Abstract:patU, one of the genes specifically found in filamentous cyanobacteria, is required for the pattern formation in heterocyst-forming species. In Anabaena sp. strain PCC 7120, patU is split into patU5 and patU3, and only patU3 is involved in heterocyst patterning. Here, we report that PatU3 is also involved in control of cell size. A patU3 deletion mutant showed remarkably smaller cell size and much higher heterocyst frequency than the wild type. Yeast two-hybrid and pulldown assays demonstrated a direct interaction between PatU3 and the cell division protein Ftn6. Without the N-terminal 16-amino-acid (aa) portion (MQERFQAVIKRRLQIH [the identified octapeptide is underlined]), PatU3 was no longer able to interact with Ftn6. This portion of PatU3 is also required for the interaction with PatN, a protein related to heterocyst differentiation/patterning. Addition of the 16-aa peptide or AVIKRRLQ-containing peptides restored the cell size and heterocyst frequency of a patU3 deletion mutant to normal or nearly wild-type levels. PatU3(1-16aa)-GFP, the N-terminal 16-aa sequence fused with green fluorescent protein (GFP), formed polar aggregates and peripheral patches in heterocysts of Anabaena sp. strain PCC 7120, whereas PatU3(1-198aa)-GFP showed a homogeneous distribution in the cytoplasm of all cells. The N-terminal AVIKRRLQ-containing sequence may function in intact PatU3, as a separate peptide, or both. IMPORTANCE PatU (or split into PatU5 and PatU3) is distributed in almost all filamentous cyanobacteria, including those that do not form heterocysts (except Pseudanabaena); however, its functions other than heterocyst differentiation/patterning have not been reported before. In this study, we found that PatU3 in Anabaena sp. strain PCC 7120 is involved in cell size determination. The N-terminal 16-aa sequence of PatU3 is required for the control of cell size and interaction with the cell division protein Ftn6, and an octapeptide (aa 7 to aa 14) within the 16-aa sequence can restore the cell size (and heterocyst frequency) of a patU3 deletion mutant to normal. Such a peptide, if generated from PatU or PatU3 in vivo, may promote intercellular coordination in filamentous cyanobacteria.
Specific Downregulation of the Bacterial-Type PEPC Gene by Artificial MicroRNA Improves Salt Tolerance in Arabidopsis

Abstract:Although phosphoenolpyruvate carboxylases (PEPC) are reported to be involved in fatty acid accumulation, nitrogen assimilation, and salt and drought stresses, knowledge on the function of PEPC genes is still limited, particularly on the bacterial-type PEPC gene. To investigate the physiological functionality of Atppc4 , an Arabidopsis bacterial-type PEPC gene, Atppc4 , was specifically suppressed by artificial microRNA (amiRNA) in Arabidopsis . Transgenic plants with constitutively expressed Atppc4 -amiRNA exhibited substantially decreased accumulation of Atppc4 transcripts, whereas other three plant-type PEPC genes, Atppc1 , Atppc2 and Atppc3 , were significantly upregulated in roots. The PEPC activity was improved about 5.1 times in roots of Atppc4 -amiRNA transgenic lines. This result indicates that transcription of bacterial-type and plant-type PEPC genes in plants is interacted with each other in plants. The bacterial-type PEPC genes, Atppc4 , may play an important role in modulating the transcription of plant-type PEPC genes. The effects of Atppc4 on seed lipid content and fatty acid composition were not detected in this research. This indicated that Atppc4 might be independent of plant lipid accumulation. However, the root development was found to be related with Atppc4 . Root elongation of transgenic plants was significantly inhibited. The inhibition can be partially relieved by salt treatment. The results showed that specific downregulation of the bacterial-type PEPC gene, Atppc4 , by artificial microRNA improved salt tolerance in Arabidopsis . The improved salt tolerance may be related with the improved PEPC activity.
patD, a Gene Regulated by NtcA, Is Involved in the Optimization of Heterocyst Frequency in the Cyanobacterium Anabaena sp. Strain PCC 7120

