Abstract:Abstract Background To fully elucidate the roles and mechanisms of plant hormones in leaf senescence, we adopted an integrated analysis of both non-senescing and senescing leaves from red maple with transcriptome and metabolome data. Results Transcription and metabolite profiles were generated through a combination of deep sequencing, third-generation sequencing data analysis, and ultrahigh-performance liquid chromatograph Q extractive mass spectrometry (UHPLC-QE-MS), respectively. We investigated the accumulation of compounds and the expression of biosynthesis and signaling genes for eight hormones. The results revealed that ethylene and abscisic acid concentrations increased during the leaf senescence process, while the contents of cytokinin, auxin, jasmonic acid, and salicylic acid continued to decrease. Correlation tests between the hormone content and transcriptional changes were analyzed, and in six pathways, genes closely linked with leaf senescence were identified. Conclusions These results will enrich our understanding of the mechanisms of plant hormones that regulate leaf senescence in red maple, while establishing a foundation for the genetic modification of Acer in the future.

Abstract:Plants have evolved the adaptive capacity to mitigate the negative effect of external adversities at chemical, molecular, cellular, and physiological levels. This capacity is conferred by triggering the coordinated action of internal regulatory factors, in which sugars play an essential role in the regulating chloroplast degradation and leaf senescence under various stresses. In this review, we summarize the recent findings on the senescent-associated changes in carbohydrate metabolism and its relation to chlorophyl degradation, oxidative damage, photosynthesis inhibition, programmed cell death (PCD), and sink-source relation as affected by abiotic stresses. The action of sugar signaling in regulating the initiation and progression of leaf senescence under abiotic stresses involves interactions with various plant hormones, reactive oxygen species (ROS) burst, and protein kinases. This discussion aims to elucidate the complex regulatory network and molecular mechanisms that underline sugar-induced leaf senescence in response to various abiotic stresses. The imperative role of sugar signaling in regulating plant stress responses potentially enables the production of crop plants with modified sugar metabolism. This, in turn, may facilitate the engineering of plants with improved stress responses, optimal life span and higher yield achievement.

Abstract:Leaf senescence is an important process of growth and development in plant, and it is a programmed decline controlled by a series of genes. In this study, the biochemical properties and transcriptome at five maturity stages (M1∼M5) of tobacco leaves were analyzed to reveal the dynamic changes in leaf senescence of tobacco. A total of 722, 1,534, 3,723, and 6,933 genes were differentially expressed (DEG) between M1 and M2, M1 and M3, M1 and M4, and M1 and M5, respectively. Significant changes of nitrogen, sugars, and the DEGs related to metabolite accumulation were identified, suggesting the importance of energy metabolism during leaf senescence. Gene Ontology (GO) analysis found that DEGs were enriched in biosynthetic, metabolic, photosynthesis, and redox processes, and especially, the nitrogen metabolic pathways were closely related to the whole leaf senescence process (M1∼M5). All the DEGs were grouped into 12 expression profiles according to their distinct expression patterns based on Short Time-series Expression Miner (STEM) software analysis. Furthermore, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis found that these DEGs were enriched in pathways of carbon metabolism, starch and sucrose metabolism, nitrogen metabolism, and photosynthesis among these expression profiles. A total of 30 core genes were examined by Weight Gene Co-expression Network Analysis (WGCNA), and they appeared to play a crucial role in the regulatory of tobacco senescence. Our results provided valuable information for further functional investigation of leaf senescence in plants.

Abstract:Leaf senescence in plants is a coordinated process that involves remobilization of nutrients from senescing leaves to sink tissues. The molecular events associated with nutrient remobilization are however not well understood. In this study the tobacco system with a source-sink relationship between different leaf positions was used in analyzing the spatiotemporal changes of 76 metabolites from leaves at 3 different stalk positions and 8 developmental stages. The metabolomic data was then compared with RNA-seq data from the same samples to analyze the activities of the metabolic pathways that are important for nutrient remobilization. Integrative analyses on metabolites accumulation and expression changes of enzyme-encoding genes in corresponding metabolic pathways indicated a significant up-regulation of the tricarboxylic acid cycle and related metabolism of sugars, amino acids and fatty acids, suggesting the importance of energy metabolism during leaf senescence. Other changes of the metabolism during tobacco leaf senescence include increased activities of the GS/GOGAT cycle which is responsible for nitrogen recycling, and increased accumulation of nicotine. The results also suggested that a number of compounds seemed to be transported from senescing leaves at lower positions to sink leaves at upper positions. Some of these metabolites could play a role in nutrient remobilization.

