene and bioactive molecule immobilization. Taming the high reactivity of the magnetic core was achieved by various functionalization techniques, producing MNPs tailored for the diagnosis and treatment of cardiovascular or neurological disease, tumors and cancer. Superparamagnetic iron oxide nanoparticles (SPIONs) are established at the core of drug-delivery systems and could act as efficient agents for MFH (magnetic fluid hyperthermia). Depending on the functionalization molecule and intrinsic morphological features, MNPs now cover a broad scope which the current review aims to overview. Considering the exponential expansion of the field, the current review will be limited to roughly the past three years.

Abstract:Magnetic nanoparticle-based hyperthermia has emerged as a promising therapeutic modality for treating malignant solid tumors that exhibit resistance to conventional cancer treatments, including chemotherapy and radiation. Despite the clinical approval of superparamagnetic iron oxide nanoparticles (SPIONs) for the adjunct treatment of recurrent glioblastoma, their therapeutic potential is undercut by chemical synthesis-inherent limitations such as low saturation magnetization, superparamagnetic characteristics, and a wide nanoparticle size distribution. Here, we introduce an micromagnetic modelling-based SAF-MDP design with in-plane magnetization, optimized through specific uniaxial anisotropy adjustments to avert the spin-flop phenomenon and eliminate hysteresis-free hard-axis magnetization loops, paired with a mechanofluidic modeling approach to assess the alignment of the SAF-MDP to the applied alternating magnetic field (AMF). Magnetic Force Microscopy characterization provides unprecedented insights into the particle switching behaviour on a single particle scale. This comprehensive strategy spanning micromagnetics and advanced magnetic characterization enables the design of particles with heating efficiencies to approach the theoretical maximum, dictated by the saturation magnetization of the utilized materials and limited solely by the biologically acceptable frequencies and amplitudes of the oscillating magnetic field. Our work not only addresses the limitations encountered by previous methodologies but also sets the stage for the development of advanced SAF-MDP designs and alignment techniques. This opens a new avenue to hyperthermia-based cancer therapy, delineated only by the boundaries of physical laws and biological safety standards.

Abstract:Magnetic nanoparticles (MNPs) are of great scientific interest for the researchers from a wide range of biomedicine applications, including clinical magnetic resonance imaging (MRI), cancer magnetic hyperthermia, triggered drug release, and catalysis. Especially, the high performance of these MNPs, such as controllable size and morphology, superior magnetism, high magnetically induced heating effects, favorable biocompatibility, accurate targeting ability, and long circulation, is crucial for their effective application in clinical diagnosis and therapy of disease. In a brief overview, the controlled chemical design and synthesis, the formation mechanism, as well as the surface functional strategy of MNPs are firstly presented. High-quality MNPs are typically prepared through a thermal decomposition of organometallic compounds in high-boiling organic solvent containing surfactants, in which the shape, size, composition, surface states, magnetic characteristics, biocompatibility, and some other important features are synthetically controlled. Understanding the interplay of these parameters is critical for the effective imaging and therapeutics, which include MRI relaxivity, heat emission, and attractive forces. We then discuss approaches to constructing versatile MNP probes as MRI contrast agents with high sensitivity, with which the sensitive and target-specific observation of biological imaging at the molecular and cellular levels is possible. Finally, we present an overview of recent breakthroughs in the development of MNP-based magnetic hyperthermia. The purpose of this section is to address different factors that affect the targeting, heating, and biodistribution to safely control the therapeutic efficacy of targeted magnetic hyperthermia (TMH).

