Abstract:Selective anion recognition remains a key challenge in supramolecular chemistry: only a very small number of systems that can function in water are known, and these nearly always preferentially bind hydrophobic anions. In this work, we report three robust hexa-cationic cages that can be prepared on scales up to 14 g in two simple and high-yielding steps from commercially-available materials. One of these cages displays unusually strong sulfate binding in water (Ka = 12,000 M–1), and demonstrates high selectivity for this anion over H2PO4–/HPO42– in DMSO/buffer mixtures. These results demonstrate that relatively large, three-dimensional supramolecular hosts can be prepared in high yields and on large scales, and can be highly potent receptors.

Abstract:Solute binding proteins can capture strongly hydrated substrates from water with high affinity. Notable examples are the sulfate binding proteins (SBPs), which use neutral NH and OH groups to stabilize highly charged sulfate anions without salt-bridge formation. Synthetic receptors are yet to achieve comparable capabilities solely by hydrogen bonds or other ion-dipole interactions. Here we report charge-neutral urea cages which have strong affinities for CrO42–, SO42– and Cl– up to micromolar and sub-micromolar levels in water, with tunable anion selectivity based on size match. This is despite the fact that these urea cages are much smaller than proteins and have their anion binding sites exposed to water, demonstrating synthetic receptors’ hydrogen bonding potency that emulates nature’s anion binders.

Abstract:A tricationic cage was synthesized via click chemistry. The cage has a preorganized cavity, which is surrounded by six relatively acidic C-H bonds. In organic solvent, the cage can recognize a variety of anions, among which the Cl- anion has the highest binding affinity due to size selectivity. The cage is also able to selectively recognize the Cl- anion in water, even though the binding affinity is just moderate.

Abstract:Strongly hydrated anions are challenging targets for molecular receptors. In nature, sulfate binding proteins use multiple neutral NH hydrogen bonds to surround the guest, resulting in sub-millimolar to sub-micromolar sulfate affinities. Synthetic receptors, however, have not yet achieved comparable efficiency and selectivity for sulfate solely by relying on hydrogen bonds or other ion-dipole interactions. Here we report charge-neutral macrotricyclic hexa-urea cages which exhibit strong sulfate affinities (6000 to 9000 M–1) in water with exceptional selectivity over hydrophobic anions. Unlike sulfate binding proteins where a low-polarity microenvironment enhances sulfate binding, the urea cages achieve high sulfate affinities with the urea binding sites exposed to water. The work demonstrates that synthetic receptors operating solely by neutral hydrogen bond donors can emulate the efficiency and selectivity of naturally occurring systems.

Abstract:Interlocked coordination cages are a class of multi-cavity architectures with applications in selective anion recognition, adaptive sensing, and catalysis. Controlling the partitioning of their cavities through ligand design and appropriate anion templates is critical to their guest binding scope, yet remains a challenge. Here, we present a thermodynamically stable [Pd 2 L 4 ](BF 4 ) 4 cage assembled from a bis-monodentate ligand featuring a semi-flexible, non-coordinating bis-pyrazole backbone. As a result of its idealized dimensions, NMR, ESI-MS, and X-ray analyses reveal that halides can trigger the interpenetration of this cage into a [X@Pd 4 L 8 ] 7+ dimer (X = Cl ‒ or Br ‒ ) where the halide template resides only in the central pocket. The anion-cation pattern of this interlocked host facilitates exceptional binding affinity for the bisulfate anion in its two outer pockets (up to 10 6 M -1 in MeCN), strongly outcompeting other tetrahedral anions of similar size.

