Abstract:Salt stress is one of the most serious limiting factors in worldwide agricultural production, resulting in huge annual yield loss. Since 1995, melatonin (N-acetyl-5-methoxytryptamine)—an ancient multi-functional molecule in eukaryotes and prokaryotes—has been extensively validated as a regulator of plant growth and development, as well as various stress responses, especially its crucial role in plant salt tolerance. Salt stress and exogenous melatonin lead to an increase in endogenous melatonin levels, partly via the phyto-melatonin receptor CAND2/PMTR1. Melatonin plays important roles, as a free radical scavenger and antioxidant, in the improvement of antioxidant systems under salt stress. These functions improve photosynthesis, ion homeostasis, and activate a series of downstream signals, such as hormones, nitric oxide (NO) and polyamine metabolism. Melatonin also regulates gene expression responses to salt stress. In this study, we review recent literature and summarize the regulatory roles and signaling networks involving melatonin in response to salt stress in plants. We also discuss genes and gene families involved in the melatonin-mediated salt stress tolerance.

Abstract:Melatonin, a small molecular weight indoleamine molecule, is involved in various biological processes and responses to environmental cues in plants. However, its function in abiotic stress response and the underlying mechanisms is less clear. In this study, we investigated the effect of melatonin on wheat seedlings growth under salt stress condition. Exogenous melatonin pretreatment partially mitigated the salt-induced inhibition of whole-plant growth as judged from shoot dry weight, IAA content, leaf photosynthesis rate, maximum photochemistry efficiency of photosystem II, and chlorophyll. The mitigation was also observed in reduced accumulation of H2O2 in melatonin-pretreated wheat seedlings exposed to salt stress. Exogenous melatonin increased endogenous melatonin content by evaluating the levels of TaSNAT transcript, which encodes a key regulatory enzyme in the melatonin biosynthetic pathway. Furthermore, melatonin increased polyamine contents by accelerating the metabolic flow from the precursor amino acids arginine and methionine to polyamines; melatonin also decreased the degradation of salt-induced polyamines. Taken together, these results provide the evidence that melatonin mitigates salt stress mainly through its regulation on polyamine metabolism of wheat seedlings.

Abstract:Melatonin, an indoleamine widely found in animals and plants, is considered as a candidate phytohormone that affects responses to a variety of biotic and abiotic stresses. In plants, melatonin has a similar action to that of the auxin indole-3-acetic acid (IAA), and IAA and melatonin have the same biosynthetic precursor, tryptophan. Salt stress results in the rapid accumulation of melatonin in plants. Melatonin enhances plant resistance to salt stress in two ways: one is via direct pathways, such as the direct clearance of reactive oxygen species; the other is via an indirect pathway by enhancing antioxidant enzyme activity, photosynthetic efficiency, and metabolite content, and by regulating transcription factors associated with stress. In addition, melatonin can affect the performance of plants by affecting the expression of genes. Interestingly, other precursors and metabolite molecules associated with melatonin can also increase the tolerance of plants to salt stress. This paper explores the mechanisms by which melatonin alleviates salt stress by its actions on antioxidants, photosynthesis, ion regulation, and stress signaling.

Abstract:The role of melatonin in plant growth and response to environmental stress has been widely demonstrated. However, the physiological and molecular regulation of salt tolerance in wheat seedlings by melatonin remains unclear. In this study, we investigated changes in phenotype, physiology, photosynthetic parameters, and transcript levels in wheat seedlings to reveal the role of melatonin in the regulation of salt tolerance in wheat. The results indicate that the application of exogenous melatonin significantly alleviates growth inhibition, reactive oxygen species accumulation, and membrane oxidative damage induced by salt stress in wheat. Additionally, exogenous melatonin increased antioxidant enzyme activity and regulated photosynthetic gas exchange. Transcriptomic data showed a significant up-regulation of genes encoding light-harvesting chlorophyll protein complex proteins in photosynthesis and genes related to chlorophyll and carotenoid biosynthesis under the influence of melatonin. These results suggest that exogenous melatonin improves salt tolerance in wheat seedlings by enhancing the antioxidant, photoprotective, and photosynthesis activities.

