Abstract:Purpose: The goal of this study was to assess the effectiveness of a bicanalicular intubation procedure for the treatment of acquired punctual stenosis (APS). Methods: A prospective study was performed on consecutive patients who accepted bicanalicular intubation to treat their APS. The intubation lasted for three to six months. Follow up time was for more than one year after removal of the Crawford tube. At follow-up, patients' signs and symptoms were carefully evaluated as well as the condition of their lacrimal punctum. Results: In total, 48 patients (67 eyes) met the criteria for acquired punctual stenosis. There were 39 females and nine males. The mean age was 48 +/- 15 years (range, 13-73 years). The initial complaint for all patients was watery eyes. Both the upper and lower punctum were closed up in 61 eyes and a single punctum was missing in six eyes. The mean duration of symptoms was 14.9 +/- 5.8 months (range 6-48 months). The mean follow up time was 12.5 +/- 3.8 months after pulling out the Crawford tube. At the final follow-up, the treatment was anatomically successful for all 67 eyes. Of the total number of eyes, 63 (94%) showed a significant improvement in symptoms. Two eyes were relieved of indoor epiphora, and two eyes had minimal epiphora outdoors but only with wind or cold. One patient had lacrimal punctum granulation and subsequently underwent granulation excision, one patient had punctum dehiscence. Conclusions: Bicanalicular intubation is an effective, safe, simple, and relatively non-invasive treatment strategy for the management of epiphora secondary to APS.

Abstract:Objective: Repair of medial canthus injury involving canaliculus is an emergency indication for canalicular intubation to restore lacrimal drainage. Herein, the author has described an innovative but simple technique for this reconstruction. Method and result: A small, blunt pigtail probe was gently passed through the opposite canaliculus in a rotational manner. A silicon stent was threaded inside canaliculi by reverse rotation of the pigtail in an atraumatic way. The technique was used on 4 pediatric cases without any postoperative complication or epiphora. Conclusions: This technique of intubation is simple, cheap, and useful in canalicular emergencies, including "distal" canaliculus lacerations.

Abstract:Objectives: To evaluate the efficacy and safety of the self-fixing and self-detachable drainage stent in pancreaticojejunostomy and to provide supportive data for the follow clinical trials. Methods: This is an experimental research in animals which completed from February 2022 to September 2022. A self-fixing and self-detachable pancreaticojejunostomy drainage stent was designed for Hong's pancreaticojejunostomy technique based on the theory of "fistula healing" in pancreaticojejunostomy. Ten biocompatibility tests were completed in vitro before this study. Twenty-five Bama minipigs were selected and double-ligated in the neck of the pancreas to dilate the distal main pancreatic duct. Twenty-three of them were successfully modelled and divided into three groups by a stratified random method: pancreaticojejunostomy drainage stent group (referred to as stent group) with 11 pigs, pancreatic duct to jejunal mucosa anastomosis group (referred to as manual suture group) with 8 pigs, sham operation group with 4 pigs. The anastomic time,amylase content in postoperative abdominal drainage fluid and the tolerable pressure value of pancreaticojejunostomy were compared between the stent group and the manual suture group. An abdominal X-ray fluoroscopy examination was adopted to detect the detach time of the stent. A postoperative pathological examination was performed to verify the healing time,the type of treatment and the stricture rate of pancreaticojejunostomy. Quantitative data was analyzed by independent sample t-test. The classified data were analyzed by Fisher's exact test. Results: There were no significant differences in the diameter of the pancreatic duct and pancreatic texture,the time of pancreaticojejunostomy,the amylase content in postoperative peritoneal drainage fluid,and the tolerable pressure value of the pancreaticojejunostomy between the stent group and the manual suture group(all P>0.05). Abdominal X-ray fluoroscopy showed that the stents gradually detached and were removed from the body 21 days after operation,and all stents were detached in the follow 3 months after operation. Pancreaticojejunostomy healed 7 days after operation based on fistula formation in the stent group,and 14 days in the manual suture group. The incidence of anastomotic stricture within 35 days after operation was 2/8 in the stent group and 6/8 in the manual suture group (Fisher's exact test: P=0.132). Conclusion: The stent method is safer and simpler than the manual suture method in pancreaticojejunostomy of Bama minipigs, with shorter anastomotic healing time and lower stricture rate.
Bicanalicular Silicone Intubation for the Management of Punctal Stenosis and Obstruction in Patients With Allergic Conjunctivitis

