Abstract:A greener and straightforward synthesis of potent antimicrobial hybrid molecules bearing three pharmacophores has been demonstrated with excellent yields. The synthesis has been achieved by a reaction using microwave and ultrasonic irradiation methods in the presence of a green catalyst and solvent. Molecular docking and ADMET revealed that the synthesized hybrids may constitute a better platform for antimicrobial drug development. To achieve environmentally benign synthesis and develop pharmacologically active compounds, a library of novel thiophene‐clubbed pyrazole‐1,2,3‐triazole hybrids was designed and successfully synthesized using L‐ascorbic acid as a green acid catalyst and ethanol as a green solvent. The reaction was carried out using microwave and ultrasonic irradiation methods to achieve a highly efficient reaction route. All title compounds were characterized and evaluated for their potential in vitro antimicrobial activity using Ciprofloxacin and Nystatin as standard drugs, in which compounds with chloro and naphthyl substitutions exhibited excellent antibacterial activity at concentrations of 25 μg/mL and compounds with methyl and methoxy substitutions exhibited excellent antifungal activity at concentrations of 100 μg/mL. Additionally, the molecular docking study showed that all compounds examined had excellent binding energies ranging from −9.3 to −10.3 kcal/mol, and naphthyl substitution displayed the highest binding energy (−10.3 kcal/mol) on the target protein of E. coli DNA gyrase subunit B. Furthermore, the final moieties were evaluated for in silico ADMET prediction to examine their drug‐likeness profile and toxic effects. The present study revealed that this structural information will be helpful in future antimicrobial drug design and development.

Abstract:Background: The increasing prevalence of antimicrobial resistant highlights the urgent need for the new therapeutic agents. This study aimed to design and synthesize fused tricyclic benzimidazole–thiazinone derivatives (CS1–CS10) through a convenient method and evaluate their antimicrobial activity against various microorganisms. Methods: A series of fused tricyclic benzimidazole–thiazinone derivatives was rationally designed and synthesized in one pot by the reaction between trans substituted acrylic acids and 1H-benzo[d]imidazole-2-thiol using coupling reagent TBTU (2-(1H-benzotriazol-1-yl)-1,1,3,3-tetramethyluronium tetrafluoroborate). The structure of these compounds was confirmed through various spectroscopic techniques like IR, 1H and 13C NMR, the DEPT and 2D-HMQC NMR techniques were also performed to confirm the relation of both carbon and proton. Further, the compounds were in vitro evaluated for their effectiveness against the Candida species and a panel of standard bacterial isolates. Results: The synthesized compounds showed moderate antimicrobial activity. Among all of the compounds, CS4 exhibited potent inhibition against Pseudomonas aeruginosa and Escherichia coli at 256 and 512 μg/mL concentrations, respectively. Additional research indicated that compound CS4 demonstrated a synergistic effect after combining with the standard antibacterial drug ciprofloxacin. Conclusions: These results suggest that CS4 is the best-synthesized antibacterial agent particularly in combination therapies. These findings highlight its promise for further development as a novel antibacterial agent.

Abstract:A library of some novel 1,4-disubstituted-1,2,3-triazole-thiosemicarbazone hybrid molecules were designed and synthesized from (4-Prop-2-ynyloxy-benzylidene)-thiosemicarbazone and aryl azides under Cu(I)-catalyzed cycloaddition reaction. All newly synthesized [4-(1-Benzyl-1H-[1,2,3]triazol-4-ylmethoxy)-benzylidene] -thiosemicarbazone hybrid molecules were efficiently characterized by IR, 1H NMR, 13C NMR, HRMS and structure of alkynes 3 & 12 were finally supported by X-ray crystallographic data. Compounds 5c, 5d, 9c, 9d 13c and 13d demonstrated excellent potency results for B. Subtilis and P. Aeruginosa bacterial strains with MIC values 0.0141, 0.0152, 0.0562, 0.0608, 0.0141, 0.0608, 0.0141, 0.0304, 0.0281, 0.0304, 0.0281, 0.0304, respectively as compared to reference drug Ciprofloxacin. Abstract:A series of new benzimidazole derivatives containing 1,3,4-thiadiazole, 1,3,4-oxadiazole, thiazolidine, or 1,2,4-triazole ring have been synthesized. All newly synthesized compounds were characterized by IR, 1 H and 13 C NMR, and LC/MS spectroscopy and were tested for their antibacterial and antioxidant activities. Among the synthesized compounds, 2-{2-[2-(cyanomethyl)-1 H -benzimidazol-1-yl]acetyl}- N -phenylhydrazinecarboxamideshowed the best antioxidant activity, and N ′-(4-bromobenzylidene)-2-[2-(cyanomethyl)-1 H -benzimidazol-1-yl]acetohydrazide exhibited the lowest minimum inhibitory concentration against Enterobacter aerogenes .