Abstract:Melatonin is an antioxidant that is widely distributed in plants and animals. It is a conservative molecule. In early studies, scientists often used isolation and identification techniques to observe whether the endogenous melatonin cycle in plants was related to the external photoperiod, plant growth, and development cycles, including seed germination, plant rooting, and floral induction. With the development of isolation and identification technology, there is ample evidence that plants possess a variety of melatonin-synthesis pathways. The comprehensive application of molecular biology, genomics, and computational biology has also led to a comprehensive understanding of the physiological functions of plant melatonin. In this paper, we not only highlight the candidate genes from Arabidopsis thaliana and Oryza sativa that might be contributing to increasing plant endogenous melatonin but also elucidate and characterize the role of melatonin in plant growth and development in response to biotic and abiotic stresses.

Abstract:Plant mineral nutrition is critical for agricultural productivity and for human nutrition; however, the availability of mineral elements is spatially and temporally heterogeneous in many ecosystems and agricultural landscapes. Nutrient imbalances trigger intricate signalling networks that modulate plant acclimation responses. One signalling agent of particular importance in such networks is phytomelatonin, a pleiotropic molecule with multiple functions. Evidence indicates that deficiencies or excesses of nutrients generally increase phytomelatonin levels in certain tissues, and it is increasingly thought to participate in the regulation of plant mineral nutrition. Alterations in endogenous phytomelatonin levels can protect plants from oxidative stress, influence root architecture, and influence nutrient uptake and efficiency of use through transcriptional and post-transcriptional regulation; such changes optimize mineral nutrient acquisition and ion homeostasis inside plant cells and thereby help to promote growth. This review summarizes current knowledge on the regulation of plant mineral nutrition by melatonin and highlights how endogenous phytomelatonin alters plant responses to specific mineral elements. In addition, we comprehensively discuss how melatonin influences uptake and transport under conditions of nutrient shortage.
Melatonin inhibits seed germination by crosstalk with abscisic acid, gibberellin, and auxin in Arabidopsis

Abstract:Seed germination, an important developmental stage in the life cycle of seed plants, is regulated by complex signals. Melatonin is a signaling molecule associated with seed germination under stressful conditions, although the underlying regulatory mechanisms are largely unknown. In this study, we showed that a low concentration (10 µM or 100 µM) of melatonin had no effect on seed germination, but when the concentration of melatonin increased to 500 µM or 1000 µM, seed germination was significantly inhibited in Arabidopsis. RNA sequencing analysis showed that melatonin regulated seed germination correlated to phytohormones abscisic acid (ABA), gibberellin (GA), and auxin. Further investigation revealed that ABA and melatonin synergistically inhibited seed germination, while GA and auxin antagonized the inhibitory effect of seed germination by melatonin. Disruption of the melatonin biosynthesis enzyme gene serotonin N‐acetyltransferase (SNAT), or N‐acetylserotonin methyltransferase (ASMT) promoted seed germination, while overexpression of ASMT inhibited seed germination. Taken together, our study sheds new light on the function and mechanism of melatonin in modulating seed germination in Arabidopsis.

Abstract:Melatonin is a ubiquitous signaling plant hormone that plays a crucial role in regulating the growth and development of plants under stress conditions. Since a few species have been investigated to unveil the effect of exogenous melatonin on salt stress, in the current research study, we investigated the effects of melatonin by measuring the photosynthetic characteristics and antioxidant defense system of maize seedling under salt stress (200 mM NaCl), along with different concentrations of melatonin (MT1—30, MT2–60, and MT3–90 µM) treatments. Salt stress reduced the plant growth characteristics and photosynthetic efficiency by increasing the ROS accumulation and reducing the antioxidant enzyme activities of maize seedling. However, pretreatment with melatonin on roots alleviated NaCl-induced decrease in photosynthetic rate and oxidative stress in a dose-dependent manner. Our results revealed that exogenous application of melatonin at an optimum concentration (60 µM) under salt stress conditions significantly increased the growth of plants, chlorophyll content, photosynthetic efficiency, antioxidant enzymes activities, i.e., superoxide dismutase (SOD), peroxide (POD), catalase (CAT) and ascorbate peroxidase (APX), and reduced the reactive oxygen species (ROS). This study unraveled the crucial role of melatonin in salt stress mitigation and thus can be implicated in the management of salinity in maize seedling.

