Abstract:Recent studies have demonstrated nanosized titanium dioxide (nano-TiO2)-induced fertility reduction and ovary injury in animals. To better understand how nano-TiO2 act in mice, female mice were exposed to 2.5, 5, and 10 mg/kg nano-TiO2 by intragastric administration for 90 consecutive days; the ovary injuries, fertility, hormone levels, and inflammation-related or follicular atresia-related cytokine expression were investigated. The results showed that nano-TiO2 was deposited in the ovary, resulting in significant reduction of body weight, relative weight of ovary and fertility, alterations of hematological and serum parameters and sex hormone levels, atretic follicle increases, inflammation, and necrosis. Furthermore, nano-TiO2 exposure resulted in marked increases of insulin-like growth factor-binding protein 2, epidermal growth factor, tumor necrosis factor-α, tissue plasminogen activator, interleukin-1β, interleukin -6, Fas, and FasL expression, and significant decreases of insulin-like growth factor-1, luteinizing hormone receptor, inhibin α, and growth differentiation factor 9 expression in mouse ovary. These findings implied that fertility reduction and ovary injury of mice following exposure to nano-TiO2 may be associated with alteration of inflammation-related or follicular atresia-related cytokine expressions, and humans should take great caution when handling nano-TiO2.

Abstract:Due to the increased application of titanium dioxide nanoparticles (TiO2 NPs) in the food industry and daily life, their potential toxic effects in humans and animals have been investigated. However, very few studies have focused on testicular oxidative stress and/or apoptosis. In order to understand the possible molecular mechanisms of testicular lesions following exposure to TiO2 NPs, male mice were exposed to 2.5, 5, or 10 mg/kg body weight TiO2 NPs for 90 consecutive days. Testicular oxidative stress and apoptosis were then evaluated, and the testicular mRNA expression of several genes and their proteins involved in oxidative stress and/or apoptosis was investigated. TiO2 NPs entered Sertoli cells and caused severe testicular oxidative damage and/or apoptosis, accompanied by excessive production of reactive oxygen species and peroxidation of lipids, proteins and DNA as well as a significant reduction in antioxidant capacity. Furthermore, exposure to TiO2 NPs resulted in the up-regulation of caspase-3, Nrbp2, and cytochrome c expression, and caused down-regulation of SOD, CAT, GPx, GST, GR, Cyp1b1, Car3, Bcl-2, Acaa2, and Axud1 expression in mouse testis. TiO2 NPs entered Sertoli cells via the blood-testis barrier and were deposited in mouse seminiferous cord and/or Sertoli cells, causing oxidative damage and apoptosis.

Abstract-In order to assess the biological safety of Fe3O4 nanoparticles, oxidative damage of which was studied. Mouse hepatic cells were exposed to different dose (50 μg/mL, 100 μg/mL, 200 μg/mL, 400 μg/mL) of Fe3O4 nanoparticles in vitro for 1 hour. Methods of 2, 7-dichlorohydrofluoroscein diacetate (DCFH-DA), 5, 5'-dithiobis (2-nitrobenzoic acid) (DTNB), thiobarbituric acid (TBA) were used to detect reactive oxygen species (ROS) level, glutathione (GSH) and malondialdehyde (MDA) content, respectively. The levels of ROS were all increased in exposure groups, especially in the group of 200 μg/mL (p<;0.05) and 400 μg/mL (p<;0.01). GSH content presented a slight increase compared with the control group but no significant difference in the dose of 50 μg/mL. Then GSH content began to decrease. When the concentration of Fe3O4 nanoparticles increased to 200 μg/mL, there was a significant difference compared with the control group (p<;0.05). However, the MDA concentration in hepatic cells showed a dose-dependent decrease after Fe3O4 nanoparticles exposure (p<;0.05, p<;0.01). These results suggested that Fe3O4 nanoparticles could cause oxidative damage on mouse liver in vitro, and might have toxic effects on human body under high concentration (>;200 μg/mL).

