Abstract:Bicyclo[1.1.1]pentanes (BCPs) are important bioisosteres of aryl, tert-butyl groups, and internal alkynes that can impact key physicochemical properties on drug candidates. Herein, we describe a novel and efficient reaction to synthesize alkyl-alkynyl-substituted BCP derivatives by synergistic photoredox and copper catalysis at room temperature. The mild reaction conditions, simple protocol, broad functional group tolerance, and high efficiency of this procedure make it a valuable strategy for accessing alkynyl-substituted BCPs.

Abstract:Herein, a visible light‐mediated multicomponent radical cross‐coupling reaction of [1.1.1]propellane with N‐heterocycles and disulfides is described. The mild conditions enabled the installation of the BCP motif in a wide variety of compounds in the absence of photocatalyst, additive and metal catalyst, with excellent functional group tolerance, This methodology opens a approach for the synthesis of potentially bioactive 1‐arylthiol‐3‐heteroaryl bicyclo[1.1.1]pentanes in moderate to good yield.

Abstract:Bicyclo[1.1.1]pentanes (BCPs) are widely utilized in drug design as sp3-rich bioisosteres for tert-butyl, internal alkynes, and aryl groups. A general and mild method for radical acylation of [1.1.1]propellane with aldehydes has been developed. The protocol provides straightforward access to bicyclo[1.1.1]pentane ketones with a broad substrate scope. The synthetic utility of this methodology is demonstrated by the late-stage modification of bioactive molecules and the versatile transformation of bicyclo[1.1.1]pentane ketones, making it useful for drug discovery.

Abstract:A novel and practical copper-catalyzed strategy for intermolecular cyanosulfonylation and cyanotrifluoromethylation of unactivated alkenes in water-containing solvents is described. The methodology developed provides an efficient and convenient access to a variety of β-sulfonyl nitriles and β-trifluoromethyl nitriles, which would have wide applications in chemical and pharmaceutical industries.

Abstract:As the magic methyl effect is well acknowledged in pharmaceutical molecules, the development of simple and efficient methods for the installment of methyl groups on complex molecules is highly coveted. Hence, we provide a general strategy for radical cascade cyclization of N-(o-cyanobiaryl)acrylamides by utilizing sulfonium salts as the sources of methyl radical and merging photoredox and copper catalysis. This novel protocol can access a wide variety of methylation or remote thioether-substituted benzo-fused N-heterocycle derivatives, which can be easily transformed into diverse highly valuable sulfone and sulfoximine compounds via late-stage diversification. Moreover, to further demonstrate the synthetic utility of this conversion, the methyl(phenyl)sulfide, which serves as both raw material and byproduct, can be recovered and reused in this transformation. The scale-up experiment for the one-pot two-step process directly offers the target product in good yield under the standard conditions.

Abstract:A Cu‐catalyzed oxidative radical [2+2+1] annulation reaction of phenol‐linked 1,7‐enynes with α‐active methylene nitrile is described, affording cyclopenta[c]chromene derivatives bearing several functional groups in excellent yields. The wide substrate range, good group tolerance, and easy amplification experiments indicate the practicability of this synthesis strategy. This process provides an alternative convenient route for the synthesis of cyclopenta[c]chromene derivatives.

Abstract:Bicyclo[1.1.1]pentane (BCP), recognized as a bioisostere for para-disubstituted benzene, has gained widespread interest in drug development due to its ability to enhance the physicochemical properties of pharmaceuticals. In this work, we introduce a photoinduced, halogen bonding-initiated, metal-free strategy for synthesizing various BCP derivatives. This method involves the generation of nucleophilic α-aminoalkyl radicals via halogen-bonding adducts. These undergo selective radical addition to [1.1.1]propellane, yielding electrophilic BCP radicals that subsequently participate in polarity-matched additions, culminating in the difunctionalization of bicyclopentane. The versatility and practicality of this metal-free approach are underscored by its broad substrate scope, which includes late-stage functionalization and a series of valuable transformations, all conducted under mild reaction conditions.

Abstract:Radical reactions play an important role in modern organic synthetic chemistry. The generation of carbon radicals from the homolytic cleavage of carbon–metal bonds has been widely studied as a fundamental step in organometallic chemistry. However, the implementation of this phenomenon in catalysed cascade reactions, that is, an organometallic-radical relay, is a highly challenging undertaking due to transient radicals generated from these thermodynamically disfavoured endergonic processes. Here we disclose a set of catalytic alkene 1,1-difunctionalization reactions conceptually based on an organometallic-radical relay. This strategy enables a diversity of sp 3 / sp 2 fragments and aryl groups to be simultaneously introduced to the same carbon of both terminal and internal alkenes with outstanding chemo- and regioselectivity. This study provides a concept for the generation of functionalized benzylic radicals, which are difficult to access by classical protocols, from non-radical feedstock alkenes and arylboronic acids.