Abstract:Melatonin functions as a negative regulatory signal of seed oil and anthocyanin accumulation during the maturation of A. thaliana seeds. Previous studies have clearly demonstrated that the putative phytohormone melatonin functions directly in many aspects of plant growth and development. In Arabidopsis (Arabidopsis thaliana), the role of melatonin in seed oil and anthocyanin accumulation, and corresponding underlying mechanisms, remain unclear. Here, we found that serotonin N-acetyltransferase1 (SNAT1) and caffeic acid O-methyltransferase (COMT) genes were ubiquitously and highly expressed and essential for melatonin biosynthesis in Arabidopsis developing seeds. We demonstrated that blocking endogenous melatonin biosynthesis by knocking out SNAT1 and/or COMT significantly increased oil and anthocyanin content of mature seeds. In contrast, enhancement of melatonin signaling by exogenous application of melatonin led to a significant decrease in levels of seed oil and anthocyanins. Further gene expression analysis through RNA sequencing and reverse-transcription quantitative PCR demonstrated that the expression of a series of important genes involved in fatty acid and anthocyanin accumulation was significantly altered in snat1-1 comt-1 developing seeds during seed maturation. We also discovered that SNAT1 and COMT significantly regulated the accumulation of both mucilage and proanthocyanidins in mature seeds. These results not only help us understand the function of melatonin and provide valuable insights into the complicated regulatory network controlling oil and anthocyanin accumulation in seeds, but also divulge promising gene targets for improvement of both oil and flavonoids in seeds of oil-producing crops and plants.
Roles of Endogenous Melatonin in Resistance to Botrytis cinerea Infection in an Arabidopsis Model

Abstract:Melatonin is an important bioactive molecule in plants. Two synthetases, N-acetylserotonin methyltransferase (ASMT) and serotonin N-acetyltransferase (SNAT) are involved in the final two steps of melatonin synthesis. Melatonin participates in responses to a variety of biotic and abiotic stresses in plants, but few studies have addressed the roles of endogenous melatonin in pathogen resistance. We investigated the role of endogenous melatonin in resistance to Botrytis cinerea infection in an Arabidopsis thaliana model system. Plant lines that overexpressed ASMT or SNAT through genetic manipulation showed upregulated expression of resistance genes PR1 and PR5 , transcription factor gene WRKY33 , and jasmonic acid (JA) defense pathway marker gene PDF1.2 , and downregulated transcription factor gene MYC2 in JA signaling pathway. Higher melatonin content also enhanced the activity of antioxidant enzymes superoxide dismutase (SOD) and peroxidase (POD), increased JA content, reduced plant disease symptoms, and reduced lesion size in leaves. These findings indicate that endogenous melatonin enhances plant resistance to B. cinerea infection. In contrast, ASMT and SNAT gene silencing lines showed opposite results and were more susceptible to B. cinerea . Thus, it can be demonstrated that melatonin functions as an effective regulator of plant stress resistance at the genetic level. A schematic model is presented for its role in resistance to B. cinerea infection. Our findings also helped to elucidate the associated signal transduction pathways and interactions between melatonin and other plant hormones.