Abstract:Purpose: To evaluate the use of bicanalicular silicone intubation for the management of punctal stenosis and obstruction in patients with allergic conjunctivitis. Methods: A retrospective interventional case series of patients with acquired epiphora due to stenotic or obstructed puncta as a result of allergic conjunctivitis was performed. Punctal dilation and bicanalicular silicone intubation were performed in all patients. Munk Scale for grading of epiphora along with grading of fluorescein dye disappearance test was used to evaluate the functional improvement. Grading of punctal stenosis using Kashkouli's grading system was applied to evaluate the anatomical improvement. Patients having canalicular or nasolacrimal duct obstruction, punctal stenosis, and obstruction due to other causes were excluded. Results: Fifty-one patients met the inclusion criteria with male-female ratio of 1:1.55 and an average age of 46 years at presentation. High patient tolerances without complications with the use of tubes were reported. Significant improvement 6 months after tube removal in comparison to preoperative period was found with anatomical and functional success rate of 91.83% and 87.75%, respectively. Two patients had a recurrence of the punctal stenosis many months after stent removal because of the early tube prolapse and exacerbation of the allergic conjunctivitis. Conclusions: Bicanalicular silicone intubation seems to be a well-tolerated and effective tool in the management of acquired punctal stenosis or obstruction secondary to allergic conjunctivitis. Bicanalicular silicone intubation appears to be a good option as patients with allergic conjunctivitis typically present with bilateral involvement of both puncti.

Abstract:Objective To evaluate the clinical effect of KTP laser combined with silicone tube for treatment of punctual stenosis and obliteration of lacrimal punctum.Methods Totally 70 patients(76 eyes)of punctual stenosis and obliteration with epiphora were treated by punctum dilation followed by laser therapy and silicone tube insertion into the lacrimal puncture.The tubes were kept for one month and then removed,and all the cases were followed up for 2-3 months.Results There were no complications during or after operation.The patients were followed up for 3-4 months.Fifty-seven patients(58 eyes)were cured,8 patients(10 eyes)were improved,and 5 patients(8 eyes)failed to have any effects.Conclusion KTP laser combined with silicone tube is a safe,effective treatment for the punctual stenosis and obliteration;it is easy to perform,economical,and with high success rate.(Shanghai Med J,2010,33:761-763)

Abstract:Objective To observe the effect of using endoscope-assisted silicone tube double-passage intubation in anastomosis for lacrim · al canalicular laceration.Methods Twenty-seven cases (27 eyes) of traumatic lacrimal canalicular laceration,under operating microscope implementation of lacrimal duct anastomosis,using endoscope-assisted silicone tube for stent,silicone tube inserted through the lacrimal punctum using the double-passage model.Results Silicone tube was removed 3～ 6 months postoperative,eyelid shape remained normal.No obvious discomfort occurred.After 3 ～ 6 months followed-up,3 cases of lacrimal duct flushing was not free and clear tears overflowed,2 cases of lacrimal duct flowed but still with mild overflow tears,24 cases of lacrimal duct flow had no tears overflow.Conclusions Using endoscope-assisted silicone tube double-passage intubation in anastomosis for lacrimal canalicular laceration,to avoid the traditional silicone tube catheter anastomosis insufficiency,make the effect of lacrimal duct anastomosis more accurate and minimally invasive,is an ideal treatment of lacrimal canalicular laceration.
Application of the Sodium Hyaluronate in Mitomycin C Trabeculectomy

Abstract:Objective To study the application of sodium hyaluronate (SH)in mitomycin C (MMC) trabeculectomy.Methods 98 cases (116 eyes)were operated with MMC trabeculectomy.SH was injected into the anterior chamber and under the sutured scleral flaps in 52 cases(62 eyes) ,and the other 46 cases(54 eyes) as control group were jnjected into BSS.The postoperative visual acuity, intraocular pressure (IOP), filtration, forming of anterior chamber and the postoperative complications were compared between the two groups. Results The postoperative visual acuity, the forming of anterior chamber or the postoperative complications in the treatment group was better than that in the control group(P<0.01 or P<0.05).The postoperative IOP of the treatment group was(8.3±1.4)mmHg(second day),or(7.5±1.6)mmHg(1week later),or(13.2±2.4) mmHg(half a year later)averagely and that of the control group was(5.6±1.0)mmHg(P<0.05),or(6.8±1.2)mmHg(P>0.05), or(17.6±3.8)mmHg(P<0.05).All cases were followed up for 6 months to 3 years(18 months averagely).The rate of functional filtering bubles was 96.8% in the treatment group,and that in the control groupwas 85.2%(P<0.05).The successful rate was 95.2% and 81.5% respectively in two groups(P<0.05).Conclusins sodium hyaluronate using in MMC trabeculectomy can decrease the postoperative complications and elevate the successful rate.
Treatment of Upper and Lower Lacrimal Punctal Occlusion Using Retrograde Canaliculotomy and Punctoplasty

