Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.

Abstract:Skin wound healing is an urgent problem in clinics and military activities. Although significant advances have been made in its treatment, there are several challenges associated with traditional methods, for example, limited donor skin tissue for transplantation and inflammation during long-term healing time. To address these challenges, in this study we present a method to fabricate Poly(ethylene-co-vinyl alcohol) (EVOH) nanofibres encapsulated with Ag nanoparticle using electrospinning technique. The fibres were fabricated with controlled diameters (59 nm–3 μm) by regulating three main parameters, that is, EVOH solution concentration, the electric voltage, and the distance between the injection needle tip (high-voltage point) and the fibre collector. Ag was added to the nanofibres to offer long-term anti-inflammation effect by slow release of Ag nanoparticles through gradual degradation of EVOH nanofibre. The method developed here could lead to new dressing materials for treatment of skin wounds.

Abstract:Abstract For effective application of electrospinning and electrospun fibrous meshes in wound dressing, we have in situ electrospun poly(vinyl pyrrolidone)/iodine (PVP/I), PVP/poly(vinyl pyrrolidone)-iodine (PVPI) complex, and poly(vinyl butyral) (PVB)/PVPI solutions into fibrous membranes by a handheld electrospinning apparatus. The morphologies of the electrospun fibers were examined by SEM, and the hydrophobicity, gas permeability, and antibacterial properties of the as-spun meshes were also investigated. The flexibility and feasibility of in situ electrospinning PVP/I, PVP/PVPI, and PVB/PVPI membranes, as well as the excellent gas permeabilities and antibacterial properties of the as-spun meshes, promised their potential applications in wound healing.

Abstract:Antibacterial wound dressing can benefit the wound healing by preventing bacterial infection, especially for the electrospun ones due to their porous structures and easily loading antibacterial drugs. However, it is challenging to apply the antibacterial electrospun wound dressing to covering the wound conveniently and safely. Here, we presented one step fabrication and application of antibacterial electrospun zein/cinnamon oil wound dressing via a handheld electrospinning setup. The prepared zein/cinnamon oil wound dressing showed gas permeability of (76.1–5.45) mm/s, hydrophilicity with zero body fluid contact angle, swelling stability after 24 h as well as antibacterial zones over 5 cm against both E. coli and S. aureus bacteria. Moreover, in situ electrospinning process can deposit the electrospun zein/cinnamon oil fibers directly onto the wound, meantime forming a wound dressing. The mice cut-wound model experiment demonstrated that the one step in situ fabrication and application of zein/cinnamon oil wound dressing could nearly heal the wound within 11 d.

Abstract:Abstract Background Electrospun (e-spun) nanofibers for wound dressing have attracted wide attention due to its large specific surface area, large porosity and breathability. Compared with solution electrospinning (e-spinning), melt e-spinning is more bio-friendly without toxic solvent participation, which provides the possibility of in situ e-spinning on wounds directly. However, previously reported melt e-spinning devices were usually bulky and cumbersome due to their necessary heating unit, and different components were separated to avoid electrostatic interference. Results In this article, we report on a self-powered hand-held melt e-spinning gun which can work without any external power supply (outdoors). The problem of electrostatic interference for this integrated device was solved by using a special high heat transfer insulation unit. The apparatus is easy and safe to operate by a single hand due to its small volume (24 × 6 × 13 cm 3 ) and light weight (about 450 g). Some biodegradable polymers, for example, polycaprolactone (PCL) fibers were successful e-spun onto wounds directly by using this dressing gun. Conclusions PCL fibrous membrane has good biocompatibility and can be in situ electrospun to wound surface as a wound dressing by the portable melt e-spinning gun. Besides wound dressing, this hand-held melt e-spinning gun may be used in 3D printing and experimental teaching demonstration aids.