Abstract:Melatonin (N-acetyl-5-methoxy-tryptamine) is a universal molecule that is present in animals and plants. It has been detected in different kinds of plants and organs in different levels. Melatonin in plants shares the same initial biosynthesis compound with auxin, and therefore functions as indole-3-acetic acid like hormones. Moreover, melatonin is involved in regulating plant growth and development, protecting plants against biotic and abiotic stresses, such as salt, drought, cold, heat and heavy metal stresses. Melatonin improves the stress tolerance of plants via a direct pathway, which scavenges reactive oxygen species directly, and indirect pathways, such as increasing antioxidate enzymes activity, photosynthetic efficiency and metabolites content. In addition, melatonin plays a role in regulating gene expression, and hence affects performance of plants. In this review, the biosynthesis pathway, growth and development regulation, and the environment stress response of melatonin in plants are summarized and future research directions and priorities of melatonin in plants are speculated.

Chloroplastic biosynthesis of melatonin and its involvement in protection of plants from salt stress

Abstract:Within the chloroplasts reactive oxygen species (ROS) are generated during photosynthesis and stressful conditions. Excessive ROS damages chloroplasts and reduces photosynthesis if not properly detoxified. In this current study, we document that chloroplasts produce melatonin, a recently-discovered plant antioxidant molecule. When N-acetylserotonin, a substrate for melatonin synthesis, was fed to purified chloroplasts, they produced melatonin in a dose-response manner. To further confirm this function of chloroplasts, the terminal enzyme for melatonin synthesis, N-acetylserotonin-O-methyltransferase (ASMT), was cloned from apple rootstock, Malus zumi. The in vivo fluorescence observations and Western blots confirmed MzASMT9 was localized in the chloroplasts. A study of enzyme kinetics revealed that the Km and Vmax of the purified recombinant MzASMT9 protein for melatonin synthesis were 500 μM and 12 pmol/min·mg protein, respectively. Arabidopsis ectopically-expressing MzASMT9 possessed improved melatonin level. Importantly, the MzASMT9 gene was found to be upregulated by high light intensity and salt stress. Increased melatonin due to the highly-expressed MzASMT9 resulted in Arabidopsis lines with enhanced salt tolerance than wild type plants, as indicated by reduced ROS, lowered lipid peroxidation and enhanced photosynthesis. These findings have agricultural applications for the genetic enhancement of melatonin-enriched plants for increasing crop production under a variety of unfavorable environmental conditions.
Exogenous melatonin alleviates salt stress by improving leaf photosynthesis in rice seedlings

Abstract:Melatonin (MT), an important antioxidant existing in plants and animals, has been widely reported to participate in the process of plants coping with stress. In this study, we demonstrated the mechanism of MT enhancing photosynthesis in rice under salt stress. The results showed that MT treatment increased relative water content, sucrose and starch content of rice under salt stress. This was mainly owing to the fact that MT enhanced the net photosynthetic rate and enhanced the absorption and transmission of light energy. The effect of MT on photosynthesis of rice under salt stress conditions was mainly due to the regulation of three processes: maintaining low ROS status by improving the total antioxidant capacity, promoting the xanthophyll cycle and increasing the xanthophyll pool size to dissipate excess light energy, increasing the activities of key photosynthetic enzymes. Taken together, these results provide a mechanism for MT to improve the photosynthetic capacity of rice under salt stress.

Abstract:Melatonin is an important phytohormone in plant development and stress responses. However, the functions of melatonin in rice salt tolerance during seed germination is largely undetermined. In this study, we investigated its potential molecular mechanism during rice seed germination under salinity through comprehensive transcriptome sequencing, metabolome profiling, and physiological assays. Transcriptome analysis revealed 4794 differentially expressed genes (DEGs) after melatonin pretreatment under salinity, including 2843 upregulated and 1951 downregulated genes. Functional annotation showed that many DEGs were associated with antioxidative activity and phytohormone biosynthesis and signal transduction. Consistent with the transcriptome results, physiological analysis revealed enhanced activity of antioxidant capacity, increased indole-3-acetic acid (IAA) level, and reduced abscisic acid (ABA) level under melatonin pretreatment. The enhanced antioxidative activity was also supported by metabolome assay that melatonin pretreatment increased the levels of non-enzymatic antioxidant organic acids and amino acids. The results demonstrated that exogenous melatonin can enhance rice seed tolerance to salinity during germination via activating antioxidants and modulating phytohormones.