Abstract:In the filamentous multicellular cyanobacterium Anabaena sp. strain PCC 7120, 5 to 10% of the cells differentiate into heterocysts, which are specialized in N 2 fixation. Heterocysts and vegetative cells are mutually dependent for filament growth through nutrient exchange. Thus, the heterocyst frequency should be optimized to maintain the cellular carbon and nitrogen (C/N) balance for filament fitness in the environment. Here, we report the identification of patD , whose expression is directly activated in developing cells by the transcription factor NtcA. The inactivation of patD increases heterocyst frequency and promotes the upregulation of the positive regulator of heterocyst development hetR , whereas its overexpression decreases the heterocyst frequency. The change in heterocyst frequency resulting from the inactivation of patD leads to the reduction in competitiveness of the filaments under combined-nitrogen-depleted conditions. These results indicate that patD regulates heterocyst frequency in Anabaena sp. PCC 7120, ensuring its optimal filament growth. IMPORTANCE Microorganisms have evolved various strategies in order to adapt to the environment and compete with other organisms. Heterocyst differentiation is a prokaryotic model for studying complex cellular regulation. The NtcA-regulated gene patD controls the ratio of heterocysts relative to vegetative cells on the filaments of Anabaena sp. strain PCC 7120. Such a regulation provides a mechanism through which carbon fixation by vegetative cells and nitrogen fixation by heterocysts are properly balanced to ensure optimal growth and keep a competitive edge for long-term survival.

A Gene Cluster That Regulates Both Heterocyst Differentiation and Pattern Formation in Anabaena Sp. Strain PCC 7120

Abstract:Wild-type Anabaena sp. strain PCC 7120, a filamentous nitrogen-fixing cyanobacterium, produces single heterocysts at semi-regular intervals. asr0100 (patU5) and alr0101 (patU3) are homologous to the 5' and 3' portions of patU of Nostoc punctiforme. alr0099 (hetZ) overlaps the 5' end of patU5. hetZ, patU5 and patU3 were all upregulated, or expressed specifically, in proheterocysts and heterocysts. Mutants of hetZ showed delayed or no heterocyst differentiation. In contrast, a patU3 mutation produced a multiple contiguous heterocyst (Mch) phenotype and restored the formation of otherwise lost intercalary heterocysts in a patA background. Decreasing the expression of patU3 greatly increased the frequency of heterocysts in a mini-patS strain. Two promoter regions and two principal, corresponding transcripts were detected in the hetZ-patU5-patU3 region. Transcription of hetZ was upregulated in a hetZ mutant and downregulated in a patU3 mutant. When mutants hetZ::C.K2 and hetZ::Tn5-1087b were nitrogen-deprived, P(hetC)-gfp was very weakly expressed, and in hetZ::Tn5-1087b, P(hetR)-gfp was relatively strongly expressed in cells that had neither a regular pattern nor altered morphology. We conclude that the hetZ-patU5-patU3 cluster plays an important role in co-ordination of heterocyst differentiation and pattern formation. The presence of homologous clusters in filamentous genera without heterocysts is suggestive of a more general role.
A proteolytic pathway coordinates cell division and heterocyst differentiation in the cyanobacterium Anabaena sp. PCC 7120

Abstract:The filamentous, multicellular cyanobacterium Anabaena sp. PCC 7120 ( Anabaena ) is a prokaryotic model for the study of cell differentiation and cell-cell interactions. Upon combined-nitrogen deprivation, Anabaena forms a particular cell type, heterocyst, for aerobic nitrogen fixation. Heterocysts are semiregularly spaced among vegetative cells. Heterocyst differentiation is coupled to cell division, but the underlying mechanism remains unclear. This mechanism could be mediated by the putative protease HetF, which is a divisome component and is necessary for heterocyst differentiation. In this study, by suppressor screening, we identified PatU3, as a negative regulator acting downstream of HetF for cell division and heterocyst development. The inactivation of patU3 restored the capacity of cell division and heterocyst differentiation in the Δ hetF mutant, and overexpression of patU3 inhibited both processes in the wild-type background. We demonstrated that PatU3 was a specific substrate of the protease activity of HetF. Consequently, PatU3 accumulated in the hetF -deficient mutant, which was responsible for the resultant mutant phenotype. The cleavage site of PatU3 by HetF was mapped after the Arg117 residue, whose mutation made PatU3 resistant to HetF processing, and mimicked the effect of hetF deletion. Our results provided evidence that HetF regulated cell division and heterocyst differentiation by controlling the inhibitory effects of PatU3. This proteolytic pathway constituted a mechanism for the coordination between cell division and differentiation in a prokaryotic model used for studies on developmental biology and multicellularity.