Abstract:Mesenchymal stem cell (MSC) therapies have been brought forward as a promising treatment modality for cutaneous wound healing. However, current approaches for stem cell delivery have many drawbacks, such as lack of targetability and cell loss, leading to poor efficacy of stem cell therapy. To overcome these problems, in the present study, we developed an in situ cell electrospinning system as an attractive approach for stem cell delivery. MSCs have a high cell viability over 90% even with a high applied voltage of 15 kV post‐cell electrospinning process. In addition, cell electrospinning does not show any negative effect on surface marker expression and differentiation capacity of MSCs. In vivo studies demonstrated that in situ cell electrospinning treatment could promote cutaneous wound healing through direct deposition of bioactive fish gelatin fibers and MSCs onto wound sites, leading to a synergic therapeutic effect. Our approach enhanced extracellular matrix remodeling by increasing collagen deposition, promoted angiogenesis by increasing the expression of VEGF and forming small blood vessels, and dramatically reduced the expression of IL‐6 during the wound healing. The use of in situ cell electrospinning system potentially provides a rapid, no touch, personalized treatment for cutaneous wound healing. This article is protected by copyright. All rights reserved

Abstract:Electrospun fibrous membranes have the potential to be effective wound dressings for promoting wound healing. However, the fabrication and application of the common electrospun fibrous wound dressings are usually complicated and separated. Here, electrospun zein/clove essential oil (CEO) fibrous membranes are fabricated and applied as a potential wound dressing through in situ electrospinning process by a portable electrospinning device. The in situ electrospinning process can directly electrospin zein/CEO membranes onto a wound site to cover the wound well and improve the convenience and comfort in use. The as‐spun zein/CEO membranes show a porous structure and exhibit higher gas permeability at 168.2 ± 43.3 mm s−1, with superhydrophilicity to absorb the wound exudate and good biocompatibility as well as antibacterial effects to protect from infection. Moreover, the mice wound model study suggests that in situ electrospun zein/CEO promotes the wound healing process.
In situ cell electrospun using a portable handheld electrospinning apparatus for the repair of wound healing in rats

Abstract:Slow or non-healing wounds caused by full-thickness skin wounds of various origins have become a difficult challenge in clinical wound treatment. In particular, large full-thickness skin wounds often lead to serious chronic skin wounds that do not heal. Electrospinning technology and stem cell treatment for wound repair have attracted much attention due to its unique advantages. In the current study， we electrospun polyvinyl alcohol (PVA) and bone marrow-derived stem cells (BMSCs) by a handheld electrospinning device, the distribution and interaction of cells and fibres were determined by light and electron microscopy and the cell viability and proliferation were determined by live/dead cell staining. The tissues were analysed by histology with Haematoxylin and Eosin (H&E) and Masson staining and immunohistochemical staining. We found that the fibres were distributed uniformly and BMSCs were distributed between the fibres. Cytotoxicity and cell proliferation tests proved its good biocompatibility. Histological staining shows it can accelerate wound healing and appendages regeneration by promoting granulation tissue repair. The instant PVA/stem cell fibres prepared by a handheld electrospinning device strongly promote the repair of full-thickness skin wounds in rats. The proposed electrospinning technology is expected to have great potential in household, outdoor and battlefield first aid.
In situ MgO nanoparticle-doped Janus electrospun dressing against bacterial invasion and immune imbalance for irregular wound healing

Abstract:Owing to the unpredictable size of wounds and irregular edges formed by trauma, nanofibers' highly customizable and adherent in situ deposition can contribute to intervention in the healing process. However, electrospinning is limited by the constraints of conventional polymeric materials despite its potential for anti-inflammatory and antimicrobial properties. Here, inspired by the Janus structure and biochemistry of nanometal ions, we developed an in situ sprayed electrospinning method to overcome bacterial infections and immune imbalances during wound healing. The bilayer fiber scaffold has a hydrophobic outer layer composed of polycaprolactone (PCL) and a hydrophilic inner layer composed of gelatin, poly(L-lactic acid) (PLLA), and magnesium oxide nanoparticles, constituting the PCL/PLLA-gelatin-MgO (PPGM) electrospun scaffold. This electrospun scaffold blocked the colonization and growth of bacteria and remained stable on the wound for continuous anti-inflammatory properties to promote wound healing. Furthermore, PPGM electrospinning modulated collagen deposition and the inflammatory microenvironment in the full-thickness skin model, significantly accelerating vascularization and epithelialization progression. This personalized Janus electrospun scaffold has excellent potential as a new type of wound dressing for first aid and wound healthcare.

