Abstract:After years of intensive research and numerous important observations, our understanding of the early stages of crystallization is still limited due to the complexity of the underlying processes and their elusive character. In the present work, we provide a detailed view on the nucleation of calcium sulfate mineralization – an abundant mineral with broad use in construction industry – in aqueous systems at ambient conditions. As experimental basis, a co-titration procedure with potentiometric, turbidimetric and conductometric detection was developed, allowing solution speciation and the formation of crystallization precursors to be monitored quantitatively as the level of nominal (super)saturation gradually increases. The nature and spatiotemporal evolution of these precursors was further elucidated by time-resolved small-angle X-ray scattering (SAXS) and analytical ultracentrifugation (AUC) experiments, complemented by cryogenic transmission electron microscopy (cryo-TEM) as a direct imaging technique. The results reveal how ions associate into nanometric primary species, which subsequently aggregate and develop anisotropic order by intrinsic structural reorganization. Our observations challenge the common understanding of fundamental notions such as the nucleation barrier or the meaning of supersaturation, with broad implications for mineralization phenomena in general and the formation of calcium sulfate in geochemical settings and industrial applications in particular.

Abstract:Crystallisation is a widely used separation step for both isolation and purification of products, for a wide range of chemical processes. Nevertheless, there remains a great challenge in controlling the onset of crystallisation — nucleation. Heterogeneous nucleation has been investigated over the last three decades to determine physico-chemical relationships between nucleant and solute properties, which may be key for controlling nucleation. This can lead to rational design of nucleants facilitating selective crystallisation of particles with desired properties. In this opinion, a review on recent advances in developing mechanistic understanding of the role of interfacial properties, mainly nano-scale surface topography, surface chemistry and the combination of both features, in controlling nucleation is presented. An outlook for the development of next generation nucleants and the potential application areas are presented.
Investigating the early stages of mineral precipitation by potentiometric titration and analytical ultracentrifugation

Abstract:Despite the importance of crystallization for various areas of research, our understanding of the early stages of the mineral precipitation from solution and of the actual mechanism of nucleation is still rather limited. Indeed, detailed insights into the processes underlying nucleation may enable a systematic development of novel strategies for controlling mineralization, which is highly relevant for fields ranging from materials chemistry to medicine. In this work, we describe experimental aspects of a quantitative assay, which relies on pH titrations combined with in situ metal ion potentiometry and conductivity measurements. The assay has originally been designed to study the crystallization of calcium carbonate, one of the most abundant biominerals. However, the developed procedures can also be readily applied to any compound containing cations for which ion-selective electrodes are available. Besides the possibility to quantitatively assess ion association prior to nucleation and to directly determine thermodynamic solubility products of precipitated phases, the main advantage of the crystallization assay is the unambiguous identification of the different stages of precipitation (i.e., prenucleation, nucleation, and early postnucleation) and the characterization of the multiple effects of additives. Furthermore, the experiments permit targeted access to distinct precursor species and intermediate stages, which thus can be analyzed by additional methods such as cryo-electron microscopy or analytical ultracentrifugation (AUC). Regarding ion association in solution, AUC detects entities significantly larger than simple ion pairs, so-called prenucleation clusters. Sedimentation coefficient values and distributions obtained for the calcium carbonate system are discussed in light of recent insights into the structural nature of prenucleation clusters.

Abstract:Crystallization is one of the most important phase transformations of first order. In the case of metals and alloys, the liquid phase is the parent phase of materials production. The conditions of the crystallization process control the as-solidified material in its chemical and physical properties. Nucleation initiates the crystallization of a liquid. It selects the crystallographic phase, stable or meta-stable. Its detailed knowledge is therefore mandatory for the design of materials. We present techniques of containerless processing for nucleation studies of metals and alloys. We demonstrate the power of these methods for crystal nucleation of stable solids but in particular also for investigations of crystal nucleation of metastable solids at extreme undercooling. This concerns the issue of heterogeneous versus homogeneous nucleation and non-equilibrium conditions. The results are analyzed within classical nucleation theory, where the activation energy of homogeneous nucleation depends on the interfacial energy and the difference of Gibbs free energies of solid and liquid. The interfacial energy acts as barrier for the nucleation process. Its experimental determination is difficult in the case of metals. In the second part of this work we therefore explore the use of colloidal suspensions as models for the crystallization process. Their nucleation process is observed in situ by optical techniques and ultra-small angle X-ray diffraction using high intensity synchrotron radiation. It allows an unambiguous discrimination of homogeneous and heterogeneous nucleation as well as the determination of the interfacial free energy of the solid-liquid interface. Our results are used to construct Turnbull plots of colloids, which are discussed in relation to Turnbull plots of metals and support the hypothesis that colloids are useful model systems to investigate crystal nucleation.

