Abstract:Mentha haplocalyx is derived from the aerial part of Mentha haplocalyx Brip. Caffeic acid, hesperidin, rosmarinic acid, buddleoside and diosmetin are active constituent of Mentha haplocalyx used in many traditional Chinese medicines. This paper describes a sensitive and specific assay for the determination of five bioactive compounds in Mentha haplocalyx extracts. In this paper, the five components were separated on an Agilent Zorbax Eclipse XDB-C-18 column (250 x 4,6 mm, 5 mu m) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.5 %, v/v) and (B) acetortitrile using a gradient elution. Analytes were performed at 30 degrees C with a flow rate of 1 mL/min and UV deteCtion at 340 nm. All calibration curves showed good linear regression (r(2) >= 0.9999) within tested ranges. The LOD and LOQ were 031-5.93. and 2.98-57 mu g/mL, respectively. Overall intra-day and inter-day variations were less than 2.19 % and the average recoveries were 98.23-100.04 % for the analytes. This newly established method is validated as simple, precise and accurate. It can be used as a valid analytical method for intrinsic quality control of Mentha haplocalyx. In conclusion, the proposed method would be-sensitive enough and reliable for comprehensive quality control for Clinical use and modernization of traditional Chinese medicines.

Abstract:In this study, the HPLC-UV-MS method for the simultaneous determination of eight active ingredients of Shengxuebao Mixture were developed based on the concept of quality by design（QbD）with a stepwise optimization approach. After the analytical target profile（ATP）had been defined, albiflorin, paeoniflorin, 2, 3, 5, 4'-tetra-hydroxy-stilbene-2-O-β-D-glucopyranoside, specnuezhenide, ecliptasaponin D, emodin, calycosin-7-glucoside, and astragaloside Ⅳ were identified as the indicator components. The resolution and the signal-to-noise ratio of indicator components were then selected as critical method attributes (CMA) for the first step optimization. According to the results collected from fractional factorial design, critical method parameters (CMP) were determined with a multiple linear regression method, which included the amount of acid addition in the mobile phase, temperature, gradient, and wavelength. After that, the amount of acid addition and the wavelength were optimized to improve the resolution and the signal-to-noise ratio of the indicator components. The peak symmetry factors of specnuezhenide and emodin were then set as CMA for the second step optimization. The Box-Behnken designed experiments were conducted. The temperature and gradient were optimized after modelling. The design space were calculated and verified. The optimized analytical method was validated, and the results showed a good precision, accuracy and stability, which means that it can be used for the quantification of the indicator components in Shengxuebao Mixture.

Abstract:Previously reported HPLC-evaporative light scattering detection methods for terpene trilactone determination in Ginkgo biloba leaf extract (EGBL) have complicated sample preparation steps and are time-consuming. Thus, in this work, an HPLC-MS method for the determination of terpene trilactones in EGBL was developed with a novel analytical quality by design approach to provide robust and simple measurements. For this purpose, analytical target profiles and systematic risk analyses were performed to identify potential critical method attributes and critical method parameters. After screening experiments, a Box-Behnken design approach was utilized to investigate the relationships between critical method attributes and critical method parameters. A hypercube design space obtained by a Monte Carlo method was used for choosing the analytical control strategy. Then, verification experiments were performed within the design space, and the models were found to be accurate. After that, the optimized method was verified and successfully used for quality control analysis of EGBL from different manufacturers, and the results were almost the same as those determined by HPLC-evaporative light scattering detection. To our knowledge, this is the first study to establish a robust HPLC-MS method for determination of terpene trilactones in EGBL based on a novel analytical quality by design concept, which can improve the quality control of commercial EGBL.
Chemical profiles and quality evaluation of Buddleja officinalis flowers by HPLC-DAD and HPLC-Q-TOF-MS/MS