Abstract:The nanofiber materials of three-dimensional spatial structure synthesized by electrospun have the characteristics of high porosity, high specific surface area, and high similarity to the natural extracellular matrix (ECM) of the human body. These are beneficial for absorbing wound exudate, effectively blocking the invasion of external bacteria, and promoting cell respiration and proliferation, which provides an ideal microenvironment for wound healing. Moreover, electrospun nanofiber dressings can flexibly load drugs according to the condition of the wound, further promoting wound healing. Recently, electrospun nanofiber materials have shown promising application prospects as medical dressings in clinical. Based on current research, this article reviewed the development history of wound dressings and the principles of electrospun technology. Subsequently, based on the types of base material, polymer-based electrospun nanofiber dressing and electrospun nanofiber dressing containing drug-releasing factors were discussed. Furthermore, the application of electrospun nanofiber dressing on skin tissue is highlighted. This review aims to provide a detailed overview of the current research on electrospun nanomaterials for wound healing, addressing challenges and suggesting future research directions to advance the field of electrospun dressings in wound healing.

Novel SA@Ca2+/RCSPs Core–shell Structure Nanofibers by Electrospinning for Wound Dressings

Abstract:Therapeutic drugs remain of great significance for the absorption of wound blood, the closure of wounds and rapid wound healing. Hence, we propose a novel composite nanofiber membrane with the above characteristics as a wound healing material. We utilize the reaction of calcium ion and alginate gel, sodium alginate (SA) and Rana chensinensis skin peptides (RCSPs) extracted from discarded Rana chensinensis skin; these two natural substances were successfully used to prepare composite nanofibers by coaxial electrospinning. The composite nanofibers are named SA@Ca2+/RCSPs nanofibers. SA@Ca2+/RCSPs nanofibers exhibited that the nanofibers contact with the liquid is unmelted, instead become gel, when compared to nanofibers of does not contain calcium ions, and the absorption rate reached 179.87%. SA@Ca2+/RCSPs nanofibers conform to the quasi-first-order dynamics model and the Ritger-Peppas release model. In vivo wound healing experiments showed that the wound-healing rate of SA@Ca2+/RCSPs nanofiber-treated wounds was 46.65% and 97.46% on days 5 and 15, respectively. In addition, SA@Ca2+/RCSPs nanofibers promoted collagen deposition and enhanced epidermal regeneration. The present study showed that composite nanofibers could quickly undergo hemostasis and effectively promote wound healing.

Abstract:Electrospinning has been recognized as an efficient technique for the fabrication of polymer nanofibers. Various polymers have been successfully electrospun into ultrafine fibers in recent years. These electrospun biopolymer nanofibers have potential applications for wound dressing based upon their unique properties. In this paper, a comprehensive review is presented on the researches and developments related to electrospun biopolymer nanofibers including processing, structure and property, characterization, and applications. Information of those polymers together with their processing condition for electrospinning of ultrafine fibers has been summarized in the paper. The application of electrospun natural biopolymer fibers in wound dressings was specifically discussed. Other issues regarding the technology limitations, research challenges, and future trends are also discussed.

Abstract:The incidence of chronic wound healing is promoted by the growing trend of elderly population, obesity, and type II diabetes. Although numerous wound dressings have been studied over the years, it is still challenging for many wound dressings to perfectly adapt to the healing process due to the dynamic and complicated wound microenvironment. Aiming at an optimal reproduction of the physiological environment, multifunctional electrospinning nanofibrous membranes (ENMs) have emerged as a promising platform for the wound treatment owing to their resemblance to extracellular matrix (ECM), adjustable preparation processes, porousness, and good conformability to the wound site. Moreover, profiting from the booming development of human-machine interaction and artificial intelligence, a next generation of intelligent electrospinning nanofibrous membranes (iENMs) based wound dressing substrates that could realize the real-time monitoring of wound proceeding and individual-based wound therapy has evoked a surge of interest. In this regard, general wound-related biomarkers and process are overviewed firstly and representative iENMs stimuli-responsive materials are briefly summarized. Subsequently, the emergent applications of iENMs for the wound healing are highlighted. Finally, the opportunities and challenges for the development of next-generation iENMs as well as translating iENMs into clinical practice are evaluated.