Abstract:1,2,4‐Triazolo/benzimidazolo‐pyrimidine‐linked 1‐benzyl‐4‐[(p‐tolyloxy)methyl]‐1,2,3‐triazole derivatives were synthesized via facile tetrabutylammonium bromide‐catalyzed green multicomponent synthesis. The resulting compounds showed potent antibacterial, antitubercular, and antimalarial activities. The multifaceted biological potential of these dihydropyrimidine derivatives represents their importance in medicinal chemistry. This study reports the design and synthesis of novel 1,2,4‐triazolo/benzimidazolo‐pyrimidine linked 1‐benzyl‐4‐[(p‐tolyloxy)methyl]‐1,2,3‐triazole derivatives as potent antimicrobial agents according to their in vitro antibacterial, antifungal, antitubercular as well as antimalarial activities. An efficient, ecologically benign, and facile multicomponent synthesis was employed to synthesize these derivatives. The synthesis is accelerated with the mild and eco‐friendly organocatalyst tetrabutylammonium bromide, providing a yield of 82%–96% within the short reaction time of 0.5–1.5 h. Compared with the MIC values of ciprofloxacin and ampicillin on the respective strains, compound d2 showed better activity against Escherichia coli and Streptococcus pyogenes and compound d8 showed better MIC against Staphylococcus aureus. Additionally, compounds d3, d4, and d5 showed potent MIC values against Pseudomonas aeruginosa. All triazolo‐pyrimidine derivatives d1–d8 showed potent inhibitory action against Gram‐positive strains. Compound e3 showed good potency against Mycobacterium tuberculosis H37Rv. The IC50 values of d3 and e2 indicated better activity against Plasmodium falciparum. Collectively, these derivatives depict potent multifaceted activity and provide promising access for further antimicrobial and antimalarial investigations.

Abstract:A series of 5-aryl-3-(4-oxo-2-phenylquinazolin-3(4H)-yl)-2-(pyridin-4-yl)thiazolidin-4-ones were synthesized and evaluated for their antibacterial and antifungal activities. Various spectral techniques e.g., IR, 1H NMR, 13C NMR and Mass spectrometry were used to determine structure of novel synthesized compounds. The title compounds showed good to excellent inhibition potency for respective Gram-positive bacterial strains and Gram-negative bacterial strains. These compounds exhibited a broad spectrum of inhibitory activity. Molecular docking studies against microbial DNA gyrase sub unit B could provide valuable insights into the binding affinity of these molecules and their plausible mechanism of antimicrobial action. Compounds 5a, 5d, 5f and 5h, 5i, 5j, 5o exhibited excellent activity against bacterial and fungal strains respectively.Graphical

