Abstract:Polymeric nanostructured materials (PNMs), which are polymeric materials in nanoscale or polymer composites containing nanomaterials, have become increasingly useful for biomedical applications. In specific, advances in polymer-related nanoscience and nanotechnology have brought a revolutionary change to produce new biomaterials with tailored properties and functionalities for targeted biomedical applications. These materials, including micelles, polymersomes, nanoparticles, nanocapsules, nanogels, nanofibers, dendrimers and nanocomposites, have been widely used in drug delivery, gene therapy, bioimage, tissue engineering and regenerative medicine. This review presents a comprehensive overview on the various types of PNMs, their fabrication methods and biomedical applications, as well as the challenges in research and development of future PNMs.

Abstract:AbstractOver the past few decades, interest in designing and developing polymeric nanoparticles has undergone considerable explosion. Indeed, these nanoparticulated polymer-based systems provide potential solution to improve therapeutic efficacy and diagnosis sensitivity. In this chapter, general properties, production, and characterization of polymer nanoparticles are introduced. Specifically, the development and application of polyhydroxyalkanoate (PHA)-based nanoparticles are emphasized because of the good biocompatible, biodegradable properties, as well as their mechanical flexibility. These PHAs nanoparticles can serve as targeting drug delivery carriers and protein purification and immobilization matrices. The perspective outlook in the last section highlights the future application of polymer nanoparticles in translational science.

Abstract:Nanosized particles have appealing attributes, which have gotten impressive consideration in the last few decades. Polymeric nanoparticles (PNP)s are particles or particulate materials with size at least in the range of 10–100 nm in one dimension. Due to the extremely small size, high volume-surface area ratio, tunable pore, polymer nanoparticles have been used for various applications in different fields,for example, in drug delivery, biosensors, stimuli responsive cargo delivery, nanocomposites, agricultural and environmental applications. The recent development of incorporation of inorganic nanoparticles to polymers permits the alteration of the polymer's physical as well as chemical properties just as the execution of new highlights in the polymer network. Conjugated polymer nanoparticles are profoundly flexible nano-organized materials that can conceivably discover applications in different branches of science and engineering, for example, optoelectronics, photonics, bio-imaging, bio-detecting and nanomedicine. Their desired sizes and unique properties, biocompatibility and low-toxicity make these materials profoundly appealing for the previously mentioned applications. Polymeric nanoparticles (NPs) are one of the most used nanomaterials for application in nanomedicine. Extreme intrigue lies in the capability of polymeric NPs to alter present-day medication. Usages of polymeric NPs incorporate medication delivery, for example, conjugation and entanglement of medications, prodrugs, stimuli responsive systems, imaging modalities, and theranostics. Biodegradable polymeric nano-structures (BPN) have indicated extraordinary guarantee in various remedial applications, for example, analysis, imaging, sedate delivery, beautifying agents, organ embeds and tissue designing. BPNs display better properties over their separate bulk materials, conquering the restrictions of customary polymers to address numerous significant clinical issues. Besides, the recent developments in research in various area of polymers can empower explicit functionalities of the polymeric nanostructures to be balanced for tunable applications. This concise review emphasizes the various preparative methods of polymeric nanoparticles developed in the last few decades and their uses in various branches of science, e.g., therapeutic, optoelectronic, catalytic and magnetic applications along with several challenging issues concerning the commercialization of emerging polymer-based therapeutics. Abstract:Tremendous developments in nanotechnology have revolutionized the impact of nanoparticles (NPs) in the scientific community and, more recently, in society. Nanomaterials are by their definition materials that have at least one dimension in range of 1 to 100 nm. Nanoparticles are found in many types of different technological and scientific applications and innovations, from delicate electronics to state-of-the-art medical treatments. Medicine has recognized the importance of polymer materials coated with NPs and utilizes them widely thanks to their excellent physical, chemical, antibacterial, antimicrobial, and protective properties. Emphasis is given to their biomedical application, as the nanoscale structures are in the range of many biological molecules. Through this, they can achieve many important features such as targeted drug delivery, imaging, photo thermal therapy, and sensors. Moreover, by manipulating in a "nano-scale" range, their characteristic can be modified in order to obtain the desired properties needed in particular biomedical fields, such as electronic, optical, surface plasmon resonance, and physic-chemical features.