Abstract:As a pleiotropic signal molecule, melatonin is ubiquitous throughout the animal and plant kingdoms and plays important roles in the regulation of plant growth, development, and responses to environmental stresses. In this study, we quantified the endogenous melatonin levels in upland cotton (Gossypium hirsutum L.), using high-performance liquid chromatography–tandem mass spectrometry. The melatonin concentrations in root, stem, and leaf were 150.60, 37.92, and 40.58 ng g fresh weight− 1, respectively. The effects of exogenous melatonin (1 µM) on plant growth, photosynthesis, antioxidant enzyme activity, and ion homeostasis in upland cotton seedlings exposed to 100 mM NaCl treatment were determined. Pretreatment (prior to exposure to salt stress) of seedlings with exogenous melatonin significantly alleviated plant growth inhibition by salt stress and maintained an improved photosynthetic capacity. The application of melatonin also significantly reduced the salt-induced oxidative damage, possibly through the accumulation of osmotic regulatory substances and the activation of antioxidant enzymes. We also showed that exogenous melatonin regulated the expression of stress-responsive and ion-channel genes under salinity, which could contribute to improved salt tolerance in cotton. Taken together, our study provides evidence that cotton contains endogenous melatonin, and it may have unraveled crucial evidence of the role of melatonin in cotton against salt stress.

Abstract:Serotonin N-acetyltransferase 1 (SNAT1), the penultimate enzyme for melatonin biosynthesis has shown N-acetyltransferase activity toward multiple substrates, including histones, serotonin, and plastid proteins. Under two different light conditions such as 50 or 100 μmol m-2 s-1, a SNAT1-knockout (snat1) mutant of Arabidopsis thaliana ecotype Columbia (Col-0) exhibited small size phenotypes relative over wild-type (WT) Arabidopsis Col-0. Of note, the small phenotype is stronger when growing at the 50 μmol m-2 s-1, exhibiting a dwarfism phenotype and delayed flowering. The snat1 Arabidopsis Col-0 accumulated less starch than the WT Col-0. Moreover, snat1 exhibited lower Lhcb1, Lhcb4, and RBCL protein levels, compared with the WT Col-0, but no changes in the corresponding transcripts, suggesting the involvement of melatonin in chloroplast protein quality control (CPQC). Accordingly, caseinolytic protease (Clp) and chloroplast heat shock proteins (CpHSPs), two key proteins involved in CPQC, as well as ROS defense were suppressed in snat1. In contrast, exogenous melatonin treatment induced expression of Clp, CpHSP, APX1, and GST, but not other growth-related genes such as DWF4, KS, and IAA1. Finally, the induction of ClpR1, APX1, and GST1 in response to melatonin was inhibited in the mitogen-activated protein kinase (MAPK) knockdown Arabidopsis (mpk3/6), suggesting that melatonin-mediated CPQC was mediated, in part, by the MAPK signaling cascade. These results suggest that melatonin is involved in CPQC, which plays a pivotal role in starch synthesis in plants.
Integrative Analyses of Transcriptome, Microrna-Seq and Metabolome Reveal Insights into Exogenous Melatonin-Mediated Salt Tolerance During Seed Germination of Maize

Abstract:Melatonin is an important phytohormone influencing plant growth and defense responses. However, the mechanism mediating the regulatory effects of melatonin on the salt tolerance of germinating maize seeds remains unexplored. In this study, the application of exogenous melatonin enhanced the salt tolerance of germinating maize seeds. A transcriptome analysis indicated that complex regulatory pathways may be associated with the melatonin-mediated salt tolerance. Remarkably, antioxidant activities, transcriptional regulation, and phytohormone (e.g., cytokinin and auxin) pathways were induced following the exogenous application of melatonin. The microRNA (miRNA)-seq analysis result indicated that exogenous melatonin obviously altered the expression of a set of miRNAs. Notably, many differentially expressed miRNAs and their target genes, including several transcription factor genes, were revealed to contribute to salt stress responses. According to metabolite profiles, the abundance of diverse metabolites, like secondary metabolites, nucleotides, cofactors, and vitamins, increased significantly after using exogenous melatonin. The combined analysis of the transcriptome and metabolome indicated that several gene–metabolite networks related to amino acid metabolism and secondary metabolite biosynthetic pathways are essential for melatonin-mediated maize tolerance to salt stress. Consistent with these findings, exogenously applied melatonin altered the phytohormone levels and increased the antioxidant enzyme activities and energy supply. Our results reflect the significance of melatonin for enhancing the salt tolerance of germinating maize seeds, which is achieved through the regulation of antioxidant capacity, phytohormone content, and metabolic adaptation.