Abstract:FtsZ is a tubulin-like GTPase that polymerizes to initiate the process of cell division in bacteria. Heterocysts are terminally differentiated cells of filamentous cyanobacteria that have lost the capacity for cell division and in which the ftsZ gene is downregulated. However, mechanisms of FtsZ regulation during heterocyst differentiation have been scarcely investigated. The patD gene is NtcA-dependent and involved in the optimization of heterocyst frequency in Anabaena sp. PCC 7120. Here we report that the inactivation of patD caused the formation of multiple FtsZ-rings in vegetative cells, cell enlargement, and the retention of peptidoglycan synthesis activity in heterocysts, whereas its ectopic expression resulted in aberrant FtsZ polymerization and cell division. PatD interacted with FtsZ, increased FtsZ precipitation in sedimentation assays, and promoted the formation of thick straight FtsZ bundles that differ from the toroidal aggregates formed by FtsZ alone. These results suggest that in the differentiating heterocysts, PatD interferes with the assembly of FtsZ. We propose that in Anabaena FtsZ is a bifunctional protein involved in both vegetative cell division and regulation of heterocyst differentiation. In the differentiating cells PatD-FtsZ interactions appear to set an FtsZ activity that is insufficient for cell division but optimal to foster differentiation.This article is protected by copyright. Effects of Various Nitrogen Sources and Phytohormones on Growth and Content of Lutein in Chlorella Sp USTB01

Abstract:The effects of three nitrogen sources such as ammonium chloride, urea and potassium nitrate, and two phytohormones such as indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA), on the growth and the content of lutein in Chlorella sp were investigated under light condition, respectively. The results showed that when glucose and potassium nitrate were used as the sole carbon and nitrogen sources, the rapid and continuous growth of Chlorella sp was achieved. If urea was used as the sole nitrogen source, the growth of Chlorella sp was distinctly slow. Using ammonium chloride as the sole nitrogen source, the growth of Chlorella sp could be apparently inhibited by the rapid decrease of pH of culture solution. Compared with urea and ammonium chloride, potassium nitrate was the best nitrogen source to promote biosynthesis of lutein by Chlorella sp, and the yield of lutein in dry algal cells could reach 0.85mg/g. In the presence of glucose and potassium nitrate, the growth of Chlorella sp could not be accelerated, on the contrary, the biosynthesis of lutein was inhibited by IAA or IBA, respectively.
PatU3 Plays a Central Role in Coordinating Cell Division and Differentiation in Pattern Formation of Filamentous Cyanobacterium Nostoc Sp. PCC 7120.

Abstract:Spatial periodic signal for cell differentiation in some multicellular organisms is generated according to Turing's principle for pattern formation. How a dividing cell responds to the signal of differentiation is addressed with the filamentous cyanobacterium Nostoc sp. PCC 7120, which forms the patterned distribution of heterocysts. We show that differentiation of a dividing cell was delayed until its division was completed and only one daughter cell became heterocyst. A mutant of patU3, which encodes an inhibitor of heterocyst formation, showed no such delay and formed heterocyst pairs from the daughter cells of cell division or dumbbell-shaped heterocysts from the cells undergoing cytokinesis. The patA mutant, which forms heterocysts only at the filament ends, restored intercalary heterocysts by a single nucleotide mutation of patU3, and double mutants of patU3/patA and patU3/hetF had the phenotypes of the patU3 mutant. We provide evidence that HetF, which can degrade PatU3, is recruited to cell divisome through its C-terminal domain. A HetF mutant with its N-terminal peptidase domain but lacking the C-terminal domain could not prevent the formation of heterocyst pairs, suggesting that the divisome recruitment of HetF is needed to sequester HetF for the delay of differentiation in dividing cells. Our study demonstrates that PatU3 plays a key role in cell-division coupled control of differentiation.
Study on Photoheterotrophic Growth of Recombinant Anabaena Sp. PCC7120