Abstract:Polymeric materials have been used in a range of pharmaceutical and biotechnology products for more than 40 years. These materials have evolved from their earlier use as biodegradable products such as resorbable sutures, orthopaedic implants, macroscale and microscale drug delivery systems such as microparticles and wafers used as controlled drug release depots, to multifunctional nanoparticles (NPs) capable of targeting, and controlled release of therapeutic and diagnostic agents. These newer generations of targeted and controlled release polymeric NPs are now engineered to navigate the complex in vivo environment, and incorporate functionalities for achieving target specificity, control of drug concentration and exposure kinetics at the tissue, cell, and subcellular levels. Indeed this optimization of drug pharmacology as aided by careful design of multifunctional NPs can lead to improved drug safety and efficacy, and may be complimentary to drug enhancements that are traditionally achieved by medicinal chemistry. In this regard, polymeric NPs have the potential to result in a highly differentiated new class of therapeutics, distinct from the original active drugs used in their composition, and distinct from first generation NPs that largely facilitated drug formulation. A greater flexibility in the design of drug molecules themselves may also be facilitated following their incorporation into NPs, as drug properties (solubility, metabolism, plasma binding, biodistribution, target tissue accumulation) will no longer be constrained to the same extent by drug chemical composition, but also become in-part the function of the physicochemical properties of the NP. The combination of optimally designed drugs with optimally engineered polymeric NPs opens up the possibility of improved clinical outcomes that may not be achievable with the administration of drugs in their conventional form. In this critical review, we aim to provide insights into the design and development of targeted polymeric NPs and to highlight the challenges associated with the engineering of this novel class of therapeutics, including considerations of NP design optimization, development and biophysicochemical properties. Additionally, we highlight some recent examples from the literature, which demonstrate current trends and novel concepts in both the design and utility of targeted polymeric NPs (444 references).

Abstract:The use of biodegradable polymeric nanoparticles (NPs) for controlled drug delivery has shown significant therapeutic potential. Concurrently, targeted delivery technologies are becoming increasingly important as a scientific area of investigation. Abstract:Polymeric nanoparticles (NPs) are particles within the size range from 1 to 1000 nm and can be loaded with active compounds entrapped within or surface-adsorbed onto the polymeric core. The term “nanoparticle” stands for both nanocapsules and nanospheres, which are distinguished by the morphological structure. Polymeric NPs have shown great potential for targeted delivery of drugs for the treatment of several diseases. In this review, we discuss the most commonly used methods for the production and characterization of polymeric NPs, the association efficiency of the active compound to the polymeric core, and the in vitro release mechanisms. As the safety of nanoparticles is a high priority, we also discuss the toxicology and ecotoxicology of nanoparticles to humans and to the environment.

Abstract:The potential of nanoparticles as effective drug delivery systems combined with the versatility of fibers has led to the development of new and improved strategies to help in the diagnosis and treatment of diseases. Nanoparticles have extraordinary characteristics that are helpful in several applications, including wound dressings, microbial balance approaches, tissue regeneration, and cancer treatment. Owing to their large surface area, tailor-ability, and persistent diameter, fibers are also used for wound dressings, tissue engineering, controlled drug delivery, and protective clothing. The combination of nanoparticles with fibers has the power to generate delivery systems that have enhanced performance over the individual architectures. This review aims at illustrating the main possibilities and trends of fibers functionalized with nanoparticles, focusing on inorganic and organic nanoparticles and polymer-based fibers. Emphasis on the recent progress in the fabrication procedures of several types of nanoparticles and in the description of the most used polymers to produce fibers has been undertaken, along with the bioactivity of such alliances in several biomedical applications. To finish, future perspectives of nanoparticles incorporated within polymer-based fibers for clinical use are presented and discussed, thus showcasing relevant paths to follow for enhanced success in the field.