Abstract:The complex leaf senescence process is governed by various levels of transcriptional and translational regulation.Several features of the leaf senescence process are similar across species,yet the extent to which the molecular mechanisms underlying the process of leaf senescence are conserved remains unclear.Currently used experimental approaches permit the identification of individual pathways that regulate various physiological and biochemical processes;however,the large-scale regulatory network underpinning intricate processes like leaf senescence cannot be built using these methods.Here,we discovered a series of conserved genes involved in leaf senescence in a common horticultural crop(Solanum lycopersicum),a monocot plant(Oryza sativa),and a eudicot plant(Arabidopsis thaliana)through analyses of the evolutionary relationships and expression patterns among genes.Our analyses revealed that the genetic basis of leaf senescence is largely conserved across species.We also created a multi-omics workflow using data from more than 10000 samples from 85 projects and constructed a leaf senescence-associated co-functional gene network with 2769 conserved,high-confidence functions.Furthermore,we found that the mitochondrial unfolded protein response(UPRmt)is the central biological process underlying leaf senescence.Specifically,UPRmt responds to leaf senescence by maintaining mitostasis through a few cross-species conserved transcription factors(e.g.NAC13)and metabolites(e.g.ornithine).The co-functional network built in our study indicates that UPRmt figures prominently in cross-species conserved mechanisms.Generally,the results of our study provide new insights that will aid future studies of leaf senescence.

Abstract:Pinellia ternata (Thunb.) Breit. (Araceae), a significant medicinal plant, has been used to treat various diseases for centuries. Terahertz radiation (THZ) is located between microwaves and infrared rays on the electromagnetic spectrum. THZ possesses low single-photon energy and a spectral fingerprint, but its effects on plant growth have not yet been investigated. The study's primary objective was to examine the transcriptome and metabolome databases of the SY line to provide a new perspective for identifying genes associated with resistance and growth promotion and comprehending the underlying molecular mechanism. Variations in the biological characteristics of P. ternata grown under control and experimental conditions were analyzed to determine the effect of THZ. Compared with the control group, phenotypic variables such as leaf length, petiole length, number of leaves, leaf petiole diameter, and proliferation coefficient exhibited significant differences. P. ternata response to THZ was analyzed regarding the effects of various coercions on root exudation. The experimental group contained considerably more sugar alcohol than the control group. The transcriptome analysis revealed 1,695 differentially expressed genes (DEGs), including 509 upregulated and 1,186 downregulated genes. In the KEGG-enriched plant hormone signaling pathway, there were 19 differentially expressed genes, 13 of which were downregulated and six of which were upregulated. In the metabolomic analysis, approximately 416 metabolites were uncovered. There were 112 DEMs that were downregulated, whereas 148 were upregulated. The P. ternata leaves displayed significant differences in phytohormone metabolites, specifically in brassinolide (BR) and abscisic acid (ABA). The rise in BR triggers alterations in internal plant hormones, resulting in faster growth and development of P. ternata . Our findings demonstrated a link between THZ and several metabolic pathway processes, which will enhance our understanding of P. ternata mechanisms.

Abstract:Senescence affects the remobilization of nutrients and adaption of the plant to the environment. Combined stresses can result in premature senescence in plants which exist in the field. In this study, transcriptomic analysis was performed on mature leaves and leaves in three stages of premature senescence to understand the molecular mechanism. With progressive premature senescence, a declining chlorophyll (chl) content and an increasing malonaldehyde (MDA) content were observed, while plasmolysis and cell nucleus pyknosis occurred, mitochondria melted, thylakoid lamellae were dilated, starch grains in chloroplast decreased, and osmiophilic granules increased gradually. Moreover, in total 69 common differentially expressed genes (DEGs) in three stages of premature senescing leaves were found, which were significantly enriched in summarized Gene Ontology (GO) terms of membrane-bounded organelle, regulation of cellular component synthesis and metabolic and biosynthetic processes. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis suggested that the plant hormone signal transduction pathway was significantly enriched. The common DEGs and four senescence-related pathways, including plant hormone signal transduction, porphyrin and chlorophyll metabolism, carotenoid biosynthesis, and regulation of autophagy were selected to be discussed further. This work aimed to provide potential genes signaling and modulating premature senescence as well as the possible dynamic network of gene expression patterns for further study.
Contribution of ABA Metabolism and ROS Generation to Sugar Starvation-Induced Senescence of Rice Leaves