Amorphous aggregates with a very wide size distribution play a central role in crystal nucleation

Abstract:There is mounting evidence that crystal nucleation from supersaturated solution involves the formation and reorganization of prenucleation clusters, contradicting classical nucleation theory. Here, a range of amino acids and peptides is investigated using light scattering, mass spectrometry, and in situ terahertz Raman spectroscopy, showing that the presence of amorphous aggregates is a general phenomenon in supersaturated solutions. Significantly, these aggregates are found on a vast range of length scales from dimers to 30-mers to the nanometer and even micrometer scale, suggesting a continuous distribution throughout this range. Larger amorphous aggregates are sites of spontaneous crystal nucleation and act as intermediates for laser-induced crystal nucleation. These results are shown to be consistent with a nonclassical nucleation model in which barrierless (homogeneous) nucleation of amorphous aggregates is followed by the nucleation of crystals from solute-enriched aggregates. This provides a novel perspective on crystal nucleation.

Abstract:Nucleation and growth of solids from solutions impacts many natural processes and are fundamental to applications in materials engineering and medicine. For a crystalline solid, the nucleus is a nanoscale cluster of ordered atoms, which forms through mechanisms that are still poorly understood. These mechanisms have important consequences on the morphology and nucleation rates of the resultant crystals but it is unclear whether a nucleus forms spontaneously from solution in a single step or through multiple steps. Using in-situ electron microscopy, we observe and quantify how gold and silver nanocrystals nucleate from a supersaturated aqueous gold and silver solution in three distinct steps: (I) spinodal decomposition into solute-rich and solute-poor liquid phases, (II) nucleation of amorphous gold nanoclusters within the gold-rich liquid phase, followed by (III) crystallization of these amorphous clusters. Our ab-initio calculations on gold nucleation suggest that these steps might be associated with strong gold-gold atom coupling and water-mediated metastable gold complexes. The understanding of intermediate steps in nuclei formation has important implications for the formation and growth of both crystalline and amorphous materials.

Abstract:Nucleation is the key step of crystallization and is also the most difficult part to observe, as it evolves in very short temporal and spatial resolution. Conflicts between the features of classical nucleation theory and the observed nucleation behaviors inspire hypotheses of nonclassical nucleation mechanisms. In this Review, experimental evidence and molecular dynamics simulations involving the structure and formation mechanisms of a series of precursors and intermediate species, such as prenucleation clusters, liquid precursors, and amorphous precursors that in some cases coexist in a single system and successively transform into one another, are summarized to advance the understanding on the nucleation mechanisms of crystalline phases. What’s more, recent progress of building two-dimensional materials avoiding the nucleation barrier and accommodating nonclassical nucleation within the classical nucleation framework is outlined, which provides a new perspective on crystal nucleation: a classical view on nonclassical nucleation.

Abstract:A fundamental assumption of the classical theories of crystal nucleation is that the individual molecules from the "old" phase associate to an emerging nucleus individually and sequentially. Numerous recent studies of crystal nucleation in solution have revealed nonclassical pathways, whereby crystal nuclei are hosted and fed by amorphous clusters pre-formed in the solution. A sizable knowledge gap has persisted, however, in the definition of the molecular-level parameters that direct a solute towards classical or nonclassical nucleation. Here we construct a suspension of colloid particles of hydrodynamic diameter 1.1 μm and monitor their individual motions towards a quasi-two-dimensional crystal by scanning confocal microscopy. We combine electrostatic repulsion and polymer-induced attraction to obtain a simple isotropic pair interaction potential with a single attractive minimum of tunable depth between 1.2 kBT and 2.7 kBT . We find that even the smallest aggregates that form in this system structure as hexagonal two-dimensional crystals and grow and mature by the association and exchange of single particles from the solution, signature behaviors during classical nucleation. The particles in the suspension equilibrate with those in the clusters and the volume fractions of suspensions at equilibrium correspond to straightforward thermodynamic predictions based on depth of the interparticle attraction. These results demonstrate that classical nucleation is selected by particles interacting with a minimal potential and present a benchmark for future modifications of the molecular interactions that may induce nonclassical nucleation.