Abstract:Polymeric nanoparticles (NPs) play a key role in nanoscale formulations for bioimaging, cancer treatment and theranostics. In this work, we designed and synthesized a series of hydrophobic polymers (P1-6) with different pendent groups via one-step multicomponent Passerini reaction. These polymers possessed similar molecular structures and various biomedical functions. Interestingly, they could self-assemble into stable NPs in aqueous media. All formed NPs were redox-sensitive because of the existence of disulfide bonds in the backbone. The stability of NPs in aqueous media with or without glutathione was systematically evaluated and compared. The optical performance, including fluorescence resonance energy transfer (FRET), was characterized in different conditions for those polymers with fluorescent components. Importantly, all formed NPs showed good cytocompatibility towards HeLa cells, and different biological functions, including drug loading and delivery, bioimaging with variable fluorescence, and photodynamic activity, as evidenced by experiments in vitro and in vivo. These results demonstrate that the great potential of multicomponent reaction to customize versatile polymeric nanoparticles for biomedical applications.

Abstract:Hybrid inorganic colloidal particles have attracted a great attention in the last years, and they have been largely used in various applications and more particularly in biomedical nanotechnology. Recently, they are used as carriers for biomolecules, and exploited for use in microsystems, microfluidics and in lab-on-a chip based bionanotechnology. Various kinds of hybrid particles can be listed starting from classical inorganic nanoparticles such as silica, gold, silver, iron oxide and those exhibiting intrinsic properties such as semiconducting nanoparticles (e.g. quantum dots). As a general tendency, to be conveniently used in biomedical applications, the encapsulation of the inorganic nanoparticles in a polymer matrix is incontestably needed. Consequently, various chemistry-based encapsulation processes have been developed and showed promising results as compared to the encapsulation using preformed polymers.

Abstract:The formulation of nanoemulsions by low-energy strategies, particularly by the phase inversion composition method, and the use of these nanoemulsions as templates for the preparation of polymer nanoparticles for biomedical applications are reviewed. The methods of preparation, nature of the components in the formulation, and their impact on the physicochemical properties, drug loading, and drug release are discussed. We highlight the utilization of ethyl cellulose, poly(lactic-co-glycolic acid), and polyurethane/polyurea in the field of nanomedicine as potential drug delivery systems. Advances are still needed to achieve better control over size distribution, nanoparticle concentration, surface functionalization, and the type of polymers that can be processed.

Abstract:The recent decade has witnessed a remarkable surge in the field of nanoparticles, from their synthesis, characterization, and functionalization to diverse applications. At the nanoscale, these particles exhibit distinct physicochemical properties compared to their bulk counterparts, enabling a multitude of applications spanning energy, catalysis, environmental remediation, biomedicine, and beyond. This review focuses on specific nanoparticle categories, including magnetic, gold, silver, and quantum dots (QDs), as well as hybrid variants, specifically tailored for biomedical applications. A comprehensive review and comparison of prevalent chemical, physical, and biological synthesis methods are presented. To enhance biocompatibility and colloidal stability, and facilitate surface modification and cargo/agent loading, nanoparticle surfaces are coated with different synthetic polymers and very recently, cell membrane coatings. The utilization of polymer‐ or cell membrane‐coated nanoparticles opens a wide variety of biomedical applications such as magnetic resonance imaging (MRI), hyperthermia, photothermia, sample enrichment, bioassays, drug delivery, etc. With this review, our goal is to provide a comprehensive toolbox of insights into polymer or cell membrane‐coated nanoparticles and their biomedical applications, while also addressing the challenges involved in translating such nanoparticles from laboratory benchtops to in vitro and in vivo applications. Abstract:Polymeric nanoparticles (NPs) play an important role in controlled cancer drug delivery. Anticancer drugs can be conjugated or encapsulated by polymeric nanocarriers, which are known as polymeric nanomedicine. Polymeric nanomedicine has shown its potential in providing sustained release of drugs with reduced cytotoxicity and modified tumor retention, but until now, few delivery systems loading drugs have been able to meet clinical demands, so more efforts are needed. This research reviews the current state of the cancer drug-loading system by exhibiting a series of published articles that highlight the novelty and functions from a variety of different architectures including micelles, liposomes, dendrimers, polymersomes, hydrogels, and metal-organic frameworks. These may contribute to the development of useful polymeric NPs to achieve different therapeutic purposes.