Abstract:This is a retrospective, noncomparative analysis of a case series to explore the safety and effectiveness of retrograde canaliculotomy and punctoplasty for treating epiphora due to upper and lower lacrimal punctal occlusion. During the procedure, the horizontal portion of the normal lower canaliculus was identified; the corresponding punctum was reconstructed via retrograde canaliculotomy and punctoplasty. Intubation was performed to prevent postoperative reocclusion. Patients were followed up for 12 to 24mo. A total of 16 patients with unilateral upper and lower lacrimal punctal occlusion were included. Satisfactory outcomes were achieved: all 16 patients exhibited improvement of epiphora; 31 rebuilt punctal openings and canaliculi achieved recanalization. Only one upper punctal opening could not be reconstructed because the corresponding canaliculus exhibited severe injury. No significant complications occurred as a result of the treatments. Retrograde canaliculotomy and punctoplasty appears to effective, safe, and minimally invasive for treatment of upper and lower punctal occlusion.

Abstract:We continue our review of on punctal stenosis by providing a detailed discussion of management modalities, their complications, and outcomes. There is a significant change in the understanding of punctal and peripunctal anatomy, puncto-canalicular junction, and the lacrimal pump mechanisms. While the snip punctoplasty procedures are still practiced, there is an increasing trend toward nonincisional procedures. The nonincisional procedures in select cases appear to be equally effective as the incisional ones. Although simple to use, punctal plugs never became the mainstay of treatment because of design issues and the inability to address the coexisting canalicular stenosis. Placing stents only in the lower punctum in cases of upper and lower punctal stenosis should be discouraged, and management needs to address punctal stenosis and not which punctum is involved. Several types of stents are used in the management of punctal stenosis, mostly based on surgeon's preference. The benefits of adjuvant mitomycin C are uncertain. In view of literature on how stent biofilms can themselves cause chronic inflammation, placing them for prolonged periods should be reviewed and debated. Enhanced understanding of the molecular pathogenesis of punctal stenosis and addressing the current controversies in management would help standardize the therapeutic interventions available in the lacrimal armamentarium.

Long-Term Effects of Simultaneous Subconjunctival and Subscleral Mitomycin C Application in Repeat Trabeculectomy

Abstract:Purpose: To assess the efficacy and safety of simultaneous mitomycin C application under conjunctival and scleral flaps in patients with repeat trabeculectomy.Methods: A total of 44 patients (44 eyes) with previous failed filtering surgery were randomized to one of two groups. The both-flaps group comprised 22 patients (22 eyes) with trabeculectomy and intraoperative mitomycin C application under conjunctival and scleral flaps, whereas the subconjunctival group comprised 22 patients (22 eyes) with subconjunctival application of mitomycin C. Particular attention was paid to intraocular pressure, postoperative medications, visual acuity, filtering bleb appearance, and complications. The mean follow-up time was 38.18 +/- 12.48 months.Results: The mean preoperative intraocular pressure decreased from 39.1 +/- 7.3 mm Hg to the postoperative level of 15.6 +/- 4.8 mm Hg in the both-flaps group (P = 0.014), and from 39.4 +/- 8.4 to 18.7 +/- 5.8 mm Hg in the subconjunctival group (P = 0.018). There was a statistically significant difference in intraocular pressure at all follow-up times, except at 1 week and 1 month postoperatively. Kaplan-Meier survival analysis showed there was no significant difference in total success rate (complete plus qualified success) between the two group, (P = 0.622, log-rank test). However, the two survival curves for the complete success subgroups (without additional medications) confirmed that mitomycin C applications under both flaps had a higher success rate than subconjunctival application (P = 0.043, log-rank test). No statistically significant difference in medications was present between the two groups, and no severe complications developed in either group.Conclusions: Trabeculectomy augmented with mitomycin C application at both sites could produce a greater lowering of intraocular pressure with low incidence of postoperative complications, and could provide an increased chance of long-term success. The procedure is effective and safe in patients with repeat trabeculectomy.