Abstract:We present herein a modular photoredox strategy for the difunctionalization of alkenes employing arylsulfonyl acetate as the bifunctional reagents. Under photoredox conditions, the electrophilic carbon-centered radical, generated from bifunctional reagent, add to alkenes followed by 1,4-aryltranlocation, providing a robust alternative to synthetic valuable γ,γ-diaryl and γ-aryl ester compounds. This method features mild reaction conditions, high atom- and step- economy, excellent functional group compatibility and great structural diversity. A complementary oxidative bifunctional reagents activation mode governs the radical cascade reactions, facilitating the simultaneous incorporation of aryl and carboxylate bearing alkyl groups into the alkenes.

Abstract:Under heating conditions, an iron(II)‐catalyzed cascade cyanoalkylsulfonylation/cyclization reaction is developed to producing cyanoalkylsulfonylated pyrrolidones bearing with a free CN moiety. This alkyl radical disconnection and cascade annulation protocol enjoys chemo‐ and regioselectivity with the tolerance toward a range of functional groups. The reactive cyanoalkyl radical, which was originated from the ring‐opening reaction of a cyclic imine radical, was found to be capable of capturing the in‐situ generated "SO2" species from DABSO, thus leading to the production of a cyanoalkyl sulfonyl radical and the subsequent alkene annulation sequences.

Abstract:Density functional theory investigations revealed that the pyridine-boryl radical generated in situ using 4-cyanopyridine and bis(pinacolato)diboron could be used as a bifunctional "reagent", which serves as not only a pyridine precursor but also a boryl radical. With the unique reactivity of such radicals, 4-substituted pyridine derivatives could be synthesized using α,β-unsaturated ketones and 4-cyanopyridine via a novel radical addition/C-C coupling mechanism. Several controlled experiments were conducted to provide supportive evidence for the proposed mechanism. In addition to enones, the scope could be extended to a wide range of boryl radical acceptors, including various aldehydes and ketones, aryl imines and alkynones. Lastly, this transformation was applied in the late-stage modification of a complicated pharmaceutical molecule.

Abstract:Bicyclo[1.1.1]pentylamines (BPCAs), emerging as sp 3 -rich surrogates for aniline and its derivatives, demonstrate unique structural features and physicochemical profiles in medicinal and synthetic chemistry. In recent years, compared with conventional synthetic approaches, the rapid development of radical chemistry enables the assembly of valuable bicyclo[1.1.1]pentylamines scaffold directly through the amination transformation of highly strained [1.1.1]propellane. In this review, we concisely summarize the emerging role of radical chemistry in the construction of BCPAs motif, highlighting two different and powerful radical-involved strategies including C -centered and N -centered radical pathways under appropriate conditions. The future direction concerning BCPAs is also discussed at the end of this review, which aims to provide some inspiration for the research of this promising project.

Abstract:A simple and low-cost tandem sulfonylation/cyclization of 1,5-diene, aryldiazonium salt, and DABCO·(SO2)2 is disclosed. This base-promoted multicomponent reaction can provide a “green” and economic synthesis of sulfonylated pyrrolidones under transition-metal-free and moisture/oxygen-insensitive reaction conditions, thus delivering a wide range of sulfonylated pyrrolidones in moderate to high yields with excellent functional group compatibility. A plausible mechanism involving a radical process is proposed, which demonstrates highly chemoselective trapping of the aryl radical with “SO2” species, and a regioselective sulfonylation/cyclization protocol in this reaction.

Abstract:The photoredox electron donor–acceptor (EDA) complex-mediated radical coupling reaction has gained prominence in the field of organic synthesis, finding widespread application in two-component coupling reactions. However, EDA complex-promoted multi-component reactions are not well developed with only a limited number of examples have been reported. Herein, we report a photoinduced and EDA complex-promoted highly chemoselective three-component radical arylalkylation of [1.1.1]propellane, which allows the direct functionalization of C(sp3)–H with bicyclo[1.1.1]pentanes (BCP)-aryl groups under mild conditions. A variety of unnatural α-amino acids, featuring structurally diversified 1,3-disubstituted BCP moieties, were synthesized in a single-step process. Notably, leveraging the high tension release of [1.1.1]propellane, the highly unstable transient aryl radical undergoes rapid conversion into a relatively stable tertiary alkyl transient radical, and consequently, the competing side-reaction of two-component coupling was entirely suppressed. The strategic use of this transient radical conversion approach would be useful for the design of diverse EDA complex-mediated multi-component reactions. It is noteworthy that the highly chemoselective late-stage incorporation of the 1,3-disubstituted BCP pharmacophores into peptides was achieved both in liquid-phase and solid-phase reactions. This advancement is anticipated to have significant application potential in the future development of peptide drugs.