Abstract:Polylactic acid (PLA)—based polymers are ubiquitous in the biomedical field thanks to their combination of attractive peculiarities: biocompatibility (degradation products do not elicit critical responses and are easily metabolized by the body), hydrolytic degradation in situ, tailorable properties, and well-established processing technologies. This led to the development of several applications, such as bone fixation screws, bioresorbable suture threads, and stent coating, just to name a few. Nanomedicine could not be unconcerned by PLA-based materials as well, where their use for the synthesis of nanocarriers for the targeted delivery of hydrophobic drugs emerged as a new promising application. The purpose of the here presented review is two-fold: on one side, it aims at providing a broad overview of PLA-based materials and their properties, which allow them gaining a leading role in the biomedical field; on the other side, it offers a specific focus on their recent use in nanomedicine, highlighting opportunities and perspectives.

Abstract:Nanoscale materials with a broad spectrum of applications are providing a new foundation for technological integration and innovation. In this article, we review various polymers being used, their polymeric properties, nanoparticle (NP) fabrication, and mechanisms and kinetics of drug release. Specific information is given on each polymer regarding transportation, fate, and delivery issues. Nanoparticles have been developed to deliver conventional drug molecules, peptides and proteins, vaccines, and genes or nucleotides. Nanoparticles have wide application in fields such as cancer therapeutics and targeted drug delivery, including transbarrier brain delivery, dermal and transdermal delivery, intraocular delivery, parenteral delivery, and imaging and diagnostics. In this review, we focus on the potentials of nanotechnology in medicine, and we discuss different nanoparticulate drug delivery systems, including polymeric NPs, metal and metal oxide NPs, ceramics, quantum dots, carbon nanotubes, polymeric micelles, and dendrimers along with their applications in therapeutics, imaging, and diagnostics. In addition, we identify several interesting developments that will affect the future of nanotechnology and nanomedicine.

Abstract:Magnetic nanoparticles (NPs) are emerging as an important class of biomedical functional nanomaterials in areas such as hyperthermia, drug release, tissue engineering, theranostic, and lab-on-a-chip, due to their exclusive chemical and physical properties. Although some works can be found reviewing the main application of magnetic NPs in the area of biomedical engineering, recent and intense progress on magnetic nanoparticle research, from synthesis to surface functionalization strategies, demands for a work that includes, summarizes, and debates current directions and ongoing advancements in this research field. Thus, the present work addresses the structure, synthesis, properties, and the incorporation of magnetic NPs in nanocomposites, highlighting the most relevant effects of the synthesis on the magnetic and structural properties of the magnetic NPs and how these effects limit their utilization in the biomedical area. Furthermore, this review next focuses on the application of magnetic NPs on the biomedical field. Finally, a discussion of the main challenges and an outlook of the future developments in the use of magnetic NPs for advanced biomedical applications are critically provided.

Abstract:Due to recent advancements in the field of science and technology, it becomes easier to synthesize and produce nanomaterials. These nanomaterials are now better choice to the researchers for the development of various products. Their use in the development of variety of products including biomedical devices and drug delivery systems has much attention to the researchers. Due to change in the physical properties at molecular level, it becomes easier to overcome the hurdles in the development of drug delivery systems with better therapeutic benefit and ease of convenience to the patients. Different carrier systems are useful for the delivery of drugs to the site of action, but nanocarriers systems have attracted due to their particle size, shape, strength and huge surface area. Among these carrier systems, nanocomposite polymeric materials have greater attention for their use in the development of various drug delivery and targeting devices. In many recent studies, their applications in the design of biomedical devices and bone tissue engineering are observed. In this review, the various types nanocomposite polymeric materials, their behaviour during synthesis, evaluation parameters and various applications with critical findings in the field of pharmaceuticals and biomedical sciences are discussed.