Abstract:Nanoparticles have emerged as versatile tools in the realm of thermal therapy, offering precise control and feedback mechanisms for targeted treatments. This review explores the intersection of nanotechnology and thermal therapy, focusing on the utilization of nanoparticles for image-guided interventions and temperature monitoring. Beginning with an examination of local temperature dynamics versus whole-body responses, we delve into the landscape of nanomaterials and their pivotal role in nanomedicine. Various physical stimuli employed in therapy and imaging are scrutinized, laying the foundation for nanothermal therapies and the accompanying challenges. A comprehensive analysis of nanomaterial architecture ensues, delineating the functionalities of magnetic, plasmonic, and luminescent nanomaterials within the context of thermal therapies. Nano-design intricacies, including core-shell structures and monodisperse properties, are dissected for their impact on therapeutic efficacy. Furthermore, considerations in designing in vivo nanomaterials, such as hydrodynamic radii and core sizes at sub-tissue levels, are elucidated. The review then delves into specific modalities of thermally induced therapy, including magnetically induced hyperthermia and luminescent-based thermal treatments. Magnetic hyperthermia treatment is explored alongside its imaging and relaxometric properties, emphasizing the implications of imaging formulations on biotransformation and biodistribution. This review also provides an overview of the magnetic hyperthermia treatment using magnetic nanoparticles to induce localized heat in tissues. Similarly, optical and thermal imaging techniques utilizing luminescent nanomaterials are discussed, highlighting their potential for light-induced thermal therapy and cellular-level temperature monitoring. Finally, the application landscape of diagnosis and photothermal therapy (PTT) is surveyed, encompassing diverse areas such as cancer treatment, drug delivery, antibacterial therapy, and immunotherapy. The utility of nanothermometers in elucidating thermal relaxation dynamics as a diagnostic tool is underscored, alongside discussions on PTT hyperthermia protocols. Moreover, the advancements in nanoparticle magnetic imaging and implications of imaging formulations especially in creating positive MRI contrast agents are also presented. This comprehensive review offers insights into the evolving landscape of nanoparticle-based image-guided thermal therapies, promising advancements in precision medicine and targeted interventions, underscoring the importance of continued research in optimization for the full potential of magnetic hyperthermia to improve its efficacy and clinical translation.

Abstract:The number of cancer patients is rapidly increasing worldwide. Among the leading causes of human death, cancer can be regarded as one of the major threats to humans. Although many new cancer treatment procedures such as chemotherapy, radiotherapy, and surgical methods are nowadays being developed and used for testing purposes, results show limited efficiency and high toxicity, even if they have the potential to damage cancer cells in the process. In contrast, magnetic hyperthermia is a field that originated from the use of magnetic nanomaterials, which, due to their magnetic properties and other characteristics, are used in many clinical trials as one of the solutions for cancer treatment. Magnetic nanomaterials can increase the temperature of nanoparticles located in tumor tissue by applying an alternating magnetic field. A very simple, inexpensive, and environmentally friendly method is the fabrication of various types of functional nanostructures by adding magnetic additives to the spinning solution in the electrospinning process, which can overcome the limitations of this challenging treatment process. Here, we review recently developed electrospun magnetic nanofiber mats and magnetic nanomaterials that support magnetic hyperthermia therapy, targeted drug delivery, diagnostic and therapeutic tools, and techniques for cancer treatment.

Abstract:Localized heat generation using nanoparticles is a promising supplementary technique to the well-established cancer treatments, such as chemotherapy and radiotherapy. Here, we demonstrate that iron carbide (Fe5C2) nanoparticles with a thin carbon shell have the collective magnetothermal and photothermal effects based on the ferromagnetic and photonic properties. When the Fe5C2 nanoparticle suspension is irradiated with a NIR laser (808 nm), it yields unprecedented heating effects. Further, owing to the observed high magnetization and coercivity, the Fe5C2 nanoparticle suspension on exposure to an alternating magnetic field (ACMF) exhibits an enhanced specific absorption rate (SAR) as compared to Fe5C2 nanoparticles of the same size. This significant improvement in the SAR arises from the cooperative contribution from the hysteresis and susceptibility losses. This work also gives quantitative information about the ACMF effects on heating ability as well as provides some guidelines for obtaining enhanced heating activity in nanoparticle suspensions of a given magnetic material. From Micromagnetic to In Silico Modeling of Magnetic Nanodisks for Hyperthermia Applications