Abstract:Silicone stents are routinely used for the maintenance of patent mucosal passages in patients with nasolacrimal disorders. A common complication associated with the use of silicone stents is lateral migration or displacement of the tubes, which can be difficult to correct. This report describes a modified Quickert-Dryden approach with fixation of the tubes by an intrasac suture. From 1990 to 1996, 53 patients had silicone stents placed by this method with no complications related to tube displacement. The intrasac fixation suture has distinct advantages over other fixation methods.

Safety and Efficacy of Mitomycin C: Augmented Trabeculectomy: Subtenon's Injection versus Sponge Application

Abstract:Objective: The objective of the study was to study the safety and efficacy of subtenon injection of mitomycin C (MMC) versus sponge application of MMC during trabeculectomy. Materials and methods: Thirty-seven patients having primary glaucoma warranting trabeculectomy were enrolled in the study and their forty eyes were alternately allocated into either of the two groups: subtenon injection (ST) of 0.1 mL of 0.01% of MMC or sponge application (SP) of 0.02% of MMC and were operated by a single surgeon and followed for 3 months. The outcome was analyzed primarily based on reduction in intraocular pressure (IOP) and bleb morphology. Results: Similar outcome in terms of complete success (ST - 90% and SP - 85%), qualified success (ST - 5% and SP - 5%), and failure rate (ST - 5% and SP - 10%) was seen at the end of 3 months. The absolute reduction in IOP from the baseline was -10.00 ± 3.67 mmHg (-41.2% ± 12.30) in ST versus -8.90 ± 5.56 mmHg (-35.9% ± 16.1) in the SP group at the end of 3 months. At the end of 3 months, blebs in the ST group had low-to-medium height and in the SP group had low height. Blebs in both the groups were diffuse with mild vascularity. Antiglaucoma medications required postoperatively were 0.20 ± 0.62 versus 0.40 ± 1.10 in the ST and SP group, respectively. The duration of surgery was 19.85 ± 0.75 min in the ST group versus 22.50 ± 0.51 min in the SP group. Conclusion: Subtenon injection of MMC is as efficacious and safe as the conventional sponge application technique.
Clinical and scintigraphic comparison of silicone and polyvinylpyrrolidone coated silicone perforated plugs

Abstract:Background/aim: Isolated stenosis of the lacrimal punctum is a frequent cause of epiphora. Treatment relies on surgical opening or dilatation with bi- or monocanalicular prosthesis. Recently, silicone perforated punctum plugs (PPP) were proposed. The drawback with these silicone PPP was that secretions accumulate in the central orifice blocking the spontaneous flow of tears. A modification of the surface of the PPP using polyvinylpyrrolidone (PVP) was thus proposed. The aim was to compare silicone PPP with new PVP surface treated PPP. Methods: A prospective study was conducted of 20 patients with dilatable stenosis of the lacrimal puncta who developed epiphora. Epiphora, tolerance, implantation of the PPP, and lacrimal drainage were evaluated using scintigraphy of the lacrimal ducts. Results: The raw data and statistical analysis showed evidence of a superior performance of PVP surface treated PPP. Conclusion: Long term evaluation of the advantages or risks of PVP plugs and comparison with microsurgical punctoplasty are warranted.
Endoscopic endonasal dacryocystorhinostomy with adjunctive mitomycin C in nasolacrimal duct obstruction

Abstract:Objective To explore the outcome and safety of endonasal dacryocystorhinostomy(EN-DCR) with the use of adjunctive mitomycin C(MMC) in nasolacrimal duct obstruction.Methods Fifty-six consecutive patients(81eyes)underwent a EN-DCR procedure,with an endonasal approach to the lacrimal sac,surgical removal of nasal mucosa,lacrimal bone,and a fragment of the frontal process of the maxilla.The medial wall of the lacrimal sac was removed completely,and a cottonoid soaked in MMC at 0.5mg/ml was placed at the osteotomy site for 10 minutes.Results The EN-DCR procedure with adjunctive MMC was successful in 212(95.4%) cases.No complications involving MMC occured during 1-3 year(mean 1.6 years) follow-up.Conclusions Results suggest that EN-DCR with MMC is a safe and successful procedure for the treatment of nasolacrimal duct obstruction.