Abstract:Two robust hexacationic cages incorporating either urea or isophthalamide motifs were synthesized via a short and high-yielding synthetic pathway using hydrazone condensation reactions in water for the cage forming step. Stability testing revealed that the cages are stable to a range of stimuli in water and in organic solvents. The urea containing cage can bind anions in pure water, and displays strong and selective binding of SO42– over HPO42–. The isophthalamide containing cage binds SO42– only weakly in 1:1 D2O:d6-DMSO but displays strong and cooperative binding of two HPO42– anions. Combined quantum mechanical/annealed molecular dynamics simulations suggest that the remarkable differences in anion selectivity are largely a result of the differing flexibilities of the two cages.

Abstract:Hydrogen bonding is prevalent in biological systems, dictating a myriad of life-sustaining functions in aqueous environments. Leveraging hydrogen bonding for molecular recognition in water encounters significant challenges in synthetic receptors on account of the hydration of their functional groups. Herein, we introduce a water-soluble hydrogen bonding cage, synthesized via a dynamic approach, exhibiting remarkable affinities and selectivities for strongly hydrated anions, including sulfate and oxalate, in water. We illustrate the use of charge-assisted hydrogen bonding in amide-type synthetic receptors, offering a general molecular design principle that applies to a wide range of amide receptors for molecular recognition in water. This strategy not only revalidates the functions of hydrogen bonding but also facilitates the effective recognition of hydrophilic anions in water. We further demonstrate an unconventional catalytic mechanism through the encapsulation of the anionic oxalate substrate by the cationic cage, which effectively inverts the charges associated with the substrate and overcomes electrostatic repulsions to facilitate its oxidation by the anionic MnO 4 − . Technical applications using this receptor are envisioned across various technical applications, including anion sensing, separation, catalysis, medical interventions, and molecular nanotechnology.

Abstract:A C–H bonding trap for chloride Part of the reason salt dissolves so well in water is that the polarized O–H bonds attract the negatively charged chloride ions. It has therefore been common to include O–H or N–H bonds in molecular receptors designed to capture anions such as chloride. Liu et al. report the surprising finding that sufficiently polarized C–H bonds can work even better (see the Perspective by Bowman-James). Molecular Cages as Encapsulants for the Stabilization of Reactive Iron-Sulfur Cofactor Models

Abstract:Iron-sulfur clusters are desirable targets for bioinspired synthesis due to their versatility in promoting a range of catalytic reactions (N2 reduction to NH3, CO2 reduction to hydrocarbons, and selective C–H functionalizations) and ability to serve as model systems for the systematic interrogation of structure and mechanism. However, a challenge in replicating the activity of clusters remains their instability outside of the protein scaffold. Previous efforts to mitigate this challenge include incorporation into polymeric and aerogel networks and artificial metalloenzymes or installation of bulky, strong-field ligands, but none of these retain both the native weak-field primary ligand sphere and capacity for molecular-level tuning of the cofactor and second coordination sphere. Herein, we present a new strategy for stabilizing weak-field synthetic clusters and conferring second coordination sphere functionality via encapsulation inside molecular cages. Thus, clusters [Fe4S4(SR)4]2– ([1a-c]2–: R = Ph, CH2CH2OH, tBu) are encapsulated inside tetrahedral naphthalene diimide (NDI)-functionalized cages [M4L6]8+ (M = Fe, Ni, Zn) via a robust and generalizable electrostatic binding strategy. 1H and DOSY NMR, ESI-MS, and 57Fe Mössbauer studies confirm immediate and quantitative binding and suggest the cluster electronic structure is minimally perturbed by the cage. At the same time, the cage’s redox-switching ability is demonstrated in CV and 57Fe Mössbauer studies demonstrating that NDI reduction renders the encapsulated cofactors [1a-b]2– significantly more reducing, consistent with their effective charge donation to the cofactor via pi-pi stacking and H-bonding interactions. Importantly, cofactor reactivity is maintained as demonstrated in diphenylacetylene reduction trials, showing that the encapsulated clusters are not only structural but also functional cofactor models. Combined, these results demonstrate that molecular cages can function as protein-like encapsulants to site isolate, tune, and deliver substrate to functional weak-field models of bioinorganic cofactors, and support the use of molecular host-guest complexes as functional enzyme mimics for both catalytic and structure-function studies.