Abstract:Some studies have found that nano-sized titanium dioxide (nano-TiO2) has adverse effects on the male reproductive system. Blood-testis barrier (BTB), as one of the tightest blood-tissue restriction, is crucial to the male reproductive system. However, the potential effects on BTB and signaling pathway changes in testis tissue induced by nano-TiO2 remain poorly understood. Therefore, in this study, 60 Institute of Cancer Research mice were divided randomly into four groups (per group = 15). The mice of four groups were intragastrically administered with 0, 10, 50, and 100 mg/kg BW nano-TiO2 respectively for 30 days to analyze the changes of BTB structure, BTB-related proteins, and MAPK signal pathways. Besides, testosterone level, estradiol level, and sperm parameter (sperm count, sperm motility, and sperm malformation rate) changes were also studied in this research. The results indicated that nano-TiO2 could induce the BTB structural damage and accompanied by the BTB main protein (ZO-1, Claudin-11, and F-actin) elevation of irritability. Nano-TiO2 could also activate the MAPK signaling pathways (p38, JNK, and ERK) of mice testis tissue. The testosterone and estradiol levels in serum reduced. Besides when the mice were administered with nano-TiO2, we also found the sperm motility rate decreased, and sperm malformation increased. The above changes may be associated with BTB damage and the activation of MAPK signaling pathways, thereby causing male reproductive dysfunction.

Abstract:Although numerous studies have described the accumulation of titanium dioxide nanoparticles (TiO(2) NPs) in the liver, kidneys, lung, spleen, and brain, and the corresponding damage, it is unclear whether or not TiO(2) NPs can be translocated to the ovary and cause ovarian injury, thus impairing fertility. In the current study, ovarian injury and gene-expressed characteristics in female mice induced by intragastric administration of TiO(2) NPs (10mg/kg) for 90 consecutive days were investigated. Our findings indicated that TiO(2) NPs can accumulate in the ovary and result in ovarian damage, cause an imbalance of mineral element distribution and sex hormones, decrease fertility or the pregnancy rate and oxidative stress in mice. Microarray analysis showed that in ovaries from mice treated with TiO(2) NPs compared to controls, 223 genes of known function were up-regulated, while 65 ovarian genes were down-regulated. The increased expression of Cyp17a1 following TiO(2) NPs treatment suggested that the increase in estradiol biosynthesis may be a consequence of increased TiO(2) NPs. In addition, the elevated expression of Akr1c18 implied that progesterone metabolism was accelerated, thus causing a decrease in the progesterone concentration. Taken together, the apparent regulation of key ovarian genes supports the hypothesis that TiO(2) NPs directly affects ovarian function.

Abstract:Titanium dioxide nanoparticles (nano-TiO2) are widely used in food, textiles, coatings and personal care products; however, they cause environmental and health concerns. Nano-TiO2 can accumulate in the reproductive organs of mammals in different ways, affect the development of the ovum and sperm, damage reproductive organs and harm the growth and development of offspring. The oxidative stress response in germ cells, irregular cell apoptosis, inflammation, genotoxicity and hormone synthesis disorder are the main mechanisms of nano-TiO2 toxicity. Possible measures to reduce the harmful effects of nano-TiO2 on humans and nontarget organisms have emerged as an underexplored topic requiring further investigation.

Reproductive toxicity of nanoscale graphene oxide in male mice

Abstract:In the past few years, much work has been performed to explore the biomedical applications and toxicity of nano-graphene and its derivatives. However, the reproductive toxicity of those carbon nanomaterials has been rarely studied. In this study, we report on the male reproductive toxicity of nanoscale graphene oxide (GO) using a mouse model. The results showed that the adult male mice injected with high dosages of GO (25 mg/kg mouse) via the tail vein exhibited normal sex hormone secretion and retained normal reproductive activity. All untreated female mice mated with the GO-treated male mice could produce healthy pups. There were no significant differences in pup numbers, sex ratio, weights, pup survival rates or pup growth over time between the GO-treated and control groups. Furthermore, these GO-treated male mice could produce a second, third, fourth and even fifth litter of healthy offspring when they lived with the untreated female mice. The testicular and epididymal histology as well as the activities of several important epididymal enzymes including α-glucosidase, lactate dehydrogenase, glutathione peroxidase and acid phosphatase were not affected by GO treatment. In addition, no damaging effects were seen at high dose rates of GO (total 300 mg/kg male mouse, 60 mg/kg every 24 h for 5 days) via intra-abdominal injection. Thus, GO showed very low or nearly no toxicity for male reproduction. This work will greatly enable future investigations of GO nanosheets for in vivo biomedical applications.