Abstract:The dried flowers and inflorescences of Buddleja officinalis Maxim are used as traditional medicines in China, and aqueous extracts of the flowers have also been used since ancient times as a yellow rice colorant at local festivals. In this study, HPLC-Q-TOF-MS/MS was used to determine the overall chemical composition of this medicine-food plant. A total of 54 compounds, including 23 flavonoids, 19 phenylethanoid glycosides, 7 alkaloids and 5 other compounds, were detected in the methanol extracts of the herb using this method. Among them, 35 compounds were found firstly in this herb. HPLC fingerprints were also developed, together with a method for the simultaneous quantification of 11 constituents that could be used for quality evaluation of B. officinalis. Fingerprint analysis, using 28 characteristic fingerprint peaks, was used to assess the similarities among 12 samples collected from different geographic areas and showed that the similarity was >0.900. Simultaneous quantification of 11 markers in B. officinalis was then performed to determine consistency of quality. Additionally, the total phenolic content and antioxidant capacity of extracts of the 12 samples of B. officinalis flowers were measured using spectroscopic methods. B. officinalis was found to have good antioxidant capacity and to be a potential natural antioxidant. The highest antioxidant capacity was found in the samples from Guizhou, Sichuan and Guangxi Province. Our results provide valuable information for further understanding and exploiting the herb.
Quantitative Determination of Verbascoside in Buddleja Officinalis by RP-HPLC

Abstract:Object To establish an HPLC method for the quantitative determination of verbascoside in Buddleja officinalis Maxim. for its quality control. Methods To determine the content of verbascoside in B. officinalis by RP HPLC. Chromatographic condition: C 18 column (250 mm×4 6 mm, 5 μm), a mobile phase of methanol acetonitrile water containing 1% acetic acid (8∶16∶76) and wavelength of 254 nm. The condition for the extraction of verbascoside was optimized by the orthogonal test as: continuous extraction by 20 times of hot 70% ethanol solution for 1 h. Results This chromatographic condition was applicable for the quantitative determination of verbascoside in B. officinalis. Verbascoside was separated successfully from other constituents, and the retention time was 14 25 min. The average recovery rate of verbascoside was 100 71% with RSD 1 41% (n=6). The correlation coefficient of standard curve was 0 999 9 (n=7). The verbascoside content in B. officinalis from ten different locations was determined to be in the range of 0 79%—2 30%. Conclusion This analytical method is proved to be sensitive, quick, simple, and accurate for the quantity determination of verbascoside with good repeatability, reproducibility, and stability. The recovery rate and the linear correlation are perfect. This RP HPLC method is reliable and recommendable for the quality control of both material and preparation of B. officinalis.
Simultaneous Determination of 10 Bioactive Components of Lophatherum gracile Brongn by HPLC-DAD

Abstract:A high-performance liquid chromatography method coupled with diode array detection (HPLC-DAD) was developed for simultaneous determination of two coumarins and eight flavonoids in Lophatherum gracile Brongn (Gramineae), namely 5-O-coumaroylquinic acid (i), 4-O-coumaroylquinic acid (ii), luteolin 6-C-β-d-galactopyranosiduronic acid (1→2)-β-d-glucopyranoside (iii), 7-O-β-d-glucopyranosyl-6-C-α-l-arabinopy ranoside (iv), isoorientin (v), swertiajaponin (vi), luteolin 6-C-β-d-galactopyranosiduronic acid (1→2)-α-l-arabinopyranoside (vii), Saponaretin (viii), swertisin (ix) and apigenin 6-C-β-d-galactopyranosiduronic acid (1→2)-α-l-arabinopyranoside (x). The analysis was performed on Cosmosil MS-IIⅡ C18 column (250 × 4.6 mm, 5 µm) with gradient elution of 0.1% aqueous acetic acid and acetonitrile. The detection wavelength was 330 nm. The developed method was able to determine the bioactive compounds with excellent resolution, precision and recovery. The validated method was successfully applied for the analysis of the 10 bioactive compounds in n samples from different cultivated regions. The results indicated that the developed method can be used as a suitable quality control method for L. gracile.
Studies on Fingerprinting of Flos Buddleja by RP-HPLC