Abstract:The interplay between densification and positional ordering during the process of crystal nucleation is a greatly investigated topic. Even for the simplest colloidal model -- hard spheres -- there has been much debate regarding the potential foreshadowing of nucleation by significant fluctuations in either local density or local structure. Considering anisotropic particles instead of spheres adds a third degree of freedom to the self-organization process of crystal nucleation: orientational ordering. Here, we investigate the crystal nucleation of hard triangular prisms. Using Monte Carlo simulations, we first carefully determine the crystal-fluid coexistence values and calculate the nucleation barriers for two degrees of supersaturation. Next, we use brute force simulations to obtain a large set of spontaneous nucleation events. By studying the time evolution of the local density, positional ordering, and orientational ordering in the region in which the nucleus first arises, we demonstrate that all local order parameters increase simultaneously from the very start of the nucleation process. We thus conclude that we observe no precursor for the crystal nucleation of hard triangular prisms.

Porous crystals in charged sphere suspensions by aggregate-driven phase separation

Abstract:The kinetics of phase transition processes often governs the resulting material microstruc-ture. Using optical microscopy, we here investigate the formation and stabilization of a po-rous crystalline microstructure forming in low-salt suspensions of charged colloidal spheres containing aggregates comprising some 5-10 of these colloids. We observe the transformation of an initially crystalline colloidal solid with homogeneously incorporated aggregates to indi-vidual, compositionally refined crystallites of perforated morphology coexisting with an ag-gregate-enriched fluid phase filling the holes and separating individual crystallites. A prelimi-nary kinetic characterization suggests that the involved processes follow power laws. We show that this route to porous materials is neither restricted to nominally single component systems nor to a particular microstructure to start from. However, it necessitates an early rapid solidification stage during which the aggregates become trapped in the bulk of the host-crystals. The thermodynamic stability of the reconstructed crystalline scaffold against melt-ing under increased salinity was found comparable to that of pure phase crystallites grown very slowly from a melt. Future implications of this novel route to porous colloidal crystals are discussed.

From adsorption to crystallization of proteins: Evidence for interface-assisted nucleation

Abstract:Protein crystallization is among the key processes in biomolecular research, but the underlying mechanisms are still elusive. Here, we address the role of inevitable interfaces for the nucleation process. Quartz crystal microbalance with dissipation monitoring (QCM-D) with simultaneously optical microscopy, confocal microscopy, and grazing-incidence small angle X-rays scattering (GISAXS) were employed to investigate the temporal behavior from the initial stage of protein adsorption to crystallization. Here we studied the crystallization of the Human Serum Albumin (HSA), the most abundant blood protein, in the presence of a charged surface and a trivalent salt. We found evidence for interface-assisted nucleation of crystals. The kinetic stages involved are initial adsorption followed by enhanced adsorption after longer times, subsequent nucleation, and finally crystal growth. The results highlight the importance of interfaces for protein phase behavior and in particular for nucleation.
Evidence for out-of-equilibrium crystal nucleation in suspensions of oppositely charged colloids

Abstract:We report a numerical study of the rate of crystal nucleation in a binary suspension of oppositely charged colloids. Two different crystal structures compete in the thermodynamic conditions under study. We find that the crystal phase that nucleates is metastable and, more surprisingly, its nucleation free energy barrier is not the lowest one. This implies that, during nucleation, there is insufficient time for sub-critical nuclei to relax to their lowest free-energy structure. Such behavior is in direct contradiction with the common assumption that the phase that crystallizes most readily is the one with the lowest free-energy barrier for nucleation. The phenomenon that we describe should be relevant for crystallization experiments where competing solid structures are not connected by an easy transformation.