Abstract:A series of 1,3‐thiazolidine‐ amide derivatives were synthesized via two different methods (microwave irradiation and conventional). These derivatives exhibited in vitro good to excellent antimicrobial activity. Molecular docking study performed on the crucial antifungal target 4WMZ. Quantitative structure toxicity relationship (QSTR) study pointed out the slight toxic nature of compounds. In silico ADME prediction of these compounds revealed the good drug‐likeness nature and followed Lipinski's rule of five. In this article, we described synthesis of 1,3‐thiazolidine ‐ amide hybrid derivatives by two different methods from (S)‐2‐amino‐3‐(4‐(benzyloxy)phenyl)‐1‐(thiazolidin‐3‐yl)propan‐1‐one (4). In first method, catalytic amidation was carried out under microwave irradiation using Ceric Ammonium Nitrate (CAN) as a green catalyst where as in second conventional method; CDI was used as a significant coupling reagent to optimize reaction condition and yield of product. All new compounds were well characterized by 1H NMR, 13C NMR, IR and ESMS spectral techniques and evaluated in vitro antibacterial and antifungal activity. The molecular docking study revealed that the designed compounds snuggly fit in the active site of 4WMZ protein. In addition, the QSTR study of new compounds were carried out with the help of Toxicity Estimation Software Tool (T.E.S.T). The results showed slight toxic nature of new compounds. In‐silico ADME studies significant values of pharmacokinetic parameters and demonstrated good drug like characteristics based on Lipinski's rule of five.

Abstract:Previously unknown hybrid compounds comprising a thiazole and a triazole moieties were synthesized with excellent yields and characterized by IR and NMR spectroscopy, mass spectrometry, and elemental analysis. The antibacterial activity of the produced hybrid compounds was tested against bacterial ( E. coli , S. aureus ) and fungal ( C. albicans , A. niger ) strains, and the minimal inhibitory concentrations were calculated. The majority of the newly synthesized hybrids showed a moderate to an excellent antibacterial activity.

Synthesis and in Silico Studies of Some New 1,2,3-Triazolyltetrazole Bearing Indazole Derivatives as Potent Antimicrobial Agents

Abstract:1,2,3-Triazole and tetrazole derivatives bearing pyrrolidines are found to exhibit notable biological activity and have become useful scaffolds in medicinal chemistry for application in lead discovery and optimization. Novel indazole bearing 1,2,3-triazolyltetrazoles were designed as potential antimicrobial candidates. The structure of duel heterocyclics was validated by a spectroscopic technique of infrared (IR), nuclear magnetic resonance (1 H and 13 C NMR), and mass spectral data. Compounds 4b, 4c, 4d, and 4h were found to have a stronger antibacterial effect against Gram-positive (S. aureus, B. subtilis, M. Luteus) and Gram-negative (E. coli, P. aeruginosa) microorganisms with MICs ranging from 5±0.03-18±0.02 μM, respectively. Moreover, scaffolds 4a, 4h showed potent antifungal activity against A. flavus, M. gypsuem strains with MIC values of 10±0.02, 11±0.01 μM, which are similar activity that of the standard Itraconazole (MIC=8±0.02, 10±0.01 μM). The binding mode for compound 4 inside the catalytic pocket of S. aureus complexed with nicotinamide adenine dinucleotide phosphate and trimethoprim and produced a network of hydrophobic and hydrophilic interactions (3FRE). From in silico results, 4b demonstrated highly stable hydrogen binding amino acids Leu62(X) [N18…O, 2.47 Å], Arg44(X) [N17…N, 3.11 Å], Thr96(X) [N10…OG1, 3.05 Å], Gly94(X) [F7…N, 2.82 Å], and Gly43(X) [F7…N, 2.90 Å], which are plays a crucial role in ensuring efficient binding of the ligand in a crystal structure of antibacterial receptor. Furthermore, the physicochemical and ADME filtration molecular properties, estimation of toxicity, and bioactivity scores of these novel scaffolds were evaluated by using SwissADME and ADMETlab2.0 online protocols. Thus, the significant antimicrobial activity of indazole linked to duel heterocyclic compounds can be used for development of new antimicrobial agents with further modifications.