Abstract:OBJECTIVE:To establish fingerprinting of Flos Buddleja by using RP-HPLC for the quality control.METHOD:The HPLC condition was as follows: Inertsil ODS-3 C18 analytical column (4.6 mm x 250 mm, 5 microm), gredient eluation with MeCN (0.1% TFA)-H2O (0.1%TFA), flow rate 1.0 mL x min(-1), detection wavelength 254 nm. 10 commercial samples were analyzed to establish a fingerprinting.RESULT:Among the obtained fingerprinting, most of the detected peaks were separated effectively. The accuracy, repeatability and stability of this method were satisfied. The RSDs of relative retention time and area of aimed peaks which existed in all samples wereless than 5%. Theresults were in accordance with the request of fingerprinting.CONCLUSION:The established fingerprinting can be used for the quality control of Flos Buddleja.

Abstract:To clarify and quantify the chemical profiling of Huang-Lian-Jie-Du decoction (HLJDD) rapidly, a feasible and accurate strategy was developed by applying high speed LC combined with hybrid quadrupole-orbitrap mass spectrometer (Q-Exactive) and UHPLC-triple quadruple mass spectrometer (UHPLC-QqQ MS). 69 compounds, including iridoids, alkaloids, flavonoids, triterpenoid, monoterpene and phenolic acids, were identified by their characteristic high resolution mass data. Among them, 18 major compounds were unambiguously detected by comparing with reference standards. In the subsequent quantitative analysis, 17 representative compounds, selected as quality control markers, were simultaneously detected in 10 batches of HLJDD samples by UHPLC-QqQ MS. These samples were collected from four different countries (regions). Icariin, swertiamarin and corynoline were employed as internal standards for flavonoids, iridoids and alkaloids respectively. All the analytes were detected within 12min. Polarity switching mode was used in the optimization of multiple reaction monitoring (MRM) conditions. Satisfactory linearity was achieved with wide linear range and fine determination coefficient (r(2)>0.9990). The relative standard deviations (RSD) of inter- and intra-day precisions were less than 5.0%. This method was also validated by repeatability, stability (8h) and recovery, with respective RSDs less than 4.6%, 5.0% and 6.3%. This research established a high sensitive and efficient method for the integrating quality control, including identification and quantification of Chinese medicinal formulas.
Quantitative determination of 10 phenylpropanoid and lignan compounds in Lancea tibetica by high-performance liquid chromatography with UV detection

Abstract:An HPLC method was developed for simultaneous determination of one phenylpropanoid glycoside, verbascoside (1), and nine lignans, including lantibeside (2), phillyrin (3), lantibeside B (4), lantibeside C (5), tibeticoside A (6), styraxjaponoside C (7), sylvatesmin (8), (+)-piperitol (9), and horsfieldin (10), from the Tibetan medicinal plant Lancea tibetica Hook. F. et Thoms. The analysis was performed within 45 min. The extraction method was optimized with different solvent systems. The HPLC method was validated for linearity, repeatability, accuracy, limits of detection, and limits of quantification. The limits of detection and limits of quantification of 10 analytes were found to be less than 0.1 and 0.5 µg/mL, respectively. The RSD for intra- and inter-day analyses was less than 4.2 %, and the recovery efficiency was 90-105 %. The method was used to analyze different populations of L. tibetica collected in China. HPLC profiles showed that the concentrations of analytes were different in samples collected from different areas of China. Verbascoside was the dominant component in three out of five plant samples; compounds 2, 3, 6, and 8 accounted for over 62 % yields in total lignan contents. The method is useful for identification, quality assurance, and quality control of L. tibetica and its related products.
Development and validation of an ultra-performance liquid chromatography tandem mass spectrometry for target determination of bioactive compounds in Dendrobium spp