Abstract:We describe a technique used in enucleated pig eyes to create a uniformly hardened lens nucleus and an anterior capsule similar to the cataractous human lens. After treatment with microwave heat and a fixative agent, the pig eyes have clear corneas, anterior capsules with less tension and elasticity, and harder lens nuclei, with a consistency similar to that of a nuclear sclerosis grade I to V in the Emery-Little classification. The eyes can be used by residents in training to practice various phacoemulsification techniques, including continuous curvilinear capsulorhexis, hydrodelineation, sculpting, divide and conquer, phaco chop, and nondividing phacoemulsification.
Evaluation of lacrimal duct probing in adults with 0.02% and 0.04% mitomycin-C in primary acquired nasolacrimal duct obstruction: A randomized comparative pilot study

Abstract:PURPOSE: The purpose of the study is to evaluate the safety and efficacy of adjunctive use of mitomycin-C (MMC) using two different concentrations 0.2 mg/ml and 0.4 mg/ml for lacrimal duct probing to treat the nasolacrimal duct obstruction (NLDO) in adults. SUBJECTS AND METHODS: Prospective, an interventional comparative randomized pilot study of lacrimal duct probing conducted in the two study groups 0.02% MMC group ( n = 30) and 0.04% MMC group ( n = 30) in confirmed primary acquired NLDO of <1-year duration. Patency of lacrimal duct probing confirmed by syringing was compared at 1, 3, and 6-month follow-up in the two study groups, and corresponding subjective improvement of watering was appraised according to Kraft and Crawford grading. RESULTS: Patency of lacrimal duct probing in 0.02% MMC versus 0.04% MMC group was 66.66%/73.33% ( P = 0.71) at 1 month, 46.66%/66.66% ( P = 0.09) at 3 months and 46.66%/66.66% ( P = 0.03) at 6-month follow-up, respectively. Subjective improvement of watering (no watering and mild watering) observed in 0.02% MMC versus 0.04% MMC group was 46.66%/73.33% ( P = 0.03), at 1 month, 66.66%/83.33% ( P = 0.13) at 3 months and 56.66%/73.33% ( P = 0.17) at 6-month follow-up. CONCLUSION: Adult lacrimal duct probing with 0.04% MMC was associated with significant higher objective success rate than adult lacrimal duct probing with 0.02% MMC, without added concurrently side effects.

Abstract:Objective To observe the clinical efficacy of silicone punctal plug for the treatment of acquired lacrimal puncta stenosis. Methods The data of 46 eyes of 23 cases with acquired lacrimal stenosis from Apr. 2017 to Apr. 2019 were analyzed retrospectively. The observation group, 23 operative eyes, received punctum dilation followed by silicone punctal plug inserted into lacrimal punctum under surgjcal microscope and silicone punctal plug extraction after 2 months. The control group, 23 non-operative eyes, treated with lacrimal point dilator and lacrimal duct irrigation once a week for 2 months. Results No complication such as displacement or exfoliation of silicone punctual plug occurred in the observation group. The cure rate at 1,2 and 3 months after silicone punctal plug extraction were 100％, 90. 5％, 80. 9％,and the apparent improvement rate was 100％. In the control group, the diameter of puncta was unchanged after treatment, and the improvement rate was only 21. 7％. Conclusion Silicone punctal plug for the treatment of acquired lacrimal puncta stenosis is an effective and safe procedure.

Abstract:Purpose: To investigate gel stent implantation with and without intraoperative sustained-release mitomycin C (MMC SR) in a rabbit model for gel stent implantation, and to examine aqueous humor outflow (AHO) postimplantation. Methods: Four groups of rabbits were included. Group 1 was untreated (control). Groups 2, 3, and 4 received the gel stent without MMC, with MMC solution (subconjunctival injection), and with MMC SR (subconjunctival injection), respectively. Intraocular pressure (IOP) and AHO were assessed via tonometry and indocyanine green-based angiography, respectively. The main efficacy measure was change in IOP from baseline. Results: Following gel stent implantation, Groups 2, 3, and 4 maintained ≥20% IOP reduction (response) for a median duration of 1 week, 6.5 weeks, and 30 weeks, respectively. Angiography showed normal aqueous humor drainage (Group 1) beginning at the perilimbal trabecular plexus and continuing posteriorly to episcleral outflow vessels. Following implantation, drainage occurred preferentially and directly into the subconjunctival bleb. Conclusions: Gel stent implantation with MMC SR was most effective in achieving sustained, long-term IOP reduction in the rabbit model, compared with implantation with or without MMC solution. Bleb presence and the postimplantation aqueous angiography results indicated redirection of the AHO to the subconjunctival vasculature and presumed lymphatics, suggesting efficient glaucoma filtration to lower IOP in this model. This rabbit model and aqueous angiography may help refine understanding of the mechanism of action of minimally invasive glaucoma surgeries and ultimately translate to improved surgical devices and procedures for patients with glaucoma.
Clinical Effectiveness of Monocanalicular Silicone Intubation for Congenital Nasolacrimal Duct Obstruction Under Nasal Endoscopic Visualization of the Terminal End of the Obstructed Nasolacrimal Duct