Abstract:At present, the clinical strategies for treating chronic wounds are limited, especially when it comes to pain relief and rapid wound healing. Therefore, there is an urgent need to develop alternative treatment methods. This paper provides a systematic review on recent researches on how electrospun nanofiber scaffolds promote wound healing and how the electrospinning technology has been used for fabricating multi-dimensional, multi-pore and multi-functional nanofiber scaffolds that have greatly promoted the development of wound healing dressings. First, we provide a review on the four stages of wound healing, which is followed by a discussion on the evolvement of the electrospinning technology, what is involved in electrospinning devices, and factors affecting the electrospinning process. Finally, we present the possible mechanisms of electrospun nanofibers to promote wound healing, the classification of electrospun polymers, cell infiltration favoring fiber scaffolds, antibacterial fiber scaffolds, and future multi-functional scaffolds. Although nanofiber scaffolds have made great progress as a type of multi-functional biomaterial, major challenges still remain for commercializing them in a way that fully meets the needs of patients.

Abstract:The modern bioactive wound dressing of electrospinning. In recent years, there has been a significant focus on bioactive dressings suitable for treating chronic and acute wounds. Electrospinning nanofibers are considered advanced dressing options due to their high porosity and permeability to air and water, effective barrier properties against external pathogens, and excellent resemblance to the extracellular matrix for wound healing and skin regeneration. This article reviews the recent advancements in the application of electrospinning nanofibers for bioactive wound healing. The review begins with an overview of the wound healing process and electrospinning methods. It then explores the advantages and disadvantages of different synthetic and natural polymers used in the preparation of electrospinning wound dressings. The natural polymers discussed in this review include collagen, gelatin, silk fibroin, chitosan, hyaluronic acid, and sodium alginate. Additionally, the review delves into commonly used synthetic polymers like polyvinyl alcohol, polyvinyl chloride, polyethylene lactone, polylactide, and polyurethane for wound dressing applications. Furthermore, the review examines the blending of natural and synthetic polymers to create high‐performance wound dressings. It also explores the incorporation of functional additives, such as antimicrobial agents, growth factors, and natural extracts, into electrospinning nanofibers to expedite wound healing and tissue repair. In conclusion, electrospinning is an emerging technology that provides unique opportunities for designing more effective wound dressings and care products.

Abstract:Near-field direct-writing electrospinning technology can be used to produce ordered micro/nanofiber membrane dressings. The application of this technology can simply realize the control of dressing porosity, compound different functional substances, and adjust their distribution, thus improving the defects of common dressings such as insufficient breathability, poor moisture retention performance, and single function. Herein, a novel multifunctional wound dressing was prepared to utilize near-field direct-writing electrospinning technology, in which calf skin collagen type I (CSC-I) and polycaprolactone (PCL) were used as the composite matrix, Hexafluoroisopropanol (HFIP) as the solvent, and erythromycin (ERY) as an anti-infective drug component. The results show that the micro/nanofiber membranes prepared by near-field direct-writing electrospinning technology can all present a complete mesh structure, excellent thermal stability, and good moisturizing properties. Moreover, the composite fiber membrane loaded with ERY not only had obvious antibacterial properties against E. coli and S. thermophilus but also a better slow-release function of drugs (it is rare to have both in traditional wound dressings). Therefore, this experimental design can provide relevant theories and an experimental foundation for preparing a new type of medical dressing with drug loading and has good guiding significance for the application and promotion of near-field direct-writing electrospinning in medical dressings.