Abstract:The current research contains synthesis, DNA-interaction analysis, in vitro anti-oxidant evaluation, and antibacterial assay of triazole-azomethine hybrids. Consequently, the synthesized hybrids were subjected to molecular docking (MD), density functional theory (DFT) guided experimental DNA-interaction studies, in vitro DPPH quenching, and bactericidal assay. Subsequently, the screened hybrids displayed higher DNA binding abilities in DNA-interaction studies. Notably, the hybrids bearing 4-hydroxy-), 2-hydroxy-4-methoxy-), and 4-bromo-) groups displayed higher binding ability in the DNA grooves with respective interaction strengths in the range of 3.43–6.43 × 10 –3 M −1 higher than the reference Cis-platin (3.142 × 10 –3 M −1 ) augmented by MD and DFT analysis. Similarly, DFT-guided UV–visible spectroscopy-based anti-oxidant assay revealed potent oxidative nature of the screened hybrids. DPPH free radical scavenging ability in terms of IC 50 values falls within a narrow range of 22.30–34.22 μM. The screened hybrids bearing 4-hydroxy-), 3,4-dihydroxy-), and 2,3,4-trihydroxy-) groups display highest DPPH scavenging potential with respective IC 50 values of 34.22 μM, 32.11 μM, and 29.30 μM. Moreover, the synthesized hybrids were evaluated against gram-negative and positive bacterial strains, indicating their inhibitory potential as zone of inhibition (ZOI). Briefly, the screened hybrid bearing 2,3,4-trihydroxy-) group displayed ZOI (21 mm) against Pseudomonas aeruginosa (gram-negative strain) higher than Benzylpenicillin (18 mm). The remaining hybrids possess inhibitory potential comparable to the reference. Conversely, the screened hybrids showed varied responses against Staphylococcus aureus (gram-positive strain), with highest ZOI (16 mm) displayed by the hybrid with 4-hydroxy-) group. Thus, the obtained ZOI suggests the potent hybrids to be explored further as effective motifs against gram-negative and positive bacterial strains.

Abstract:Background: Antimicrobial resistance has become a critical health concern, and quorum-sensing exacerbates the resistance by facilitating cell-to-cell communication within the microbial community, leading to severe pathogenic outbreaks. Methods & results: Novel 1-(2-((5 H -[1,2,4]-triazino[5,6- b ]indol-3-yl)thio)acetyl)indoline-2,3-diones were synthesized. The title compounds exhibit outstanding anti-quorum-sensing efficacy, and compound 7g demonstrated the maximum proficiency (IC 50 = 0.0504 μg/ml). The hybrids displayed potent antioxidant action, and compound 7c showed the highest antioxidant ability (IC 50 = 40.71 μg/ml). Molecular docking of the isatin hybrids against DNA gyrase and quorum-sensing receptor CviR validated the observed in vitro findings. The befitting pharmacokinetic profile of the synthesized drug candidates was ascertained through absorption, distribution, metabolism, excretion and toxicity screening. Conclusion: The remarkable biocompetence of the synthesized triazinoindoles may help to combat drug-resistant infections.

Abstract:Thiazole derivatives ( 7a-f and 9a-i ) were synthesized in good yields (up to 98%). The synthetic protocol is achieved through a two-step reaction. The synthesized candidates were then evaluated for their antimicrobial efficacies against gram-positive and gram-negative bacteria. 4-methyl-2-(2-(3-(pentyloxy)benzylidene)hydrazinyl)-5-(p-tolyldiazenyl)thiazole showed significant antimicrobial activity against Staphylococcus aureus (ATCC: 13565) compared to ampicillin. On the other hand, 2-(2-(3-(hexyloxy)benzylidene)hydrazinyl)-4-(4-methoxyphenyl)thiazole shows potent activity against Klebsiella pneumonia (ATCC: 10031) compared to gentamicin as a standard antibiotic. The ADME profile of the prepared compounds indicated favorable pharmacological behaviors. Additionally, docking and in-silico ADMET analyses were performed for the promising derivatives and the results revealed strong antimicrobial activity. DFT calculations were performed, which provided additional insight into the thiazoles' structure and formation.