Abstract:Studies on the effects of organic carbon sources as well as L_arginine and growth hormones 2,4_D and KT on the growth of recombinant Anabaena sp. PCC7120 were carried out. On the basis of research of carbon sources, the optimization of photoheterotrophic conditions were carried out too. The results show that saccharose and glucose can promote the growth of recombinant Anabaena sp. PCC7120. Optimum composition of carbon and nitrogen sources are 6 g/L glucose and 1.5 g/L NaNO 3 under high_pressure disinfection, that L_arginine does not support the photoheterotrophic growth of recombinant Anabaena sp. PCC7120, and that suitable content of 2,4_D and KT can stimulate the latter growth of recombinant Anabaena sp. PCC7120 under the photoheterotrophic conditions.
Differential Effects of Paenibacillus Spp. on Cucumber Mycorrhizas

Abstract:The effects of 17 Paenibacillus strains on root colonization by Glomus intraradices or Glomus mosseae and plant growth parameters (shoot and root weight) of mycorrhizal cucumber plants were examined. The Paenibacillus strains were originally isolated from mycorrhizal ( G. intraradices ) and non-mycorrhizal cucumber rhizosphere and/or hyphosphere, except for strain EJP73, which originated from a Pinus sylvestris-Lactarius rufus ectomycorrhiza. Root colonization of cucumber plants by G. intraradices or G. mosseae was unaffected by all seven strains of Paenibacillus polymyxa , but was decreased or increased by four strains of Paenibacillus macerans and strain EJP73 of Paenibacillus sp. Overall, shoot dry weight of cucumber grown in symbioses with either G intraradices or G. mosseae was unaffected by inoculation with all of the Paenibacillus strains, except for strain MB02-429 of P. macerans , which increased the shoot dry weight in the cucumber- G. mosseae symbiosis. On the other hand, several Paenibacillus strains caused altered root growth. Three strains of P. polymyxa and four strains of P. macerans increased the root fresh weight of the cucumber– G. intraradices symbiosis, whereas three strains of P. polymyxa and one strain of P. macerans as well as Paenibacillus sp. EJP73, decreased the root fresh weight of the cucumber– G. mosseae symbiosis. In conclusion, our results show that bacteria from several species of Paenibacillus differentially affect cucumber mycorrhizas.
Effects of the Helicobacter pylori Virulence Factor CagA and Ammonium Ion on Mucins in AGS Cells

Abstract:Purpose: To investigate the effects of Helicobacter pylori (H. pylori)-CagA and the urease metabolite NH₄⁺ on mucin expression in AGS cells. Materials and methods: AGS cells were transfected with CagA and/or treated with different concentrations of NH₄CL. Mucin gene and protein expression was assessed by qPCR and immunofluorescence assays, respectively. Results: CagA significantly upregulated MUC5AC, MUC2, and MUC5B expression in AGS cells, but did not affect E-cadherin and MUC6 expression. MUC5AC, MUC6, and MUC2 expression in AGS cells increased with increasing NH₄⁺ concentrations until reaching a peak level at 15 mM. MUC5B mRNA expression in AGS cells (NH₄⁺ concentration of 15 mM) was significantly higher than that at 0, 5, and 10 mM NH₄⁺. No changes in E-cadherin expression in AGS cells treated with NH₄⁺ were noted, except at 20 mM. The expression of MUC5AC, MUC2, and MUC6 mRNA in CagA-transfected AGS cells at an NH₄⁺ concentration of 15 mM was significantly higher than that at 0 mM, and decreased at higher concentrations. The expression of MUC5B mRNA increased with increases in NH₄⁺ concentration, and was significantly higher compared to that in untreated cells. No significant change in the expression of E-cadherin mRNA in CagA-transfected AGS cells was observed. Immunofluorescence assays confirmed the observed changes. Conclusion: H. pylori may affect the expression of MUC5AC, MUC2, MUC5B, and MUC6 in AGS cells via CagA and/or NH₄⁺, but not E-cadherin.
Growth, photosynthetic and nutrition characteristics of Pyropia haitanensis in response to the effects of increased CO2 and chloramphenicol