Abstract:While numerous studies have focused on the impact of chirality on some magic amino acid clusters, this article investigates the effects of steric isomerization using 4-hydroxyproline octamers as a model system. Through mass spectrometry, infrared photodissociation spectroscopy, and theoretical calculation, it was demonstrated that the cis-4-hydroxy-L-proline octamer can selectively cage potassium, rubidium, or caesium Ions through stable cis-hydroxyl locks, while the trans-form cannot. The results highlight the importance of hydroxyl group orientation in designing biocompatible membrane transporters with high ion-selectivity.

Abstract:Here, a capsule-shaped cage comprising three monocationic arms was efficiently self-assembled by condensing a triscationic trisaldehyde and a trisamino linkage in water. Multivalence endows the cage with thermodynamic stability in water. Despite its triscationic nature, the cage is able to use its trisimino residue to coordinate a silver cation. As a comparison, other cations lead to cage decomposition or no coordination. The cage and Ag+-coordinated complex were both characterized and confirmed by NMR spectroscopy, mass spectrometry, and theoretical calculations. The metal-ligand complex exhibits a pale-yellow color that can be detectable by the naked eye. The Ag+-coordinated complex undergoes decoordination upon the addition of NaCl, during which the cage containing imine bonds remains intact. Such stability implies that the cage might be potentially employed in silver detection and mining.

Abstract:An ester–imine cage was synthesized from hexapyrrolic tripodal precursor and characterized with NMR spectroscopy and X–ray crystallography. This novel molecular cage displays good stability in wet organic solvents and exhibits moderate to strong affinity (10 3 –10 6 M– 6 ) for anions in chloroform. Theoretical calculations support the relative strong binding between the cage and SO 4 2− .

Abstract:Exploration of metal clusters (MCs) adaptive to both aqueous and oil phases without disturbing their size is promising for a broad scope of applications. The state-of-the-art approach via ligand-binding may perturb MC size due to varied metal-ligand binding strength when shuttling between solvents of different polarity. Herein, we applied physical confinement of a series of small noble MCs (less than 1 nm) inside ionic organic cages (I-Cages), which by means of anion exchange enables reversible transfer of MCs between aqueous and hydrophobic solutions without varying their ultrasmall size. Moreover, the MCs(at)I-Cage hybrid serves as a recyclable, reaction-switchable catalyst featuring high activity in liquid-phase catalysis.

Abstract:The allosteric effect is omnipresent in proteins with two distinct binding sites communicating over a distance to control biological functions and complex processes, which drives an everlasting innovation of molecular-scale devices and machines with allosteric functions. Herein, we report a protein-mimicking metallo-amine cage (MOC-68) with excellent tolerance for harsh acid-base conditions, presenting a proof-of-concept example of proton-driven allosteric regulation of multi-state transitions and molecular motions controlled by a water effector as in the natural system. Owing to its multiple N groups for proton transfer, 8 water-binding allosteric sites for remote control, and one endo-cavity and six exo-cavities for guest exchange, the acid-base or solvent switching protocols of cage conformation, charge-state, amphoteric property, phase transfer, guest dynamics, and, unprecedentedly, ring-on-cage sliding and capping/uncapping motions have been demonstrated; this not only provides a unique allosteric cage model for molecular machines and smart biomaterials but also promises potential in drug delivery and release.

Abstract:A novo triangular covalent organic cage with reduced symmetry was synthesized employing 2,4,6‐triethylbenzene derivatives as caps and 8‐amino‐2‐quinoline carboxyl group as edges. The molecular cage could bind nitrate, acetate, and fluoride anions with moderate affinity. Organic molecular cages with low symmetry are promising to generate complex chemical environments for molecular recognition and catalysis. Herein, a new triangular prism cage with three rigid 8‐aminoquinoline‐2‐carboxylic acid derivatives as edges were synthesized. This molecular cage possesses reduced symmetry and contains both secondary amine and amide as hydrogen donating groups for molecular recognition. It could bind anions, such as NO3−, AcO−, and F− with moderate affinity.