Abstract:Melatonin is a pleiotropic molecule with multiple functions in plants. Since the discovery of melatonin in plants, numerous studies have provided insight into the biosynthesis, catabolism, and physiological and biochemical functions of this important molecule. Here, we describe the biosynthesis of melatonin from tryptophan, as well as its various degradation pathways in plants. The identification of a putative melatonin receptor in plants has led to the hypothesis that melatonin is a hormone involved in regulating plant growth, aerial organ development, root morphology, and the floral transition. The universal antioxidant activity of melatonin and its role in preserving chlorophyll might explain its anti‐senescence capacity in aging leaves. An impressive amount of research has focused on the role of melatonin in modulating postharvest fruit ripening by regulating the expression of ethylene‐related genes. Recent evidence also indicated that melatonin functions in the plant's response to biotic stress, cooperating with other phytohormones and well‐known molecules such as reactive oxygen species and nitric oxide. Finally, great progress has been made towards understanding how melatonin alleviates the effects of various abiotic stresses, including salt, drought, extreme temperature, and heavy metal stress. Abstract:Melatonin (N-acetyl-5-methoxytryptamine) is a biological hormone involved in many important physiological processes in plants. To better understand the protective roles of melatonin in plants against salt stress, we determined the effects of exogenous melatonin (1 µM) on plant growth, photosynthesis, antioxidant enzyme activity and ion homeostasis in maize plants under 100 mM NaCl. Results showed that salt stress decreased plant dry matter accumulation, net photosynthetic rate (Pn) and chlorophyll contents. However, 1 µM melatonin application significantly alleviated this growth inhibition and enhanced Pn by 19 %. Melatonin application also enhanced the activities of antioxidative enzymes of salt-stressed maize leaves, and decreased their electrolyte leakage and MDA content by 25 and 22 %, respectively. In addition, melatonin application significantly increased K+ contents and K+/Na+ ratios in salt-stressed maize shoots by 18 and 52 %, respectively. However, the Na+ content was decreased significantly in melatonin-treated leaves under salinity. The results suggested that the melatonin enhanced maize salt tolerance in maize were most likely due to the improvement of photosynthetic capacity, antioxidative capacity and ion homeostasis in leaves. This study provides, for the first time, the evidence that support the protective roles of exogenous melatonin in maize against salinity.

Abstract:Melatonin mediates many physiological processes in plants. We investigated its role in regulating growth, potassium uptake, and root system architecture under three types of stress: salinity or a deficiency of all nutrients in Malus hupehensis Rehd., as well as a K deficiency in Malus rockii Rehd. Each treatment caused a reduction in growth rates and disrupted the absorption of potassium. However, pretreatment with 0.1 mmol/L melatonin significantly alleviated such inhibitions. The addition of melatonin also upregulated genes for antioxidant enzymes involved in the ascorbate-glutathione cycle (MdcAPX, MdDHAR1, MdDHAR2, MdMDHAR, and MdcGR) and helped decrease the accumulation of H2 O2 while improving the expression of K transporters and genes for the CBL1-CIPK23 pathway. These results indicated that melatonin can regulate the ROS signal and activate the CBL1-CIPK23 pathway to regulate the expression of a potassium channel protein gene, thereby promoting the absorption of potassium ions. Our findings demonstrate that inducing melatonin production is an important mechanism for plant defenses that can serve as a platform for possible applications in agricultural or related fields of research.