Abstract:To clarify the relationship of sugar starvation with ABA-induced ROS generation during leaf senescence, the genotype-dependent differences in sugar concentration, endogenous ABA content, and ROS generation as well as their temporal patterns during leaf senescence were investigated by using two rice genotypes, namely, Zhehui7954 and its corresponding mutant with the premature senescence of flag leaves ( psf ). Meanwhile, the interplay between sugar starvation and ABA metabolism in the induction of leaf senescence was examined using detached leaves. The results showed that the psf mutant differed evidently from its wild type (Zhehui7954) in the temporal pattern of soluble sugar, sucrose and hexose (fructose and glucose) contents during leaf senescence, with the rapidly dropping concentrations of total soluble sugar, and sucrose, and hexose for the senescing leaves of psf mutant. Sugar starvation evidently accelerated leaf senescence and significantly enhanced the ABA concentration and malonaldehyde (MDA) accumulation in detached leaves, while exogenous sugar supply severely suppressed the ABA concentration and ROS level in detached leaves, thereby the delayed leaf senescence for the detached leaves treated by exogenous sugar supply. Correspondingly, ABA biosynthesis inhibitor (NDGA) effectively retarded the sugar starvation-induced leaf senescence, while ABA catabolism inhibitor (DNCZ) obviously accelerated leaf senescence by enhancing the endogenous ABA concentration in senescent leaves. Furthermore, sugar starvation severely repressed the transcripts of several key genes related to ABA biosynthesis and its degradation ( NCED1 , NCED4, NCED5, ABA8ox2 and ABA8ox3 ), with the significantly lower amount of their transcriptional expression in the senescent leaves of psf mutant relative to its wild type during leaf senescence. Hence, the disequilibrium between ABA biosynthesis and catabolism was strongly responsible for sugar starvation-induced leaf senescence, which was derived from the suppression of ABA degradation, rather than the enhancement of ABA biosynthesis.
Investigation of sugar signaling behaviors involved in sucrose-induced senescence initiation and progression in N. tabacum

Abstract:Sugar is involved in initiating leaf senescence. However, its regulatory role, especially as a signal in the senescence process, is unclear. Therefore, this study was designed to illustrate how sugar stimulates the onset of leaf senescence and controls sugar homeostasis through the T6P-SnRK (sucrose non-fermenting (SNF)-related kinase) and HXK (hexokinase) signaling pathways. We used a leaf disc system detached from fully expanded leaves of Nicotiana tabacum cv. K326 and designed a time-course study (days 3, 5, 7, and 9) with exogenously gradient concentrations (0, 30, 60, 90, 120, and 150 mM) of sucrose (Suc) treatment to identify how Suc application affects sugar metabolism and induces senescence. Our results revealed that early decreases of Fv/Fm and increases in electrolyte leakage responded to Suc on day 3. Furthermore, a substantial increase in lipid peroxidation and up-regulated expression of senescence marker genes (NtSAG12) (except 60 mM on day 3) responded sequentially by day 5. The glucose, G6P, and HXK contents were first induced by Suc on day 3 and then repressed from day 5 to day 7. However, exogenous Suc treatment significantly improved the TPS content and the subsequent precursor T6P from day 3 to day 7. Following exogenous Suc treatments, the transcript level of NtSnRK1 was markedly down-regulated from day 3 to day 7. On the other hand, a linear regression analysis demonstrated that the T6P-NtSnRK1 signaling pathway was strongly associated with senescence initiation, and was accompanied by membrane degradation and NtCP1/NtSAG12 up-regulation by day 3. The T6P-NtSnRK1 signaling pathway experienced membrane and chloroplast degradation by day 5. HXK functioned as a metabolic enzyme promoting Glc-G6P and as a Glc sensor, accelerating the initiation of senescence through the HXK-dependent pathway by repressing PSII by day 3 and the senescence process through the Glycolytic pathway by day 7. These physiological, biochemical, and molecular analyses demonstrate that exogenous Suc regulates T6P accumulation, inducing senescence through the NtSnRK signaling pathway. These results illustrate the role of Suc and the transition of the sugar signaling pathway during the progression of senescence initiation.