Abstract:Magnetic nanodisks have been recently proposed as biomedical tools for therapeutics at the nanoscale level, with a special focus on hyperthermia for cancer cure. Here we present a detailed study of permalloy nanodisks to be used in alternative to superparamagnetic iron oxide nanoparticles, as efficient heating agents that release heat via magnetic hysteresis. A micromagnetic modeling analysis is carried out to identify sizes and ac field parameters that maximize the specific loss power (SLP), guaranteeing the fulfillment of biophysical constraints (Hergt–Dutz limit) and vortex state at remanence (reduced agglomeration effects). The highest SLP (790 W g−1) is found for 100 nm diameter and 20 nm thickness nanodisks, excited at a frequency of 75 kHz. Further analysis elucidates the influence of magnetostatic interactions and local nanodisk‐field orientation on the SLP of nanodisk clusters, which originate from the deposition in target tissues. At high concentrations, magnetostatic interactions can lead to a reduction of 40–50% in the hysteresis losses. From thermal simulations, we finally demonstrate that in a murine model temperature increments comparable to that obtained in calorimetric measurements under quasi‐adiabatic conditions can be achieved only by using an order of magnitude larger dosage of nanodisks, due to blood perfusion effects. A micromagnetic modeling analysis is performed to explore permalloy nanodisks as heating agents in magnetic hyperthermia and identify sizes and field parameters that maximize energy losses, enabling biophysical constraint fulfillment and agglomeration effect reduction. Magnetostatic interactions in nanodisk clusters are also investigated. Thermal simulations in a murine model are finally carried out to provide indications about effective dosage and side‐effects.

Abstract:The use of magnetic nanoparticles (MNPs) to locally increase the temperature at the nanoscale under the remote application of alternating magnetic fields (magnetic particle hyperthermia, MHT) has become an important subject of nanomedicine multidisciplinary research, focusing among other topics on the optimization of the heating performance of MNPs and their assemblies under the effect of the magnetic field. We report experimental data of heat released by MNPs using a wide range of anisometric shapes and their assemblies in different media. We outline a basic theoretical investigation, which assists the interpretation of the experimental data, including the effect of the size, shape and assembly of MNPs on the MNPs' hysteresis loops and the maximum heat delivered. We report heat release data of anisometric MNPs, including nanodisks, spindles (elongated nanoparticles) and nanocubes, analysing, for a given shape, the size dependence. We study the MNPs either acting as individuals or assembled through a magnetic-field-assisted method. Thus, the physical geometrical arrangement of these anisometric particles, the magnetization switching and the heat release (by means of the determination of the specific adsorption rate, SAR values) under the application of AC fields have been analysed and compared in aqueous suspensions and after immobilization in agar matrix mimicking the tumour environment. The different nano-systems were analysed when dispersed at random or in assembled configurations. We report a systematic fall in the SAR for all anisometric MNPs randomly embedded in a viscous environment. However, certain anisometric shapes will have a less marked, an almost total preservation or even an increase in SAR when embedded in a viscous environment with certain orientation, in contrast to the measurements in water solution. Discrepancies between theoretical and experimental values reflect the complexity of the systems due to the interplay of different factors such as size, shape and nanoparticle assembly due to magnetic interactions. We demonstrate that magnetic assembly holds great potential for producing materials with high functional and structural diversity, as we transform our nanoscale building blocks (anisometric MNPs) into a material displaying enhanced SAR properties.

Abstract:The scientific community has made great efforts in advancing magnetic hyperthermia for the last two decades after going through a sizeable research lapse from its establishment. All the progress made in various topics ranging from nanoparticle synthesis to biocompatibilization and in vivo testing have been seeking to push the forefront towards some new clinical trials. As many, they did not go at the expected pace. Today, fruitful international cooperation and the wisdom gain after a careful analysis of the lessons learned from seminal clinical trials allow us to have a future with better guarantees for a more definitive takeoff of this genuine nanotherapy against cancer. Deliberately giving prominence to a number of critical aspects, this opinion review offers a blend of state-of-the-art hints and glimpses into the future of the therapy, considering the expected evolution of science and technology behind magnetic hyperthermia.