Abstract:A base-promoted palladium-catalyzed cascade reaction is described to access the trifluoromethylated dipyridodiazepinone derivatives in an aqueous system (1,4-dioxane-H2O). This methodology uses simple chemicals, broad substrate scope, is waste minimized (E-factor = 0.3-0.9) and produces 11-CF3-tethered dipyridiodiazepinone derivatives in good to excellent yield. All the synthesized analogues were preliminary examined for antibacterial activity against E. coli and S. aureus and compared to the reference drugs. Further, inhibition of peptide deformylase enzyme and antibiofilm studies was performed and compound 5i exhibited the best inhibitory effect than the other analogues. Furthermore, these analogues were in silico analysed viz. molecular docking, molecular simulation, drug-likeness, physicochemical and ADMET studies. Results from biological evaluation and computational studies revealed that compound 5i could be used as a lead molecular structure for the development of novel antibacterial agents. In conclusion, green metrics evaluation of the defined protocol provides advantages in the synthesis of biologically active compounds.

Abstract:We have developed an efficient divergent strategy to access various functionalized 5‐substituted 1,2,4‐oxadiazole‐3‐yl benzoxazole derivatives in good to excellent yields by one pot reaction involving nucleophilic addition reaction of amidoxime with aryl or alkyl acid chloride, followed by cyclodehydration in presence of base. A wide range of functional groups were tolerated during this process. Discovery of novel antimicrobial drugs is one of the hottest topics in the field of medicinal chemistry. An efficient and catalyst free synthesis of novel fluorinated 5‐substituted 1,2,4‐oxadiazole‐3‐yl benzoxazole derivatives is described. A series of novel compounds have been synthesized and characterized by different spectroscopic techniques such as FTIR, 1H NMR, 13C NMR, and LCMS. When tested for antibacterial activity, the synthesized benzoxazole derivatives exhibited outstanding to good activities in comparison to the conventional medications. Moreover, the theoretical forecasts grounded in molecular docking against microbial DNA gyrase may offer a glimpse into the conceivable mode of operation and establish a connection between the detected antimicrobial efficacy and the binding affinity, illuminating particular thermodynamic (bonded and nonbonded) interactions that regulate the activity. The produced compounds also exhibited promising qualities to develop into viable oral medication candidates when their absorption, distribution, metabolism, and excretion were examined. In summary, the research proposes that these novel compounds have emerged as interesting contender for the development of next generation antimicrobial drugs. Our protocol demonstrates broad substrate scope, dramatic decrease of synthesis time, and energy consumption with excellent yields.

Abstract:Antimicrobial Resistance (AMR) due to non-responding viruses, fungi, bacteria and parasites leads to discovery of new antimicrobial medicines which can control the risk of disease spread, severe illness, disability and death. Therefore, compounds such as 1-(Chloromethyl)-1H-Benzotriazole, 1; 1-((1-H-benzo[d][1,2,3]triazol-1-yl)methyl)phenyl hydrazine, 2; 1-((1-H-benzo[d][1,2,3]triazol-1-yl)methyl)hydrazine, 3; and N-(benzo[e][1,2,4]triazin-4(3-H)-ylmethylbenzenamine, 4 were designed, and synthesized through conventional and microwave-assisted methods. All of these novel benzotriazoles were explored through in-vitro antimicrobial studies and in silico studies. Antimicrobial activity was carried out against bacterial strains Escherichia coli, Bacillus subtilis, and fungal strains Aspergillus niger and Candida albicans at concentrations 5, 10 and 15 mg/ml. In silico studies was carried out with 4CAW: Aspergillus fumigatus N-myristoyl transferase in complex with myristoyl CoA and a pyrazole sulphonamide ligand. Our antimicrobial and molecular docking studies revealed that all of the derivatives showed promising activity, moreover molecular docking gave significant values of ligand posed energy and docking run elapsed time which further endorsed the astonishing characteristic of benzotriazole derivatives esp. N-(benzo[e]a[1,2,4] triazin-4(3-H)-ylmethylbenzenamine for biological and therapeutic leads.
Design, one-pot synthesis, in silico ADMET prediction and molecular docking of novel triazolyl thiadiazine and thiazole derivatives with evaluation of antimicrobial, antioxidant and antibiofilm inhibition activity