Abstract:Abstract Senescence is a degenerative process triggered by intricate and coordinated regulatory networks, and the mechanisms of age-dependent senescence and stress-induced premature senescence still remain largely elusive. Thus we selected leaf samples of developmental senescence (DS) and premature senescence (PS) to reveal the regulatory divergence. Senescent leaves were confirmed by yellowing symptom and physiological measurement. A total of 1171 and 309 genes (DEGs) were significantly expressed respectively in the whole process of DS and PS. Up-regulated DEGs in PS were mostly related to ion transport, while the down-regulated DEGs were mainly associated with oxidoreductase activity and sesquiterpenoid and triterpenoid biosynthesis. In DS, photosynthesis, precursor metabolites and energy, protein processing in endoplasmic reticulum, flavonoid biosynthesis were notable. Moreover, we found the vital pathways shared by DS and PS, of which the DEGs were analyzed further via protein–protein interaction (PPI) network analysis to explore the alteration responding to two types of senescence. In addition, plant hormone transduction pathway was mapped by related DEGs, suggesting that ABA and ethylene signaling played pivotal roles in formulating the distinction of DS and PS. Finally, we conducted a model containing oxidative stress and ABA signaling as two hub points, which highlighted the major difference and predicted the possible mechanism under DS and PS. This work gained new insight into molecular divergence of developmental senescence and premature senescence and would provide reference on potential mechanism initiating and motivating senescence for further study.

Abstract:The quality of tea is highly related with the maturity of the fresh tea leaves at harvest. The present study investigated the proteomic and transcriptomic profiles of tea leaves with different maturity, using iTRAQ and RNA-seq technologies. A total of 4455 proteins and 27 930 unigenes were identified, with functional enrichment analyses of GO categorization and KEGG annotation. The compositions of flavonoids (catechins and flavonols) in tea leaves were determined. The total content of flavonoids decreased with leaf maturity, in accordance with the protein regulation patterns of shikimate, phenylpropanoid, and flavonoid pathways. The abundance of ANR had a positive correlation with epi-catechin content, while LAR abundance was positively related with catechin content (P < 0.05). The biosynthetic network of flavonoid biosynthesis was discussed in combination with photosynthesis, primary metabolism, and transcription factors. Bud had the lowest activities of photosynthesis and carbon fixation but the highest flavonoid biosynthesis ability in opposite to mature leaf. SUS-INV switch might be an important joint for carbon flow shifting into the follow-up biochemical syntheses. This work provided a comprehensive overview on the functional protein profile changes of tea leaves at different growing stages and also proposed a research direction regarding the correlations between primary metabolism and flavonoid biosynthesis.
Comprehensive investigation of tobacco leaves during natural early senescence via multi-platform metabolomics analyses

Abstract:Abstract Senescence is the final stage of leaf growth and development. Many different physiological activities occur during this process. A comprehensive metabolomics analysis of tobacco middle leaves at 5 different developmental stages was implemented through multi-platform methods based on liquid chromatography, capillary electrophoresis and gas chromatography coupled with mass spectrometry. In total, 412 metabolites were identified, including pigments, sterols, lipids, amino acids, polyamines, sugars and secondary metabolites. Dramatic metabolic changes were observed. Firstly, membrane degradation and chlorophyll down-regulation occurred after the 50% flower bud stage. Levels of major membrane lipids decreased, including those of the glycolipids in chloroplast thylakoids and phospholipids in membrane envelopes. Clear decreases in free sterols and acylated sterol glucosides were detected along with the accumulation of sterol esters. The accumulation of alkaloids was found. The amino acid levels were significantly decreased, particularly those of N-rich amino acids (glutamine and asparagine), thus reflecting N translocation. Subsequently, the antioxidant system was activated. Sugar alcohols and polyphenols accumulated when the lower leaves turned yellow. These results comprehensively revealed the metabolic changes that occur during tobacco leaf development and senescence under natural conditions.

Abstract:Leaf senescence is the last stage of leaf development, manifested by leaf yellowing due to the loss of chlorophyll, along with the degradation of macromolecules and facilitates nutrient translocation from the sink to the source tissues, which is essential for the plants' fitness. Leaf senescence is controlled by a sophisticated genetic network that has been revealed through the study of the molecular mechanisms of hundreds of senescence-associated genes (SAGs), which are involved in multiple layers of regulation. Leaf senescence is primarily regulated by plant age, but also influenced by a variety of factors, including phytohormones and environmental stimuli. Phytohormones, as important signaling molecules in plant, contribute to the onset and progression of leaf senescence. Recently, peptide hormones have been reported to be involved in the regulation of leaf senescence, enriching the significance of signaling molecules in controlling leaf senescence. This review summarizes recent advances in the regulation of leaf senescence by classical and peptide hormones, aiming to better understand the coordinated network of different pathways during leaf senescence.