Abstract:Magnetic hyperthermia, an alternative anticancer modality, is influenced by the composition, size, magnetic properties, and degree of aggregation of the corresponding nanoparticle heating agents. Here, we attempt to evaluate the AC magnetic field heating response of Fe-based nanoparticles prepared by solar physical vapor deposition, a facile, high-yield methodology. Nanoparticle systems were grown by evaporating targets of Fe and Fe3O4 with different stoichiometry. It is observed that Fe3O4 nanoparticles residing in the magnetic monodomain region exhibit increased heating efficiency together with high specific loss power values above 0.9 kW/g at 765 kHz and 24 kA/m, compared with that of 0.1 kW/g for zero-valent Fe nanoparticles under the same conditions. The enhanced performance of Fe3O4 nanoparticles under the range of field explored (12–24 kA/m) may be attributed to the activation of a magnetic hysteresis loss mechanism when the applied AC field surpasses the particle anisotropy field at H ≥ 0.5HA. This is also illustrated by the smaller coercivity of Fe3O4 nanoparticles compared with that of their Fe counterparts. Therefore, understanding the interconnection between intrinsic parameters (composition, size and magnetic properties), the dosage (concentration, volume) and the intensity and frequency of the AC field can lead to essential design guidelines for in vitro, in vivo, and clinical applications of magnetic nanoparticles for hyperthermia.

Abstract:In the past decades, cancer has become the most serious threat for people’s health. The traditional treating methods for cancer including chemotherapy and radiation therapy would cause severe side effects, which restrict the treating effectiveness for tumors. Recently, magnetic induction hyperthermia (MIH) was proved to have great potential for cancer therapy owing to their advantages including excellent targeting ability and high safety. With the development of nanotechnology, magnetic nanoparticles such as iron oxide and their composites have been widely applied for MIH, and improved treating effectiveness was obtained. Besides, great progress was also made for the combination of MIH and other treating methods. In this review, we firstly summarized the methods for improving the heating generation efficiency of magnetic NPs, which is the key to the treating effectiveness. Then we summarized recent development for MIH and their combination therapy based on the current reports. Finally, the current challenge and prospects were proposed.Graphical abstract

Abstract:Hyperthermia is a noninvasive method that uses heat for cancer therapy where high temperatures have a damaging effect on tumor cells. However, large amounts of heat need to be delivered, which could have negative effects on healthy tissues. Thus, to minimize the negative side effects on healthy cells, a large amount of heat must be delivered only to the tumor cells. Magnetic hyperthermia (MH) uses magnetic nanoparticles particles (MNPs) that are exposed to alternating magnetic field (AMF) to generate heat in local regions (tissues or cells). This cancer therapy method has several advantages, such as (a) it is noninvasive, thus requiring surgery, and (b) it is local, and thus does not damage health cells. However, there are several issues that need to achieved: (a) the MNPs should be biocompatible, biodegradable, with good colloidal stability (b) the MNPs should be successfully delivered to the tumor cells, (c) the MNPs should be used with small amounts and thus MNPs with large heat generation capabilities are required, (d) the AMF used to heat the MNPs should meet safety conditions with limited frequency and amplitude ranges, (e) the changes of temperature should be traced at the cellular level with accurate and noninvasive techniques, (f) factors affecting heat transport from the MNPs to the cells must be understood, and (g) the effect of temperature on the biological mechanisms of cells should be clearly understood. Thus, in this multidisciplinary field, research is needed to investigate these issues. In this report, we shed some light on the principles of heat generation by MNPs in AMF, the limitations and challenges of MH, and the applications of MH using multifunctional hybrid MNPs.