Abstract:A facile one pot multi-component synthesis of novel 1,3,4-thiadiazine and thiazole derivatives from the reaction of (methyl-1-(p-tolyl)-1H-1,2,3-triazol-4-yl)ethan-1-one, thiocarbohydrazide/or thiosemicarbazide, and aromatic aldehydes. The synthesized compounds were evaluated for antimicrobial, antibiofilm, and antioxidant activity. The results revealed the marked potency as antimicrobial agents. Thiazole 6e has the highest antimicrobial activity. Furthermore, most compounds inhibited biofilms produced by Pseudomonas aeruginosa. Promising thiazole 6c and thiadiazine derivatives (4c and 4 g) exhibited superior antioxidant activity and remarkable scavenging activity with DPPH, (86.99 ± 1.1, 81.54 ± 3.4 and 78.25 ± 1.8) and ABTS (80.06 ± 0.12, 62.29 ± 0.31 and 44.38 ± 0.20) respectively. The molecular docking simulation showed lower binding energy with different types of interaction at the active site of Dihydropteroate synthase, Sortase A, LasR, and Penicillin-binding proteins pockets indicating that these compounds could inhibit the enzyme and cause promising antimicrobial effects. Also, Molecular docking to the active sites of Peroxidase enzyme as an antioxidant receptor revealed that thiadiazine derivatives (4c and 4d) displayed minimal binding energy and have a good affinity toward the active pocket of each enzyme.Graphical Abstract

Abstract:Optimization of green synthesis for a novel pyrazolo[1,5- a ]pyrimidines 3a-i, 7a-f, triazolo[4,3- a ]pyrimidine 8 and benzo[ Citation 4 , Citation 5 ]imidazo[1,2- a ]pyrimidine 9 were synthesized from the reaction of 1-(4-aminophenyl)-3-(thiophen-2-yl)prop-2-en-1-one 1 with 4-arylazo-3,5-diaminopyrazoles 2a-i, 5-amino- N -aryl-3-(phenylamino)-1 H -pyrazole-4-carboxamides 6a-f, 3-amino-1,2,4-triazole and 2-aminobenzoimidazole, respectively. A simple approach was used under various conditions to achieve the best conditions for the synthesis. Some new compounds were evaluated for antibacterial activity in vitro, and when compared to traditional antibiotics, they showed good efficacy. The MIC value for pyrazolo[1,5- a ]pyrimidine 3d , 3i and 7d in Escherichia coli, Klebsiella pneumonia and Acinetobacter baumannii equal 31.25 μg/L. Better binding patterns were revealed by molecular docking studies for 3d , 3i and 7d with the active site of Escherichia coli DHFR co-crystalized with the active site of trimethoprim [PDB coded 6XG5]. The physicochemical characteristics and toxicity hazards by Osiris methodology besides Swiss ADME predications were performed for all the synthesized compounds exhibiting better biological and antibacterial characters.

Abstract:Herein, we report design, synthesis and characterization of a new library of 7‐azaindole N‐ethyl linked 1,2,3‐triazoles containing ethylene as a spacer unit, and evaluation of all the synthesized compounds for their antimicrobial properties. Antibacterial potential was checked against two Gram positive (B. subtilis and S. aureus) and two Gram negative (E. coli and P. aeruginosa) bacterial strains while antifungal potential was assayed against two fungal strains (C. albicans and A. niger). All the tested compounds showed satisfactory antibacterial potency in comparison to reference drug ciprofloxacin with MIC values ranging from 0.0108 to 0.0432 μmol/mL. Interestingly, except two, all the target compounds showed better antifungal property as compared to the reference drug fluconazole with MIC values less than 0.0408 μmol/mL. One of the compounds exhibited two‐fold better antifungal potential in comparison to fluconazole. Furthermore, in‐silico ADMET and DFT studies reported drug likeness behavior and chemical reactivity parameters, respectively. The cytotoxicity results on substrate azide 3 and most potent 1,2,3‐triazoles (5d and 5l) were found to be non‐toxic.