Abstract:Pinellia ternata (Thunb.) Breit. is an important traditional Chinese medicinal herb and very sensitive to high temperatures. To gain a better understanding of flavonoid biosynthesis under heat stress in P. ternata , we performed integrated analyses of metabolome and transcriptome data. P. ternata plants were subjected to a temperature of 38 °C, and samples were collected after 10 d of treatment. A total of 502 differential accumulated metabolites and 5040 different expressed transcripts were identified, with flavonoid biosynthesis predominantly enriched. Integrated metabolomics and transcriptome analysis showed that high temperature treatment upregulated the expression of CYP73A and downregulated the expression of other genes (such as HCT, CCoAOMT , DFR1, DFR2 ), which might inhibit the biosynthesis of the downstream metabolome, including such metabolites as chlorogenic acid, pelargonidin, cyanidin, and (-)-epigallocatechin in the flavonoid biosynthesis pathway. The transcription expression levels of these genes were validated by real-time PCR. Our results provide valuable insights into flavonoid composition and accumulation patterns and the candidate genes participating in the flavonoid biosynthesis pathways under heat stress in P. ternata .

Abstract:Sugar metabolism and flavonoid biosynthesis vary with the development of tea leaves. In order to understand the regulatory mechanisms underlying the associations between them, a comprehensive transcriptomic analysis of naturally growing tea leaves at different stages of maturity was carried out. Based on weighted gene coexpression network analysis, the key gene modules (Modules 2 and 3) related to the varying relationship between sugar metabolism and flavonoid biosynthesis as well as the corresponding hub genes were obtained. KEGG (Kyoto Encyclopedia of Genes and Genomes) enrichment analysis showed that the transcription factors (TFs) in Modules 2 and 3 were mainly enriched in the pathway of plant hormone signal transduction. An in vitro study showed that the transcriptional levels of ERF1B-like TF for hexokinase inhibitor and sucrose treatments were upregulated, being respectively 28.1- and 30.2-fold higher than in the control, suggesting that ERF1B-like TFs participate in the sugar-induced regulation of flavonoid biosynthesis. The results of yeast one-hybrid and dual-luciferase assays demonstrated that CsF3'H, encoding flavonoid 3'-hydroxylase, was the target flavonoid biosynthetic gene for CsERF1B-like TF. Our study identified the potential key regulators participating in the metabolism of sugars and flavonoids, providing new insights into the crosstalk between sugar metabolism and flavonoid biosynthesis in tea plants.

Abstract:Non-coding RNAs (ncRNAs) are a special class of functional RNA molecules that are not translated into proteins. ncRNAs have emerged as pivotal regulators of diverse developmental processes in plants. Recent investigations have revealed the association of ncRNAs with the regulation of leaf senescence, a complex and tightly regulated developmental process. However, a comprehensive review of the involvement of ncRNAs in the regulation of leaf senescence is still lacking. This manuscript aims to summarize the molecular mechanisms underlying ncRNAs-mediated leaf senescence and the potential applications of ncRNAs to manipulate the onset and progression of leaf senescence. Various classes of ncRNAs, including microRNAs (miRNAs), small interfering RNAs (siRNAs), long noncoding RNAs (lncRNAs), and circular RNAs (circRNAs), are discussed in terms of their regulatory mechanisms in leaf senescence. Furthermore, we explore the interactions between ncRNA and the key regulators of senescence, including transcription factors as well as core components in phytohormone signaling pathways. We also discuss the possible challenges and approaches related to ncRNA-mediated leaf senescence. This review contributes to a further understanding of the intricate regulatory network involving ncRNAs in leaf senescence.