Abstract:Natural transport channels (or carriers), such as aquaporins, are a distinct type of biomacromolecule capable of highly effective transmembrane transport of water or ions. Such behavior is routine for biology but has proved difficult to achieve in synthetic systems. Perhaps most significantly, the enantioselective transmembrane transport of biomolecules is an especially challenging problem both for chemists and for natural systems. Herein, a group of homochiral zirconium metal-organic cages with four triangular opening windows have been proposed as artificial biomolecular channels for enantioselective transmembrane transport of natural amino acids. These structurally well-defined coordination cages are assembled from six synthetically accessible BINOL-derived chiral ligands as spacers and four n-Bu3-Cp3Zr3 clusters as vertices, forming tetrahedral-shaped architectures that feature an intrinsically chiral cavity decorated with an array of specifically positioned binding sites mediated from phenol to phenyl ether to crown ether groups. Fascinatingly, the transformation of single-molecule chirality to global supramolecular chirality within the space-restricted chiral microenvironments accompanies unprecedented chiral amplification, leading to the enantiospecific recognition of amino acids. By virtue of the highly structural stability and excellent biocompatibility, the orientation-independent cages can be molecularly embedded into lipid membranes, biomimetically serving as single-molecular chiral channels for polar-residue amino acids, with the properties that cage-1 featuring hydroxyl groups preferentially transports the l-asparagine, whereas cage-2 attaching crown ether groups spontaneously favor transporting d-arginine. We therefore develop a new type of self-assembled system that can potentially mimic the functions of transmembrane proteins in nature, which is a realistic candidate for further biomedical applications.

Abstract:Chemically and thermally stable permanently porous coordination cages are appealing candidates for separations, catalysis, and as the porous component of new porous liquids. However, many of these applications have not turned to microporous cages as a result of their poor solubility and thermal or hydrolytic stability. Here we describe the design and modular synthesis of iron and cobalt cages where the carboxylate groups of the bridging ligands of well-known calixarene capped coordination cages have been replaced with more basic triazole units. The resultingly higher M-L bond strengths afford highly stable cages amenable to the same modular synthetic approach and potential functionalization or modification. Owing to the robust nature of these cages, they are highly processable and are isolable in various physical states with tunable porosity depending on the solvation methods used. As the structural integrity of the cages is maintained upon high activation temperatures, apparent losses in porosity can be mediated by resolvation and crystallization or precipitation.

Abstract:Discrete molecular cages, as artificial mimics of protein cavities, are central to various applications. Anionocages have emergered in the past two decades as a new type of molecular cages, utilizing anion coordination (hydrogen-bonding in nature) as the driving force of self-assembly rather than the widely utilized matal coordination. In this Chemistry Frontiers paper, we will introduce the historical background of anionocages and summarize their critical features, highlighting the high sensitivity of the assemblies in response to external stimuli. Finally, advances of anionocages are introduced in terms of the coordination nodes and critical features of organophosphates-directed anionocages.

Abstract:A triangular-prism shaped cage was self-assembled by the formation of a dynamic covalent bond, namely a hydrazone, in acidic aqueous solution. The hexacationic host bears a number of relatively acidic protons pointing inside the cage cavity, which is able to accommodate an iodide anion selectively in water over other halide anions such as F- , Cl- , and Br- , which are more hydrated. As a comparison, a macrocycle analogue bearing fewer positive charges and fewer hydrogen bond donors shows no anion binding ability. As inferred from the single-crystal structure and theoretical calculations, the ability of the cage to selectively recognize iodide anions results from the combination of electrostatic forces, C-H⋅⋅⋅I- hydrogen bonds, and the hydrophobic effect.