Abstract:Pyropia haitanensis was cultured under two CO 2 (410 (LC), 1000 (HC) μL L -1 ) concentrations and six chloramphenicol (CAP)-methanol solutions (0, 0+methanol, 10, 50, 100, 250 μg mL -1 ) to investigate the effects of elevated CO 2 and CAP on its growth, photosynthesis and biochemical characteristics. HC had no obvious effects on the growth rate (RGR) with CAP in the range of 10 to 100 μg mL -1 , but the decrease of RGR by HC was statistically significant with the CAP dosage at 250 μg mL -1 . HC had no significant effect on net photosynthetic rates (P n ) in the present of CAP (10-250 μg mL -1 ). CAP greatly reduced net photosynthesis as well as the maximal photochemical yield (F v /F m ) and photosynthetic efficiency (α ETR ). In contrast, the maximum relative electron transport rates (rETR m ) were almost constant with the CAP dosage from 10 to 100 μg mL -1 . HC significantly increased the energy fluxes (per RC) for absorption (ABS/RC), trapping (TR o /RC) and transport fluxes (ET o /RC) with the dosage of CAP at 250 μg mL -1 . Principal component analysis (PCA) indicated that CAP was positively correlated with the synthesis of free amino acids (FAA), contents of umami-, sweet- and essential AA were significantly enhanced with the interaction of HC and higher CAP dosage at 100 μg mL -1 , which led to the variation of flavor in algae. Furthermore, phycobiliproteins and soluble protein (SP) contents were remarkably reduced by CAP. Contents of chlorophyll a (Chl a ), carotenoids (Car), soluble carbohydrates (SC) and C/N ratios were almost unchanged among treatments. The study indicates that future ocean acidification has no obvious effects on the biomass productivity of P. haitanensis , maintained steady photosynthetic activities with the CAP (within 100 μg mL -1 ) and induces better flavor. The data obtained have important theoretical relevance for in-depth understanding of algal responses to global changes and oceanic contamination.

Characteristics of Inositol Phosphorylceramide Synthase and Effects of Aureobasidin A on Growth and Pathogenicity of Botrytis Cinerea

Abstract:Inositol phosphorylceramide (IPC) synthase is the key enzyme with highly conserved sequences, which is involved in fungal sphingolipid biosynthesis. The antibiotic aureobasidin A (AbA) induces the death of fungi through inhibiting IPC synthase activity. The mutations of AUR1 gene coding IPC synthase in fungi and protozoa causes a resistance to AbA. However, the mechanism of AbA resistance is still elusive. In this paper, we generated two mutants of Botrytis cinerea with AbA-resistance, BcAUR1a and BcAUR1b, through UV irradiation. BcAUR1a lost an intron and BcAUR1b had three amino acid mutations (L197P, F288S and T323A) in the AUR1 gene. AbA strongly inhibits the activity of IPC synthase in wild-type B. cinerea, which leads to distinct changes in cell morphology, including the delay in conidial germination, excessive branching near the tip of the germ tube and mycelium, and the inhibition of the mycelium growth. Further, AbA prevents the infection of wild-type B. cinerea in tomato fruits via reducing oxalic acid secretion and the activity of cellulase and pectinase. On the contrary, AbA has no effect on the growth and pathogenicity of the two mutants. Although both mutants show a similar AbA resistance, the molecular mechanisms might be different between the two mutants.
Antibacterial effect and mechanism against Escherichia coli of polysaccharides from Armillariella tabescens mycelia

Abstract:The objectives of this study were (1) to prepare Armillariella tabescens mycelia polysaccharides (PAT) with remarkably growth inhibitory effect on typical food-borne pathogenic bacteria using a green and efficient polyamide method and (2) to explore the antibacterial mechanism of PAT for use as a natural antibacterial agent. The sugar and uronic acid contents of PAT were 93.41% and 12.24%, respectively. PAT could inhibit the growth of Escherichia coli, Proteus vulgaris, Bacillus subtilis, and Staphylococcus aureus cells, with minimum inhibitory concentrations of 0.5, 1.0, 4.0, and 4.0 mg/mL, respectively. Ultra-high-resolution field emission scanning electron microscopy and high-resolution transmission electron microscopy analysis revealed cell wall and membrane rupture of E. coli treated with PAT. Further, 0.5–4.0 mg/ml PAT was found to significantly (P < 0.01) and concentration-dependently increase the conductivity of the broth, exudation of the intracellular protein, and alkaline phosphatase and β-galactosidase activities. Confocal laser scanning microscopy revealed morphological changes in E. coli DNA after PAT treatment and intracellular reactive oxygen species accumulation; flow cytometry revealed E. coli cell apoptosis. Our findings provide a theoretical basis and technical support for the development of PAT as a natural antibacterial product.