Abstract:As a medicine-food homology herb, Dendrobium spp. has versatile applications in modern medicine and food industry. Herein, an ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) based method was established for simultaneous quantification of six active components, including gigantol, erianin, naringenin, quercetin, rutin, and p-coumaric acid in Dendrobium spp., on the basis of optimized sample preparation, mass spectrometry conditions, and chromatography conditions. Sample extraction was carried out using methanol at a temperature of 60 °C, followed by separation on a T3 C18 column utilizing a gradient eluting program. The results demonstrated excellent linearity (r > 0.999) for the six active components within a specified concentration range. The average recovery rates ranged from 84.7 % to 106.9 %, and the precision (RSD) was within 7.4 %. The detection and quantification limits of this method ranged from 0.34 to 4.17 ng mL-1 and 1.12-13.91 ng mL-1, respectively. The established method demonstrates high accuracy and reliability and is applicable in practical sample detection. Different Dendrobium spp. exhibit specific variations in compound composition, with D. fimbriatum Hook. having a higher content of benzyl compounds and D. crystallinum. Rchb. f. having a higher content of flavonoids. This study provides experimental evidence for the quality and safety regulation of Dendrobium spp.

Abstract:A high-performance liquid chromatography/mass spectrometry (HPLC–MS) method for the quality control of Shuang-Huang-Lian oral liquid, an antimicrobial and antipyretic herbal preparation, has been developed. Pure compounds are subjected to tandem mass spectrometry (MSn) analysis to clarify their fragmentation rules. Then, the sample of Shuang-Huang-Lian was analyzed by on-line LC–MSn. A total of 27 compounds, including seven phenylethanoid glycosides, three lignans, seven quinic acids, six saponins and four flavonoids, in the extract of Shuang-Huang-Lian oral liquid have been identified or tentatively characterized. It is expected to develop a comprehensive quality control method of this commonly used herbal preparation.
[Quality control of Tianwang Buxin Pills based on UPLC fingerprint and multi-component quantification]

Abstract:Tianwang Buxin Pills have demonstrated therapeutic effects in clinical practice, whereas there is a serious lack of comprehensive quality control to ensure the safety and effectiveness of clinical medication. In this study, ultra-performance liquid chromatography(UPLC) was employed to establish the fingerprint and the method for simultaneously determining the content of seven components of Tianwang Buxin Pills. Furthermore, chemometrics was employed to identify the key factors for the stable quality, which provided a reference for the comprehensive quality control and evaluation of this preparation. There were 25 common peaks in the UPLC fingerprints of 15 batches of Tianwang Buxin Pills, from which thirteen compounds were identified. A quantitation method was established for seven pharmacological components(α-linolenic acid, salvianolic acid B, glycyrrhetinic acid, schisandrin A, β-asarone, 3,6'-disinapoylsucrose, and ligustilide). The principal component analysis(PCA) and partial least square discriminate analysis(PLS-DA) were performed to determine the key pharmacological components for controlling the quality stability of Tianwang Buxin Pills, which included 3,6'-disinapoylsucrose, α-linolenic acid, and β-asarone. The established fingerprint and multi-component content determination method have strong specificity, stability, and reliability. In addition, 3,6'-disinapoylsucrose, α-linolenic acid, and β-asarone are the key pharmacological components that ensure the quality stability between batches and can be used to comprehensively control the quality of Tianwang Buxin Pills. The findings provide a scientific basis for the quality evaluation and standard establishment of Tianwang Buxin Pills.

Abstract:The Jin-Gu-Lian capsule, a Chinese Miao herbal compound, is widely used to treat rheumatoid arthritis. In this study, a rapid, selective, and sensitive UHPLC-Orbitrap Exploris 240 MS method was developed to analyze the chemical composition of Jin-Gu-Lian capsules. A total of 88 compounds were identified, including 23 flavonoids, 23 organic acids, 14 phenylpropanoids, 12 phenols, eight alkaloids, four terpenes, three quinones, and one ketone. Among these, 21 compounds were clearly detected based on a comparison with reference standards and selected as quality control markers. Thereafter, these compounds were simultaneously determined in the Jin-Gu-Lian capsules. The established method was successfully validated and applied for the simultaneous determination of 21 biologically active compounds in Jin-Gu-Lian capsules of 27 sample batches. Quantitative data of the analytes were analyzed using multivariate statistical analysis to determine the quality of the Jin-Gu-Lian capsules. Four compounds (JGLC6 [salidroside], JGLC8 [chlorogenic acid], JGLC12 [liriodendrin], JGLC19 [quercetin]) were identified as chemical markers for quality control of Jin-Gu-Lian capsules. Altogether, the established method was validated as a novel and efficient tool, that can be used for rapid analysis of Jin-Gu-Lian capsules. Accordingly, this study serves as a reference for scientific research on traditional Chinese and ethnic medicine.

