Abstract:With advances in radioactive particle implantation in clinical practice, Iodine-125 ( 125 I) seed brachytherapy has emerged as a promising treatment for cholangiocarcinoma (CCA), showing good prognosis; however, the underlying molecular mechanism of the therapeutic effect of 125 I seed is unclear. To study the effects of 125 I seed on the proliferation and apoptosis of CCA cells. CCA cell lines, RBE and HCCC-9810, were treated with reactive oxygen species (ROS) scavenger acetylcysteine (NAC) or the p53 functional inhibitor, pifithrin-α hydrobromide (PFTα). Cell counting kit-8 (CCK-8) assay, 5-bromo-2-deoxy-uridine (BrdU) staining, and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay and flow cytometry assay were performed to test the radiation-sensitivity of 125 I seed toward CCA cells at different radiation doses (0.4 mCi and 0.8 mCi). 2,7-dichlorofluorescein diacetate (DCF-DA) assay, real-time quantitative polymerase chain reaction (RT-qPCR), and western blot analysis were performed to assess the effect of 125 I seed on the ROS/p53 axis. A dose-dependent inhibitory effect of 125 I seeds on the proliferation of CCA cells was observed. The 125 I seed promoted apoptosis of CCA cells and induced the activation of the ROS/p53 pathway in a dose-dependent manner. NAC or PFTα treatment effectively reversed the stimulatory effect of 125 I seed on the proliferation of CCA cells. NAC or PFTα suppressed apoptosis and p53 protein expression induced by the 125 I seed. 125 I seed can inhibit cell growth mainly through the apoptotic pathway. The mechanism may involve the activation of p53 and its downstream apoptotic pathway by up-regulating the level of ROS in cells.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore the effects of iodine - 125 (125I) particles on the proliferation and apoptosis of cholangiocarcinoma (CCA) cells and their potential molecular mechanisms. Specifically, the researchers hope to experimentally verify whether 125I particles can inhibit the proliferation of CCA cells and promote their apoptosis by up - regulating the level of reactive oxygen species (ROS) and activating the p53 signaling pathway. This provides new experimental evidence and possible treatment strategies for the treatment of cholangiocarcinoma. ### Main research contents: 1. **Effects of 125I particles on the proliferation of CCA cells**: - The inhibitory effects of different doses (0.4 mCi and 0.8 mCi) of 125I particles on the proliferation of CCA cell lines (RBE and HCCC - 9810) were detected by CCK - 8 method and BrdU staining method. - The results showed that 125I particles inhibited the proliferation of CCA cells in a dose - dependent manner. 2. **Effects of 125I particles on the apoptosis of CCA cells**: - The induction effect of 125I particles on the apoptosis of CCA cells was detected by TUNEL staining and flow cytometry. - The results indicated that 125I particles promoted the apoptosis of CCA cells in a dose - dependent manner, and the effect of the high dose (0.8 mCi) was more significant. 3. **Activation effect of 125I particles on the ROS/p53 pathway**: - The changes in the ROS level in CCA cells after treatment with 125I particles were detected by using DCFH - DA probe. - The changes in the expression of p53 gene and protein were detected by RT - qPCR and Western blot analysis. - The results showed that after treatment with 125I particles, the ROS level increased significantly, and the expression of p53 gene and protein also increased significantly. 4. **Role of ROS in 125I - particle - induced apoptosis**: - CCA cells were pretreated with ROS scavenger NAC to observe its effects on 125I - particle - induced apoptosis and proliferation inhibition. - The results showed that NAC could effectively reverse the induction effect of 125I particles on CCA cell apoptosis and the inhibition effect on proliferation, indicating that ROS plays a key role in this process. 5. **Role of p53 in 125I - particle - induced apoptosis**: - CCA cells were pretreated with p53 function inhibitor PFTα to observe its effects on 125I - particle - induced apoptosis and proliferation inhibition. - The results showed that PFTα could partially reverse the induction effect of 125I particles on CCA cell apoptosis and the inhibition effect on proliferation, further confirming the importance of p53 in this process. ### Research conclusions: 125I particles inhibit the proliferation of CCA cells and promote their apoptosis in a dose - dependent manner by up - regulating the ROS level and activating the p53 signaling pathway. These findings provide new experimental evidence and potential treatment strategies for the treatment of cholangiocarcinoma.