Abstract:Purpose: To evaluate the cause of nasolacrimal duct obstruction through confirmation by nasal endoscopic findings of Hasner's valve and to report the success rate of monocanalicular silicone intubation (MCI) for the management of congenital nasolacrimal duct obstruction (CNLDO). Methods: Seventy-seven eyes of 56 patients with CNLDO underwent MCI under nasal endoscopic visualization of the terminal end of the obstructed nasolacrimal duct at the Korea University Ansan Hospital and Guro Hospital from October 2008 to March 2013. The following demographic information was analyzed: age, sex, endoscopic findings of Hasner's valve during operation, complications, and outcomes. The main outcome measures were disappearance of epiphora symptoms beginning the first 2 months after removal of the silicone tube. The silicone tube was removed under topical anesthesia in the office between 2 and 3 months, postoperatively. Results: The mean (± SD) age of the study population was 29.8 (± 26.9) months (range: 6 months to 12 years). Under nasal endoscopic view, 45 ducts (58.4%) had a thin membranous obstruction or had a simple stenotic opening of Hasner's valve. Seven ducts (9.1%) had thick obstructing membranes with probe passage under the lateral nasal mucosa. Twenty-three ducts (29.9%) showed the probe tip protruding through balloon-like nasal mucosa. Two ducts (2.6%) appeared to have probe protrusion through a stretchable valve. The overall success rate was 89.6% (69/77). The incidence of tube prolapse and tube loss was 18.2% (14 eyes) and 13.0% (10 eyes), respectively. No other complications were observed. Conclusions: Monocanalicular silicone intubation under nasal endoscopic visualization of the terminal end of the obstructed nasolacrimal duct can be an effective procedure for the management of CNLDO.
Antifibrotic role of low-dose mitomycin-c-induced cellular senescence in trabeculectomy models

Abstract:We assessed whether mitomycin-C (MMC) has different antifibrotic mechanisms in trabeculectomy wound healing.We identified 2 concentrations of MMC as "low-dose" by using WST-1 assay, Lactic dehydrogenase assay, and fluorescence-activated cell sorting flow cytometry. Senescence-associated β-galactosidase (SA-β-gal) and fibrotic gene expression was examined through immunocytochemistry, flow cytometry, real-time quantitative reverse transcription polymerase chain reaction, Western blotting, zymography, and modified scratch assay in vitro. In vivo, 0.1 mL of MMC or normal saline was injected to Tenon's capsule before trabeculectomy in a rabbit model. SA-β-gal expression, apoptotic cell death, and collagen deposition in sites treated and not treated with MMC were evaluated using terminal dUTP nick end labeling assay and histochemical staining. Bleb function and intraocular pressure (IOP) levels were examined 3, 7, 14, 21, 28, and 35 days after trabeculectomy.In vitro, human Tenon's fibroblast (HTF) senescence was confirmed by observing cell morphologic change, SA-β-gal accumulation, formation of senescence-associated heterochromatin, increased p16INK4a and p21CIP1/WAF1 expression, lower percentage of Ki-67-positive cells, and decreased COL1A1 release. Increased expression of α-SMA, COL1A1, and Smad2 signaling in TGF-β1-induced stress fibers were passivated in senescent HTFs. In addition, cellular migration enhanced by TGF-β1was inactivated. In vivo, histological examination indicated increased SA-β-gal accumulation, lower apoptosis ratios, and looser collagen deposition in sites treated with 0.2 μM MMC. Low-dose MMC-induced cellular senescence prolonged trabeculectomy bleb survival and reduced IOP levels in a rabbit model.Low-dose MMC-induced cellular senescence is involved in the antifibrotic mechanism of trabeculectomy wound healing.