c-di-GMP Homeostasis Is Critical for Heterocyst Development in Anabaena sp. PCC 7120

Abstract:c-di-GMP is a ubiquitous bacterial signal regulating various physiological process. Anabaena PCC 7120 ( Anabaena ) is a filamentous cyanobacterium able to form regularly-spaced heterocysts for nitrogen fixation, in response to combined-nitrogen deprivation in 24h. Anabaena possesses 16 genes encoding proteins for c-di-GMP metabolism, and their functions are poorly characterized, except all2874 ( cdgS ) whose deletion causes a decrease in heterocyst frequency 48h after nitrogen starvation. We demonstrated here that c-di-GMP levels increased significantly in Anabaena after combined-nitrogen starvation. By inactivating each of the 16 genes, we found that the deletion of all117 5 ( cdgSH ) led to an increase of heterocyst frequency 24h after nitrogen stepdown. A double mutant ΔcdgSHΔcdgS had an additive effect over the single mutants in regulating heterocyst frequency, indicating that the two genes acted at different time points for heterocyst spacing. Biochemical and genetic data further showed that the functions of CdgSH and CdgS in the setup or maintenance of heterocyst frequency depended on their opposing effects on the intracellular levels of c-di-GMP. Finally, we demonstrated that heterocyst differentiation was completely inhibited when c-di-GMP levels became too high or too low. Together, these results indicate that the homeostasis of c-di-GMP level is important for heterocyst differentiation in Anabaena .

Overexpression of phytochelatin synthase (pcs) enhances abiotic stress tolerance by altering the proteome of transformed Anabaena sp. PCC 7120

Abstract:The present study provides data on the insertion of an extra copy of phytochelatin synthase (alr0975) in Anabaena sp. PCC 7120. The recombinant strain (AnFPN-pcs) compared to wild type showed approximately 22.3% increase in growth rate under UV-B, NaCl, heat, CuCl2, carbofuran, and CdCl2. It also registered 2.25-fold enhanced nitrogenase activity and 5-fold higher phytochelatin production. A comparison of the protein profile of wild type with the recombinant strain revealed that recombinant strain accumulated proteins belonging to the following categories: (i) detoxification (nutrient stress induced DNA binding protein, Mn-SOD, Alr0946 (CalA)), (ii) protein folding and modification (molecular chaperone DnaK, FKBP-type peptidyl-prolyl cis-trans isomerase), (iii) nucleotide and amino acid biosynthesis (dihydroorotase and Ketol-acid reductoisomerase), (iv) photosynthesis and respiration (coproporphyrinogen III oxidase, phycocyanin alpha chain, ferredoxin-NADP+ reductase), and (v) transport (sugar transport ATP-binding protein). Thus, it can be concluded that, above category proteins with their respective role in scavenging reactive oxygen species, proper folding of unfolded proteins, and protection of protein from degradation, sustained carbon fixation and energy pool and active transport of sugar together conceivably help the recombinant cyanobacterium (AnFPN-pcs) to cope with abiotic stress employed in the present study. Such recombinant strains have potential for future use as biofertilizer.

Abstract:Results show that Microcystis aeruginosa can utilize both dissolved organic phosphorus (DOP) and orthophosphate (DIP) even under low phosphorus (P) conditions to sustain its growth. Total P concentrations decreased markedly in all three P source treatments. Alkaline phosphatase activity (APA) in the different P sources tested changed in response to the DOP and DIP. The APA of DOP groups remained low after decreasing significantly, but the APA in the DIP treatments remained high during the period of culture. Changes in APA at different PO4(3-)-P concentrations in a culture medium revealed negative correlations between APA and DIP. However, a positive relationship was observed between APA and DOP under low P concentrations. These findings indicate that M. aeruginosa can regulate its physiological metabolism to acclimate to low ambient DIP environments.