Abstract:Biomineralization is a typical interdisciplinary subject attracting biologists, chemists, and geologists to figure out its potential mechanism. A mounting number of studies have revealed that the classical nucleation theory is not suitable for all nucleation process of biominerals, and phase-separated structures such as polymer-induced liquid precursors (PILPs) play essential roles in the non-classical nucleation processes. These structures are able to play diverse roles biologically or pathologically, and could also give inspiring clues to bionic applications. However, a lot of confusion and dispute occurred due to the intricacy and interdisciplinary nature of liquid precursors. Researchers in different fields may have different opinions because the terminology and current state of understanding is not common knowledge. As a result, our team reviewed the most recent articles focusing on the nucleation processes of various biominerals to clarify the state-of-the-art understanding of some essential concepts and guide the newcomers to enter this intricate but charming field.

Can Ordered Precursors Promote the Nucleation of Solid Solutions?

Abstract:Crystallization often proceeds through successive stages that lead to a gradual increase in organization. Using molecular simulation, we determine the nucleation pathway for solid solutions of copper and gold. We identify a new nucleation mechanism (liquid$\to$$L1_2$~precursor$\to$solid solution), involving a chemically ordered intermediate that is more organized than the end product. This nucleation pathway arises from the low formation energy of $L1_2$ clusters which, in turn, promote crystal nucleation. We also show that this mechanism is composition-dependent since the high formation energy of other ordered phases precludes them from acting as precursors.

Crystal Nucleation in Al-Ni Alloys: an Unsupervised Chemical and Topological Learning Approach

Abstract:Crystallization represents a fundamental process engendering solidification of a material and determines its microstructure. Driven by complex phenomena at the atomic scale, its understanding for alloys still remains elusive. The present work proposes a large scale molecular dynamics simulation study of the homogeneous crystal nucleation pathways of prototypical undercooled Al-Ni binary alloys. An unsupervised topological learning analysis shows that the nucleation sets in first from a chemical ordering, followed by a bond-orientational ordering of the underlying crystal phase. Our results indicate also a different polymorph selection that depends on composition. While the nucleation pathway of Al50 Ni50 displays a single step with the emergence of B2 short-range order, a step-wise nucleation toward the L12 phase is seen for Al25 Ni75 . The influence of the nucleation of pure Al and Ni counterparts is further discussed.

Abstract:We study low-salt, binary aqueous suspensions of charged colloidal spheres of size ratio Phi = 0.57, number densities below the eutectic number density n_E, and number fractions of p = 1.00-0.40. The typical phase obtained by solidification from a homogeneous shear-melt is a substitutional alloy of body centred cubic structure. In strictly gas-tight vials, the polycrystalline solid is stable against melting and further phase transformation for extended times. For comparison, we prepare the same samples also by slow and mechanically undisturbed deionization in commercial slit cells. These cells feature a complex but well reproducible sequence of global and local gradients in salt concentration, number density and composition as induced by successive deionization, phoretic transport and differential settling of the components, respectively. Moreover, they provide an extended bottom surface suitable for heterogeneous nucleation of the beta-phase. We give a detailed qualitative characterization of the crystallization processes using imaging and optical microscopy. By contrast to the bulk samples, the initial alloy formation in slit cells is not volume-filling, and we now observe also alpha- and beta-phases with low solubility of the odd component. In addition to the initial homogeneous nucleation route, the interplay of gradients opens various further crystallization and transformation pathways leading to a great diversity of microstructures. Upon subsequent increase in salt concentration, crystals melt again. Wall-based, pebble-shaped beta-phase crystals and facetted alpha-crystals melt last. Our observations suggest that the substitutional alloys formed in bulk experiments by homogeneous nucleation and subsequent growth are mechanically stable in the absence of solid-fluid interfaces but thermodynamically metastable.
Evidence for liquid-liquid phase separation during the early stages of Mg-struvite formation