Abstract:Electrospun nanofibers have been widely used in various biomedical applications. However, electrospinning methods generally focus on monolithic and coaxial fibers while ignoring Janus fibers. In this study, Janus-structured nanofibers for wound healing were fabricated via uniaxial electrospinning based on the phase separation of polyvinylpyrrolidone and polycaprolactone in solution. To enhance the bioactivity and antibacterial activity of wound dressings based on the Janus nanofibers, Rana chensinensis skin peptides (RCSPs) and silver nanoparticles (Ag-NPs) were successfully loaded into the Janus nanofibers. The Janus nanofibers exhibited a bead-free surface and a characteristic side-by-side interior structure. The thermal properties, wettability, and mechanical properties of the Janus nanofibers were studied. The Janus nanofibers were found to have low cytotoxicity and antibacterial activity and were able to accelerate wound healing, which was typical of re-epithelialization, fibroblast proliferation, and regeneration of capillaries and hair follicles within 14 days. The findings indicate that Janus nanofibers loaded with RCSPs and Ag-NPs are promising wound dressing for skin tissue engineering.

Abstract:Ag/Zn dots are deposited on the thermoplastic polyurethane (TPU) layer of TPU/cotton nonwovens (Cotton). The hydro‐activated Ag/Zn's electrical stimulation (ES) can promote the migration of fibroblast cells and show excellent antibacterial activity against E. coli and S. aureus. The exudate management coupled with ES can accelerate wound healing at multiple stages by promoting re‐epithelization, collagen deposition, and vascularization. Exudate management and bioelectric signals are critical to promoting wound healing. In this study, the thermoplastic polyurethane (TPU) nanofiber is directly electrospun on the cotton microfibers (Cotton) to prepare a Janus dressing (TPU/Cotton), which can unidirectionally drain the excess exudate. Then, Ag/Zn dot electrodes are deposited on the TPU layer, which can produce an electric field penetrating the wound when activated by wound exudate. The hydro‐activated Ag/Zn's electrical stimulation (ES) can promote the migration of fibroblast cells and shows excellent antibacterial activity against E. coli and S. aureus. Furthermore, in vivo experiments show that the exudate management coupled with ES accelerated wound healing at multiple stages by promoting re‐epithelization, collagen deposition, and vascularization. Moreover, the Ag/Zn@TPU/Cotton dressing shows excellent biocompatibility in the vitro cytotoxicity experiments. These findings may shed some light on developing the next generation of wound dressings with self‐activated electrical stimulation and exudate management.

Abstract:The novel multifunctional electrospun textiles were fabricated by incorporating sheet-like kaolinite and silver nanoparticles (AgNps) into a polyurethane (PU) textile by using electrostatic spinning to promote wound-healing process. Threedimensional network of PU electrospun textiles offered an appropriate framework for loading kaolinite nanosheets and AgNps. Moreover, the kaolinite nanosheets healed bleeding wounds by accelerating plasma absorption, increasing blood cell concentrations, and stimulating coagulation factors. Furthermore, the AgNps killed microbes by destroying the cell membrane, while the deleterious effects were controlled by incorporation into the electrospun textile. The therapeutic effects of multifunctional electrospun textile in treating full-thickness abdominal wall defect were explored. The wound healing process could be accelerated via the textile by restoring the abdominal physiological environment, reducing the inflammatory response, and promoting collagen deposition, angiogenesis, and epithelization.

Abstract:The Janus structure with asymmetric wettability has unique unidirectional water transport properties, which can be utilized to manage moisture in the wound and subsequently help the wound heal. Electrospinning technology allows flexible control of the thickness and pore size of nanofibers, and through the selection of different wettability materials, the desired Janus dressings can be prepared in a highly controllable manner. The preparation of Janus dressings using electrostatic spinning technology is receiving increasing attention. However, a comprehensive review of functional Janus dressings is lacking. This paper introduces the preparation method of Janus dressing based on electrospinning and discusses the liquid transport mechanism and influencing factors. The combination of Janus structure dressings with the function of antibacterial, anti-inflammatory/anti-oxidative, angiogenesis, stimulus-responsive, wound monitoring, electrical stimulation, and hemostatic are summarized and discussed. Finally, this review identifies the remaining challenges and recommendations for future research.