Abstract:Leaf senescence, the final stage of leaf development, is one of the adaptive mechanisms formed by plants over a long period of evolution. Leaf senescence is accompanied by various changes in cell structure, physiological metabolism, and gene expressions. This process is controlled by a variety of internal and external factors. Meanwhile, the genes and plant hormones involved in leaf aging affect the quality, yield and stress resistance in horticultural plants. Leaf senescence mediated by plant hormones affected plant quality at both pre-harvest and post-harvest stages. Exogenous plant growth regulators or plant hormone inhibitors has been applied to delay leaf senescence. Modification of related gene expression by over-expression or antisense inhibition could delay or accelerate leaf senescence, and thus influence quality. Environmental factors such as light, temperature and water status also trigger or delay leaf senescence. Delaying leaf senescence could increase chloroplast lifespan and photosynthesis and thus improve source strength, leading to enhanced yield. Accelerating leaf senescence promotes nutrient redistribution from old leaves into young leaves, and may raise yield under certain circumstances. Many genes and transcriptional factors involved in leaf senescence are associated with responses to abiotic and biotic stresses. WRKY transcriptional factors play a vital role in this process and they could interact with JA signalling. This review summarized how genes, plant hormones and environmental factors affect the quality, yield. Besides, the regulation of leaf senescence holds great promise to improving the resistance to plant biotic and abiotic stresses.

Abstract:Leaf senescence is an essential physiological process that is accompanied by the remobilization of nutrients from senescent leaves to young leaves or other developing organs. Although leaf senescence is a genetically programmed process, it can be induced by a wide variety of biotic and abiotic factors. Accumulating studies demonstrate that senescence-associated transcription factors (Sen-TFs) play key regulatory roles in controlling the initiation and progression of leaf senescence process. Interestingly, recent functional studies also reveal that a number of Sen-TFs function as positive or negative regulators of plant immunity. Moreover, the plant hormone salicylic acid (SA) and reactive oxygen species (ROS) have been demonstrated to be key signaling molecules in regulating leaf senescence and plant immunity, suggesting that these two processes share similar or common regulatory networks. However, the interactions between leaf senescence and plant immunity did not attract sufficient attention to plant scientists. Here, we review the regulatory roles of SA and ROS in biotic and abiotic stresses, as well as the cross-talks between SA/ROS and other hormones in leaf senescence and plant immunity, summarize the transcriptional controls of Sen-TFs on SA and ROS signal pathways, and analyze the cross-regulation between senescence and immunity through a broad literature survey. In-depth understandings of the cross-regulatory mechanisms between leaf senescence and plant immunity will facilitate the cultivation of high-yield and disease-resistant crops through a molecular breeding strategy.
Participation of ABA Metabolism and ROS Generation in Sugar Starvation-Induced Senescence of Rice Flag Leaves

Abstract:Abstract Background: Both sucrose and abscisic acid (ABA) play pivotal role in the regulation of plant leaf senescence. However, the exact mechanism by which sugar starvation , ABA, and reactive oxygen species (ROS) interact with each other during leaf senescence remains largely unknown. In this study, the genotype-dependent alteration in temporal patterns of sugar concentration during leaf senescence and its relation to ABA metabolism and ROS generation were investigated by using the premature senescence of flag leaf ( psf ) mutant and its wild type. Results: Results showed that sugar starvation-induced leaf senescence was closely associated with the endogenous ABA concentration and ROS level in senescent leaves. Sugar starvation accelerated leaf senescence, concomitantly with the marked increase in ABA concentration and malonaldehyde (MDA) accumulation in detached leaves. Conversely, exogenous sugar treatment significantly suppressed the ABA concentration ad ROS level in detached leaves, thus leaf senescence was delayed by exogenous sugar supply. Pharmacological tests revealed that ABA biosynthesis inhibitor (NDGA) delayed the sugar starvation-induced leaf senescence, while ABA catabolism inhibitor (DNCZ) accelerated leaf senescence and significantly increased the endogenous ABA content in senescent leaves. For the expression patterns of ABA synthesis and catabolism related genes induced by sugar starvation, exogenous sucrose supply, NDGA and DNCZ. sugar starvation up-regulated the OsABA8ox1 transcript, while exogenous sucrose and NDGA down-regulated the transciptional expressions of OsNCED1 , OsNCED4 and OsNCED5 and OsABA8ox2 and OsABA8ox3 e by sugar starvation and DNCZ, while the transcript of was increased. Conclusion: Together, our results demonstrated that the rise in endogenous ABA content during sugar starvation-induced leaf senescence is mostly caused by the suppression of ABA catabolism, rather than the enhancement of ABA biosynthesis, and the expression of ABA metabolic genes determines the equilibrium between ABA biosynthesis and catabolism that eventually influence cross-talk between endogenous factors. The breaking for the equilibrium between ABA biosynthesis and catabolism was strongly responsible for sugar starvation-induced leaf senescence, which was resulted from the suppression of ABA catabolism, rather than the enhancement of ABA biosynthesis .