The presenting author of this paper has the option to appeal this decision by contacting TPII@ieee.org. Trichlorfon (TCF) is a widely used broad-spectrum agricultural organophosphorus (OP) pesticide. Few studies have evaluated the effects of TCF on reproductive toxicity after low level exposure, especially long-term exposure. In this study we assessed the direct effects of TCF on estrous cycle in female mice and developmental outcomes in near-term fetuses after consecutive 30 days of maternal exposure to 2, 10 and 50 mg/kg body weight/day TCF via drinking water. Both male and female fetuses in the 50 mg/kg/day TCF treated group had significantly reduced body weight but not in 2 mg/kg/day and 10 mg/kg/day. There was no difference in estrous cycle between the control and TCF-treated groups. No increased incidence of fetal external malformations was observed in the TCF-treated groups. The lack of effects of TCF of 2 mg/kg/day and 10 mg/kg/day on any in vivo reproductive and developmental endpoints above suggested that for TCF, a hazard of reproductive toxicity below 10 mg/kg/d may not be expected. However, a significant decrease in fetal body weight was observed in the 50 mg/kg/day TCF treated group, suggesting that potential adverse health effects of OP pesticides exposure should continue to be carefully studied in the future.

Abstract:BACKGROUND:Numerous studies have demonstrated that titanium dioxide nanoparticles (TiO2 NPs) induced nephrotoxicity in animals. However, the nephrotoxic multiple molecular mechanisms are not clearly understood.METHODS:Mice were exposed to 2.5, 5 and 10 mg/kg TiO2 NPs by intragastric administration for 90 consecutive days, and their growth, element distribution, and oxidative stress in kidney as well as kidney gene expression profile were investigated using whole-genome microarray analysis technique.RESULTS:Our findings suggest that TiO2 NPs resulted in significant reduction of renal glomerulus number, apoptosis, infiltration of inflammatory cells, tissue necrosis or disorganization of renal tubules, coupled with decreased body weight, increased kidney indices, unbalance of element distribution, production of reactive oxygen species and peroxidation of lipid, protein and DNA in mouse kidney tissue. Furthermore, microarray analysis showed significant alterations in the expression of 1, 246 genes in the 10 mg/kg TiO2 NPs-exposed kidney. Of the genes altered, 1006 genes were associated with immune/inflammatory responses, apoptosis, biological processes, oxidative stress, ion transport, metabolic processes, the cell cycle, signal transduction, cell component, transcription, translation and cell differentiation, respectively. Specifically, the vital up-regulation of Bcl6, Cfi and Cfd caused immune/ inflammatory responses, the significant alterations of Axud1, Cyp4a12a, Cyp4a12b, Cyp4a14, and Cyp2d9 expression resulted in severe oxidative stress, and great suppression of Birc5, Crap2, and Tfrc expression led to renal cell apoptosis.CONCLUSIONS:Axud1, Bcl6, Cf1, Cfd, Cyp4a12a, Cyp4a12b, Cyp2d9, Birc5, Crap2, and Tfrc may be potential biomarkers of kidney toxicity caused by TiO2 NPs exposure.

Abstract:Titanium dioxide nanoparticles (TiO2-NPs) are used intensively. Thanks to their extremely small size (1–100 nm), TiO2-NPs are more absorbable by living organisms; consequently, they can cross the circulatory system and then be distributed in various organs including the reproductive organs. We have evaluated the possible toxic effect of TiO2-NPs on embryonic development and the male reproductive system using Danio rerio as an organism model. TiO2-NPs (P25, Degussa) were tested at concentrations of 1 mg/L, 2 mg/L, and 4 mg/L. TiO2-NPs did not interfere with the embryonic development of Danio rerio, however, in the male gonads the TiO2-NPs caused an alteration of the morphological/structural organization. The immunofluorescence investigation showed positivity for biomarkers of oxidative stress and sex hormone binding globulin (SHBG), both confirmed by the results of qRT-PCR. In addition, an increased expression of the gene responsible for the conversion of testosterone to dihydrotestosterone was found. Since Leydig cells are mainly involved in this activity, an increase in gene activity can be explained by the ability of TiO2-NPs to act as endocrine disruptors, and, therefore, with androgenic activity.

Abstract:Objective:To study the effects ofnano silica (Nano-SiO2) on the estrous cycle of female mice.Methods:Six ICR pregnant rats at 8 weeks were selected and randomly divided into two groups:one group was injected with nano silica in E5.5-E11.5 every other day,while the other group was injected with PBS.Three offspring female mice were taken daily by PAP staining examination of the estrous cycle in each group when they grow up to 7 weeks were.Results:Injection ofNano-SiO2 of offspring female mice compared with injection of PBS of offspring female mice,there were no significant differences in total time of the estrous cycle of offspring female mice (P＞0.05); proestrus time was significantly shorter and metestrus time was significantly prolonged (P＜0.05); although the estrous period was shortened,but the difference was not significant (P＞0.05).Conclusion:Prenatal exposure to Nano-SiO2 did not result in offspring female mice estrous cycle disorder,but may have a certain influence on the reproductive function.