Abstract:Baizhu Shaoyao San (BSS) is a famous traditional Chinese medicinal prescription, which is composed of Rhizome atractylodis macrocephalae, Radix Paeoniae Alba, Pericarpium Citri Reticulatae and Radix Saposhnikovia. In this study, a simple and sensitive high-performance liquid chromatography coupled with diode array detection method was established for the simultaneous determination of 10 marker compounds including atractylenolide I, gallic acid, albiflorin, paeoniflorin, narirutin, hesperidin, nobiletin, cimifugin, prim-O-gluco-sylcimifugin, 4'-O-beta-D-glucosyl-5-O-methylvismminol in BSS and its single herbs. The chromatographic separation was performed using a YMC C18 column with a gradient elution system of acetonitrile and 0.1% formic acid aqueous solution at a flow rate of 1 mL/min. The results demonstrated that the validated method was simple, reliable and successfully applied to evaluate the selected compounds in BSS and its single herbs for quality control. Moreover, the experimental data showed that the contents of two major active components detected in BSS decoction were significantly higher than those in the single herbs, which indicated that the combination of single herbs could result in the difference in the amount of active constituents.
Study on UPLC–MS/MS Method for Four Compositions of Cangzhu

Abstract:Background: The legal quality control of Cangzhu is used the single component as an evaluation indicator, which would not be enough to reflect the authenticity of quality. It is necessary to select several typical chemical components as the basis for quality control of medicinal materials and decoction pieces to reflect the content of effective ingredients. Purpose: Based on UPLC-MS/MS, the study aimed to build the content determination method of atractyloidin, atractylidin A, atractylenolide I, and atractylenolide III in Cangzhu from different sources. Materials and Methods: Agilent SB-C18 (2.1 × 100 μm, 1.8 μm) was used to be the stationary phase. The mobile phase system was formed by water-formic acid (1000:1) and acetonitrile-formic acid (1000:1) in gradient elution. The column temperature set at 40°C, and the flow rate was 0.3 mL min –1 . Results: The contents of atractyloidin, atractylidin A, atractylenolide I, and atractylenolide III had good linearity in a certain range (r 2 ≥ 0. 9990). The RSD of the precision, stability and repetitive test results were less than 3.00%. In addition, the content of the four components were some different among different patches of Cangzhu. Conclusion: The method was simple, rapid and highly sensitive, which was suitable for the determination of atractyloidin, atractylidin A, atractylenolide I, and atractylenolide III in Cangzhu.

Abstract:A high-performance liquid chromatographydiode array UV detectionelectrospray ionization tandem mass spectrometry (HPLCDADESI-MS) method was established for the simultaneous qualitative and quantitative analysis of the major bioactive constituents in Qing-Luo-Fang (QLF), an anti-angiogenesis herbal formula. The separation was performed on a Phenomenex Gemini C18 column using gradient elution. The mobile phase A was ammonium acetate (10mM) in water; and mobile phase B was methanol-acetonitrile (1:3, v/v) containing 10mM ammonium acetate and 0.2% ammonia. Twelve compounds were identified by online ESI-MS, and eight of which were quantitated by HPLCDAD. The eight compounds are major alkaloid compounds present in QLF, including magnoflorine, oxymatrine, oxysophocarpine, sinomenine, sophocarpine, matrine, palmatine, and berberine. The method was validated and the linearities (r20.999) and recoveries (ranged from 95.24 to105.50%) were acceptable. The limits of detection (LOD) and the limits of quantitation (LOQ) of these alkaloids ranged from 0.63 to 6.70ng/mL and from 2.01 to 20.69ng/mL, respectively. The intra- and inter-day precision was less than 3% and the accuracy ranged from 97.4% to 102.7%. These results prove that this established method can be used to identify the structure of active constituents responsible for the pharmacological effects of QLF and to control the quality of QLF.
Development and Validation of a UHPLC-PDA-MS Method for the Quantitative Analysis of Anthraquinones in Bulbine natalensis Extracts and Dietary Supplements