Abstract:The precipitation of struvite, a magnesium ammonium phosphate hexahydrate (MgNH4PO4 · 6H2O) mineral, from wastewater is a promising method for recovering phosphorous. While this process is commonly used in engineered environments, our understanding of the underlying mechanisms responsible for the formation of struvite crystals remains limited. Specifically, indirect evidence suggests the involvement of an amorphous precursor and the occurrence of multi-step processes in struvite formation, which would indicate non-classical paths of nucleation and crystallization. In this study, we use synchrotron-based in situ x-ray scattering complemented by cryogenic transmission electron microscopy to obtain new insights from the earliest stages of struvite formation. The holistic scattering data captured the structure of an entire assembly in a time-resolved manner. The structural features comprise the aqueous medium, the growing struvite crystals, and any potential heterogeneities or complex entities. By analysing the scattering data, we found that the onset of crystallization causes a perturbation in the structure of the surrounding aqueous medium. This perturbation is characterized by the occurrence and evolution of Ornstein-Zernike fluctuations on a scale of about 1 nm, suggesting a non-classical nature of the system. We interpret this phenomenon as a liquid-liquid phase separation, which gives rise to the formation of the amorphous precursor phase preceding actual crystal growth of struvite. Our microscopy results confirm that the formation of Mg-struvite includes a short-lived amorphous phase, lasting >10 s.
Controlling crystallization: What liquid structure and dynamics reveal about crystal nucleation mechanisms

Abstract:Over recent years, molecular simulations have provided invaluable insights into the microscopic processes governing the initial stages of crystal nucleation and growth. A key aspect that has been observed in many different systems is the formation of precursors in the supercooled liquid that precedes the emergence of crystalline nuclei. The structural and dynamical properties of these precursors determine to a large extend the nucleation probability as well as the formation of specific polymorphs. This novel microscopic view on nucleation mechanisms has further implications for our understanding of the nucleating ability and polymorph selectivity of nucleating agents, as these appear to be strongly linked to their ability in modifying structural and dynamical characteristics of the supercooled liquid, namely liquid heterogeneity. In this perspective, we highlight recent progress in exploring the connection between liquid heterogeneity and crystallization, including the effects of templates, and the potential impact for controlling crystallization processes.

Abstract:A critical analysis was carried out for the purpose of understanding the role of subcolloidal (nanosized) (alumino)silicate precursor species in the early stage of crystallization of zeolites in heterogeneous systems (hydrogels). The formation and evolution of these subcolloidal species in both the solid and the liquid phases were investigated by various experimental methods such a scanning electron microscopy (SEM, FE-SEM), transmission electron microscopy, atomic force microscopy, particle size analysis, pH measurement, atomic absorption spectroscopy, and dynamic light scattering, after careful separation of intermediates from reaction mixture by two-step centrifugation treatment. The results revealed that a chain of processes (i) the formation of low-molecular-weight (LMW) silicate species, by dissolution of Al-enriched amorphous silica, and their aggregation into about 3 nm sized primary precursor species (PPSs), (ii) the formation of larger (∼3 to ∼15 nm sized) silicate precursor species (LSPSs) by a rapid aggregation/coalescence of PPSs, (iii) the formation of "gel" (primary amorphous precursor) by a random aggregation of LSPSs at room temperature, and (iv) the formation of the worm-like particles (secondary amorphous precursor) occurred in the solid phase during heating of the reaction mixture (hydrogel) from room temperature to 170 °C. It is interesting that almost the same processes occur in the liquid phase but with decreased rate according to the relative low concentration of LMW silicate species. With the above described findings, it is highly expected that the manipulation of crystallization pathway through controlling the formation/evolution of precursor species in the initial stage of the process can be achieved.

Abstract:Growing crystals by attaching particles Crystals grow in a number a ways, including pathways involving the assembly of other particles and multi-ion complexes. De Yoreo et al. review the mounting evidence for these nonclassical pathways from new observational and computational techniques, and the thermodynamic basis for these growth mechanisms. Developing predictive models for these crystal growth and nucleation pathways will improve materials synthesis strategies. These approaches will also improve fundamental understanding of natural processes such as biomineralization and trace element cycling in aquatic ecosystems.