Abstract:Abstract Background Thiazole-based Schiff base compounds display significant pharmacological potential with an ability to modulate the activity of many enzymes involved in metabolism. They also demonstrated to have antibacterial, antifungal, anti-inflammatory, antioxidant, and antiproliferative activities. In this work, conventional and green approaches using ZnO nanoparticles as catalyst were used to synthesize thiazole-based Schiff base compounds. Results Among the synthesized compounds, 11 showed good activities towards Gram-negative E. coli (14.40 ± 0.04), and Gram-positive S. aureus (15.00 ± 0.01 mm), respectively, at 200 μg/mL compared to amoxicillin (18.00 ± 0.01 mm and 17.00 ± 0.04). Compounds 7 and 9 displayed better DPPH radical scavenging potency with IC 50 values of 3.6 and 3.65 μg/mL, respectively, compared to ascorbic acid (3.91 μg/mL). The binding affinity of the synthesized compounds against DNA gyrase B is within − 7.5 to − 6.0 kcal/mol, compared to amoxicillin (− 6.1 kcal/mol). The highest binding affinity was achieved for compounds 9 and 11 (− 6.9, and − 7.5 kcal/mol, respectively). Compounds 7 and 9 displayed the binding affinity values of − 5.3 to − 5.2 kcal/mol, respectively, against human peroxiredoxin 5. These values are higher than that of ascorbic acid (− 4.9 kcal/mol), in good agreement with the experimental findings. In silico cytotoxicity predictions showed that the synthesized compounds Lethal Dose (LD 50 ) value are class three (50 ≤ LD 50 ≤ 300), indicating that the compounds could be categorized under toxic class. Density functional theory calculations showed that the synthesized compounds have small band gap energies ranging from 1.795 to 2.242 eV, demonstrating that the compounds have good reactivities. Conclusions The synthesized compounds showed moderate to high antibacterial and antioxidant activities. The in vitro antibacterial activity and molecular docking analysis showed that compound 11 is a promising antibacterial therapeutics agent against E. coli, whereas compounds 7 and 9 were found to be promising antioxidant agents. Moreover, the green synthesis approach using ZnO nanoparticles as catalyst was found to be a very efficient method to synthesize biologically active compounds compared to the conventional method.

Abstract:Chalcone and triazole scaffolds have demonstrated a crucial role in the advancement of science and technology. Due to their significance, research has proceeded on the design and development of novel benzooxepine connected to 1,2,3-triazolyl chalcone structures. The new chalcone derivatives produced by benzooxepine triazole methyl ketone 2 and different aromatic carbonyl compounds 3 are discussed in this paper. All prepared compounds have well-established structures to a variety of spectral approaches, including mass analysis, 1H NMR, 13C NMR, and IR. Among the tested compounds, hybrids 4c, 4d, 4i, and 4k exhibited exceptional antibacterial susceptibilities with MIC range of 3.59-10.30 μM against the tested S. aureus strain. Compounds 4c, 4d displayed superior antifungal activity against F. oxysporum with MIC 3.25, 4.89 μM, when compared to fluconazole (MIC = 3.83 μM) respectively. On the other hand, analogues 4d, 4f, and 4k demonstrated equivalent antitubercular action against H37Rv strain with MIC range of 2.16-4.90 μM. The capacity of ligand 4f to form a stable compound on the active site of CYP51 from M. tuberculosis (1EA1) was confirmed by docking studies using amino acids Leu321(A), Pro77(A), Phe83(A), Lys74(A), Tyr76(A), Ala73(A), Arg96(A), Thr80(A), Met79(A), His259(A), and Gln72(A). Additionally, the chalcone‒1,2,3‒triazole hybrids ADME (absorption, distribution, metabolism, and excretion), characteristics of molecules, estimations of toxicity, and bioactivity parameters were assessed.