Abstract:Pateamines act as inhibitors of the RNA helicase eIF4A and exhibit antiviral and anticancer properties. Recently, we observed that inhibition of eIF4A by rocaglates affects the immune response. To investigate whether the observed immunomodulatory effects are specific to rocaglates or the inhibition of eIF4A, a comprehensive study was conducted on the influence of pateamines that exhibit the same inhibitory mode of action as rocaglates on various immune cells. The effects of pateamine A (PatA) and des-methyl des-amino pateamine A (DMDA) on the expression of surface markers, release of cytokines, cell proliferation, inflammatory mediators and metabolic activity in primary human monocyte-derived macrophages (MdM), T cells and B cells were assessed. Additionally, safety and bioavailability profiles were determined. DMDA revealed almost no immunomodulatory effects within the tested concentration range of 0.5–5 nM. PatA reduced B cell activation, as shown by reduced immune globulin release and decreased chemokine release from macrophages, while T cell function remained unaffected. Both DMDA and PatA showed low permeability in Caco-2 and Calu-3 cell barrier assays and no mutagenic potential. However, 10 nM PatA exhibited genotoxic potential, as shown by the micronucleus assay. In conclusion, DMDA had a good safety profile but exhibited low permeability, whereas PatA had a poor safety profile and also low permeability. The observed immunomodulatory effects of elF4A inhibitors on B cells appear to be target-specific.

Insights into the Metabolic Response of Lactiplantibacillus plantarum CCFM1287 upon Patulin Exposure

Abstract:Patulin (PAT) is a common mycotoxin in the food industry, and is found in apple products in particular. Consumption of food or feed contaminated with PAT can cause acute or chronic toxicity in humans and animals. Lactiplantibacillus plantarum CCFM1287 is a probiotic strain that effectively degrades PAT in PBS and food systems. In this study, it was found that the concentration of PAT (50 mg/L) in MRS medium decreased by 85.09% during the first stages of CCFM1287 growth, and this change was consistent with the first−order degradation kinetic model. Meanwhile, the regulation of oxidative stress by L. plantarum CCFM1287 in response to PAT exposure and metabolic changes that occur during PAT degradation were investigated. The degree of intracellular damage was attenuated after 16 h of exposure compared to 8 h. Meanwhile, metabolomic data showed that 30 and 29 significantly different metabolites were screened intracellularly in the strain after 8 h and 16 h of PAT stress at 50 mg/L, respectively. The results of pathway enrichment analysis suggested that the purine metabolic pathway was significantly enriched at both 8 h and 16 h. However, as is consistent with the performance of the antioxidant system, the changes in Lactiplantibacillus diminished with increasing time of PAT exposure. Therefore, this study helps to further explain the mechanism of PAT degradation by L. plantarum CCFM1287.

Abstract:Bacterial cell shape is determined by the peptidoglycan (PG) layer. The cyanobacterium Anabaena sp. PCC 7120 ( Anabaena ) is a filamentous strain with ovoid-shaped cells connected together with incomplete cell constriction. When deprived of combined nitrogen in the growth medium, about 5–10% of the cells differentiate into heterocysts, cells devoted to nitrogen fixation. It has been shown that PG synthesis is modulated during heterocyst development and some penicillin-binding proteins (PBPs) participating in PG synthesis are required for heterocyst morphogenesis or functioning. Anabaena has multiple PBPs with functional redundancy. In this study, in order to examine the function of PG synthesis and its relationship with heterocyst development, we created a conditional mutant of mraY , a gene necessary for the synthesis of the PG precursor, lipid I. We show that mraY is required for cell and filament integrity. Furthermore, when mraY expression was being limited, persistent septal PG synthetic activity was observed, resulting in increase in cell width. Under non-permissive conditions, filaments and cells were rapidly lysed, and no sign of heterocyst development within the time window allowed was detected after nitrogen starvation. When mraY expression was being limited, a high percentage of heterocyst doublets were found. These doublets are formed likely as a consequence of delayed cell division and persistent septal PG synthesis. MraY interacts with components of both the elongasome and the divisome, in particular those directly involved in PG synthesis, including HetF, which is required for both cell division and heterocyst formation.
