Optimization and characterization of tomato seed protein isolate: a sustainable source of functional protein

Abstract:The project aims to optimise the extraction of proteins from waste tomato seeds of Indian cultivar using Box-Behnken design of response surface methodology (RSM) with variables viz. pH (9.5–11.5), meal to solvent ratio (1:10–1:20) and extraction duration (2–4 h). Protein yield of 20.28% was achieved with a pH of 11.5, meal-to-solvent ratio of 1:15 and 2 h 34 min of extraction time, yielding 83.95% protein and a lightness value of 47.51. Tomato seed protein isolate (TSPI) was found to have an isoelectric precipitation point at pH 3.9. The physicochemical, functional and bioactive properties of TSPI were quantified at optimal points. Morphological images indicate that TSPI exhibited a tendency to develop thin, irregular shaped sheets. The particle size of TSPI showed the presence of monomodal distribution. Differential thermal calorimetry indicated the temperature and energy for denaturation of TSPI. FTIR spectroscopy showed the presence of amide I and amide II band in the TSPI. Rheological studies of TSPI gels showed non-Newtonian and a shear-thinning flow behaviour with G' > G" indicating stable gel. The potential uses of agricultural waste as a non-conventional protein source for the isolation of protein isolate are highlighted by this study. TSPI will be a sustainable source for the functional protein.