Abstract:A UHPLC-photodiode array-MS method was developed and validated for the quantification of one chromone and six anthraquinone type of compounds from Bulbine natalensis plant samples and dietary supplements. Metabolites 1: - 7: were identified based on their retention times and electrospray ionization-MS spectra compared with a mix of previously isolated compounds. The quantification of 1: - 7: was based on photodiode array detection. The optimized separation was achieved using a CORTECS C18 column with a gradient of water/acetonitrile as the mobile phase. Seven compounds were separated within 15 minutes with detection limits of 50 pg on the column. The analytical method was validated for linearity, repeatability, accuracy, limits of detection, and limits of quantification. The relative standard deviations for intra- and inter-day experiments were less than 5% and the recovery efficiency was 98 - 101%. Nine dietary supplements labeled as containing B. natalensis were examined. Anthraquinone-type compounds were detected in only five out of nine dietary supplements, with the total amount ranging from 11.3 to 90.4 mg per daily dose. The analytical method is simple, economic, rapid, and can be applied for quality assessment of B. natalensis and dietary supplements. Electrospray ionization-MS was used for the identification of these compounds in plant samples and dietary products.

Abstract:Sanjoin-tang (SJIT) is an ancient oriental medicine prescription listed in the Jinguiyaolue that is mainly used for the treatment of primary insomnia. This study was conducted to develop and validate an ultra-performance liquid chromatography with tandem mass spectrometry (UPLC–MS/MS) simultaneous analysis method for the quality control of SJIT using 18 target compounds. The 18 analytes were separated on an Acquity UPLC BEH C18 column maintained at 45 °C using a mobile phase composed of distilled water and acetonitrile. The MS system was used to simultaneously detect all analytes using the multiple reaction monitoring (MRM) method of Xevo TQ-XS coupled with an electrospray ionization source. The concentrations of the 18 analytes investigated in the SJIT samples ranged from below the limit of detection to 9.553 mg/g. In conclusion, the validated UPLC–MS/MS MRM analysis method can be used to obtain basic data to establish chemical-nonclinical linkage efficacy and for the clinical research and quality evaluation of SJIT.

Abstract:To characterize the chemical constituents of Huanbei Zhike Prescription by ultra-high performance liquid chromatography-time of flight mass spectrometry( UPLC-Q-TOF-MS/MS). A Thermo Syncronls C18 column( 2. 1 mm×100 mm,1. 7 μm) was used with methanol( A)-0. 1% formic acid solution( B) as the mobile phase for gradient elution. The injection volume was 2 μL; the column temperature was 40 ℃; the flow rate was 0. 3 m L·min-1; and electrospray ionization( ESI) source was used to collect data in positive and negative ion modes. The ion scanning range was m/z 50-1 200,with capillary voltage of 3 000 V,ion source temperature of100 ℃,atomization gas flow rate of 50 L·h-1,desolvent gas flow rate of 800 L·h-1,desolvent temperature of 400 ℃,cone hole voltage of 40 V,with argon as the collision gas and the collision energy was 20-35 V. The excimer ion peak information was analyzed by Waters UNIFI data processing software. The molecular formula with error within 1×10-5 was compared with the data in database to identify the compounds. The secondary fragment ion information of the target compound was selected,and then compared with the retention time and fragmentation patterns provided by the database and the existing literature to further confirm the compositions and structures of the compounds. A total of 68 main compounds in Huanbei Zhike Prescription were identified,including 38 flavonoids,10 organic acids,6 terpenoids and 10 nitrogen-containing compounds,of which 12 compounds were verified by the control substances. This method is rapid and accurate,which provides a new strategy for the qualitative analysis of the chemical constituents of Huanbei Zhike Prescription,and lays a foundation for the further study and quality control of the compound pharmacodynamic substance.