Abstract:The clinical applications of magnetic hyperthermia therapy (MHT) have been largely hindered by the poor magnetic-to-thermal conversion efficiency of MHT agents. Herein, we develop a facile and efficient strategy for engineering encapsulin-produced magnetic iron oxide nanocomposites (eMIONs) via a green biomineralization procedure. We demonstrate that eMIONs have excellent magnetic saturation and remnant magnetization properties, featuring superior magnetic-to-thermal conversion efficiency with an ultrahigh specific absorption rate of 2390 W/g to overcome the critical issues of MHT. We also show that eMIONs act as a nanozyme and have enhanced catalase-like activity in the presence of an alternative magnetic field, leading to tumor angiogenesis inhibition with a corresponding sharp decrease in the expression of HIF-1α. The inherent excellent magnetic-heat capability, coupled with catalysis-triggered tumor suppression, allows eMIONs to provide an MRI-guided magneto-catalytic combination therapy, which may open up a new avenue for bench-to-bed translational research of MHT.

Iron-based nanoparticles have been used in magnetic resonance imaging (MRI) to produce clearer and more detailed images, and they have therapeutic applications in magnetic fluid hyperthermia (MFH). In recent years, researchers have used clay minerals, such as ceramic materials with iron-based nanoparticles, to construct nanocomposite materials with enhanced saturation, magnetization, and thermal effects. Owing to their unique structure and large specific surface area, iron-based nanoparticles can be homogenized by adding different proportions of ceramic minerals before and after modification to enhance saturation magnetization. In this review, we assess the potential to improve the magnetic properties of iron-based nanoparticles and in the preparation of multifunctional composite materials through their combination with ceramic materials. We demonstrate the potential of ferromagnetic enhancement and multifunctional composite materials for MRI diagnosis, drug delivery, MFH therapy, and cellular imaging applications.

Abstract:The use of non-invasive alternating magnetic field (AMF) on biocompatible, small sized iron oxide nanoparticles can be used for heat generation in magnetic hyperthermia or for contrast enhancement of biological tissue. However, the behavior of magnetic nanoparticle in the presence of magnetic field is restricted by their size, shape, surface defects, and coatings. Hence, it becomes imperative to closely monitor the magnetic properties of the nanoparticles as current novel formulations of nanoparticles being developed for tissue targeting involves conjugating a magnetic nanoparticle with a site specific ligand or a peptide. Thus, in this review article we have reviewed the effect of size, shape, doping, and surface coating on the magnetic properties of the nanoparticle. Finally we have concluded with the clinical status of magnetic nanoparticle in the field of magnetic fluid hyperthermia.

Abstract:Nanomaterials, in addition to their small size, possess unique physicochemical properties that differ from the bulk materials, making them ideal for a host of novel applications. Magnetic nanoparticle (MNP) is one important class of nanomaterials that have been widely studied for their potential applications in nanomedicine. Due to the fact that MNPs can be detected and manipulated by remote magnetic fields, it opens a wide opportunity for them to be used in vivo. Nowadays, MNPs have been used for diverse applications including magnetic biosensing (diagnostics), magnetic imaging, magnetic separation, drug and gene delivery, and hyperthermia therapy, etc. This review aims to provide a comprehensive assessment of the state-of-the-art biological and biomedical applications of MNPs. In addition, the development of high magnetic moment MNPs with proper surface functionalization has progressed exponentially over the past decade. Herein, we also reviewed the recent advances in the synthesis and surface coating strategies of MNPs. This review is not only to provide in-depth insights into the different synthesis, biofunctionalization, biosensing, imaging, and therapy methods but also to give an overview of limitations and possibilities of each technology.

Abstract:Nowadays, magnetic hyperthermia constitutes a complementary approach to cancer treatment. The use of magnetic particles as heating mediators, proposed in the 1950s, provides a novel strategy for improving tumor treatment and, consequently, patient quality of life. This review reports a broad overview about several aspects of magnetic hyperthermia addressing new perspectives and the progress on relevant features such as the ad hoc preparation of magnetic nanoparticles, physical modeling of magnetic heating, methods to determine the heat dissipation power of magnetic colloids including the development of experimental apparatus and the influence of biological matrices on the heating efficiency.
